지골피(地骨皮)가 $H_{2}O_{2}$에 의한 $LLC-PK_1$ 세포의 Redox Status 및 $NF-{\kappa}B$ Signaling에 미치는 영향

Objectives : This study was aimed to verify the cytoprotective function, antioxidative effect and inflammation genes inhibitory effects of Lycium chinense Milie. Therefore the generation of superoxide anion radical ( $O_2\;^-$), peroxynitrite ($ONOO^-$), nitric oxide (NO) and prostaglandin $E_2$ $(PGE_2)$ was investigated in the renal epithelial cells of mouse. Effects of Lycium chinense Milie on the expression of inflammation-related proteins, $IKK-{\alpha}$. $p-IKK-\alpha\beta$, $p-I{\kappa}B-\alpha$, $NF-{\kappa}B$ (p50, p65), COX-2 and iNOS, were examined by western blotting. Methods : For this study, the fluorescent probes were used, namely dihydrorhodamine 123 (DHR 123), 4.5-diaminofluorescein (DAF-2) and 2',7'-dichlorodihydrofluorescein diacetate (DCFDA). Western blotting was performed using anti-$IKK-\alpha$, anti-phospho $IKK-\alpha\beta$, anti-phospho $I{\kappa}B-\alpha$, anti-$NF-{\kappa}B$ (p50, p65), anti-COX-2 and anti-iNOS, respectively. Results : Lyciutn chinense Milie reduced $H_{2}O_{2}$-induced cell death dose-dependently. It inhibited the generation of $O_2\;^-$, $ONOO^-$, NO and $PGE_2$ in the $H_{2}O_{2}$-treated renal epithelial cells of mouse in vitro. Lycium chinense Milie inhibited the expression of $IKK-\alpha$, $p-IKK-\alpha\beta,\;p-I{\kappa}B-\alpha$, COX-2 and iNOS genes by means of decreasing activation of $NF-{\kappa}B$. Conclusions : According to above results. Lycium chinense Milie recommended to be applied in treatment for the inflammatory process and inflammation-related diseases.
구안괘사(口眼?斜)환자의 일반적 특성 및 비수(肥瘦)에 따른 임상적 고찰

Objective : This study was aimed to investigate the general features and differences between obesity and skinniness of patients with Bell's palsy. Methods : We measured the sex, age. BMI. pulse diagnosis and HBGS (House-Brackmann Grading System) of 234 patients who were diagnosed with Bell's palsy. Results and Conclusions : The results with statistical significance were as follows (1) The distribution of age revealed that 40s was the most at 30.8 %: (2) The improvement period in facial palsy patients with sub-paralysis was shorter than whole-paralysis. And in one part the more we treated, the shorter the improvement period was: (3) In distribution of fat rate in facial palsy patients, obesity was the most at 61.37%, low weight 15.88%. So we found that the fatter the patients was. the higher the onset rate was: (4) In distribution of pulse diagnosis in facial palsy patients with obesity. the ratio of Xu mai (虛脈) was 67.06%. Shi mai (實脈) 32.94%. The Xu mai was similar to Qi xu (氣虛). So we found that the facial palsy patients with obesity were more Qi xu than with low weight. In distribution of pulse diagnosis in facial palsy patients with skinniness, the ratio of Chi mai (遲脈) was none. Shuo mai (數脈) was most: (5) In distribution of region in facial palsy patients with obesity-Xu mai. the ratio of left was 45.10%, right 54.90%, but this result was not statistically significant.
사용자 인증을 위한 심전도 기저선 영점 조정 기법 연구

최근 지문과 얼굴 인증에 비해 위•변조에 강인한 심전도를 이용하여 사용자 인증 기술이 활발하게 연구되고 있다. 심전도 신호는 전기적 신호로 여러 잡음이 포함되어 측정된다. 특히 피험자 호흡으로부터 발생되는 기저선 변동 잡음이 심할수록 비선형 잡음에 의해 형태학적 특징 변형으로 사용자 인증 성능이 저하된다. 기저선 변동 잡음을 제거하기 위해 주파수 필터를 이용하지만 심전도의 형태학적 특징을 왜곡시키는 문제가 있다. 본 논문에서는 심전도에서 T-P파, P-R파 중앙값을 이용한 1차 회귀분석으로 기저선 변동을 추정하고, 추정된 기저선을 영점으로 조정한다. 실험결과, 기저선 영점 조정 후 개인 심전도 10주기에서 기존 방법보다 평균 유사도가 최대 4.6% 향상되고, 기저선 영점 조정 전 사용자 인증 최대 EER 성능은 1.82%였지만 기저선 영점 조정 후 최대 성능은 1.11%로 향상됨을 확인하였다. Recently, user authentication technology using ECG, which is stronger in forgery and alteration than fingerprint and face authentication, has been actively studied. The ECG signal is an electrical signal that contains several noises. In particular, as the baseline fluctuation noise generated from the subject"s breathing becomes more severe, user authentication performance is degraded due to morphological characterization due to nonlinear noise. Although frequency filters are used to remove baseline fluctuation noise, there is a problem of distorting the morphological characteristics of the ECG. In this paper, baseline variation is estimated by first-order regression analysis using median T-P wave and P-R wave in ECG, and the estimated baseline is adjusted to zero. Experimental results showed that the average similarity was improved by up to 4.6% in 10 cycles of ECG after baseline zeroing. The maximum EER performance of user authentication before baseline zeroing was 1.82%, but the maximum performance after baseline zeroing was improved to 1.11%.
사인추출물의 인슐린 유사 성장인자-I의 합성과 피부 노화 개선에 대한 효과

본 연구에서 프로모터-리포터 분석방법을 통해 인슐린 유사 성장인자-I의 프로모터를 자극하는 천연물을 선별한 결과 사인 추출물이 가장 좋은 프로모터 자극효과를 나타냈으며, 무모생쥐에서 패쇄첩포 후 RT-PCR로 실험한 결과 IGF-1 mRNA를 35% 증가시키는 것으로 나타났다. 사인 추출물의 피부 주름개선 효과를 알아보기 위하여 인체 섬유아세포에서는 type-I collagen과 MMP-1 합성 변화를 관찰하였으며, 무모생쥐에서는 콜라젠의 증가와 진피 두께를 관찰하였다. 그 결과, 동위원소를 이용한 콜라젠 증가실험에서 type-I collagen은 38% 증가하였으며 무모생쥐에서 실시한 RT-PCR 결과에서는 mRNA가 21% 증가하는 것으로 관찰되었다. MMP-1 효소발현의 경우 ELISA 분석을 통해서 63%의 높은 발현저해능을 확인하였고 Western blot에서도 발현이 저해되는 것을 확인하였다 추출물을 무모생쥐에 패쇄첩포 하였을 경우 대조군에 비해 진피 두께가 두꺼워지고 콜라젠 양도 증가되는 것으로 조직염색 관찰을 통해 확인하였다. 이상의 결과를 통해 사인추출물은 피부 주름개선에 좋은 효과를 나타내는 것으로 보이며, 여기에는 인슐린 유사성장 인자-I의 발현증가와 관련된 기전이 관여하는 것으로 판단된다. We screened several materials to stimulate IGF-1 promoter activity using luciferase reporter assay and found that Amomi Semen extract (ASE) among them is the most powerful stimulator. We also studied about the anti-wrinkle effect of ethanolic extract of Amoni Semen in vitro and in vivo. Semi-quantitative RT-PCR showed that the extract elevated the presence level of IGF-1 mRNA. And [<sup>3</sup>H] proline incorporation and semi-quantitative RT-PCR showed that the extract increased the expression of type-I collagen compared with vehicle in vitro and in vivo, respectively. Significant inhibition of MMP-1 expression was determined by ELISA and Western blot. Finally, topical treatment of the extract on hairless mouse's dorsal skin expanded the volume of collagen and dermal thickness. These results suggest that Amomi Semen may be a good candidate for improving extracellular matrix through the increase of collagen expression and inhibition of MMP-1 expression. Moreover, this study enables us to guess that IGF-1 stimulated by the extract may be involved in the mechanism of anti-wrinkle effect of it.

Object : This study was designed to evaluate the effects of oriental medicine therapy on a lung cancer patient. Methods : The patient was admitted on August 20. 2008 and remained until September 24. 2008. She was treated with herbal medicine. acupuncture and moxa therapy. Result & Conclusion : After the treatment. the symptoms and chest X-ray results improved. This result suggests that oriental medicine is an effective treatment for some lung cancer patients. Nevertheless more clinical case reports are needed.

Objectives : The aims of this study were to investigate the cytoprotective, antioxidative and inflammation genes inhibitory effects of Patriniae Radix on the mouse LLC-$PK_1$ cells (renal epithelial cells). Methods : The cytoprotective effect of Patriniae Radix was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The antioxidative effect was measured in terms of generation amount of superoxide anion radical (${\cdot}{O_2}^-$) by 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), nitric oxide (NO) by 4,5-diaminofluorescein (DAF-2), peroxynitrite ($ONOO^-$) by dihyldrorhodamine 123 (DHR 123) and prostaglandin $E_2$ ($PGE_2$) by $PGE_2$ immunoassay on $H_2O_2$-treated LLC-$PK_1$ cells. For measuring of inflammation genes inhibitory effects, western blot was performed to detect IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, NF-${\kappa}B$ (p50, p65), COX-2, iNOS, IL-$1{\beta}$ and VCAM-1 protein level in cytosol fractions from LLC-$PK_1$ cells. Results : Patriniae Radix extract reduced the $H_2O_2$-induced cell death and inhibited the amount of $H_2O_2$-induced ${\cdot}{O_2}^-$, NO, $ONOO^-$, $PGE_2$ generation dose-dependently on the mouse LLC-$PK_1$ cells in vitro. Also Patriniae Radix extract inhibited the expression of IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, COX-2, iNOS, IL-$1\beta$ and VCAM-1 genes dose-dependently by means of decreasing activation of NF-${\kappa}B$. Conclusions : According to above results, it was identified that Patriniae Radix had the cytoprotective, antioxidative and inflammation genes inhibitory effects. So it was suggested that Patriniae Radix would be effective to the treatment for the inflammatory process and inflammation-related diseases.
