Arsenic Trioxide Induces Apoptosis in Human Colorectal Adenocarcinoma HT-29 Cells Through ROS

Purpose: Treatment with arsenic trioxide (As2O3) results in a wide range of cellular effects that includes induction of apoptosis, inhibition of cell growth, promotion or inhibition of cellular differentiation, and inhibition of angiogenesis through a variety of mechanisms. The mechanisms of As2O3-induced cell death have been mainly studied in hematological cancers, and those mechanisms in solid cancers have yet to be clearly defined. In this study, the mechanisms by which As2O3 induces apoptosis in human colorectal adenocarcinoma HT-29 cells were investigated.Materials and Methods: To examine the levels of apoptosis, HT-29 cells were treated with As2O3 and then we measured the percentage of Annexin V binding cells, the amount of ROS production and the mitochondrial membrane potential. Western blot analysis was performed to identify the activated caspases after As2O3 exposure, and we compared the possible target molecules of apoptosis. As2O3 treatment induced the loss of the mitochondrial membrane potential and an increase of ROS, as well as activation of caspase-3, -7, -9 and -10.Results: As2O3 induced apoptosis via the production of reactive oxygen species and the loss of the mitochondrial membrane potential. As2O3 induced the activation of caspase-3, -7, -9 and -10. Furthermore, As2O3 treatment downregulates the Mcl-1 and Bcl-2 expressions, and the release of cytochrome c and an apoptosis-inducing factor (AIF). Pretreating the HT-29 cells with N-acetyl-L-cysteine, which is a thiol-containing antioxidant, inhibited the As2O3- induced apoptosis and caspase activation.Conclusion: Taken together, these results suggest that the generation of reactive oxygen species (ROS) by As2O3 might play an important role in the regulation of As2O3- induced apoptosis. This cytotoxicity is mediated through a mitochondria-dependent apoptotic signal pathway in HT-29 cells. (Cancer Res Treat. 2006;38:54-60)
작용과 반작용에 관한 학습에서 토론을 통한 인지갈등과 개념변화

본 연구에서는 중학생을 대상으로 자신의 생각과 불일치한 동료 한 명과 자유로운 토론을 통한 인지갈등 유발의 가능성과 그 효과를 알아보았다. 또한 토론 후에 동료의 생각으로 변화하는 정도를 분석하여 토론을 통한 개념변화의 효과를 알아보았다. 연구 대상 개념은 문헌 조사와 예비 조사를 통하여 다양한 사전개념이 존재하여 토론이 원활하게 이루어질 수 있다고 판단된 작용과 반작용 개념을 선정하였다. 연구 대상 학생들은 작용과 반작용 개념에 대해 이미 학습한 경험이 있는 여자 중학교 2학년 학생들로 108명을 대상으로 하였다. 작용과 반작용 개념에 대한 중학생들의 사전개념을 조사하였으며, 조사된 사전개념을 바탕으로 자신과 다른 개념을 가진 동료와 임의로 짝을 구성하게 하였다. 우선 동료의 생각을 단순히 듣고 기술하게 한 후에 동료와 자유로운 토론을 하게 하였으며, 토론을 마친 후 인지갈등 검사를 실시하고 개념변화 정도를 조사하였다. 동료와의 토론을 통한 인지갈등과 개념변화 정도를 분석한 결과 다음과 같은 결론을 얻을 수 있었다. 첫째, 학생들은 자신과 다른 생각을 가진 동료와의 토론을 통해 인지갈등을 일으켰다. 이것은 동료와의 토론 상황이 불일치한 상황의 하나임을 말해주는 것으로 동료와의 토론을 통해서 인지갈등이 유발될 수 있음을 보여준다. 이때 유발된 인지갈등은 학생들이 기존에 가졌던 사전개념에 따라 다르고, 또 자신과 토론한 학생의 개념 종류에 따라서도 달랐다. 둘째, 동료와의 토론 이후, 전체 학생의 57%가 여전히 자신의 생각을 고수하는 것으로 나타났으며, 동료의 생각이 옳은 지 자신의 생각이 옳은 지에 결정을 내리지 못하는 학생들이 16%, 그리고 동료의 생각으로 변화한 학생들이 27%로 나타났다. 이 때 일어난 개념변화 정도는 학생들이 기존에 가졌던 사전개념에 따라 그리고 자신과 토론한 학생의 개념의 종류에 따라 다르게 나타났다. The purpose of this study is to investigate the possibility of induction and the effect of cognitive conflict through dyadic debate with peer who had different conception about the action and reaction in the middle school. In this study, 108 eighth grade students were participated, who already had learned the action and reaction concept. The students' preconceptions about the action and reaction were examined. And two students who had different conceptions were paired randomly according to students' preconceptions. The students listened to and described idea of their partners'. Then, there was a chance to debate freely each other. The levels of cognitive conflict and conceptual change to the partner's conception were examined accordingly. In result, first, debating with peer who had different conception raised cognitive conflict in students' mind. This result indicates that the debate with peer is a kind of anomalous situation to students, and cognitive conflict can be raised by debate with peer. The levels of cognitive conflicts were different according to one's own and partner's conception. Second, some students changed their conceptions to partners' conceptions through debate with peer. The levels of conceptual change of students were also different according to one's own and partner's conception. These results lead to the conclusion that peer dyadic debate could be used to produce cognitive conflict situation in science teaching.
