        Chloroplast markers for detecting rice grain-derived food ingredients in commercial mixed-flour products

        The development of DNA techniques such as quantitative real-time PCR (qRT-PCR) has led to advancements in the field of illegal food product detection. Here, we report a PCR-based method to detect rice grain flour in commercial mixed-flour products. To select the chloroplast genes available for a rice-specific marker, we analyzed chloroplast DNA (cpDNA) polymorphisms in several gene families from five plant species, including rice, adlay, barley, maize, and wheat by using comparative sequence analysis. We found two potential rice-specific marker genes, rpoB and rpoC2, which exhibited relatively high numbers of segregating sites compared to other genes. We designed gene-specific primers for rpoB and rpoC2 on the basis of sequence differences, and identified the appropriate PCR amplification in grain flour samples derived from six Korean rice varieties using the linearity test of the qRT-PCR assay. To test the applicability of these cpDNA markers, we performed a qRT-PCR assay on total DNA obtained from different commercial food products, and successfully detected the rice-specific cpDNA region (rpoB and rpoC2) in several commercial food products that were declared to contain rice. Thus, the reported qRT-PCR assay may prove to be a useful tool for the detection of various rice flours in commercial mixedflour products such as Sunsik.

        조직시민행동과 직무소진의 관계에 따른 성별, 재직기간의 조절효과

        This paper is to investigate the relationship between organizational citizenship behavior(OCB) and job burnout, and the moderating effect of employee’s gender and tenure in Korea Polytechnic Colleges. The results of our study are as follows; First, in the relationship between OCB and job burnout, altruism and sportsmanship are negatively related with emotional exhaustion, depersonalization and the reduced personal accomplishment, and conscientiousness and civic virtue are negatively related with only depersonalization and the reduced personal accomplishment. Second, in the relationship between OCB and job burnout, gender affected negatively in the moderation of 2 relationships - between altruism and the reduced personal accomplishment, and between sportsmanship and depersonalization. And it affected positively in the moderation of 2 relationships - between civic virtue and emotional exhaustion, and between civic virtue and the reduced personal accomplishment. Third, in the relationship between OCB and job burnout, tenure affected positively in the moderation of 3 relationships - between altruism and emotional exhaustion, between sportsmanship and the reduced personal accomplishment, and between civic virtue and the reduced personal accomplishment. 외환위기와 금융위기를 경험하면서 공공기관의 취업 및 고용에 대한 역할이 강조되고 있다. 한국폴리텍대학의 높은 취업률은 구성원들의 직무역할은 물론 직무외적 역할에 대한 자발적 참여의식이 바탕이 되고 있다. 이에 대해 본 연구에서는 조직시민행동과 직무소진의 관계를 분석하였다. 분석결과, 첫째로 조직시민행동의 구성요소인 이타성과 양심성은 직무소진의 모든 구성요소인 정서적 고갈, 비인간화, 성취감 감소에 부정적 영향을, 양심성과 공익성은 비인간화, 성취감 감소에만 부정적 영향을 미치는 것으로 나타났다. 둘째로 이타성이 성취감 감소에 미치는 효과와 비불평성이 비인간화에 미치는 효과에서 여성보다 남성의 경우에서 감소효과가 크며, 공익성이 정서적 고갈과 성취감 감소에 미치는 효과에서는 남성에 비해 여성에서 감소효과가 큰 것으로 나타났다. 셋째로 조직시민행동과 직무소진의 관계에서 재직기간은 이타성과 정서적 고갈, 비불평성과 성취감 감소, 공익성과 성취감 감소에서 긍정적 영향을 미치는 것으로 나타났다.

      • 유무선 통합환경에서 QoS 향상을 위한 라우팅 알고리즘

        본 논문은 유무선 통합환경에서 QoS(Quality of Service) 향상을 위한 라우팅 알고리즘을 제안한다. 본 논문에서는 유무선 통합환경에서 QoS의 요구사항 및 특징을 분석하고 유선망에 노드 이동성을 추가하여 유선망 뿐 아니라 유무선 통합환경에서도 QoS 향상을 가능하게 해주는 M-Dijk 알고리즘을 제안하였다. 제안한 M-Dijk 알고리즘에 따라서 각 수신자는 유무선 통합환경에서 자원을 보다 효율적으로 활용할 수 있게 될 것이다. In this paper, we design a routing algorithm for QoS(Quality of Service) improvement in the unified environment of wire/wireless. The M-Dijk algorithm for QoS improvement in the wire/wireless unified environment is proposed by adding node's mobility to the wire network. According to this proposed M-Dijk algorithm, each receiver will be able to utilize network resources more efficiently.

        내적 정서의 주관적 조형화를 통한 화예작품제작 - 본인작품 ‘아버지에 대한 기억’을 중심으로 -

        본 연구는 작가의 내적 정서와 관련된 사건을 회상하고 그를 바탕으로주관적 조형화를 시도하여 화예 창작을 행한 연구이다. 연구자는 고향의사진, 관련된 시집, 소설, 논문을 조사하였으며 아버지가 즐겨 부르시던 음악을 다시 듣고, 영화 등 다양한 미디어를 통해 고향의 기억을 떠올렸고구체화했다. 또한 동생들과의 면담을 통해 아버지와의 소중한 추억들을 구체화하여 상기시켰다. 이렇게 상기된 기억을 토대로 내적 정서를 분석하고아버지의 성품을 잘 드러낼 수 있는 기억들을 선택하여 모티브를 찾아 구체화하고 발전시켰다. 구체화된 기억을 토대로 행한 다양한 스케치를 토대로 내적 정서를 잘 표현할 수 있는 재료를 찾아 조형 실험을 하였다. 다양한 실험을 행하고 형태를 변형시키는 가운데 조형적 어휘와 표현을 개발하며 화예작품으로 완성시켜 나갔다. 이 과정에서 ‘온가족 합창’, ‘무거운짐’, ‘우울한 노랫가락’, ‘머리파마’, ‘한밤중 산을 넘으심’의 5개 작품을 완성할 수 있었으며 그를 가지고 학위 청구 전을 개최하였다. This study aims to recollect events of my internal emotions, thereby attempt to make subjective figurative images, and ultimately create floral art works. I was committed to recollect all the precious memories on my hometown and father, through his favorite songs, his pictures, interviews with my brothers, poems and novels on fathers, essays, use of mass media, and films. Based on the recollection, I picked up and embodied feelings conceived as motives, which fully describe my father's characteristics and personality, and analyzed and evolved their internal emotion. Nevertheless, it would be a success to produce floral art works from my childhood and hometown memories by making the most use of living plants. It would be of great significance that I could take out the emotion on my old days to create art works and develop methodology, through these series of works. What is better, I could get healed in the process of this work

        Transcriptome analysis for identifying possible gene regulations during maize root emergence and formation at the initial growth stage

        The root plays an important role during plant development and growth, i.e., the plant body maintenance, nutrient storage, absorption of water, oxygen and nutrient from the soil, and storage of water and carbohydrates, etc. The objective of this study was attempted to determine root-specific genes at the initial developmental stages of maize by using network-based transcriptome analysis. The raw data obtained using RNA-seq were filtered for quality control of the reads with the FASTQC tool, and the filtered reads were pre-proceed using the TRIMMOMATIC tool. The enriched BINs of the DEGs were detected using PageMan analysis with the ORA_FISHER statistical test, and genes were assigned to metabolic pathways by using the MapMan tool, which was also used for detecting transcription factors (TFs). For reconstruction of the co-expression network, we used the algorithm for the reconstruction of accurate cellular networks (ARACNE) in the R package, and then the reconstructed co-expression network was visualized using the Cytoscape tool. RNA-seq. was performed using maize shoots and roots at different developmental stages of root emergence (6–10 days after planting, VE) and 1 week after plant emergence (V2). A total of 1286 differentially expressed genes (DEGs) were detected in both tissues. Many DEGs involved in metabolic pathways exhibited altered mRNA levels between VE and V2. In addition, we observed gene expression changes for 113 transcription factors and found five enriched cis-regulatory elements in the 1-kb upstream regions of both DEGs. The network-based transcriptome analysis showed two modules as co-expressed gene clusters differentially expressed between the shoots and roots during plant development. The DEGs of one module exhibited gene expressional coherence in the maize root tips, suggesting that their functional relationships are associated with the initial developmental stage of the maize root. Finally, we confirmed reliable mRNA levels of the hub genes in the potential sub-network related to initial root development at the different developmental stages of VE, V2, and 2 weeks after plant emergence.

        Resequencing of a Core Rice Mutant Population Induced by Gamma-Ray Irradiation and Its Application in a Genome-Wide Association Study

        A total of 2561 gamma ray-induced mutants (M7) were evaluated in six abiotic stress treatments, and then principal component analysis was performed to score their phenotypes. Resequencing was performed on 123 selected rice mutants and wild-type rice to assess the structural changes in the genomes of these mutants, such as single nucleotide polymorphisms (SNPs) and insertion/deletion (InDels). A total of 5160 (avg. 87.3 per plant) SNPs and 7195 (avg. 121.9) InDels were observed in the 200-Gy lines, whereas 5650 (avg. 88.3) SNPs and 7676 (avg. 119.9) InDels were found in the 300-Gy lines. We found that one candidate gene (Os02g0528900, which is similar to the gene for pleiotropic drug resistance (PDR)-like ATP-binding cassette (ABC) transporter) contained two SNPs that were included in principal component 4 via a genome-wide association study. The 300-42 line had the lowest phenotype score against heat, possibly suggesting the existence of a relationship between these SNPs and heat stress. Co-expression analysis of the candidate gene involved in responses to toxic substances and the chemical stimulation of significant biological processes. Therefore, the studied targeting induced local lesions in genome (TILLING) population could be a useful source of mutations in the breeding of new genetic resources for forward and reverse genetics.
