        Antioxidant Activity of Extracts from Akebia quinata Decne

        Antioxidant activity of Akebia quinata Decne was evaluated. Water extract (0.5 g/ 50 mL) of flowers and leaves of A. quinata were prepared and total phenol contents and radical scavenging activity of the extracts was determined for antioxidant activity. The total phenol contents of extracts from A. quinata flowers (FAQ) and leaves (LAQ) were 30.05 M and 20.23 M, while the radical scavenging activity of FAQ and LAQ were 60.51% and 52.97%, respectively. In addition, the effect of FAQ and LAQ extract on DNA damage induced by H2O2 in human lymphocytes was evaluated by comet assay. The FAQ and LAQ showed strong inhibitory effect against DNA damage induced by 200 M of H2O2. These results suggest that water extracts of A. quinata Decne flowers and leaves showed significant (p<0.05) antioxidant activity and protective effect against oxidative DNA damage.

        재활용 천연 염료를 이용한 모발의 염색성 시연

        The purpose of this study is to find the possibility of developing and using recycled natural hairdye as an alternative solution to the hair damage by the use of chemical hairdye and environmental pollution by the remnants of hairdye. First, alum and bean curd were separately added to onion skin extracts as mordant. The L-values(luminosity) of the recycled hairdyes were 49.1 and 43.9, respectively, and 46.6 for chemical hairdye. The a-values (red line) were +14.9, +14.6, and +12.1, respectively, and the b-values (yellow line) were +45.9, +29.3, and +26.3, respectively. Second, when we compare the grape skin extracts to which alum and bean curd were separately added with the chemical hairdye. The L-values were 39.2, 40.3, and 22.5, respectively. The a-values were +10.0, +12.7, and +9.7, respectively, and the b-values were +18.8, +21.8, and +4.9, respectively. Third, we compared the remnants of roasted bean coffee to which alum and bean curd were added with chemical hairdye. The L-value, a-value, and b-value of chemical hairdye were the lowest at 17.1, +2.5, and +0.6. Fourth, we compared Chinese ink to which alum and bean curd were added with chemical hairdye. The L-value, a-value, and b-value of chemical hairdye were the lowest at 16.3, +0.2, and -0.9. Like the results for remnants of roasted bean coffee, the coloring rate of chemical hairdye was high at low L-value. This shows that the chemical hairdye is effective for coloring at low L-value. The results of this study confirmed the possibility of using recycled natural hairdyes for colors with high L-value, and there were no difference between the values of recycled natural hairdyes and chemical hairdye at high luminosity. Therefore, it is believed that these recycled natural hairdyes will be useful as new hairdyes.

        Incidence of Alternaria Species Associated with Watermelon Leaf Blight in Korea

        Alternaria leaf blight is one of the most common dis- eases in watermelon worldwide. In Korea, however, the Alternaria species causing the watermelon leaf blight have not been investigated thoroughly. A total of 16 Alternaria isolates was recovered from diseased water- melon leaves with leaf blight symptoms, which were collected from 14 fields in Korea. Analysis of internal transcribed spacer (ITS) region, glyceraldehyde-3-phos- phate dehydrogenase (GAPDH), and RNA polymerase II second largest subunit (RPB2) were not competent to differentiate the Alternaria isolates. On the contrary, analysis of amplicon size of the histone H3 (HIS3) gene successfully differentiated the isolates into three Alter- naria subgroups, and further sequence analysis of them identified three Alternaria spp. Alternaria tenuissima, A. gaisen, and A. alternata. Representative Alternaria iso- lates from three species induced dark brown leaf spot lesions on detached watermelon leaves, indicating that A. tenuissima, A. gaisen, and A. alternata are all causal agents of Alternaria leaf blight. Our results indicate that the Alternaria species associated watermelon leaf blight in Korea is more complex than reported previ- ously. This is the first report regarding the population structure of Alternaria species causing watermelon leaf blight in Korea.

        한약재 추출물의 에스트로겐 유사활성 및 조골세포 증식과 분화에 미치는 영향

        본 연구는 3종의 한약재(감초(Glycyrrhizae radix), 황기(Astragali radix) 및 산약(Dioscorea rhizome)) 추출물의 에스트로겐 유사활성 및 조골세포 증식과 분화에 미치는 영향을 검토하고자 실험을 진행하였다. 에스트로겐 유사활성측정을 위하여 인체 유방암 세포주인 MCF7 세포를 luciferase 유전자를 리포터로 사용하는 에스트로겐 반응성 리포터 시스템을 가지는 세포로 형질전환하여 사용하였다. 상기 세포를 이용하여 추출물의 에스트로겐 유사활성을 측정한 결과, 한약재 추출물이 음성대조군과 비교하여 1.11배~5.73배 높은 에스트로겐 유사활성을 나타내었다. 그 중 감초 추출물이 가장 높은 에스트로겐 유사활성을 보였다. 감초 추출물의 에스트로겐 유사활성은 추출물의 농도가 50 μg/ml 및 500 μg/ml 일 때 각각 10<SUP>-8</SUP> M 및 10<SUP>-7</SUP> M 17β-estradiol과 거의 유사한 활성을 나타내었다. 조골세포주인 MC3T3-E1 세포를 이용한 실험에서는 감초 추출물의 농도가 1~1,000 μg/ml 범위일 때 독성을 나타내지 않았다. 황기 추출물은 조골세포에 있어서 유의적인 세포 증식률을 보이지 않았다. 그러나, 감초 및 산약추출물은 각각 148% 및 133%의 최대 증식률을 보였다. 또한, 감초 추출물은 대조군과 비교하여 alkaline phosphatase (ALP) 활성이 증가하였으며, 추출물의 농도가 100 μg/ml 일 때 최대 122%의 ALP 활성을 나타내었다. 또한, Alizarin red S 염색법을 이용한 석회화 형성능 측정에서도 대조군과 비교하여 석회화가 증가하였다. 이러한 결과로 미루어 보아 감초 추출물이 골 형성 및 골다공증 예방 효과가 우수한 식품 소재인 것으로 사료된다. The purpose of this study was to investigate the effect of 3 types of medicinal herbs (Glycyrrhizae radix, Astragali radix and Dioscorea rhizoma) extracts on estrogen-like activities, proliferation and differentiation in osteoblast. Human breast cancer cell line MCF7 was transfected using an estrogen responsive luciferase reporter plasmid for measure the estrogen-like activity. Estrogen-like activities of extracts were in the range of 1.11~5.73 fold to that of negative control. The extract of G. radix showed the strongest estrogen-like activities. The estrogen-like activities of 50 and 500 μg/ml extracts of G. radix were similar to that of 10<SUP>-8</SUP> and 10<SUP>-7</SUP> M standard solution (17β-estradiol), respectively. G. radix extract showed no cytotoxicity against osteoblast MC3T3-E1 cells at 1~1,000 μg/ml. The extract of A. radix showed no significant proliferation of osteoblast. However, the extract of G. radix and D. rhizome showed maximum 148% and 133% proliferation effects. The extract of G. radix also increased alkaline phosphatase activity and the maximum was 122% at 100 μg/ml compared to that of control. The nodule formation by the method of the Alizarin red S staining increased compared to control. These results suggest that G. radix is able to perform the bone formation and prevent osteoporosis.

      • Low-Level Microsatellite Instability as a Potential Prognostic Factor in Sporadic Colorectal Cancer

        <P><B>Abstract</B></P><P>Although microsatellite instability-high (MSI-H) colorectal cancers (CRCs) have been shown to exhibit a distinct phenotype, the clinical value of MSI-low (MSI-L) in CRC remains unclear. We designed this study to examine the clinicopathologic characteristics and oncologic implications associated with MSI-L CRCs.</P><P>We retrospectively reviewed data of CRC patients from 3 tertiary referral hospitals in Korea, who underwent surgical resection between January 2003 and December 2009 and had available MSI testing results. MSI testing was performed using the pentaplex Bethesda panel. Clinicopathologic features and oncologic outcomes were compared between MSI-L and microsatellite stable (MSS) CRCs; prognostic factors for survival were also examined.</P><P>Of the 3019 patients reviewed, 2621 (86.8%) were MSS, and 200 (6.6%) were MSI-L; the remaining 198 (6.6%) were MSI-H. MSI-L and MSS CRCs were comparable in terms of their clinicopathologic features, with the exception of proximal tumor location (MSI-L 30.0% vs MSS 22.1%, <I>P</I> = 0.024) and tumor size (MSI-L 5.2 ± 2.6 cm vs MSS 4.6 ± 2.1 cm, <I>P</I> = 0.001). No differences were detected in either 3-year disease-free survival (MSI-L 87.2% vs MSS 82.6%, <I>P</I> = 0.121) or 5-year overall survival (OS) (MSI-L 74.2% vs MSS 78.3%, <I>P</I> = 0.131) by univariable analysis. However, MSI-L was an independent prognostic factor for poor OS by Cox regression analysis (hazard ratio 1.358, 95% confidence interval 1.014–1.819, <I>P</I> = 0.040).</P><P>MSI-L may be an independent prognostic factor for OS in sporadic CRCs despite their clinicopathologic similarity to MSS. Further studies investigating the significance of MSI-L in the genesis and prognosis of CRCs are needed.</P>

        A Comparison of Two Techniques for Ultrasound-guided Caudal Injection: The Influence of the Depth of the Inserted Needle on Caudal Block

        Background: Caudal epidural injections have been commonly performed in patients with low back pain and radiculopathy. Although caudal injection has generally been accepted as a safe procedure, serious complications such as inadvertent intravascular injection and dural puncture can occur. The present prospective study was designed to investigate the influence of the depth of the inserted needle on the success rate of caudal epidural blocks. Methods: A total of 49 adults scheduled to receive caudal epidural injections were randomly divided into 2 groups: Group 1 to receive the caudal injection through a conventional method, i.e., caudal injection after advancement of the needle 1 cm into the sacral canal (n = 25), and Group 2 to receive the injection through a new method, i.e., injection right after penetrating the sacrococcygeal ligament (n = 24). Ultrasound was used to identify the sacral hiatus and to achieve accurate needle placement according to the allocated groups. Contrast dyed fluoroscopy was obtained to evaluate the epidural spread of injected materials and to monitor the possible complications. Results: The success rates of the caudal injections were 68.0% in Group 1 and 95.8% in Group 2 (P = 0.023). The incidences of intravascular injections were 24.0% in Group 1 and 0% in Group 2 (P = 0.022). No intrathecal injection was found in either of the two groups. Conclusions: The new caudal epidural injection technique tested in this study is a reliable alternative, with a higher success rate and lower risk of accidental intravascular injection than the conventional technique.         미국 텔레비전 시트콤 여배우들의 성격과 헤어스타일의 연관성 - Sex & the City를 중심으로 -

        < Sex & the City > which tells about the love, career, and life of women in their 20s and 30s has 4 main female characters. These 4 characters have had an influence not only on the hair and fashion styles of Korean women in their 20s and 30s but also the way they think. It seems that the desire of Korean women to accept the Western culture after the Korean War is similar to their wanting to imitate the hair styles and thought of characters in the American TV dramas or sitcoms. In this regard, this paper examines the relations between the images of women and the images that different hair styles give. The correlations between the characters and hair styles of the 4 main figures in < Sex & the City > are turned out to be as follows: First, Carrie Bradshaw, a sex columnist has a liberal and expressive character. She has her own styling so unique as to relieve stress by buying luxury shoes. And her chic fashion styles are added to her image. Accordingly, her hair style is not tied up to a certain one but shows a wide range of styles from straight to curly and diverse updos. Carrie shows the best styling fit to T.P.O(Time, Place, Occasion) and hair styles of extrovert women which are not stereotyped. Second, Samantha Jones is a brand PR director who has much self-respect. With daring fashion styles which reveal her body line and big accessories, she expresses her confidence. She sticks to medium―length hair but shows diverse hairdos from curly to straight depending on styles. Using the straight style of cold look and the curly style of mild look, she shows the extreme looks. Third, Miranda Hobbes is a lawyer of a strong image. She enjoys suits and short hair which go well with an image of a lawyer. Having the least diverse hair styles of the 4 figures, she shows changes in hair style when wearing a makeup and when doing nothing. Although she demonstrates the increased rights of women, by showing unkempt looks quite often she shows the negative side of increased women’s rights. Fourth, Charlotte York is a gallery curator of an introvert image. Having the romantic look blended with elegant elements, she sticks to the medium-length, non-layered brunette. The most glamorous look she has shown is the big curly hair. She expresses her styles using a hair band or having updos. Her non-layered hair reflects her conservative character as she is; comparing to the liberal styles of Carrie or Samantha, the neat pony-tail style Charlotte enjoys shows her character as well.
