        각종 조영제에 의한 폐색 소장조영에 관한 실험적 연구

        For the evaluation of the level and cause of small bowel obstruction, an oral barium study is usually chosen. When perforation is whether barium or a water-soluble contrast agent should be used, because barium causes foreign body reactions as well as adhesions in the extraluminal tissues. Water-soluble contrast agent, on the other hand, are less satisfactory but in general have no untoward effects in the extraluminal tissues. Because of hyperosomolartity, water-soluble contrast will likely avoid pulmonary complications due to aspiration. The author performed this experimental study to compare the radiological efficacy of different contrast media in enteric follow-through examinations of obstructed small bowel. Rate had a ligature applied to the distal ileum via loparotomy. Four contrast media were subjected to testing by instillation via oro-gastric tube immediately after laparotomy. Radiographs were exposed at 1,4,and 8 hours and evaluated later. After 24 hours the animals were sacrificed and the stomach and small bowels, free from mesentery and omental tissue, were weighted with contrasts. The progression in the owel was proportionate to the osmolarity of the contrast media. After 1 hour, the observations indicated sodium diatrizoate(Gastrografin, Schering) to be the least favorable medium with respect to sharpness of the mucosal border. After 8 hours, barium gave a poorer delineation compared to Ioxaglate(Hexabric, Guerbet) and Iopromide(Ultravist, Schering). Early deaths were noted in the sodium diatrizoate and ioxaglate groups. So we conclude that when using a rather high-volume bolus, low-osmolar non-ionic contrast media seem to have significant prospects for general diagnostic use in patients with suspected intestinal obstruction.

        ELISA 에 의한 B . t . 살충성독소 단백질의 정량

        An ELISA (Enzyme Linked Immunosorbent Assay) method was studied for rapid and sensitive detection of entomocidal toxins of Bacillus thuringiensis subsp. kurstaki and israelensis, potent microbial insecticides. Crystalline toxins were purified with a Telebrix double layer density centrifuge and antisera were prepared from rabbits, respectively. Using these antisera, both indirect and sandwich ELISA methods were examined and compared. Both ELISA systems were demonstrated to be sensitive and specific for the respectable toxins. The minimum detectable concentrations of toxins were about 2-10 ng toxin/㎖.

        미생물 살충제로서 그 가능성이 크게 안식되고 있는 Bacillus thuringiensis subsp. kurstaki와 israelensis의 결정성독소 단백질을 신속 간편하게 정량화하기 위하여 ELISA 방법을 시도하였다. Telebrix 밀도 이중층 원심분리에 의하여 각 결정성 독소단백질을 분리 정제하고, 정제된 독소 단백질에 대한 항혈청은 토끼로부터 제조하였다. 제조한 항혈청을 사용하여 indirect ELISA와 sandwich ELISA를 수행한바, 정밀하고 특이성을 갖은 독소단백질 정량이 가능하였다. indirect ELISA와 sandwich ELISA에서 그 정밀도는 거의 같았으며, 독소 단백질의 최소강지농도는 약 2-10 ng/ml 이었다.         생쥐에 있어서 사람 성장호르몬 유전자의 발현

        The structural gene of human growth hormone was fused with mouse MT-1 promoter and the fusion plasmid was designated as pMThGH. The 2.6Kb BamHI/Bst EⅡ DNA fragment of pMThGH was used as a source of human growth hormone fusion gene in these studies. For microinjection, DNA fragments were diluted with TE buffer in the range of 2-8ng/ul. Fertilized one-cell eggs were flushed from the oviducts on 22-24 hours after HCG injection. To remove cumulus cells, cumulus cell masses were treated with hyaluronidase (300units/ml) and then washed 3 times in fresh medium. The obscure pronuclei of eggs due to hybrid(C57BL/6x CBA) mouse were visible after centrifugation for 3 min. at 15,000rpm. Visualization of nuclear structures was aided by the use of a differential interference contrast microscope and microinjection was carried out under x200 magnification using a Leitz micromanipulator. The microinjected eggs were surgically transferred to the oviducts of synchronized recipients. The litters developed from microinjected eggs were weaned about 4-6 weeks after birth and a piece of tail from each mouse was cut off about 1-1.5cm to analyze foreign DNA. The genomic DNAs isolated from each tail were analyzed by dot hybridization and Southern blot to determine which mouse carried MThGH genes. The results obtained in these experiments were summarized as follows; I. Of 106 microinjected eggs cultured for 96 hours in vitro, 53 eggs(50.0%) were developed to normal blastocyst stage. 2. When microinjection was carried out in the medium with or without Cytochalasin B(5 ug/ml), survival rate of eggs was 72.9% and 50.2%, respectively. 3. 382 microinjected eggs were transferred to the oviducts of 23 recipients and 140 mice consequently developed from these eggs. 4. Of 77 mice analyzed by dot hybridization and Southern blot, 6 mice were positive for MThGH genes and each carried at least one copy of hGH gene. 5. Transgenic mice had very high levels of hGH in their serum, 450 - 6, 100mg/ml, and body weights of them were heavier 1.5-2.0 times than those of controls.
