      • NEDD4L limits cAMP signaling through ubiquitination of CREB‐regulated transcription coactivator 3

        <P>The transcription factor cAMP-responsive element-binding protein (CREB) is involved in a variety of physiologic processes. Although its activity appears to be largely correlated with its phosphorylation status, cAMP-mediated dephosphorylation and the subsequent nuclear migration of the CREB-regulated transcription factors (CRTCs) are required to stimulate CREB transcriptional activity. Among the 3 identified mammalian homologs of CRTCs, CRTC3 has been shown to be expressed predominantly in adipose tissues in response to catecholamine signals that regulate lipid metabolism. Here, we show that prolonged cAMP signaling down-regulates CRTC3 in a proteasome-dependent manner and that neural precursor cell-expressed developmentally down-regulated gene 4-like (NEDD4L), a specific ubiquitin ligase for CRTC3, is responsible for this process. By recognizing the PY motif of CRTC3, NEDD4L interacts with CRTC3 and promotes its polyubiquitination. Interaction between NEDD4L and CRTC3 is further boosted by cAMP signaling, and this enhanced interaction appears to be dependent on the cAMP-mediated phosphorylation of NEDD4L at the Ser448 site. Furthermore, we show that food withdrawal stimulates NEDD4L phosphorylation in mice, which then show a decrease of adipose tissue CRTC3 protein levels.       • 남자 대학생에서 걷기와 달리기 운동 시 체지방과 골격근량에 대한 비교

        The purpose of this study was to investigate the difference in body fat mass and skeletal muscle mass during walking and running after resistance training based on healthy college student. One male college student (age = 25 years, Height = 175.0 cm, Weight = 86.9 kg, Body fat = 27%) volunteered to participate in the study. Body composition was assessed by bioelectrical impedance (Inbody 270, Korea). Walking exercise (4.8 km/hr, 60 min) was performed for 3 weeks, followed by running exercise (7.0 km/hr, 26 min) for 3 weeks. The experimental results are as follows. Running was more effective at dropping body weight and BMI than walking, however walking was more effective at reducing body fat (%) and increasing skeletal muscle mass (kg) than running in male college student. In conclusion, both walking and running provide benefits for body composition, and the best exercise for weight and body fat loss is the one that it actually do on a regular basis.

        Clinical Characteristics and Metabolic Features of Patients with Adrenal Incidentalomas with or without Subclinical Cushing’s Syndrome

        Background: The aim of this study was to examine the clinical characteristics of adrenal incidentalomas discovered by computed tomography (CT) and to investigate metabolic features of subclinical Cushing’s syndrome (SCS) in patients with adrenal incidentalomas in a tertiary hospital in Korea. Methods: This retrospective study examined the clinical aspects of 268 patients with adrenal incidentalomas discovered by CT at Soonchunhyang University Bucheon Hospital. Clinical data and endocrine function of the patients as well as histological findings were obtained from medical records, while anatomic characteristics were analyzed by reviewing imaging studies. Hormonal tests for pheochromocytoma, Cushing’s syndrome, and aldosterone-secreting adenoma were performed. Results: Most (n=218, 81.3%) cases were nonfunctioning tumors. Of the 50 patients with functioning tumors (18.7%), 19 (7.1%) were diagnosed with SCS, nine (3.4%) with overt Cushing’s syndrome, 12 (4.5%) with primary aldosteronism, and 10 (3.7%) with pheochromocytoma. Malignant tumors (both primary and metastatic) were rare (n=2, 0.7%). Body mass index, fasting glucose, hemoglobin A1c, and total cholesterol were significantly higher in patients with SCS in comparison with those with nonfunctioning tumors. The prevalence of type 2 diabetes mellitus and hypertension were significantly higher in patients with SCS compared with those with nonfunctioning tumors. Conclusion: Functioning tumors, especially those with subclinical cortisol excess, are commonly found in patients with adrenal incidentalomas, although malignancy is rare. In addition, patients with SCS in adrenal incidentalomas have adverse metabolic and cardiovascular profiles.

      • Dopaminergic neural differentiation of human adipose-derived mesenchymal stem cells transplanted in a rat model of Parkinson's disease

        Mesenchymal stem cells constitute an potential cellular source to promote brain regeneration with Parkinson's disease. Mesenchymal stem cells have significant advantages over other stem cell types and greater potential for immediate clinical application. The purpose of this study was to investigate whether hMSCs from the human adipose tissue could be induced to differentiate into dopaminergic cells and to assess the developmental potential of hMSC for selectively replacing the midbrain dopamine neurons lost in Parkinson's disease in vitro and in vivo. MSCs were cultured under conditions that promote differentiation of dopaminergic neuron. Using media that include SHH, FGF8, and GDNF. the MSCs were induced in vitro to become dopaminergic neurons. The expressions of the LIM homeobox transcription factor 1, alpha (Lmx1a), tyrosine hydroxylase(TH) proteins were determined by immunofluorescence. Lmx1a has been shown sufficient to confer neurogenic activity on mesencephalic floor plate cells and to determine a mesencephalic dopaminergic neurons fate. This result suggests that hMSCs have the ability to differfentiate into dopaminergic neurons. hMSCs were then transplanted into the striatal in a rat model of Parkinson's disease. The rats were unilaterally lesioned in the substantia nigra with 6-hydroxydopamine and were tested for rotational apomorphine-induced behavior. Following differentiation of dopaminergic neuron, cells displayed dopaminergic morphology and that they expressed dopaminergic marks genes. Finally transplantation of hMSCs into the striatal of Parkinsonian rats resulted in improvement of their behavioral deficits by apomorphine-induced rotational behavior. The hMSCs transplanted rats were proved to be better than compared with the transplantation of PBS. Immunohistochemical analysis of grafted brains revealed that abundant hMSCs survived from the grafts and some of them displayed dopaminergic marks. Our results indicate that hMSC may serve as a good cell source for the treatment of neurodegenerative diseases and have high potential for being used in multiple applications. This cellular approach might become a restorative therapy in Parkinson's disease.

      • Surface-modified Silk Hydrogel Containing Hydroxyapatite Nanoparticle

        We define hydrogel as a polymer network containing a large amount of water or biological fluid in a 3-D structure. Because of the physical or chemical chains present in a hydrogel, it is stable in aqueous environment. Therefore, it has been used in diverse medical fields. In addition, by controlling the gelation degree of polymer solution (the state prior to hydrogel) hydrogels can be easily applied to damaged tissue area. This unique structure and properties of hydrogel shares a similarity with ECM (Extracellular matrix) in that it has a potential to be applied in tissue engineering field. Especially, the injectable property and ECM like structure can be applied to bone regeneration. Out of several polymers can be form hydrogels, silk fibroin (SF) has an excellent biocompatibility, biodegradability and it can be used to create bone regeneration scaffold in the form of hydrogel. In this study, we fabricated a SF hydrogel containing hydroxyapatite nanoparticle (HAp). To improve the dispersibility of HAp in the SF aqueous solution, we chemically modified the surface of HAp particles using hyaluronic acid (HA) – dopamine (DA) conjugate. Since SF aqueous solution has a long term gelation time, we utilized ultra-sonication method to induce a rapid gelation. Stability of HAp in SF aqueous solution was measured by ELS and TGA. Finally, FT-IR and WAXD were used to evaluate the changes of secondary structure of silk hydrogel according to concentrations of hydroxyapatite nanoparticle concentration.

        사람과 상호작용하는 소셜 로봇을 위한 인공지능 기반 스피드 퀴즈 콘텐츠의 설계와 구현

        본 논문에서는 사람과 상호작용이 가능한 소셜 로봇에서 구동할 수 있는 스피드 퀴즈 콘텐츠의 설계 및 구현 방법을 제안하고, 구현을 위해 필요한 지능 모듈을 개발하는 방법을 제시한다. 또한, 지능 모듈 블록들을 배치하고 연결하여 맵을 구성하는 과정을 통해 스피드 퀴즈 콘텐츠를 구현하는 방법을 제안한다. 최근 소프트웨어 교육이 의무화되고 프로그래밍에 대한 관심이 높아지고 있다. 하지만 프로그래밍은 프로그래밍 언어에 대한 기초 지식이 없는 학생들이 바로 접근하는 데에는 어려움이 따라, 초심자에게 적합한 블록형 프로그래밍 플랫폼에 대한 관심이 커지고 있다. 본 논문에서 사용한 블록형 프로그래밍 플랫폼은 사람과 로봇과의 상호작용을 지원하여 즉각적이고 직관적인 프로그래밍이 가능한 플랫폼이다. 본 논문에서는 스피드 퀴즈 콘텐츠를 위해 구현한 지능 모듈을 블록형 프로그래밍 플랫폼 내에서 블록화하여 사용하였다. 본 논문에서 제안하는 스피드 퀴즈 콘텐츠의 시나리오를 구현하기 위해 총 3가지의 이미지 기반 인공지능 모듈을 구현한다. 지능 모듈 외에도 다양한 기능 블록을 배치함으로써 스피드 퀴즈 콘텐츠를 구현하였다. 본 논문에서는 스피드 퀴즈 콘텐츠 시나리오를 설계한 방법과 이를 위한 지능 모듈을 구현한 방식을 제안한다. In this paper, we propose a design and implementation method of speed quiz content that can be driven by a social robot capable of interacting with humans, and a method of developing an intelligent module necessary for implementation. In addition, we propose a method of implementing speed quiz content through the process of constructing a map by arranging and connecting intelligent module blocks. Recently, software education has become mandatory and interest in programming is increasing. However, programming is difficult for students without basic knowledge of programming languages to directly access, and interest in block-type programming platforms suitable for beginners is growing. The block-type programming platform used in this paper is a platform that supports immediate and intuitive programming by supporting interactions between humans and robots. In this paper, the intelligent module implemented for the speed quiz content was used by blocking it within a block-type programming platform. In order to implement the scenario of the speed quiz content proposed in this paper, we implement a total of three image-based artificial intelligence modules. In addition to the intelligent module, various functional blocks were placed to implement the speed quiz content. In this paper, we propose a method of designing a speed quiz content scenario and a method of implementing an intelligent module for speed quiz content.

      • <i>Staphylococcus aureus</i> lipoproteins augment inflammatory responses in poly I:C-primed macrophages

        <P><B>Abstract</B></P> <P>Secondary bacterial infection contributes to severe inflammation following viral infection. Among foodborne pathogenic bacteria, <I>Staphylococcus aureus</I> is known to exacerbate severe inflammatory responses after infection with single-stranded RNA viruses such as influenza viruses. However, it has not been determined if <I>S. aureus</I> infection enhances inflammatory responses after infection with RNA enteric viruses, including rotavirus, which is a double-stranded RNA virus. We therefore investigated the molecular mechanisms by which a cell wall component of <I>S. aureus</I> enhanced inflammatory responses during enteric viral infection using poly I:C-primed macrophages, which is a well-established model for double-stranded RNA virus infection. <I>S. aureus</I> lipoproteins enhanced IL-6 as well as TNF-α production in poly I:C-primed macrophages. Pam2CSK4, a mimic of Gram-positive bacterial lipoproteins and <I>S. aureus</I> lipoproteins, also significantly enhanced IL-6 production in poly I:C-primed macrophages. While IFN-β expression was increased in poly I:C-primed macrophages treated with Pam2CSK4 or <I>S. aureus</I> lipoproteins, the level of IL-6 enhancement in poly I:C-primed macrophages was decreased in the presence of anti-IFN-α/β receptor antibody, suggesting that IFN-β plays an important role in enhanced IL-6 production. Phosphatidylinositol-3-kinase, Akt, ERK and NF-κB were also involved in the enhanced IL-6 production.         D2GSNP: a web server for the selection of Single Nucleotide Polymorphisms within human disease genes

        D2GSNP is a web-based server for the selection of single nucleotide polymorph isms (SNPs) within genes related to human diseases. The D2GSNP is based on a relational database created by downloading and parsing OMIM, GAD, and dbSNP, and merging it with positional information of UCSC Golden Path. Totally our server provides 5,142 and 1,932 non-redundant disease genes from OMIM and GAD, respectively. With the D2GSNP web interface, users can select SNPs within genes responding to certain diseases and get their flanking sequences for further genotyping experiments such as association studies.

        리좀의 특성을 적용한 묵호항 철도하역장 재생 계획

        묵호항 철도하역장은 묵호항을 개설했을 때부터 동해시 성장의 중심지였음에도 현재는 일반인의 출입 통제와 화물 및 어획량 감소로 경제·공간적 가치가 저하되어있는 상태로, 철도하역장이 이전되는 2020년쯤에는 철도하역장을 포함하여 주변의 슬럼화가 가속화되고 있다. 이런 상황에서 묵호항 철도하역장의 재생계획안을 제시함에 있어 리좀의 이론을 적용하여 표현코자 한다. 따라서 본 연구는 유·무형의 가치가 저하된 묵호항 철도하역장에 대해 리좀의 특징을 적용한 건축형태 계획안을 제시하고, 묵호항 철도하역장과 주변 구역의 이질적인 요소들을 활용하여 묵호항 철도하역장을 재생함으로써 그 가치를 높이는 것을 목적으로 한다. 이런 목적을 바탕으로 묵호항 철도하역장 재생계획안의 적용가능성과 적용한 계획안의 긍정적 효과에 대해 확인하고자 한다. 연구의 과정에서는 이론과 대상지에 적용하고자 하는 계획안 연구로 범위를 규정하여 실용적 가치 여부에 관한 연구는 본 논문의 영역에서 다루지 않고 추후의 과정으로 남겨놓고자 한다 The Mukho Port Railroad district with the receiving and shipping space has been the center of growth in Donghae city since its opening at Mukho Port. However, the value of the economy and space has been reduced due to the restriction of citizen’s access and the decrease of freight trains and fish catches, hence the slumism of Mukho port railroad district is accelerating further. In this situation, this study proposes a regeneration plan of Mukho Port railroad district with the receiving and shipping space to apply it to the Rhizome theory. Therefore, this study suggests the architectural plan that applies the characteristic of the rhizome to the railway dockyard of Mookhohang Railway which has low value of tangible and intangible. The purpose of this study is to investigate the applicability and effectiveness of the restoration plan of the Mukhoang Port Railway Dockyard based on this purpose, I will leave it as a process of the study of practical value.

        Analysis of Some Online Questions with High Frequency about Dental Treatment in Korea

        Background: The Internet has advantages in terms of accessibility and amount of information, and the search for health information over the Internet is increasing exponentially. The purpose of this study is to analyze the information generated about some dental treatment on the internet by year. Methods: Naver Knowledge (JisikIn in Korean) which is an interactive search service was selected as the first search site in Korea. Scaling, wisdom tooth extraction, and endodontic treatment that can be paid by Korean health insurance were selected. Finally, 4,729 questions about scaling, 23,963 wisdom teeth extraction questions and 17,733 endodontic treatment questions were extracted. The question contents, the information about the questioner and the answerer, and an error of answers were investigated. Frequency analysis was used and chi-square test was used if necessary. Results: The most frequently asked questions were discomfort and dissatisfaction after the treatment. The need for treatment was the second in questions of the wisdom tooth extraction and endodontic treatment, but the health insurance benefit was the second in dental scaling. Most of the questioners didn’t disclose personal information. The public answered the most in 2013∼2014, but the highest percentage of the respondents was experts in 2017. Responses were mostly personal experience, but showed a tendency to decrease with years, and professional knowledge showed an increasing tendency. The error of the answer has also gradually decreased. Conclusion: Questions about dental care over the Internet are increasing exponentially, experts are responding increasingly, and errors in answers are decreasing. Nevertheless, it is necessary to pay attention to the related expert group to prevent misinformation.
