N-Acyliminium에의 친핵성 부가에 의한 Octahydro-2,3-dioxo-cyclopenta[b]pyrrole-3a-carboxylates의 합성

5원자 고리형 ${\beta}$-keto ester와 phenylalkylamine의 탈수축합으로 얻은 enamine A의 oxalyl화 및 물이나 메탄올 부가에 의하여 octahydro-2,3-dioxo-cyclopenta[b]-pyrrole-3a-carboxylate의 6a-hydroxy- 및 6a-methoxy-유도체(1~6)를 합성하였다. 헤테로 고리의 형성은 불안정한 N-acyliminium(B)체를 경유하여 일어나며, 안정한 부가체 C (1∼6)는 endo-ene형 pyrrolinium B에 친핵체를 부가하여 얻을 수 있었다. 6a-Hydroxy- and 6a-methoxy-derivatives (1∼6) of octahydro-2,3-dioxo-cyclopenta[b]-pyrrole-3a-carboxylate were synthesized from oxalylation of enamine A, which was prepared from condensation of five-membered cyclic ${\beta}$-keto ester and phenylalkylamine, followed by addition of water or methanol. The formation of heterocyclic ring was assumed to occur by the way of unstable N-acyliminium (B). Stable adduct C (1∼6) was obtained from nucleophilic addition to the endo-ene type pyrrolinium B.
Simple Isolation of DNA from the Tissue Homogenate by Acridinium Affinity Chromatography

고염농도(1.4 M NaCl)의 어류 근조직 균질액을 아크리디늄 친화성 컬럼에 흡착시킨 후 2M $MgCl_2$로 용출시켜 고순도의 DNA ($A_{260}/A_{280}$?2.0)를 얻었다. 이 컬럼에 흡착된 DNA는 0.5M $MgCl_2$로도 용출되었으나 2M NaCl로는 용출되지 않았다. 한편, DNase-I 처리로 친화성 컬럼상의 DNA의 크로마토그래피 유형이 달라졌으며, 이 친화성 컬럼은 단일 및 이중 나선형 DNA 모두에 좋은 결합 친화성을 가진 것으로 여겨진다. 따라서, 이 방법은 고이온강도에서 생조직 균질액으로부터 DNA를 분리하는데 적합할 것이다. The homogenate of fish muscle tissue in high salt concentration (1.4 M NaCl) was applied on the acridinium affinity column, and the elution of the column with 2 M $MgCl_2$ produced the material, which turned out to be a highly pure DNA ($A_{260}/A_{280}$?2.0). The DNA bound on the column was desorbed easily with 0.5 M $MgCl_2$, but not even with 2 M NaCl. Separately, the treatment with DNase-I altered the chromatographic profile of DNA on the affinity column, and the affinity column seems to express good binding affinity toward both single and double stranded DNAs. This method would be suitable for the separation of DNA from crude tissue homogenates in high ionic strength.
수용액에서 SPE-ACF를 이용한 특정화학물질의 신속 스크리닝 기법 연구

study of the solid phase extraction (SPE) technique using activated carbon fiber (ACF) as a sorbent was carried out on the schedule chemicals in water. 14 different schedule chemicals, which are the hydrolysis products and simulants of chemical agents such as alkylphosphonate, thiodiglycol etc. were selected for the study. Pyridine was used as a promoter to increase the derivatization efficiency, especially to improve silylation of 3-quinuclidinol. To improve the recovery efficiency of amines in the water solution, 1 mL of 5% Et3N/MeOH was introduced into SPE-ACF before the water sample loading. Throughout this study, the optimum condition was established to detect rapidly above chemicals in water. 활성탄소섬유(ACF; activated carbon fiber)를 흡착제로 사용한 고상추출법(SPE; solid phase extraction)으로 수용액 시료 중에 존재하는 특정화학물질(schedule chemicals)의 분석법을 연구하였다. 본 연구에 사용된 특정화학물질은 화학작용제류의 가수분해 생성물과 유사작용제로 사용되는 알킬포스포네이트류, 티오디글리콜 등 14종이었다. 분석을 위한 추출물의 실릴 유도체화 반응에서 피리딘을 사용하여 특히 3-quinuclidinol의 유도체화 반응 효율을 높일 수 있었다. 물 시료에서 아민류의 추출 회수율을 개선하기 위하여 SPE-ACF 튜브에 시료를 부하하기 전에 5% triethylamine/MeOH(1 mL) 용액으로 SPE-ACF 튜브를 세척한 후에 시료를 SPE-ACF에 부하하는 방법으로 추출 회수율을 증대시킬 수 있었다. 이 연구를 통해서 수용액 시료에서 특정화학물질을 신속히 검출할 수 있는 최적 조건을 선정하였다.
Soybean Lipoxygenase-Catalyzed Conversion of Arachidonic Acid into Lipoxins

아라키돈산에 대두 lipoxygenase를 작용시켰을때, 300-301 nm에서 강한 흡수를 나타내는 극성 물질들이 형성되었다. 이 물질들을 UV스펙트럼, HPLC, GC/MS에 의한 분석 결과, lipoxin A와 lipoxin B 이성체로 확인되었다. 이 방법은 수백 마이크로그람의 liposin A 및 B 이성체를 얻는데 편리한 것으로, lipoxins의 생합성과 생리적 활성을 연구하는데 유용할 것으로 사료된다. These products were found to be lipoxin A and B isomers, based on UV spectrum, HPLC and GC/MS analyses. This method, convenient to prepare hundreds of micrograms of lipoxin A and B isomers, would be useful for the study on the biosynthesis and the biological activity of lipoxins.

Dermatomyositis (DM) is an inflammatory myopathy of unknown etiology, which involves predominantly the muscles and skin, but also produces pulmonary manifestations in approximately 10% of the patients. Increasing attention has been paid to progressive interstitial pneumonitis as an important pulmonary manifestation of DM that can sometimes have a serious influence on the prognosis. Pneumomediastinum has been reported as a rare complication of DM. In this report, we present a case of 40-year-old woman with DM and interstitial lung disease who developed spontaneuous pneumomediastinum and was successfully treated with cyclosporin A.
