When evaluating the extent of the impact of water pollutants on the surrounding area, we would like to present the target level (proposal) of the quality of the environment, which is the standard for evaluation. We propose the environmental quality target level for substances that need to be applied domestically by investigating overseas cases operating the integrated environmental management system and the implications of domestic environmental pollutant management. The appropriateness of the environmental quality target level reviewed in this study was determined using data from the water quality measuring network, and future improvement measures were proposed. We review the available methodologies for setting quality objectives for the environment. It proposed the environmental quality target level for 21 substances that have domestic water pollutant emission standards and do not have environmental standards, and proposed future improvement measures. If it is necessary to add quality target-level items of the environment in the future, it is believed that expansion will be possible based on the methodology presented in this study.
치커리(Cichorium intybus L.)와 쑥갓(Chrysanthemum coronarium L.)의 기내 종자발아 개선

치커리와 쑥갓에 있어 기내 파종 시 발아 및 무균묘 생산을 증진시키기 위한 적절한 방법을 제안하기 위해 일련의 실험들이 수행되었다. NaOCl의 적정 처리농도와 시간 검정을 통해 치커리 'Precole'과 'Chiavari' 및 쑥갓 'Okiku 3' 모두 Naocl 5%로 15분간 처리하는 것이 발아 및 유묘형성을 촉진시키고 오염발생을 억제하는 것으로 관찰되었다. 배지조성에 있어 MS와 1/2 MS 처리구들의 발아반응성은 뚜렷이 차이가 나타나지 않았으며, sucrose 농도가 높아짐에 따라 발아 및 유묘형성이 지연되었다. 배양용기에 따른 반응성의 차이는 치커리 'Precole'의 경우 petri-dish에서 양호한 발아율 및 유묘형성율을 보여주었지만, 치커리 'Chiavari'와 쑥갓 'Okiku 3'에서는 용기에 큰 영향을 받지 않았다. 초음파 처리는 치커리의 발아율 및 유묘형성율을 증가시켰지만, 쑥갓 'Okiku 3'에서는 초음파 처리효과가 나타나지 않았다. 따라서 치커리와 쑥갓 모두에서 Naocl 5%, 15분간 종자소독하고, sucrose가 무첨가된 1/2 MS 배지로 petri-dshi를 이용하며, 초음파 처리는 Naoci 5% 용매에서 120분간 하는 것이 기내 발아향상에 적당한조건으로 선정되었다. A series of experiments were conducted to suggest efficient in vitro germination conditions to improve germination rate and aseptic seedling production in chicory and garland chrysanthemum. For this purpose, various levels of NaOCl concentration and soaking treatment time combinations were tasted. Fifteen minutes of soaking treatment with 5% NaOCl solution significantly improved germination and seedling formation percentages and prevented contamination in both chicory 'Precole' and 'Chiavari' and garland chrysanthemum 'Okiku 3'. There was no significant difference in germination response between MS and 1/2 MS medium compositions, but germination and seedling formation were delayed as sucrose concentration increased. Although petri-dish among various culture containers gave rise to improved germination responses in chicory' Precole', there was no significant difference in gelmination responses among culture container types in chicory 'Chiavari' and garland chrysanthemum 'Okiku 3'. The ultrasonic treatment stimulated germination and seedling formation in both chicory cultivars but there was no effect in garland chrysanthemum 'Okiku 3'. The results suggested 15 minutes with 5% NaOCl for seed sterilization, 1/2 MS medium with no sucrose and petri-dish as proper medium composition and culture container, and 120 minutes with 5% NaOCI solution of ultrasonic treatment as promoting method for in vitro germination in chicory and garland chrysanthemum.
Agrobacterium tumefaciens를 이용한 γ-TMT 유전자의 치커리 형질전환

γ-tocopherol과 γ-tocotrienol을 α- 형태로 바꾸는 역할을 하는 γ-tocopherol methyltransferase(γ-TMT) 유전자를 치커리(Cichorium intybus L. var. foliosum) ‘Precole’에 도입하고자 본 연구를 수행하였다. 치커리 자엽절편체를 이용한 hygromycin 항생제 감응성 검정을 실시하였으며, 10㎎ㆍL⁻¹ 이상의 hygromycin을 첨가한 경우 적정 재분화 배지에서도 callus 및 신초의 형성이 현저하게 저하되었다. Arabidopsis thaliana에서 분리된 1,047bp 크기의 γ-TMT 유전자를 pCAMBIA1302 vector에 재조합하였고, 재조합된 vector는 LBA4404 계통의 Agrobacterium tumefaciens에 도입시켰으며, 이것을 무균발아시켜 약 7일된 치커리 자엽절편체와 함께 공동배양하여 형질전환을 실시하였다. 일부 절편체는 IAA 1㎎ㆍL⁻¹, BAP 1㎎ㆍL⁻¹, hygromycin 10 ㎎ㆍL⁻¹, cefotaxime 200㎎ㆍL⁻¹와 AgNO₃ 10㎎ㆍL⁻¹가 첨가된 MS 선발배지에서도 callus와 신초의 재분화가 이루어졌으며, 선발배지에서 재분화된 신초들은 200㎎ㆍL⁻¹ cefotaxime이 첨가된 1/2 MS 발근배지에서 정상적으로 발근되었다. 총 14개의 순화개체 중 2개체에서 γ-TMT 유전자의 발현을 확인하였으며 이를 위해 PCR, RT-PCR, Northern blot 등 다양한 분석방법이 이용되었다. The γ-tocopherol methyltransferase (γ-TMT) gene, which encodes an enzyme converting γ-tocotrienol and γ-tocopherol to α-forms, was introduced into chicory (Cichorium intybus L. var. foliosum) ‘Precole’ and the gene expression was investigated. Results from hygromycin sensitivity test showed that callus and shoot formation from cotyledon explants were completely inhibited by the supplementation of above 10 ㎎ㆍL⁻¹ hygromycin, and this level was applied for the primary selection. One-week-old cotyledon explants were co-cultivated with Agrobacterium tumefaciens strain LBA4404 carrying binary vector pCAMBIA1302 harboring 1047 bp γ-TMT gene which originated from Arabidopsis thaliana. The explants successfully produced calli and adventitious shoots on the MS selection medium with 1 ㎎ㆍL⁻¹ IAA, 1 ㎎ㆍL⁻¹ BAP, 10 ㎎ㆍL⁻¹ hygromycin, 200 ㎎ㆍL⁻¹ cefotaxime, and 10 ㎎ㆍL⁻¹ AgNO₃. New shoots regenerated on the selection medium were rooted on the 1/2MS rooting medium with only 200 ㎎ㆍL⁻¹ cefotaxime. PCR screening using γ-TMT gene-specific primers produced 2 transgenic plants among 14 putative transgenic plants and the successful expression of γ-TMT gene in these plants was confirmed by RT-PCR and Northern blot analysis.
