In this study, a rapid immuno-chromatographic assay was developed to detect Salmonella Typhimurium, This assay was based on the lateral flow immunoassay, Two different anti-Salmonella polyclonal antibody (Pab) are used in this strip assay for the double antibody sandwich immunoassay method, The strip sensor was composed of sample pad, conjugation pad, membrane, and absorbent pad. For the optimal conjugation between nano-gold and anti-Salmonella Pab, the pH value of colloidal gold solution was determined to 8.4, To prevent non-specific binding between nano gold and anti-Salmonella Pab, sample pad and membrane was treated with PBS buffer, included 2% BSA and 10% sucrose, The control and detection zones were visually detected according to antigen-antibody binding. The detection limit of the developed strip assay was 105CFU/mL within 10 min, and improved until 104CFU/mL after 12 hr.

이미지 분석을 이용한 살모넬라 신속 진단키트의 측정감도 향상

식중독을 일으키는 대표적인 원인균인 살모넬라를 검출하기 위해 측방 유동형 간이진단 키트를 제작하였다. 또한, 제작된 간이진단 키트의 검출한계를 향상시키고 살모넬라 균수를 정량적으로 분석하기 위해 이미지 분석 시스템을 적용하였다. 그 결과 간이진단 키트의 검출한계는 10? cfu/mL에서 105 cfu/mL로 향상시킬 수 있었다. 살모넬라 균수의 정량적 분석을 위해 Tpeak/Cpeak와 Tarea/Carea값을 이용하여 다중회귀분석을 수행하였으며 개발된 다중회귀방정식 중에서 가장 단순하고 결정계수가 높은 다중회귀방정식을 선정하였다. 추가로 제작된 간이진단 키트를 이용하여 개발된 다중회귀방정식을 검증한 결과 결정계수가 0.9393으로 우수한 선형성을 나타내었다. 본 연구에서 개발된 다중회귀방정식을 이용하여 더 정확하게 간이진단 키트에서 측정된 살모넬라 균수를 예측하는 것이 가능하였다. The objective of this study was to improve the detection limit of rapid detection kit for Salmonella Typhimurium by image analysis system. The rapid detection kit was comprised of four elements: sample pad, conjugate pad, nitrocellulose pad and absorbent pad. Gold nanoparticle and Salmonella antibody were used as a tag and a receptor. Salmonella antibody and goat rabbit IgG antibody were used as test and control lines on nitrocellulose membrane. The color intensity of test line began to increase from 10?CFU/mL of Salmonella sample. A multiple linear regression analysis was employed to explain the relationship between predicted and measured number of Salmonella cells. The developed model could successfully predict the cell number of Salmonella with validation against extra-experimental result.

This study was performed to develop and optimize a rapid strip kit for the dual-detection of Aflatoxin B1 (AF) and Ochratoxin A (OTA). The strip kit is composed of sample pad, conjugation pad, nitrocellulose membrane, and absorbent pad. Manually spotted mycotoxin-BSA conjugates and anti-mouse IgG on nitrocellulose membrane were used as the test line and the control line, respectively. Conditions for rapid and easy detection of mycotoxins, conjugation between toxin-MAbs and nano Au particles, pretreatment method of pads, and composition of standards solutions were optimized. Feasibility of the strip kit to detect AF and OTA were evaluated with standard samples. Test results were acquired in 10 min and the detection limits were 100 ppm for AF and OTA, respectively.
