진균에 의한 오염은 재배사에서 톱밥 배지를 이용한 표고 재배에 중대한 영향을 미치기 때문에 표고 재배사 실내 공기에 존재하는 진균에 대한 모니터링을 수행하던 중 여러 진균을 분리하였다. 분리된 진균 중에는 국내 미기록인 Aspergillus pulverulentus와 Cosmospora butyri 등 2종을 비롯하여 기록은 있으나 균학적 확증이 부족한 Lecanicillium psalliotae와 L. antillanum 등 2종이 존재하였다. 본 논문에서는 이들 균류에 대한 형태적 특성과 더불어 internal transcribed spacer (ITS) rDNA region 또는 ${\beta}$-tubulin 유전자 염기서열에 기반한 계통학적 분석 결과를 기술하였다. Mold contamination is one of the detrimental factors affecting sawdust media-based shiitake cultivation in greenhouses. During mold monitoring of indoor air of greenhouses, several fungi were isolated. Among them, Aspergillus pulverulentus and Cosmospora butyri were found to be new in Korea and Lecanicillium psalliotae and L. antillanum were known in Korea without taxonomic validation. In this study the morphological characteristics and phylogenetic analysis based on the internal transcribed spacer (ITS) rDNA region or ${\beta}$-tubulin gene of the four identified species were described.

Shiitake breeding requires the procedures of mating of two different parental strains and selection of hybrid strains that have good traits for the mushroom production. In this study, we tested the possibility of the use of chromogenic plate-based assay for extracellular enzyme production in order to assess and find good biochemical properties-possessed hybrid strains that were generated from genetic cross of the monokaryotic strains derived from two different parental strains of Lentinula edodes Sanjo-101ho and Sanjo-108ho. We observed that there was difference in the ability of producing β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease among the monokaryotic strains. We could also comparatively assess that the ability of the seven extracellular enzymes production in the hybrid strains depended on the mating combination of the monokaryotic strains. Our results demonstrate that the assessment method for extracellular enzyme production using chromogenic plate assay could be usefully applied to the assessment of the hybrid strains derived from the breeding procedure of L. edodes.
p38 Mitogen-Activated Protein Kinase의 억제를 통한 진세노사이드 Rg3의 Thromboxane A₂ 억제 효과

G-Rg3는 인삼의 성분으로서 많은 효과가 밝혀졌지만, 혈소판에서의 억제 기작에 대한 연구는 미흡하다. 본 연구에서는 G-Rg3가 혈소판에서 합성되어 분비되는 TXA₂의 생산에 관여한다고 알려진 효소인 COX-1, TXAS와 신호전달 분자인 p38 MAPK, cPLA₂의 인산화를 어떻게 조절하며 혈소판응집을 억제시키는지 규명하고자 하였다. 그 결과 G-Rg3는 thrombin이 유도한 혈소판 응집을 강력하게 억제하였고, p38 MAPK, cPLA₂의 인산화와 COX-1, TXAS의 활성을 감소시키며 arachidonic acid 분비와 TXA₂의 합성을 농도의존적으로 억제하였다. 또한 G-Rg3는 thrombin이 촉진시킨 p38의 인산화를 억제함으로써 과립방출의 지표인 ATP와 serotonin의 방출을 억제하였다. 따라서 G-Rg3는 인체혈소판에서 TXA₂의 생성을 억제함으로써 혈소판 응집억제를 나타내는 치료 및 예방약물로서 유용한 가치가 있다고 여겨진다. Thromboxane A₂ plays a central role in hemostasis and thrombus formation, which stimulates other platelets as an agonist leading intracellular signaling cascades. In this study, we tried to explain the inhibitory mechanism of 20(S)-ginsenoside Rg3 (G-Rg3) on thromboxane A₂ production by assaying the activities of TXA₂ production-associated microsomal enzymes and associated signaling molecules. Thrombin-stimulated human platelet aggregation was dose-dependently decreased by G-Rg3 and attenuated thromboxane A₂ production via down regulation of microsomal cyclooxygenase-1 and thromboxane A₂ synthase activities. In addition, arachidonic acid release, which is a precursor of TXA₂ was decreased by G-Rg3 through dephosphorylation of cytosolic phospholipase A₂ and p38 mitogen-activated protein kinase. Overall, the results revealed the specific signaling pathway of TXA₂ production via G-Rg3. Therefore, G-Rg3 is a beneficial novel compound inhibiting TXA₂ production, which may prevent platelet aggregation-mediated thrombotic disease.
인산화단백질의 억제를 통한 진세노사이드 Rg6의 혈소판 기능 및 혈전 형성 조절

G-Rg6는 인삼의 성분으로서 혈소판 억제 기작에 대한 연구는 아직 미흡하다. 본 연구에서는 G-Rg6가 platelet aggregation과 thrombus formation에 미치는 영향을 평가하였고, 이와 관련된 신호전달 분자인 IP₃RI와 VASP의 인산화를 어떻게 조절하는지 규명하고자 하였다. 그 결과 G-Rg6는 collagen이 유도한 platelet aggregation을 강력하게 억제하였고, IP3RI와 VASP의 인산화 및 세포 내 Ca<SUP>2+</SUP> 동원과 fibrinogen binding의 활성을 감소시키며 clot retraction 반응을 농도 의존적으로 억제하였다. 따라서 G-Rg6는 인체 혈소판에서 platelet aggregation과 thrombus formation에 억제 효과를 나타내는 물질이므로 치료 및 예방약물로서 잠재적 가치가 있다고 여겨진다. The root of Panax ginseng is used in traditional medicine in Eastern Asia and various studies have reported that P. ginseng has inhibitory effects on cardiovascular disease including stroke and myocardial infarction. Each factor causing cardiovascular disease is known to have various processes. Among these processes, platelets are the most important. Therefore, inhibiting the activity of platelets is an essential factor for the prevention of platelet-mediated cardiovascular diseases. This study examined the inhibitory mechanism of ginsenoside Rg6 (G-Rg6) on human platelet aggregation and thrombus formation. Collagen-induced human platelet aggregation was inhibited dose-dependently by G-Rg6, and it suppressed the collagen-induced elevation of [Ca<SUP>2+</SUP>]i mobilization. In addition, G-Rg6 suppressed collagen-induced fibrinogen binding to αIIb/β3 and clot retraction. Thus, G-Rg6 has an inhibitory effect on human platelet activation and thrombus formation, highlighting its potential as a natural substance for preventing thrombosis and cardiovascular diseases.

Button mushroom (Agaricus bisporus) strains from diverse origins were compared in this study to obtain basicin formation on their growth and biochemical properties that are helpful for breeding. Among 31 dikaryotic strains tested, most strains showed better mycelial growth on oatmeal agar than on MEA and PDA. Mycelia of the mushroom strains revealed β-glucosidase activity the most clearly among the seven extracellular enzymes tested. All the strains showed protease activity, but β-glucosidase activity was found in 27 strains and xylanase activity was found in 30 strains. The activity of avicelase, CM-cellulase, amylase, and pectinase was detected in less than 20 strains. These results implied that enzymatic characteristics could be used as a criterion of strain selection for breeding study.
