        증숙 더덕 용매별 추출물의 Nitric Oxide 생성 저해 효과 및 Acetylcholinesterase 저해활성

        Steaming is a method that has traditionally been used for medicinal plant extraction. This study investigated nitrite oxide production, ferrous ion chelating activity, α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities of ethanol, acetone and hot-water extracts of Codonopsis lanceolata prepared by steaming seven times. MTT assay showed that each extract was non-toxic up to a concentration of 700 ㎍/mL confirming that there was no cytotoxicity in all extracts. The α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities exhibited by the hot-water extract obtained from steaming seven times were higher (83.1%) than the other extracts. Higher production of nitrite oxide and better ferrous chelating activity was recorded with hot-water extract compared to ethanol and acetone extracts. These results indicated that more steaming of Codonopsis lanceolata extracts would be required to validate the possibility of developing antioxidants. Also, further study is needed to determine if the components present in the tested extracts might be useful in the prevention of Alzheimer's disease. These results showed that hot-water extracts may be useful for their antioxidant and the production inhibitory activity of nitrite oxide. It will be helpful in the investigation of the constituent analysis of the steam-processed product of Codonopsis lanceolata.

        임플란트 주위 골변화와 생존율에 대한 연구

        The success and failure of dental implants depends on various factors such as patient's systemic status, quantity and quality of surrounding bone, presence or absence of marginal infection and mechanical loading condition. The measurement of crestal bone changes around the implants is implemental to evaluate the success and long-term prognosis of the implant. This study was to evaluate the cumulative survival rate of the implants which had been placed in the Department of Periodontics, Chonnam National University Hospital between 1992 and 2003, and to observe the crestal bone loss around the implants which had at least 2 consecutive periapical radiographs after connecting the transmucosal abutment. The radiographs were scanned and digitalized, and the crestal bone levels on the mesial and distal surface of implants were measured using Image analyzer (Image Pro Plus, Media Cybernetics, USA), immediately after implant placement, at 2nd surgery, and 3 months, 6 months, 1 year, and every year thereafter. Any bone loss was not observed during the period between the 1stand 2nd surgery, and the bone loss was 0.86 ${\pm}$ 0.92 mm for the first year of loading after connecting the transmucosal abutment. After 1 year of loading, annual bone loss was 0.1 ${\pm}$ 0.27 mm, and total bone loss was 0.90 ${\pm}$ 0.80 mm (during the average follow-up periods of 22.5 ${\pm}$ 25.6 Mos), The implant, with smooth surface, in the mandible, and with the fixed bridge prosthesis showed greater bone loss, compared to those, with the rough surface, in the maxilla and with single crown. In systemically diseased patients (including DM or osteoporosis), the greater bone loss was observed. The cumulative survival rate among 432 implants was 94.10% for 7 years. Among 15 failed implants, 9 implants were removed due to mobility from disintegration of bone-implant interface. From this results, crestal bone loss around the implants were greatest during 1 year after transmucosal abutment connection, and various factors could affect peri-implant bone loss. To prevent and predict the bone loss around the implants and improve the prognosis, further comprehensive maintenance and follow-up schedules are required.

        명화감상에 기초한 이야기꾸미기 활동이 유아의 그리기표상능력과 문해능력에 미치는 효과

        본 연구는 명화감상에 기초한 이야기꾸미기 활동이 유아의 그리기표상능력과 문해능력에 미치는 효과를 분석하는데 목적이 있다. 본 연구의 대상은 어린이집 유아 40명이며, 평균 월령은 71.55개월이었다. ‘그리기표상능력 평가척도’, '구문의미 이해력 검사도구'를 사용하여 연구자료를 수집하였으며, 수집된 연구자료는 t-검증을 통해 실험처치의 효과를 분석하였다. 연구 분석 결과, 명화감상에 기초한 이야기꾸미기 활동을 실시한 실험집단 유아들의 그리기표상능력과 문해능력 점수가 비교집단 유아들보다 유의미하게 높게 나타났다. 이러한 본 연구 결과는 명화감상에 기초한 이야기꾸미기 활동이 유아의 그리기표상능력과 문해능력 증진에 긍정적인 효과가 있음을 시사하는 것이다. The purpose of this study is to develop an art appreciation program connected with story-making for young children and analyzed the effects of the program on young children's drawing representation ability and literacy ability. The subjects of this study were 40 young children who attended day-care center in S city. The mean age of the subjects was 71.55 months. It used ‘Drawing Representation Evaluation Scale’ and ‘Sentence Comprehension Evaluation Scale’ in collection of data. The experiment procedure was according to this order, a pilot study, a pretest, a four-week experiment, and a post-test. The experimental group participated in the story-making activity based on art appreciation while the comparative group participated in an art application based on art element. Both activities were totally carried out twelve times, three times a week for four weeks. The results of pretest and post-test were analyzed on the basis of t-test by SPSS Win 18.0 Program. It was revealed that there were significant differences between the experimental group and control group in the post-test scores. The scores of the experimental group in the drawing representation ability, literacy ability were significantly higher than the scores of the comparative group. In conclusion, the results of this study suggest that the Story-Making Activity based on Art Appreciation is effective for promoting young children's drawing representation ability and literacy ability.

        식이급여형태가 고지방식이를 급여한 흰쥐 지방조직의 지방합성 및 지방합성 효소활성에 미치는 영향

        본 연구는 식이급여형태를 달리하면서 고지방식이를 급여한 흰쥐의 부고환지방조직과 장간막지방조직의 지방조직 부위별 지방합성량, 지방합성에 관여하는 주된 효소인 ME, G6PDH 및 6PGDH 활성 및 지방축적의 주된 효소인 LPL 활성에 미치는 영향을 구명하였다. Sprague-Dawley 계통 수컷 흰쥐 16마리를 체중에 따라 2군으로 나누어 4주간 실시하였고, ad libitum군은 실험 식이를 무제한 급여하였으며, meal-fed군은 1일 1회 3시간씩 제한 급여하였다. 지방조직의 지방합성량은 포도당이 총지방으로 변하는 양으로 측정하였고, 지방조직의 G6PDH, 6PGDH 및 ME 활성은 NADPH 생성량으로 측정하였다. 또한 지방조직의 LPL 활성과 혈청지단백질, 중성지방 및 insulin 함량도 측정하였다. Mealfed군이 ad libitum군에 비해 식이섭취량 저하로 인한 증체량 및 부고환지방조직과 장간막지방조직의 무게가 유의하게 저하되었다. 혈청 총콜레스테롤, HDL-콜레스테롤 및 LDL-콜레스테롤 함량은 식이급여형태에 따른 유의차가 없었으나, 혈청 중성지방 함량은 meal-fed군이 ad libitum군에 비해 유의하게 증가하였고, insulin 함량은 저하되었다. 지방합성량 및 G6PDH과 6PGDH 활성은 부고환지방조직과 장간막지방조직 모두 meal-fed군이 ad libitum군에 비해 증가하였으나, ME 활성은 식이급여형태에 따른 유의차가 없었다. 지방조직의 HR-LPL과 TE-LPL 활성은 meal-fed군이 ad libitum군에 비해 유의하게 높았다. 지방조직 부위에 따른 차이는 지방함량, 지방합성량 및 G6PDH와 LPL 활성은 부고환지방조직에 비해 장간막지방조직에서 낮은 경향이었다. 이와 같이 meal feeding시 섭취한 에너지를 체지방 형태로 더 많이 보유하도록 지방조직내 지방합성량과 지방합성 관련 효소들의 활성을 증가시켜 체내 대사에 영향을 미치는 것으로 사료된다. 또한 지방조직 부위에 따라 지방대사에 미치는 영향이 다르다는 것을 알 수 있었다. This study was undertaken to investigate the effects of meal pattern on lipogenesis and activities of lipogenic enzyme in rats epididymal and mesenteric adipose tissues. A high fat diet was fed either ad libitum or in 1 meal during the last 3 h of the dark cycle for 4 weeks. Lipogenesis was measured as glucose conversion to total lipid and activities of glucose?6?phosphate dehydrogenase (G6PDH), 6?phophogluconate dehydro? genase (6PGDH) and NADP?malate dehydrogenase (ME) were determined by measuring NADPH production. Lipoprotein lipase (LPL) activity and serum lipoprotein concentrations were also measured. Meal?fed (3 h) rats had a decreased food intake, body weight and carcass fat compared with rats fed ad libitum. The serum triglyceride concentration of meal?fed rats tended to be higher than that of the ad libitum rats. However, there were no differences between meal?fed group and ad libitum group in serum concentrations of HDL? cholesterol, LDL?cholesterol and total?cholesterol. Rates of lipogenesis in both epididymal and mesenteric adipose tissues were significantly higher in the meal?fed group than that in the ad libitum group. In addition, meal?fed group showed higher G6PDH, 6PGDH and LPL activities in both epididymal and mesenteric adipose tissues, but exerted no significant effect on ME activity. These results suggest that meal?fed rats compared with ad libitum rats have marked lipogenic capacity, although such elevation probably does not result in increase in carcass fat concentration. Thus, meal?fed diet can be an important determinant of the alterations in adipose lipid metabolism.

        증숙 더덕 에탄올 추출물에 대한 항산화·항균 활성

        This study was investigated the contents of total polyphenol, flavonoids and antioxidant and antimicrobial activities of Codonopsis lanceolata extracts according to different steaming times. The contents of total polyphenol and flavonoid proportionally increased from 6.45 mgGAE/g to 18.26 mgGAE/g and 2.01 mgRE/g to 6.12 mgRE/g according to ethanol extracts at EDS7. DPPH radical scavenging activity was found to have been 15.26~65.2% and showed the highest level of antioxidant activity at EDS7 was 65.2%. The activity of ABTS radical scavenging and SOD-like activity were also the same result. DPPH and ABTS radical scavenging activity was related to the number of steaming, and the scavenging activity was increased up to 7 times of steaming. The antimicrobial activity of EDS7 had strong antioxidant activity. Antimicrobial activities were examined against 5 microorganisms related to pathogens and food poisoning. The antimicrobial Activity was different depending on the bacteria, but it was effective at the concentration of 300 mg/mL rather than 150 mg/mL. These results showed that Codonopsis lanceolata extracts with a different number of steaming would be conducted to confirm the possibility of developing antimicrobial and antioxidant. It will be helpful in the study of component analysis of Codonopsis lanceolata extracts processed products.

        증숙된 더덕 추출물의 피부기능 개선 효과

        In this study, hot water and ethanol, acetone, and hexane extracts of the steamed Deodeok were investigated for use as raw materials for natural cosmetics. Hot water and ethanol, acetone and hexane extracts of steamed Deodeok did not indicate cell toxicity up to 300 ㎍/mL concentration in the MTT assay. The tyrosinase inhibition effects of the ethanol extract were higher than the ascorbic acid. The efficacy of the steamed Deodeok's ethanol extract on the elastase inhibition was similar to that of quercetin at higher concentrations. In conclusion, water, ethanol, acetone and hexane extracts of the steamed Deodeok indicated strong possibility as a whitening and anti-wrinkle functional cosmetic ingredient. Among them, the steamed Deodeok's ethanol extract, which showed excellent activity, is highly available because it has high safety compared to other organic solvent extraction methods used in cosmetics, and has minimal irritation to the skin.
