        임플란트 생체금속들과 골조직간의 생체적합도에 관한 연구

        Screw-shaped implants of commercially pure (c.p.) titanium, c.p. niobium, c.p. zirconium, and stainless steel(Sus 304) were inserted in the rabbit tibial bone over 12 weeks of follow-up. New developed torque gauge instrument was used to evaluate the implant holding power and a image analysis program coupled to a microscope was used for histomorphometry. The three best consecutive threads of each implant were measured. Quantitative analyses at 12 weeks revealed a partial bone contact to the four kinds investigated metals. There were no obvious adverse tissue reactions to any of the biomaterials. At 12 weeks the average removal torques for titanium, niobium and zirconium were better than that needed for Sus 304 screws, on the other hand high score of bony contact ratio of titanium and niobium were showed in comparison to those of zirconium and Sus 304. There was no significant differences in the amount of interfacial bone of zirconium and Sus 304 whereas there was significant difference in the torque forces of niobium and Sus 304. Three months after implant insertion, the average removal torque was 6.64 Ncm for the titanium, 6.57 Ncm for the niobium, 6.38 Ncm for the zirconium, and 4.25 Ncm for the Sus 304. On average bone contacts there were 51.24% in the titanium, 48.19% in the niobium, 31.79% in the zirconium, 23.54% in the Sus 304. Biocompatibility of the titanium, niobium and zirconium was acceptable level in comparison to the Sus 304.

        생약 추출물이 세포성장 및 cytokine 생산에 미치는 영향

        The native connective tissue attachment of the periodontium is known to be a complex consisting of gingival fibroblasts, periodontal ligament cells, gingival epithelial cells, cementum, alveolar bone and extensive extracellular matrix (collagen, glycoprotein and proteoglycans). The purpose of this study was to evaluate the effects of natural extracts on DNA, collagen and protein synthesis and inhibition of cytokine production in the gingival and periodontal ligament fibroblasts and gingival epithelial cells. Healthy gingival tissue was obtained from orthodontic treatment patients, and gingival epithelial cells, gingival fibroblasts and periodontal ligament cells were isolated and cultured from the samples. After treated with Ginseng protein, Pluronic F-68, Scutellariae Radix, centella asiatica, PDGF, IGF, DNA synthesis, total protein and collagen synthesis, and cytokine production of gingival epithelial cell, gingival fibroblast and periodontal ligamentcells were measured. MTT method for DNA synthesis, Peterkofsky and Dingerman method for total protein and collagen synthesis, and IL-1 ELISA kit for cytokine production were used. The proliferation of epithelial cells was enhanced in Centella asiatica, Ginseng protein, Pluronic F-68 and Scutellariae Radix. The activities of PDL cells were increased in PDGF, IGF, and Pluronic F-68. Higher collagen synthesis was observed in Scutellariae Radix and total protein synthesis was increased in Scutellariae Radix and PDGF. The inhibitory effects on IL-1, IL-6, $TNF-{\alpha}$ were observed in all exrracts.

        Poly(alpha-hydroxy acids) 제제 생분해성 차폐막의 치주조직 재생유도능력에 관한 조직학적 장기관찰

        The recent trend of research and development on guided tissue regeneration focuses on the biodegradable membranes, which eliminate the need for subsequent surgical removal. They have demonstrated significant and equivalent clinical improvements to the ePTFE membranes. This study evaluate guided tissue regeneration wound healing in surgically induced intrabony periodontal defects following surgical treatment with a synthetic biodegradable membranes, made from a copolymer of glycolide and lactide, in 8 beagle dogs. After full thickeness flap reflection, exposed buccal bone of maxillary and mandibular canine and premolar was removed surgically mesiodistally and occlusoapically at $6mm{\times}6mm$ in size for preparation of periodontal defects. In experimental sites a customized barrier was formed and fitted to cover the defect. Flap was replaced slightly coronal to CEJ and sutured. Plaque control program was initiated and maintained until completion of the study. In 4, 8, 16 and 24 weeks after surgery, the animals were sacrificed and then undecalcified specimens were prepared for histologic evaluation. Histologic examination indicated significant periodontal regeneration characterized by new connective tissue attachment, cementum formation and bone formation. These membranes showed good biocompatibility throughout experiodontal period. The barriers had been completely resorbed with no apparent adverse effect on periodontal wound healing at 24 weeks. These results implicated that present synthetic biodegradable membrane facilitated guided tissue regeneration in periodontal defect.

        홍화 추출물이 치주인대세포, 조골세포 활성도에 미치는 영향

        Safflower(Carthamus tinctorius $Linn\acute{e}$ has been traditionally used for the treatment of blood stasis, and Dipsasi Radix has been used as a drug for fracture in Chinese medicine. The purpose of present study was to examine the biologic effects of safflower extract and Disasi radix extracts on the periodontal. ligament cells and osteoblastic cells and on the wound healing of rat calvarial defect. The ethanolic extract of safflower blossom, safflower seed and Dipsasi Radix(125, 250, and 500 ${\mu}g/ml$) were prepared as test group, and PDGF-BB(lOng/ml) and unsafonifiable fraction of Zea Mays L.(125, 250, and 500 ${\mu}g/ml$) were employed as positive control. The effects of each agents on the growth and survival, ALPase activity, expression of PDGF-BB receptor, chemotactic response of PDL cell and ATCC human osteosarcoma MG63 cells in vitro were examined. The tissue regenerative effect of each extracts was evaluated by histomorphometric measuring of newly formed bone on the 8mm defect in rat calvaria after oral administration of 3 different dosages groups : 0.02, 0.1 and 0.35g/kg, per day. It was also employed the same dosages of unsaponifiable fraction of Zea Mays L. as positive controls. Safflower blossom extract, safflower seed extract, and Dipsasi Radix extract stimulate the cellular activity of MG63 cells in concentration range of $125-500{\mu}g/ml$, and safflower bolssom extract and safflower seed extract stimulate also the cellular activity of periodontal ligament cells in concentration range of $250-500{\mu}g/ml$. In activity of ALPase, $250-500{\mu}g/ml$ of safflower blossom extracts showed significant stimulating effects on MG63 cells, and the same concentration range of safflower seed extracts showed significant effect on periodontal ligament cells. In the recovery on PDGF-BB receptor expression which was depressed by $IL-1{\beta}$, $125-250{\mu}g/ml$ of safflower blossom extracts and $250-500{\mu}g/ml$ of safflower seed extracts showed significant increasing effect on MG63 cells, and $500{\mu}g/ml$ of safflower blossom extract and $250-500{\mu}g/ml$ of safflower seed extracts showed significant effect on periodontal ligament cells. In chemotactic response, among all tested group, safflower seed extracts only were chemotactic to MG63 cells and periodontal ligament cells in concentration range of $125-500{\mu}g/ml$. Also in the view of bone regeneration in rat calvarial defect model, the only group that was orally administrated 0.35g/kg, day of safflower seed extract showed significant new bone formation. These results suggested that safflower extracts might have a potential possibilities as an useful drug for adjunct to treatment for regeneration of periodontal defect.

        치은섬유아세포와 치은인대세포의 형태와 화학주성에 미치는 대조추출물의 효과에 관한 연구

        The most important object of periodontal treatment is the perfect regeneration of destructed periodontal tissue. The healing of periodontal lesion is affected by several cells & factors, which result in formation of long juntional epithelium, root resorption, bony ankylosis or connective tissue attachment. And ideal healing is enhanced by epithilial exclusion or periodontal ligament cell activation. In this investigation, I studied the effect of Zizyphus Fructus extract which enhances biologic activity& collagen synthesis, on the chemotaxis & cell nature. The cells were obtained from interdental area & middle third area of the freshly extracted teeth for the orthodontic purpose. And they were fully incubated in${\alpha}-MEM$ solution containing $100{\mu]g/ml$ penicillin & $100{\mu]g/ml$ streptomycin followed by 6 generation incubation. The test cells were collected by trypsin-EDTA & centrifuge in the fully incubated cells, counted by Hernacyotmeter, incbated $5{\times}10^5/ml$ cells for 24 hours, re-incubated 24 hours in media containing natural extract and photographed. The cells were incubated for 4 hours in 48 well microchemotaxis chamber bisecting upper & lower chamber by 8ug/m pore polycarbonate membrane coating 5mg/ml gelatin solution. The migrated cells in microscope were counted, which meaned cell chemotaxis activity. The study had shown that the morphology of cell was spindle-shaped as the control group, and the subextract test groups were not significantly different. In gingival fibroblasts, the chemotaxis effect of PDGF was statistically significant compared to control group. The Zizyphus Fructus extract was more or less enhanced chemotaxis effect and in $1{\mu}g/ml$ concentration the chemotaxis effect was slightly elevated compared with $10{\mu}g/ml$ concentration. But, among the subextracts, it was not significantly defferent. In PDL cells, the chemotaxis effect of PDGF in statistically significant, and the zizyphus Fructus extract had shown the enhanced effect. The effect was slightly higher in $1{\mu}g/ml$ concentration than 10g/ml concentration,and no significance among the subextracts.

      • 치주인대세포와 치은섬유아세포의 분화에 미치는 rhBMP-2의 효과에 대한 연구

        BMP can induce ectopic bone formation when implanted into sites such as rat muscle and can greatly enhance healing of bony defects when applied exogenously. In addition, BMP stimulated osteoblastic differentiation in vitro in various types of cells. The aim of this study was to investigate the effect of recombinant human bone morphogenetic protein(rhBMP-2) on the proliferation and osteoblastic differentiation of human periodontal ligament cells and gingival fibroblasts. The cell number and alkaline phosphatase activity were measured in 3 experimental groups of human periodontal ligament cells and gingival fibroblasts (control group, rhBMP-2 50ng/ml group, and rhBMP-2 100ng/ml group) at 1 and 2 weeks after culture. At the same time, total RNA of cultured cells were extracted and reverse trascription polymerase chain reaction(RT-PCR) was performed to determine the expression of mRNA of bone matrix protein. RhBMP-2 had no effect on the cell proliferation of human periodontal ligament cells and gingival fibroblasts. Alkaline phosphatase activity was elevated significantly by rhBMP-2 in both cells. And periodontal ligament cells showed significantly higher alkaline phosphatase activity than gingival fibroblasts. ${\beta}$-actin, type I collagen, alkaline phosphatase, BMP-2 mRNA were expressed in all of the samples. Osteopontin, osteocalcin mRNA were expressed in all periodontal ligament cell groups, and rhBMP-2 50ng/ml group, rhBMP-2 100ng/ml group of 2 week culture period of gingival fibroblasts. Bone sialoprotein mRNA was only expressed in rhBMP-2 50ng/ml group and rhBMP-2 100ng/ml group of 2-week culture period. These results suggest that rhBMP-2 stimulates osteoblastic differentiation in human periodontal ligament cells and gingival fibroblasts in vitro.

        교육에 있어서 다양한 분야의 교육이 중요하겠지만 최근 진로교육이 크게 대두되고 있다. 진로를 결정 하는데 있어서 다양한 사항이 반영되어야만 하지만 학생의 의견도 무시 할 수 없다. 그렇다고 무작정 학생의 의견만 반영하기에는 현실적으로 어렵다는 애로 사항이 있다. 우리나라에서 진로는 단순히 학생의 희망 혹은 적성에만 기댈 수 없는 것이 누구나 좋은 진로나 직업을 가지기를 원하지만 성적이 따라줘야 희망하는 분야로 결정할 수 있기 때문이다. 이에 본 연구에서는 학생들의 적성 및 성격을 반영 할 수 있는 MBTI와 직업군에 대한 성적을 반영하여 학생들에게 적합한 진로를 추천해주는 시스템에 관한 연구를 하고자 한다. 이 연구를 통하여 교사는 학생들에게 적합한 진로를 추천하여 줄 수 있고 학생들 자신의 적성과 성적에 따른 진로를 추천 받을 수 있어서 본인이 나중에 어느 분야 어떠한 진로로 진출 할 것인지에 대한 고민도 줄일 수 있을 것이라고 생각한다. While education in various fields will be important in education, career education has become a big issue recently. Various factors must be reflected in determining a career path, but students’ opinions cannot be ignored. However, it is difficult to just reflect the opinions of students. In our country, career simply cannot be relied upon by students’ hopes or aptitude, as anyone wants to have a good career or job, but only when their grades are followed can they decide on the desired field. In this study, we want to do research on MBTI that can reflect students’ aptitude and personality and systems that recommend appropriate career paths to students by reflecting their performance in the vocational group. Through this study, teachers can recommend suitable career paths for students and receive recommendations based on students’ own aptitude and grades, so they can also reduce their worries about which career they will enter in the future.

      • 딥 러닝을 이용한 교원 임용후보자 경쟁시험 합격 여부 예측 적중률을 높이기 위한 방안 연구

        최근 4차 산업혁명 시대를 맞이하여 다양한 분야에서 인공지능 및 딥 러닝 머신러닝 등을 이용하여 예측 및 추천시스템을 개발 활용하고 있다. 따라서 본 연구는 사범대학 재학생들 및 교육대학원 재학생들의 가장 고민인 국내 대학생들의 가장 큰 고민인 진로 문제 즉 임용후보자 경쟁시험의 합격여부에 대한 예측을 하고 적중률을 높이기 위한 연구를 통하여 학생들의 지도 및 공부에 도움이 되고자 한다. 국·공립 중긍 교사가 되기 위해서는 사범대 및 교육대학에 진학하여 교원 자격증을 취득한 뒤 교원임용 후보자 경쟁시험이라는 일명 임용고사라는 시험을 통과하여야한다. 그러나 점차 인구수감소와 맞물려서 교육부에서 발표되는 임용T.O는 매년 줄어들고 있고 그에 비해 입학학생 수 및 재수생숫자는 늘어나 경쟁률은 가파르게 오르고 있는 사정이다. 이에 본 연구에서는 딥 러닝을 이용하여 교원 임용 후보자 경쟁시험 합격 여부 예측 모델을 구상하고 이에 예측 적중률을 높이기 위한 연구를 하고자 한다. 데이터 분석 및 딥 러닝과 같은 정보기술을 활용하여 학생들의 합격여부를 예측 하고 이에 따른 조언과 학습을 위한 방안을 제시하여 임용 후보자 경쟁시험 합격률 상승을 사료할 수 있을 것 이다. In the wake of the recent 4th Industrial Revolution, it has developed and utilized prediction and recommendation systems using artificial intelligence and deep running machine learning in various fields. Therefore, this study aims to help students guide and study through a study to predict whether they will pass the competitive examination for appointment candidates, which is the biggest concern for Korean university students, who are enrolled in the College of Education and students who are enrolled in the Graduate School of Education. In order to become a national and public-oriented teacher, he/she must go to a college of education or an education university, obtain a teacher"s license, and pass a test called a competitive examination for teachers" appointment candidates. However, the Ministry of Education"s employment T.O., which is announced by the Ministry of Education in line with the decrease in population, is decreasing every year, while the number of students enrolled and re-entered is increasing rapidly. In this study, we are going to use deep learning to devise a model for predicting whether or not a teacher candidate will pass the competitive examination and to increase the predicted hit rate. Information technology, such as data analysis and deep learning, will be used to predict whether students will pass the examination and to provide advice and learning measures accordingly to feed the increase in the rate of passing the competitive examination for appointment candidates. future.

        후박 및 은행물 추출물을 함유한 치약의 임상 및 미생물학적 효과에 관한 연구

        Previous studies have shown that Magnoliae cortex and Ginkgo biloba extracts were showed on the antimicrobial and anti-inflammatory action, in vitro. The purpose of this study was to evaluate on the effect of antimicrobial and anti-inflammatory activity of Magnoliae cortex and Ginkgo biloba extracts containing dentifrice in gingivitis. 70 subjects with gingivitis were divided into an experimental group which performed normal oral hygiene procedure with Magnoliae cortex and Ginkgo biloba extracts containing dentifrice and a control group which also performed normal oral hygiene procedure with the same dentifrice without the natural extracts and completed a doubleblind, cross-over study. At baseline and 3 weeks, subjects were assayed for clinical study by plaque index, gingival index, pocket depth, GCF rate, and microbiological study by subgingival dental plaque bacterial morphotypes by phase contrast microscopy, total anaerobes, total aerobes, Black pigmented bacteroides, A.actionomycetemcomitans, A.viscosus, C.rectus, Ssenguis; P.gingivalis, P.intennedia by bacterial culture and immunofluorescence microscopy. After 3 weeks using their respective dentifrices, reductions in the clinical indices of subjects were similar between the experimental dentifrice group and a control dentifrice group except for statistically significant much reductions in PI, GI, and GCF rate in the experimental dentifrice group as compared to control dentifrice group. Also statistically significant reductions in the motile rods and Spirochetes were found in both experimental group to compare with control group, however statistically much reduction in total anaerobes, Black pigmented bacteroides, and P.gingivalis, P.intennedia were found in the experimental dentifrice group as compared to control dentifrice group. This results indicates that Magnoliae cortex and Ginkgo biloba extracts containing dentifrice might be useful for elimination of gingival inflammation.

        E. coli lipopolysaccharides로 유도된 사람 호중구에서 CD14, Toll-like receptors, cytoskeletal inhibitors 그리고 $NF-{\kappa}B$ inhibitor가 MMP-8 분비에 미치는 영향

        Objective: MMP-8 is a neutrophil enzyme and its level increases in some inflammatory diseases, including periodontal disease. We knew that the lipopolysaccharide of E.coli(E-LPS) induced MMP-8 release from human neutrophils. E-LPS is known to induce the production and release of inflammatory cytokines through CD14, Toll-like receptor(TLR). In the present study, we investigated whether MMP-8 release by E-LPS is induced via CD14-TLR pathway and the cellular mechanism of MMP-8 release in human neutrophils. Material and methods: Human neutrophils were isolated from the peripheral blood of healthy donors and pre-incubated in medium containing antibodies against CD14, anti-TLR2 and anti-TLR4 or several inhibitors of microtubules and microfilaments and then incubated with E-LPS. The cells were treated TPCK and E-LPS simultaneously. The MMP-8amount in the culture medium was determined using ELISA. Results: E-LPS increased MMP-8release from neutrophils and its induction was inhibited by anti-CD14 and anti-TLR4 but not by anti-TLR2 antibodies. The inhibitors of microtubule and microfilament polymerization significantly decreased E-LPS-induced MMP-8release. TPCK inhibited E-LPS-induced MMP-8 release. Conclusion: These results suggest that MMP-8 release is induced by E-LPS via the CD14-TLR4 signal pathway in human neutrophils and may be depedent on microtubule and microfilament systems and $NF-{\kappa}B$ pathway.
