      • DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

        <P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

        Effects of Pilger Rolling and Heat Treatment on Microstructure and Corrosion Resistance of Ag-electroplated 304 Stainless Steel Tubes

        We introduce a cost-effective method that combines electroplating with metal cold working processes to manufacture compositetubes with robust corrosion resistance. First, a 3-step electroplating process was developed to form an adhesive anduniform Ag coating on the outer wall of a 304 stainless steel tube. The process consisted of a Ni-strike step for removing thepassivation layer of the initial 304 tube, Cu deposition for smoothing the surface by adding a buffer layer, and Ag deposition. To reinforce the interfacial adhesion between the electroplated layers and the 304 tube and increase the area of the Ag coating,a pilger rolling or pilger rolling + heat-treatment process was performed after the Ag electroplating process. Scanningelectron microscopy of the composite tubes after each process indicated that the as-deposited Ag coating changed fromhaving a rough to smooth surface after only pilger rolling and with additional heat treatment. Electron backscatter diffractionanalysis of the microstructures and textures of the tubes revealed that dynamic recrystallization occurred extensively duringpilger rolling, resulting in the formation of a relatively defect-free grain structure without heat treatment. Furthermore,electrochemical polarization curves determined that the Ag-coated composite tubes are superior to the uncoated 304 tubein terms of corrosion resistance under Cl−atmosphere, owing to the formation of a AgCl passivation layer during testing. Surface analysis of the composite tubes suggests that the properties of the AgCl passivation layer are related to the grainsize of the Ag coating and the density of plastic-deformation-induced defects.

        Environmental Microbiology / Microbial Diversity : Calcium Carbonate Precipitation by Bacillus and Sporosarcina Strains Isolated from Concrete and Analysis of the Bacterial Community of Concrete(s)

        Microbially induced calcium carbonate precipitation (CCP) is a long-standing but re-emerging environmental engineering process for production of self-healing concrete, bioremediation, and long-term storage of CO2. CCP-capable bacteria, two Bacillus strains (JH3 and JH7) and one Sporosarcina strain (HYO08), were isolated from two samples of concrete and characterized phylogenetically. Calcium carbonate crystals precipitated by the three strains were morphologically distinct according to field emission scanning electron microscopy. Energy dispersive X-ray spectrometry mapping confirmed biomineralization via extracellular calcium carbonate production. The three strains differed in their physiological characteristics: growth at alkali pH and high NaCl concentrations, and urease activity. Sporosarcina sp. HYO08 and Bacillus sp. JH7 were more alkali- and halotolerant, respectively. Analysis of the community from the same concrete samples using barcoded pyrosequencing revealed that the relative abundance of Bacillus and Sporosarcina species was low, which indicated low culturability of other dominant bacteria. This study suggests that calcium carbonate crystals with different properties can be produced by various CCP-capable strains, and other novel isolates await discovery.

      • 활동기 척추동물의 십이지장 배상세포내 점액질에 관한 조직화학적 연구

        본 실험은 다람쥐(Tamias sibiricus asiaticus Gmeli), 살모사(Agkistrodon halys Pallas), 황소개구리(Rana catesbeiana Shaw), 뱀장어(Anguilla japonica Temminck et Schlegel)등의 소장 배상세포내 점액질에 대한 성상을 밝히기 위하여 PAS reaction, Alcian blue pH2.5 및 Alcian blue pH0.4, 그리고 ABpH2.5-PAS reaction 복합염색등을 시행한 결과 다음과 같은 결론을 얻었다. 1. 사용한 각 동물에 있어서 소장 배상세포는 중성점액질, 약산성 및 강산성 황화점액질을 포함하고 있었다. 2. 중성점액질이 각 동물에 있어서 가장 안정된 분포를 보였으며 약산성 황화점액질도 비교적 고르게 분포되어 있었다. 3. 강산성 황화점액질은 다람쥐, 살모사 및 황소개구리에서는 소량 관찰되어 불안정한 분포를 보였으며 뱀장어에서는 관찰되지 않았다. 4. 각 실험동물의 십이지장 배상세포내에 포함된 점액질의 성상은 동물에 따라서 그 양과 염색성에 다소의 차이를 인정할 수 있다. In general, the goblet cells in the small intestines of experimental animals contained neutral mucosubstances, weakly and strongly acidic sulfated mucosubstances. Neutral mucosubstances show most stable distribution in the experimental animals. Mucosubstances in the small intestines of eels were composed of only neutral mucosubstances and weakly acidic sulfated mucosubstances. The contents and stain properties of mucostances produced in the intestinal goblet cells vary a little in portions within a species and in the same cell types of different species. This studies were performed in order to study composition of the mucosubstance in the goblet cells of small intestines of the various vertebrates. The specimens used in this study were obtained from the small intestines of 4 species of vertebrates, squirrels (Tamias sibiricus asiaticus Gmelin), vipers (Agkistrodon halys Palls), frogs (Rana catesbeiana Shaw), eels (Anguilla japonica Temminck et Scchlegel). All specimens were fixed in 10% neutral buffered formalin, embedded in paraffin wax, and sectioned at a thickness of 4㎛. They were stained with hematoxylin-eosin for general histological observation and with Periodic Acid-Schiff reaction, alcian blue pH2.5 and 0.4 and alcian blue pH2.5-Periodic Acid-Schiff reaction for detection of the mucosubstances. The mucous compositions in the goblet cells of the small intestines of various vertebrates examined by their staining reaction were neutral mucosubstances, weakly and strongly acidic sulfated mucosubstances.

        Halo-analogues of pyrimidine nucleoside are of medicinal interest. The best knowns are 5-Fluorouracil and Ftorafur. But 5-Fluorouracil has severe side effects, i.e., toxicity to the central nerve system, which its clinical applications restrict. To overcome such problems we synthesize some derivatives of 5-Fluorouracil : 1-Alkenyl-5 fluorouracil and 1-Hydroxyalkyl-5-Fluorouracil. These are to be used as starting compounds for another derivatives and to be tested for their antitumor activities.

        This paper presents the development of standardized modules and an integrated program which will be used for social environmental education. The study objectives are to develop modules which are based on various environmental topics, to relate the regional environmental topics to the educational modules and program, and to include the demand of students in the modules and program. Literature and questionary surveys, as well as, module development method were utilized as research tools. The titles of developed modules are "The Green Plant Earth," "Unexpected Resources: Save Resources and Recycle", "Water as the Mother of All Lives", "The Blue Sky with Clean Air", and "Soil as the Home of All Living and Non-living Things". It is hoped that the developed modules and program are utilized as course materials and curriculum for social environmental education in various educational institutes. The study results will be materialized as multimedia in near futher.

      • 조밀한 다이아몬드 막의 성장

        RF-MW 혼합프로세스를 사용하여 다이아몬드 막을 만들 때, 우선 RF 플라즈마CVD법으로 실리콘 기판 위에 다이아몬드 시드를 증착시키고, 그 시드로부터 MW 플라즈마CVD법에 의해 다이아몬드 막을 성장시켰다. RF-MW 혼합프로세스를 사용하여 성장된 다이아몬드 막은 MW 프로세스만으로 성장된 막에 비해 결정입계의 크기는 작고, 조밀해졌으며, 결정성 또한 더 좋아졌다. 성장된 다이아몬드 막은 전자현미경, X-선 회절기, Raman분광기 등으로 분석하였다. In case of making the diamond films by using RF-MW mix-process, at first. diamond seeds were deposited on silicon substrate by RF plasma CVD, and then diamond layer grown by MW plasma CVD from the seeds. The grain-size of diamond films deposited by using RF-MW mix-process was smaller and denser, in addition, crystallity of diamond films was better than MW plasma CVD process. The deposited diamond films were analyzed by SEM, XRD, and Raman spectroscopy.

        Inhibitory Effect of Cordycepin on Human Platelet Aggregation

        Cordycepin separated from Cordyceps militaris is a major physiologic active component in Cordyceps militaris. The platelet aggregation is stimulated by Ca^(2+), which is either mobilized from intracellular endoplasmic reticulum or transported from extracellular space. cGMP antagonizes the actions of Ca^(2+). Based on these facts, we have investigated the effects of cordycepin on the mobilization of Ca^(2+) and the production of cGMP on collagen (10 ㎍/ml)-induced human platelet aggregation. Cordycepin potently stimulated the human platelet aggregation induced by collagen (10 ㎍/ml) in a dose-dependent manner. Cordycepin (500 μM) inhibited also the collagen-induced human platelet aggregation in the presence both 1 mM and 2 mM of CaCl_(2). These are in accord with the results that cordycepin inhibited the Ca^(2+)-influx on collagen-induced human platelet aggregation. These results suggest that cordycepin decrease the intracellular Ca^(2+) concentration to inhibit collagen-induced human platelet aggregation. Besides, cordycepin increased the level of cGMP on collagen-induced human platelet aggregation. This result is related with the decrease of intracellular Ca^(2+) concentration, because cGMP inhibits the mobilization of Ca^(2+). In addition, cordycepin inhibited the human platelet aggregation induced by LY-83583, inhibitor of guanylate cyclase. This result suggested that cordycepin inhibit the platelet aggregation by stimulating the activity of guanylate cyclase. In conclusion, we demonstrated that cordycepin might have the antiplatelet function by inhibiting Ca^(2+)-mobilization via the stimulation of the production of cGMP.
