The anthelmintic substance of the roasted fruits of Prunus mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercarial of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethyfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at 90~110℃ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

Objective: This study aimed to determine the effects of a novel surgical technique of adenomyomectomy with transient occlusion of uterine arteries (TOUA) in patients with symptomatic diffuse uterine adenomyosis. Methods: All cases of adenomyomectomy with TOUA were performed by a single surgeon from July, 2011 to Oct. 2015. Surgical outcomes included operative time, intraoperative injury to blood vessels, nerves, and pelvic organs and operative blood loss. We assessed the degree of improvement in dysmenorrhea and menorrhagia and the recurrence of adenomyotic lesions by ultrasonography at the 6-month follow-up after adenomyomectomy with TOUA and then regular follow up with 6 month-inteval. Results: one hundred-twenty women, who were refractory to medical treatment or wanted a surgical treatment for uterine preservation, underwent adenomyomectomy with TOUA using vascular clips. The mean age was 37.4 years (range: 27-48 years). The mean total surgical time was 116.1 minutes (range, 60-300 minutes, SD: 36.7). The mean estimated blood loss was 223.74 ml (range, 50-500 ml, SD: 110.91) and there were no cases of injury to the uterine arteries or pelvic nerves. The mean time of TOUA was 7.79 minutes (range: 4-10 minutes, SD: 2.94). The mean duration of hospital stay was 5.65 days (range, 4-7 days, SD: 0.85). There were no major complications requiring reoperation or readministration during the mean follow-up period of 29.5 months. At the eight-month follow-up after adenomyomectomy with TOUA, complete remission of dysmenorrhea and menorrhagia was observed in 98.4% and 100% of patients, respectively. Conclusion: Adenomyomectomy with TOUA could be a safe and effective surgical method in women with symptomatic diffuse uterine adenomyosis to preserve fertility.
USP10 modulates estrogen receptor in Uterine Adenomyosis

Objective: Adenomyosis is one of an estrogen dependent benign disease that causes severe menstrual cramp and heavy uterine bleeding. Pathophysiology of adenomyosis is known as that it derives from invasion of endometrial tissue to the myometrium. Methods: We hypothesized that estrogen receptor was modulated by posttranslational modification by upiquitine-specific peptidase -10 (USP10). We cultivated Ishikawa cell as an in vitro model of adenomyosis and analyzed 20 patients with uterine adenomyosis who received an adenomyomectomy between December 2019 and April 2020 at our hospital. During surgeries, samples were obtained from the uterus of each patient that included the adenomyosis lesion, the myometrium without adenomyosis, and endometrial tissue. Results: The interaction between USP10 and the estrogen receptor was confirmed by immunoprecipitation assays. Cell-based transactivation assays in Ishikawa cells revealed that overexpression of wild-type USP10 stimulated estrogen receptor (ER) expression. Conversely, ER was decreased when USP10 expression knock-down by siRNAs . When comparing human uterine tissue of adenomyosis, immunnochemistry of USP10 was more increased in the glands of adenomyosis than glands in endometrium. Conclusion: The data indicate that USP10 is a new cofactor that controls degradation process of estrogen receptor. This finding underlines the role of the USP10, one of ubiquitin/proteasome system is modulating the consistency of estrogen receptor in uterine adenomyosis. This suggests that USP10 plays a significant role in estrogen dependent diseases such as adenomyosis.

A 31-year-old nulliparous woman with severe diffuse uterine adenomyosis, which replaced nearly the whole uterine myometrium, visited our hospital due to severe dysmenorrhea, menorrhagia, and a desire to have a baby. The patient had a history of two spontaneous abortions. Laparotomic adenomyomectomy with transient occlusion of uterine arteries (TOUA) was performed safely and the patient tried in vitro fertilization and achieved a intrauterine twin pregnancy after recovery time of the operation. At 31+6 weeks of gestation, a male neonate baby weighing 1,620 g and a male neonate baby weighing 1,480 g were born by transverse lower segment cesarean delivery. There was no complication after the operation. The babies were discharged after receiving routine neonatal intensive care for neonatal respiratory distress syndrome. Adenomyomectomy with TOUA technique would be an option for conservative surgical treatment in patients with severe diffuse whole uterine adenomyosis. This is the first report of twin pregnancy after diffuse whole uterine adenomyomectomy with TOUA.

Brominated flame retardants (BFRs) are present in many consumer products ranging from fabrics to plastics and electronics. Wide use of flame retardants can pose an environmental hazard, which makes it important to determine the mechanism of their toxicity. In the present study, dose-dependent toxicity of tetrabromobisphenol A (TBBPA), a flame retardant, was examined in male prepubertal rats (postnatal day 18) treated orally with TBBPA at 0, 125, 250 or 500 mg/kg for 30 days. There were no differences in body weight gain between the control and TBBPA-treated groups. However, absolute and relative liver weights were significantly increased in high dose of TBBPA-treated groups. TBBPA treatment led to significant induction of CYP2B1 and constitutive androstane receptor (CAR) expression in the liver. In addition, serum thyroxin (T4) concentration was significantly reduced in the TBBPA treated group. These results indicate that repeated exposure to TBBPA induces drug-metabolising enzymes in rats through the CAR signaling pathway. In particular, TBBPA efficiently produced reactive oxygen species (ROS) through CYP2B1 induction in rats. We measured 8-hydroxy-2'-deoxyguanosine (8-OHdG), a biomarker of DNA oxidative damage, in the kidney, liver and testes of rats following TBBPA treatment. As expected, TBBPA strongly induced the production of 8-OHdG in the testis and kidney. These observations suggest that TBBPA-induced target organ toxicity may be due to ROS produced by metabolism of TBBPA in Sprague-Dawley rats.

As the study about the non-animal tests came to be internationally active and the interest in the animal welfare gradually became increased, OECD TG (Test Guideline) 401 that has been used since 1987 was abolished in 2002. Because TG401 is acute oral toxicity method depending on survival and death of many animals, it was heavily criticized. Therefore, the three alternative methods were developed. OECD TG420 and TG423 determine the class of chemicals according to GHS (Globally Harmonized Classification System for Chemical Substances and Mixtures) classification and OECD TG425 suggests the predicted LD50 of chemicals using AOT425 program. In this study, 10 chemicals were selected. The internationally admitted TG420, TG423 and TG425 were introduced and established through the method that these chemicals were orally administrated to SD female rats and then, the results were observed. Each chemical belonged to already known GHS class in the study using TG420 and TG423 and predicted LD50 was same or higher in the study using TG425 compared to already known LD50 value. In conclusion, the result of our study confirmed the decrease in the animal number and validated. The international harmonization of the non-animal tests will be pursued through this validation study.
