Purpose: To compare the subfoveal choroidal vascularity indices (CVIs) of treatment-naïve patients with central retinal vein occlusion (CRVO) with controls. Methods: Twenty-one eyes of 21 treatment-naïve CRVO patients and 42 age- and sex-matched control eyes (of 42 subjects) lacking any retinal disease were included. The subfoveal CVIs were measured using ImageJ software. Results: The mean subfoveal CVI in eyes with treatment-naïve CRVO at the initial visit was significantly higher than that of control eyes (0.6464 ± 0.0326 vs. 0.6209 ± 0.0434 respectively; p = 0.024). In contrast, no significant difference in the mean subfoveal CVI was found between CRVO eyes 6 months after initial diagnosis and control eyes (0.6311 ± 0.0600 vs. 0.6209 ± 0.0434 respectively; p = 0.238). Of the 21 eyes with CRVO, 18 (85.7%) received intravitreal injections (bevacizumab, ranibizumab, aflibercept, or triamcinolone) and four (19.0%) underwent panretinal photocoagulation. Conclusions: The subfoveal CVI of eyes with treatment-naïve CRVO at the initial visit was significantly higher than that of control eyes, but decreased by 6 months. Further studies are needed to determine whether CRVO treatments such as intravitreal injection or panretinal photocoagulation affect choroid hemodynamics. 목적: 치료 경험이 없는(treatment naïve) 중심망막정맥폐쇄안과 정상안의 맥락막혈관지수(choroidal vascularity index)를 비교하고자 하였다. 대상과 방법: 21명의 치료 경험이 없는 중심망막정맥폐쇄 환자 21명의 21안과 나이 및 성별을 매칭한 정상 대조군 42명의 42안이 분석에 포함되었다. 맥락막혈관지수는 ImageJ 소프트웨어를 통해 측정하였다. 결과: 치료 경험이 없는 중심망막정맥폐쇄안의 평균 중심오목하 맥락막혈관지수는 0.6464 ± 0.0326, 정상 대조군의 중심오목하 맥락막혈관지수는 0.6209 ± 0.0434였으며 통계적으로 유의한 차이를 보였다(p=0.024). 6개월 경과 후, 중심망막정맥폐쇄안의 평균중심오목하 맥락막혈관지수는 0.6311 ± 0.0600이었으며, 정상 대조군과 유의한 차이를 보이지 않았다(p=0.238). 중심망막정맥폐쇄안 21안 중 18안(85.7%)은 유리체강내 bevacizumab, ranibizumab, aflibercept 또는 triamcinolone을 주입받았으며, 4안(19.0%)은 범망막레이저광응고술을 시행받았다. 결론: 치료 경험이 없는 중심망막정맥폐쇄안의 평균 중심오목하 맥락막혈관지수는 정상안에 비해 유의하게 높았으나, 6개월 후 감소하여 정상안과 차이를 보이지 않았다. 유리체강내 주사 또는 범망막광응고술 등의 중심망막정맥폐쇄에서의 치료가 맥락막의 혈역학적 변화에 영향을 주었는지에 대해서는 후속 연구가 필요하다.
인체혈청 하에서 배양한 인체지방기질줄기세포의 표면항원 및 유전자 발현

인간중간엽줄기세포는(Human mesenchymal stem cells, hMSC) 골수, 지방, 피부, 근육, 혈액에 존재하며, 뼈, 연골, 지방, 근육, 신경세포로 분화가능성이 보고되어 손상조직의 재생을 위한 재료로서뿐만 아니라 유전자치료의 매개체로 이용될 수 있는 가능성이 제안되고 있다. 인간중간엽줄기세포의 적절한 배양조건에는 소 태아혈청(fetal bovine serum, FBS)이 요구되어지므로 세포치료에는 소 태아혈청이 다수 포함되어 있을 것이며 세포배양 배지 유래 소 태아혈청의 단백질에 의한 면역거부반응이 우려된다. 이미 앞선 연구에서 자가혈청 하에서 인체지방줄기세포 분리와 계속적인 세포배양을 실시하였을 때 인체지방줄기세포의 증식능력과 다 분화 능이 유지되며 면역결핍 생쥐에 골수의 말초혈액에서 유래된 CD34세포 이식 시 안착 능을 촉진함을 보였다. 본 연구에서 인체지방줄기세포가 인체혈청 하에서 배양되었을 때 소 태아혈청 하에서 배양할 때 발현하는 표면항원을 유지함을 확인했으며 microarray를 사용하여 유전자 발현을 비교했다. 유 세포 분석을 통하여 인체혈청 하에서 계속적으로 배양된 인체유래지방줄기세포에서 HLA-DR, CD117, CD29 와 CD44 의 발현이 소 태아혈청 하에서 배양했을 때와 비슷함을 밝혔다. 그러나 인체혈청 하에서 배양된 인체지방줄기세포의 유전자 발현형태와 소 태아혈청 하에서 배양된 세포의 유전자 발현형태 간에는 상당한 차이를 보였다. 그러므로 본 연구는 인체혈청 하에서 배양된 인체지방기질줄기세포가 임상적용을 위한 선행 데이터로써 직접적인 추정을 하기 위해서는 인체지방기질줄기세포 이식연구에 in vivo 동물실험연구가 수행되어져야 함을 제시하고 있다. Human mesenchymal stem cells(hMSC), that have been reported to be present in bone marrow, adipose tissues, dermis, muscles and peripheral blood, have the potential to differentiate along different lineages including those forming bone, cartilage, fat, muscle and neuron. Therefore, hMSC are attractive candidates for cell and gene therapy. The optimal conditions for hMSC expansion require medium supplemented with fetal bovine serum(FBS). Some forms of cell therapy will involve multiple doses, raising a concern over immunological reactions caused by medium-derived FBS proteins. Previously, we have shown that hADSC can be cultured in human serum(HS) during their isolation and expansion, and that they maintain their proliferative capacity and ability for multilineage differentiation and promote engraftment of peripheral blood-derived CD34 cells mobilized from bone marrow in NOD/SCID mice. In this study we determined whether hADSC grown in HS maintain surface markers expression similar with cells grown in FBS during culture expansion and compared gene expression profile by Affymetrix microarray. Flow cytometry analysis showed that HLA-DR, CD117, CD29 and CD44 expression in HS-cultured hADSC during culture expansion were similar with that in FBS-cultured cells. However, the gene expression profile in HS-cultured hADSC was significantly different from that in FBS-cultured cells. Therefore, these data indicated that HS-cultured hADSC should be used in vivo animal study of hADSC transplantation for direct extrapolation of preclinical data into clinical application.

Kalopanax septemlobus is a deciduous medicinal tree species belonging to the family Araliaceae. It contains a number of triterpenoid saponins primarily constituted of hederagenin aglycones. Here, we presented de novo comparative transcriptome analysis using 454 pyrosequencing technology between embryogenic (EC) and nonembryogenic callus (NEC) of K. septemlobus. A total of 372,229 high-quality reads were generated by combining 168,065 EC reads and 204,164 NEC reads, which were then assembled into 80,280 and 63,128 unigenes, respectively. A comparative analysis of transcription profiles showed that 13,766 genes were differentially expressed between EC and NEC. Moreover, functional enrichment analysis revealed that a number of genes involved in the proteolysis and embryogenesis were highly expressed in EC, whereas oxidative stress-responsible genes were overpresented in NEC. Interestingly, the expression levels of genes involved in saponin biosynthesis, such as squalene epoxidase, cytochrome P450s, and glycosyl transferase, were also higher in NEC than in EC, suggesting that their expression is stress-inducible. In this study, we characterized the K. septemlobus transcriptome profiles involved in somatic embryogenesis (SE), which might enhance knowledge of SE process and provide a useful molecular resource for further study of K. septemlobus.
낙엽송 클론 채종원에서 구과 채취시기에 따른 구과특성 및 종자품질

Harvest time is one of the most important determining factors of seed quality, especially for species that produce seeds over irregular and long-term periods, such as Larix kaempferi. A cone collection plan must be established to increase seed production efficiency and stable mass production. We investigated seed qualitiessuch as seed efficiency, germination rate, and T50 (germination speed), with 7 or 8 cone collection times at a clonal seed orchard of L. kaempferi in Chungju between 2021 and 2022. A multivariate analysiswas then performed for the collected data. In early August, decreasesin the moisture contents and browning of cones were observed. These were followed by a decrease in germination rate, with a peak at the end of September, but no clear trend was observed. The later the cones were harvested, the better the seed vigor (T50). However, the seed yield and efficiency decreased owing to increases in seed scattering and the number of insect-damaged seeds. As a result, the optimal time of seed harvest for the seed orchard was in early August. To produce uniform seedlings, insect damage must be reduced through timely control and harvest cones in early September. This shows that the degree of browning and moisture content of cones can be used as indicators of the timing of cone collection in L. kaempferi seed orchards.
