        Conessine Treatment Reduces Dexamethasone-Induced Muscle Atrophy by Regulating MuRF1 and Atrogin-1 Expression

        Conessine, a steroidal alkaloid, is a potent histamine H3 antagonist with antimalarial activity. We recently reported that conessine treatment interferes with H2O2-induced cell death by regulating autophagy. However, the cellular signaling pathways involved in conessine treatment are not fully understood. Here, we report that conessine reduces muscle atrophy by interfering with the expression of atrophy-related ubiquitin ligases MuRF-1 and atrogin-1. Promoter reporter assay revealed that conessine treatment inhibits FoxO3a-dependent transcription, NF-κB-dependent transcription, and p53-dependent transcription. We also showed by quantitative RT-PCR and western blot assays that conessine treatment reduced dexamethasone-induced expression of MuRF1 and atrogin-1. Finally, we demonstrated that conessine treatment reduced dexamethasone-induced muscle atrophy using differentiated C2C12 cells. These results collectively suggest that conessine is potentially useful in the treatment of muscle atrophy.

        Analysis of Critical COVID-19 Cases Among Children in Korea

        Background: Coronavirus disease 2019 (COVID-19) is generally asymptomatic or mild in otherwise healthy children, however, severe cases may occur. In this study, we report the clinical characteristics of children classified as critical COVID-19 in Korea to provide further insights into risk factors and management in children. Methods: This study was a retrospective case series of children < 18 years of age classified as critical COVID-19. Cases were identified by the Korea Disease Control and Prevention Agency surveillance system and medical records were reviewed. Critical COVID-19 was defined as cases with severe illness requiring noninvasive (high flow nasal cannula, continuous positive airway pressure, or bilevel positive airway pressure) or invasive mechanical ventilation, extracorporeal membrane oxygenation (ECMO) or continuous renal replacement therapy (CRRT), between January 20, 2020 and October 7, 2021. Results: Among 39,146 cases diagnosed with COVID-19 in subjects < 18 years of age, eight cases (0.02%) were identified as critical COVID-19. The median age was 13 years (range 10 month–17 years) and male-to-female ratio was 1:1. Three children had underlying diseases; one child has asthma and major depressive disorder, one child had LennoxGastaut syndrome and one child had mental retardation and was newly diagnosed with type 2 diabetes mellitus with the diagnosis of COVID-19. Among the eight children, seven were obese (body mass index range [BMI] median 29.3, range 25.9–38.2, weight-for-length > 97% for infant) and one was overweight (BMI 21.3). All patients had fever, six patients had dyspnea or cough and other accompanied symptoms included sore throat, headache, lethargy and myalgia. Radiologic findings showed pneumonia within 1–8 days after symptom onset. Pneumonia progressed in these children for 2–6 days and was improved within 5–32 days after diagnosis. Among the eight critical cases, remdesivir was administered in six cases. Steroids were provided for all cases. Inotropics were administered in one case. Six cases were treated with noninvasive mechanical ventilator and three required mechanical ventilator. One case required ECMO due to acute respiratory distress syndrome. All cases were admitted to the intensive care unit and admission period ranged from 9–39 days. Among all critical COVID-19 cases < 18 years of age, there were no fatal cases. Conclusion: To develop appropriate policies for children in the COVID-19 pandemic, it is important to monitor and assess the clinical burden in this population.

        발산적 사고를 통한 아이디어 발상의 디자인 프로세스

        이 연구는 디자이너들이 직관을 이용하는 발산적 사고로 아이디어 발상을 하는 것에 능숙하다는 것에 착안하여 이러한 발상법들을 더욱 적극적으로 활용하여 아이디어 발상을 하는 디자인 프로세스를 제안하는 것을 목적으로 한다. 즉 디자인 프로세스에서 발산적 사고를 통해 아이디어 발상을 하는 방법을 제안하는 것을 목적으로 한다. 이를 위한 연구의 방법은 다음과 같다. 첫 번째 단계로 디자인프로세스에 대해 이론적 고찰을 하였다. 1세대부터 3세대 이후까지의 세대별 디자인 프로세스를 조사하여 디자인 프로세스에서 직관적 아이디어 발상이 어떻게 활용되었는지를 조사하였다. 두 번째 단계로 직관에 대한 이론적 고찰을 하였다. 직관은 철학에서 다루어지는 개념으로 베르그송과 러셀의 견해 등을 통해 직관이 객관적 지식으로 다루어 질수 있는 지를 조사하였다. 이를 토대로 세 번째 단계에서 발산적 사고에 대해 고찰하였다. 이 단계에서 발산적 사고의 개념과 종류를 조사하고 마지막 단계로 디자인 프로세스에서 발산적 사고를 통한 아이디어 발상을 하기 위한 프로세스를 제안 하였다. This study aimed at proposing a design process based on creating ideas through divergent thinking. This idea is based on that designers are well trained to do divergent thinking a kind of intuitive thinking. The following three stages were undertaken to solve the aim. The first stage constituted an examination of the existing design processes building on the previous literature. As a result of this examination a process model of divergence and convergence emphasizing on designers' intuition and creativity was selected as this study's model. The second stage covered an examination of the existing theories of intuition including the argument of Bergson that intuition also can be used as evidence. At the third stage how to derive group creativity and how to think up foresighted ideas by using divergent thinking as one of creative thinking methods were investigated. Finally a design process model based on creating intuitive ideas through divergent thinking was suggested.

        과학기술관련 사회쟁점 미디어 정보에 대한 중학생들의 평가 양상 탐색

        대중매체를 통해 전달되는 과학기술관련 사회쟁점(SSI)에 관한 미디어 정보를 비판적으로 평가하여 합리적인 의사결정을 내릴 수 있는 능력은 시민으로서 지녀야 할 과학적 소양의 한 측면이다. 본 연구는 합리적인 의사결정을 위해 선행되어야 할 학생들의 정보 평가 양상에 관한 것으로, 일상에서 접하는 SSI 미디어 정보를 과학적 요소가 강조된 수치정보와 정서적 공감이 강조된 공감정보로 구분한 후, 이러한 정보의 특성에 따라 중학생들의 정보 평가 양상이 어떻게 다른지 탐색해보았다. 본 연구에는 중학생 96명이 참여하였으며, 신문기사 형식의 SSI 자료 내 정보의 신뢰도와 설득력 있는 정도를 묻는 질문지를 도입하였다. 질문지는 세 가지 SSI 주제에 대한 찬성 또는 반대 입장에 대해 과학적 요소가 강조된 수치정보와 정서적으로 호소하여 공감을 불러일으키는 공감정보 기사 2개로 구성되었으며, 정보가 얼마나 믿을만한가를 묻는 신뢰도 평가 문항과 정보가 얼마나 설득력 있는가를 묻는 설득력 있는 정도 평가 문항을 포함하였다. 연구 결과, 첫째 전반적으로 학생들은 SSI 맥락에서 공감정보보다 수치정보를 더 신뢰하는 것으로 나타났다. 신뢰하는 정보 요소로는 수치정보에서는 과학적 증거와 데이터를, 공감정보에서는 사례, 사회 문제, 표현, 그리고 가치를 주로 신뢰하는 것으로 나타났다. 둘째, 설득력 있는 정도에 대한 평가는 수치나 공감과 같은 정보의 유형보다는 SSI 맥락이나 내용적인 측면에 따라 상이한 것으로 나타났다. 즉, 학생들은 SSI 정보를 담은 기사의 설득력을 판단할 때 정보에 담긴 가치, 근거의 풍부함, 정보의 논리성을 바탕으로 판단하였다고 볼 수 있다. Ability to make informed decisions by critically evaluating media information on socioscientific issues (SSI) is one of the crucial elements of scientific literacy that citizens should obtain. This study aims to investigate how middle school students evaluated media information about socioscientific issues (SSI) when they faced two different types of information (i.e., numerical and empathic information). To achieve the aim, 96 middle school students responded to the questionnaires asking them to evaluate reliability and persuasiveness of SSI media information. The questionnaires consisted of two sets of newspaper articles on each SSI (pro-numerical/empathic, against-numerical/empathic). After reading the articles, the students evaluated reliability and persuasiveness of each article and wrote the reasons for their evaluation. The results were as follows: First, the students believed that news articles with numerical information were more reliable than the ones with empathic information in all SSI contexts. They tended to trust scientific evidence and data from numerical information, and real cases, societal problems, expressions, and values from empathic information. In addition, they evaluated their reliability based on the logic of information, accuracy of information, breadth and depth of data, and quantity and quality of sources both numerical and empathic information. Second, in case of evaluating persuasiveness of the articles, they focused more on the values that information contained, richness and logicality of the information, rather than the types of information, regardless of the type of information.

        This paper is to study the tendency of the development of the urban comprehensive park in the modern history of China, and take the urban comprehensive park of Wuhan as an example to study the local research through the literature.. In 1928, Wuhan Urban Comprehensive Park starting stage characteristics to the ideological education of the government as the center, in 1977 the policy of reform and opening to the outside world to entertainment center, ecology design point of view, the introduction of open space and urban plaza, to human centered design, application of science and technology. 2015 in Wuhan Urban Garden Expo theme can be seen in green life as the goal, through the design of urban comprehensive park ecology and human centered design, the application of science and technology development.

      • In silico re-identification of properties of drug target proteins

        <P><B>Background</B></P><P>Computational approaches in the identification of drug targets are expected to reduce time and effort in drug development. Advances in genomics and proteomics provide the opportunity to uncover properties of druggable genomes. Although several studies have been conducted for distinguishing drug targets from non-drug targets, they mainly focus on the sequences and functional roles of proteins. Many other properties of proteins have not been fully investigated.</P><P><B>Methods</B></P><P>Using the DrugBank (version 3.0) database containing nearly 6,816 drug entries including 760 FDA-approved drugs and 1822 of their targets and human UniProt/Swiss-Prot databases, we defined 1578 non-redundant drug target and 17,575 non-drug target proteins. To select these non-redundant protein datasets, we built four datasets (A, B, C, and D) by considering clustering of paralogous proteins.</P><P><B>Results</B></P><P>We first reassessed the widely used properties of drug target proteins. We confirmed and extended that drug target proteins (1) are likely to have more hydrophobic, less polar, less PEST sequences, and more signal peptide sequences higher and (2) are more involved in enzyme catalysis, oxidation and reduction in cellular respiration, and operational genes. In this study, we proposed new properties (essentiality, expression pattern, PTMs, and solvent accessibility) for effectively identifying drug target proteins. We found that (1) drug targetability and protein essentiality are decoupled, (2) druggability of proteins has high expression level and tissue specificity, and (3) functional post-translational modification residues are enriched in drug target proteins. In addition, to predict the drug targetability of proteins, we exploited two machine learning methods (Support Vector Machine and Random Forest). When we predicted drug targets by combining previously known protein properties and proposed new properties, an F-score of 0.8307 was obtained.</P><P><B>Conclusions</B></P><P>When the newly proposed properties are integrated, the prediction performance is improved and these properties are related to drug targets.       • IL-7 Induces an Epitope Masking of<i>γ</i>c Protein in IL-7 Receptor Signaling Complex

        <P>IL-7 signaling via IL-7R<I>α</I>and common<I>γ</I>-chain (<I>γ</I>c) is necessary for the development and homeostasis of T cells. Although the delicate mechanism in which IL-7R<I>α</I>downregulation allows the homeostasis of T cell with limited IL-7 has been well known, the exact mechanism behind the interaction between IL-7R<I>α</I>and<I>γ</I>c in the absence or presence of IL-7 remains unclear. Additionally, we are still uncertain as to how only IL-7R<I>α</I>is separately downregulated by the binding of IL-7 from the IL-7R<I>α</I>/<I>γ</I>c complex. We demonstrate here that 4G3, TUGm2, and 3E12 epitope masking of<I>γ</I>c protein are induced in the presence of IL-7, indicating that the epitope alteration is induced by IL-7 binding to the preassembled receptor core. Moreover, the epitope masking of<I>γ</I>c protein is inversely correlated with the expression of IL-7R<I>α</I>upon IL-7 binding, implying that the structural alteration of<I>γ</I>c might be involved in the regulation of IL-7R<I>α</I>expression. The conformational change in<I>γ</I>c upon IL-7 binding may contribute not only to forming the functional IL-7 signaling complex but also to optimally regulating the expression of IL-7R<I>α</I>.</P>

      • Water-Soluble Polyelectrolyte-Grafted Multiwalled Carbon Nanotube Thin Films for Efficient Counter Electrode of Dye-Sensitized Solar Cells

        <P>Water-soluble, polyelectrolyte-grafted multiwalled carbon nanotubes (MWCNTs), MWCNT-g-PSSNa, were synthesized using a “grafting to” route. MWCNT-g-PSSNa thin films fabricated by an electrostatic spray (e-spray) technique were used as the counter electrode (CE) for dye-sensitized solar cells (DSSCs). The e-sprayed MWCNT-g-PSSNa thin-film-based CEs (MWCNT-CE) were uniform over a large area, and the well-exfoliated MWCNTs formed highly interconnected network structures. The electrochemical catalytic activity of the MWCNT-CE at different thicknesses was investigated. The MWCNT-g-PSSNa thin film showed high efficiency as a CE in DSSCs. The power conversion efficiency (PCE) of the DSSCs using the MWCNT-g-PSSNa thin-film-based CE (DSSC-MWCNT) was >6% at a CE film thickness of ∼0.3 μm. The optimum PCE was >7% at a film thickness of ∼1 μm, which is 20−50 times thinner than conventional carbon-based CE. The charge transfer resistance at the MWCNT-CE/electrolyte interface was 1.52 Ω cm<SUP>2</SUP> at a MWCNT-CE thickness of 0.31 μm, which is lower than that of a Pt-CE/electrolyte interface, 1.78 Ω cm<SUP>2</SUP>.
