Bacillus stearothermophilus No. 239의 돌연변이주 MNNG 8이 생산하는 CGTase를 사용하여 감자전분을 동시 액화, cyclodextrin(CD) 생산을 하였다. 고농도(30%)의 감자전분이 29%의 수율로 CD로 전환되었으며 그 때의 조건은 pH 6.0, 80℃, 4.3 mM CaCl_2, 40℃에서 1g의 전분당 3.0 DAU의 CGTase를 첨가하는 것이다. Simultaneous liquefaction and cyclodextrin (CD) production were conducted on potato starch using cyclomaltodextrin glucanotransferase (CGTase) from a mutant strain MNNG8 of Bacillus stearothermophilus No. 239. A high concentration (30%) of potato starch was converted to cyclodextrins (CDs) with 29% yield in the conditions of pH 6.0, temperature 80℃, 4.3 mM CaCl_2, CGTase addition of 3.0 dextrinizing activity unit (DAU) at 40℃/g starch.

The analytical method for the determination of phosphorus in foods was validated by inductively coupled plasma optical emission spectrometry (ICP-OES) in terms of precision, accuracy, recovery efficiency and linearity. Regression analysis revealed good correlation coefficient, higher than 0.999. Recovery efficiencies of the minerals ranged from 90.36% to 110.63%, and the limit of detection (LOD) and the limit of quantification (LOQ) were 0.0745 mg/ kg and 0.2482 mg/kg, respectively. The value of inter-day and intra-day ranged from 1.43 to 3.23% and from 0.40 to 1.77%. The recovery efficiencies ranged from 97.8 to 110.6%. The method was also compared with Molybdenum blue colorimetric method using certified and statistically significant difference was also not observed in the between two different analytical methods. The ICP-OES method was applied to phosphorus determination in commonly consumed foods. The obtained results suggest that the method verified in the present study may be used as an official analytical method for clear understanding of phosphorus database for national health promotion.
Bacillus stearothermophilus에 의한 Cyclomaltodextrin Glucanotransferase의 생산

A microorganism capable of producing high level of extracelluar cyclomaltodextrin glucanotransferase(EC 2.4.1.19; CGTase) was isolated from soil. The isolated strain No. 239 was identified as Bacillus stearothermophilus. The maximal CGTase production(about 7.0units/㎖) was observed in medium containing 2% soluble starch, 0.5% defatted soybean meal, 0.1% NaH_2 PO_4·2H_2O and 0.015% CaCl_2 with initial pH7.0. The strain was cultured at 55℃ for 48 hr with reciprocal shaking. At 0.83% substrated concentration potato starch was the optimum substrate with 50.1% conversion to cyclodextrin(CD) after the reaction at 65℃ for 24 hr (CGTase 10 unit/g starch). Using soluble starch as substrate (5% substrate concentration, CGTase 10 unit/g starch), the maximum conversion of 40% was obtained at 11 hr reaction, and the ratio of α-, β- and γ-CD production at this time were 1.0:1.3:0.4, respectively.
Cyclomaltodextrin Glucanotransferase의 생산을 위한 Bacillus stearothermophilus 균주의 돌연변이

Bacillus stearothermophilus No. 239 isolated from soil was mutated with N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) to yield a series of mutants with increasing levels of cyclomaltodextrin glucanotransferase (EC 2.4.1.19; CGTase) production. After five consecutive mutation steps, a mutant MNNG 8 with about 14 times of CGTase activity than the parent strain was obtained. The mutant had a smaller colony size and the growth rate was slower. It also had different optimum cultural conditions from those of the parent strain with sucrose, a cheap carbon source, replacing soluble starch as the best carbon source, for example. The overall enhancement of the CGTase production after optimization of the cultural conditions was about fourty times so that the mutant produced a culture broth with about 80 dextrinizing activity unit (DAU) at 40℃/㎖.
