Combined Effects of Baicalein and Docetaxel on Apoptosis in 8505c Anaplastic Thyroid Cancer Cells via Downregulation of the ERK and Akt/mTOR Pathways

Background: Anaplastic thyroid cancer (ATC) is one of the most lethal human malignancies. Docetaxel, a microtubule stabilizer, isa common chemotherapeutic agent used to treat various metastatic cancers. However, prolonged use results in various side effectsand drug resistance. Flavonoids, such as baicalein, are accepted chemotherapeutic and dietary chemopreventive agents with manyadvantages, such as greater accessibility, affordability, and lower toxicity, compared with traditional chemotherapy agents. In thisstudy, we evaluated whether baicalein enhances the effects of docetaxel on apoptosis and metastasis in 8505c ATC cells. Methods: The 8505c cells were treated with baicalein or docetaxel individually and in combination. Cell viability was measured by MTT(thiazolyl blue tetrazolium bromide) assay, and apoptosis was detected by fluorescence microscopy of Hoechst-stained cells. The expressionof apoptotic (Bax and caspase-3), anti-apoptotic (Bcl-2), angiogenic (vascular endothelial growth factor [VEGF], transforminggrowth factor β [TGF-β], E-cadherin, and N-cadherin), and signaling (extracellular signal-regulated kinase [ERK] mitogen activated proteinkinase [MAPK], Akt, and mammalian target of rapamycin [mTOR]) proteins was determined by Western blot analysis. Results: The combination of baicalein (50 or 100 μM) and docetaxel (10 nM) significantly inhibited proliferation and induced apoptosiscompared with monotherapies. The combination treatment significantly inhibited the expression of Bax, caspase-3, VEGF,TGF-β1, E-cadherin, N-cadherin, and mTOR, but decreased the expression of Bcl-2 and significantly decreased the phosphorylationof ERK and Akt. Conclusion: The combination of baicalein and docetaxel effectively induced apoptosis and inhibited metastasis in 8505c cellsthrough downregulation of apoptotic and angiogenic protein expression and blocking of the ERK and Akt/mTOR pathways in 8505ccells. These results suggest that baicalein enhances the anticancer effects of docetaxel in ATC.

사례관리 담당 사회복지사의 직무요구가 이직의도에 미치는 영향에서 소진의 다중매개효과 검증 : 일반 업무 담당 사회복지사와의 비교를 중심으로

The purpose of this study was to examine the effect of job demands on turnover intention of social workers in charge of case management and not in charge of case management in the community welfare centers, and to investigate the mediating effect of emotional exhaustion, depersonalization of the client, and reduced personal accomplishment, which are sub-dimensions of burnout, as well as to identify differences between two groups. For this purpose, 120 social workers in charge of case management in community welfare centers in Seoul, Gyeonggi and Incheon were analyzed, and 198 social workers not in charge of case management were selected as a comparison group. The analysis verified the multiple mediation model using the PROCESS Macro program. The results are as follows. First, the job demands of the social workers in charge of case management showed a statistically significant effect on the turnover intention. Second, the mediating effect of burnout was significant in the effect of job demands on turnover intention. Third, as a result of verifying the specific mediating effects of the sub-dimensions of burnout, the path of depersonalization of the client was significant in social workers in charge of case management, whereas the path of emotional exhaustion was significant in the social workers not in charge of case management. Based on these results, this study suggested that need for prevention of excessive job demands to reduce turnover intention, expansion of professional staffs, education and supervision system to prevent burnout, and management program for reducing work stressors. 본 연구의 목적은 지역의 사회복지관에 근무하는 사례관리 담당 사회복지사의 직무요구가 이직 의도에 미치는 영향과 두 변수간의 관계에서 소진의 하위차원들의(정서적 탈진, 클라이언트 비인간 화, 성취감 매개효과) 매개효과를 일반 업무 담당 사회복지사와 비교하는 데 있다. 분석대상은 서울, 경기, 인천지역의 복지관에서 사례관리업무를 담당하고 있는 사회복지사 120명이며, 일반 업무 담당 사회복지사는 198명이다. 통계분석은 PROCESS Macro 프로그램을 활용한 다중매개모형을 검증하였다. 분석 결과, 첫째, 직무요구는 소진에 정적인 영향을 미치는 것으로 나타났다. 둘째, 직무요구는 이직의도에 정적인 영향을 미치는 것으로 나타났다. 셋째, 직무요구가 이직의도 에 미치는 영향에서 소진의 매개효과가 유의하였다. 넷째, 소진의 하위차원별 특정매개효과를 검증 한 결과, 사례관리 담당 사회복지사의 경우 클라이언트 비인간화의 경로가, 사례관리 비담당 사회 복지사의 경우 정서적 탈진의 매개경로가 유의하였다. 이러한 결과를 토대로 사례관리담당 사회복 지사의 이직의도 예방을 위한 과도한 직무요구 예방, 전문인력의 확대, 소진예방을 위한 교육 및 슈퍼비전체계 구축, 업무상 스트레스요인 감소를 위한 관리프로그램이 필요함을 제언하였다.

Methimazole, a type of thionamide, is used to treat hyperthyroidism. Several adverse effects of thionamides have been reported. The representative minor adverse effects are arthralgia, skin rash, and gastric intolerance. Methimazole is reported to induce 1-6% of arthralgia cases. These patients begin to suffer from arthralgia from 1 month to 2 years after methimazole treatment. Here, we present a patient with acute onset methimazole- induced arthralgia and skin rash. At 2 days after starting methimazole treatment, a 57-year-old female developed arthralgia and a skin rash on her right leg, which subsequently spread to her left leg and right arm, and she stopped taking the medication. The patient was admitted to the rheumatology department of Ulsan University Hospital, where laboratory tests and a skin biopsy were performed to ascertain whether she had a rheumatic disorder. The skin biopsy revealed nonspecific inflammation. At 2 days after stopping methimazole treatment, the arthralgia and skin rashes had improved and methimazole treatment was recommenced. However, the same symptoms developed within 1 day. Therefore, methimazole treatment was again stopped and the symptoms disappeared.

발달장애인 자녀 어머니의 양육부담감이 우울에 미치는 영향

본 연구는 발달장애인 자녀 어머니의 양육부담감과 우울과의 관계를 살펴보고, 이 관계에서 탄력성의 조절효과를 검증하는 데 목적이 있다. 조사대상자는 경기 지역 15개소의 지역사회복지관 및 치료센터에서 발달장애인 관련 프로그램을 이용하는 어머니 200명이다. 연구결과, 첫째, 발달장애인 자녀 어머니의 우울수준을 분석한 결과, 전체 대상자 중 우울 위험군에 속하는 비율이 15%로 나타나 우울 문제가 심각한 것으로 나타났다. 둘째, 발달장애인 자녀 어머니의 양육부담감은 우울에 영향을 미쳤고, 탄력성은 양육부담감이 우울에 미치는 영향을 완화시키는 것으로 나타났다. 이러한 결과를 토대로 발달장애인 자녀를 양육하는 어머니의 우울문제를 예방하기 위한 효과적인 전략으로서 양육부담감을 완화시키는 지원 외에도 어머니의 탄력성을 강화하는 개입이 병행될 필요가 있음을 제시하였다. The purpose of this study is to examine the associations between care strain and depression among mothers of child with developmental disability and to verify the moderating effect of resilience in the relationship between these two variables. The participants was 200 mothers of children with developmental disabilities using programs related to developmental disability of 15 community welfare centers in Gyeonggi province. Major findings are as follows. First, 15% of mothers of child with developmental disabilities were depression risk group. Second, mother’s strain were associated with depression and the resilience moderated the relationship between care strain and depression. Based on these findings, it is suggested that policy makers and practitioners should expand intervention program not only for alleviating care strain but also improving mother’s resilience to prevent depression of mothers with children developmental disabilities.
