        Chirose hamster ovary 세포에서 발현된 preS2 및 S부위 함유 HBsAg의 항체유발능

        Many studies have provided evidences that hepatitis B surface antigen (HBsAg) including pres region could be an ideal candidate for a new hepatitis B virus (HBV) vaccine with higher efficacy. We established CHO cell lines, H-CHO-2 and IY-CHO-11 expressing high levels of HBsAg containing preS2 and S pretein by stable transfection method. These cell lines expressed the correct size (about 1 kb in length) of HBsAg myNA as eld)ected. The purified protein from the culture supernatants of the clones showed the same sizes as those expressed in native hepatitis B virus (24 kDa, 27 kDa, 34 kDa and 36 kDa). Antibody productivity of CHO-derived HBsAg protein at lower dose challenge was higher than the protein containing S region alone expressed in yeast system. These results indicate that CHO-derived HBsAg protein containing preS2 and S region can be effectively used for a better immune response as a HBV vaccine.

        인공 요추간판 치환술 도중과 직후에 발생한 단기추시 중 합병증

        목적: 인공 요추간판 치환술 시 저자가 경험한 합병증에 대하여 기술하고, 합병증의 발생과 관련된 요인 및 합병증의 발생이 임상 결과에 어떠한 영향을 미칠 수 있는지 알아보고자 하였다. 대상 및 방법: 2002년 5월부터 2004년 5월까지 인공 요추간판 치환술을 받은 43명의 환자를 대상으로 술 중 또는 술 후에 발생하였던 합병증을 조사하였다. 환자의 평균 연령은 45세(남자: 18명, 여자: 25명)였다. 예상되는 위험 인자들과 각각의 합병증 발생과의 관련성 여부를 조사하였다. 임상 결과의 평가는 visual analogue scale 및 Oswestry disability index (ODI)를 이용하였다. 결과: 술 중 합병증으로 정맥 혈관 파열이 2예, 복막 파열이 2예 있었다. 술 후 합병증으로 일시적인 방사통이 6예 있었고, 인공 추간판의 침강이 1예, 인공 추간판의 덮개판과 폴리에틸렌 핵의 해리가 1예 있었다. 합병증 발생과 여러 인자들과 관련성에 대한 통계적 유의성을 찾을 수 없었다(p>0.05). 합병증의 발생 유무에 따른 VAS와 ODI의 유의한 차이는 없었다. 결론: 인공 요추간판 치환술 도중 및 직후에 비교적 많은 합병증이 발생하였으나, 술 후 임상 결과에 유의한 영향을 미치지 않았다. Purpose: To investigate the perioperative complications and their effect on the clinical outcomes after a lumbar total disc replacement (TDR). Materials and Methods: Forty-three patients (18 men and 25 women) who had undergone a lumbar TDR from May 2002 and May 2004 were reviewed. Subgroup analysis was performed to determine if age, gender, previous discectomy, multilevel, learning curve of operator, or the difference between preoperative and postoperative disc height were associated with the development of intraoperative or postoperative complications. The patients rated their low back pain on a visual analogue scale and completed an Oswestry disability index (ODI) at the preoperative and post-operative follow-up. Results: There were 4 cases of intraoperative complications (2 major vein injuries and 2 peritoneal rupture) and 8 cases of postoperative complications (6 radicular pains, 1 subsidence of implant, and 1 dissociation of endplate and polyethylene core). There were no significant differences in the incidence of complications according to age, gender, prior surgery, multilevel, learning curve, or disc height difference (p > 0.05). In addition, there were no significant differences in the VAS and ODI between the complication and non-complication groups. Conclusion: The authors experienced a relatively high rate of complications (11/43 patients) following TOR and statistical analysis did not reveal any associated risk factors.

        흉강경을 이용한 측만증 교정술 후 척추 체 축상회전의 변화

        목적: 흉강경을 이용한 측만증의 전방 교정술시 효과적인 측만증의 반회전이 일어나는 지와 기기 술이 이루어진 분절 내와 그 원위 및 근위 부에서의 회전의 양상을 알아보고자 하였다. 대상 및 방법: 특발성 측만증환자 20명에서 술 전과 술 후에 척추 전장에 걸친 자기공명 검사를 시행하였고 이들에서 각각의 척추 체에서의 횡단면상을 얻어 Aaro씨 방법을 이용하여 수술 전 후의 척추 체의 축상 회전각을 측정하였다. 첨추체 부위, 기기 술 근위부위 및 윈위부위, 그리고 기기 술이 이루어지지 않은 척추 체로의 분절 이행부위에서의 척추 체 반회전각을 비교하였다. 또한 각각의 부위에서의 분절간 회전과 분절간 반회전 정도를 계산하였다. 통계적 분석은 2원 배치 분산분석 (2-way ANOVA) 및 Wilcoxon 부호순위 검정을 이용하여 분석하였다. 결과: 첨추체 부위에서의 평균 축상 회전각은 술 전 10.1도였으며 술 후 5.3도로 평균 48%의 축상 회전 교정율을 얻을 수 있었다. 첨추체 부위에서의 반회전은 기기 술 근위부 및 원위부에 비해 큰 값을 보였으며 원위부에서는 유의한 반회전은 일어나지 않았으나(p=0.023) 근위부의 경우는 의의 있는 반회전을 보였다(p<0.001). 술후 기기 숨이 이루어지지 않은 근위 및 원위부의 첫 척추체는 기기 술 끝 척추의 방향과 동일한 방향으로 의의 있게 회전하였다. 분절간 회전은 기기 술 근위부 및 원위부에서 술 전과 술 후에 모두 첨추체부위 보다 더 큰 값을 보였지만 분절간 반회전은 기기 술이 행해진 근위부 및 원위부 그리고 첨추체 부위 모두에서 균일하게 일어났다. 기기 술이 이루어 지지 않은 분절 이행 부위에서는 술 전 분절간 회전은 기기 술이 행해진 근위부나 원위부와 동등한 값을 보였지만 수술 후 분절간 반회전은 경미하였다. 결론: 흉강경을 이용한 측만증의 전방 교정술은 각 척추 체의 반회전과 분절간 반회전을 통해 효과적인 회전변형에 대한 교정이 이루어지는 것으로 생각된다. Purpose: In this study, we investigated whether thoracoscopic anterior correction can effectively derotates the scoliotic spine and we also evaluated the patterns of derotational changes inside the instrumented area and at the junction between the instrumented and un instrumented area. Materials and Methods: Preoperative and postoperative MR images with single axial cuts through each vertebral level were obtained in 20 patients who underwent thoracoscopic anterior instrumentation. Each vertebral rotation was measured by the use of Aaro's method. Vertebral axial derotation of the apical zone (AZ), upper instrumented zone (UIZ), lower instrumented zone (LIZ) and junctional segment were compared. The amount of segmental rotation and segmental derotation in each zone was calculated. Statistical analysis was performed by the use of by 2-way ANOVA and the Wilcoxon signed ranks test. Results: The average axial rotations at AZ were 10.1° preoperatively and 5.3° postoperatively with an average correction rate of 48%. Derotation of the AZ was greater than the UIZ and LIZ. For the LIZ derotation was not significant (p=0.023) while for the UIZ there was a significant derotation (p < 0.001). Postoperatively, the first uninstrumented vertebra rotated significantly in the direction of rotation of the instrumented end vertebra. Preoperative and postoperative segmental rotation was higher in the UIZ and LIZ than in the AZ. However, segmental derotation occurred uniformly in the instrumented area. At the junctional segment, preoperative segmental rotation was same as that of the UIZ and LIZ and segmental derotation was not significant. Conclusion: Thoracoscopic scoliosis correction can achieve effective axial correction by causing derotation of each of the vertebral bodies and segmental derotation.

        초등학교 도덕과 웹기반 수행평가 시스템에 관한 연구

        In elementary moral education, performance assessment should be conducted in both cognitive, affective, and behavioral domains in harmony. The present study attempts to consider the features of performance assessment in elementary moral education; classify them by the types of assessment, and; implement and apply the web-based performance assessment system, in which teachers, students, and parents all participate. The development of this web-based performance assessment system for elementary moral education allowed the students to carry out the performance assessment process by themselves and to see the result no matter when and where they are. The students became interested in their own behavior, and the parents got to have a broader range of understanding for their children by participating in personal in the assessment procedure. The parents also realized that home guidance for their children's life habits became pretty easy. It is expected that the performance assessment system carried out based on the collaboration between the school and the family will be helpful in applying the contents of the moral subject studied at school and the relevant knowledge and skills to the actual life outside the school.

        알긴산 장용 비드에 봉입한 새로운 장 표적성 경구용 장티푸스 Ty21a 백신의 개발

        To increase the viability of oral typhoid vaccine during the passage through the gastro-intestinal tract, .numerous attempts have been made including the vaccine coating. However, problems such as high death rate during the coating process and its instability in the gastric juice still remain to be solved. In this study, the oral vaccine was made as the micro-enteric beads by adding Salmcnella typhi Ty21a cells to sodium alginate solution and spraying onto calcium chloride solution (ionotropic relation method). The vaccine showed more than 90% of its original viability after treating it for 1 hour in the artificial gastric juice (37oC,300 rpm). The clearance rate of the Ty21a in the liver and spleen of the mice orally administrated with coated Ty21a was similar to that of the mice intraperitoneally administrated with uncoated Ty21a. The peripheral blood Iymphocytes (PBL) isolated from the mice orally administered with this vaccine produced 15.5 fold higher specific IgA antibody titer than that from the control mice administerd with saline solution. Fllrthermore, the mice treated with the coated Ty21a had higher survival rates (50∼87%) than the control mice treated with saline solution (0-10%) in the intraperitoneal challenge test with wild type S. typhi cells. These results suggest that the alginate-based coaling technique is effective to protect live Ty21a from acidic environments, and produces better intestinal immune responses thereby providing a potentially excellent oral typhoid vaccine.

        시판되는 에너지음료의 법랑질 침식효과에 대한 연구

        To find out the effect of commercially available energy drinks on tooth enamel erosion, analyzed pH, buffering capacity, and the content of some of the inorganic components selecting 4 energy drinks that has high affinity of the products currently being sold. In addition, by observing the degree of erosion before and after immersion in energy drink by surface microhardness and scanning electron microscope (SEM) the results were as follows: Acidity of energy drink ?Burn Intense? was the lowest as 2.78±0.01 highest on distilled water as 6.475±0.01. ?Burn Intense? buffering capacity was 3.48±0.155 at pH 5.5, 1.88±0.15 at pH 7.0 which is the highest, and ?Hot6? was 1.71±0.37, 1.23±0.35 on each of it showing the lowest points. Ca content on energy drink was the highest at ?Volt Energy? as (77.21±2.70 mg/kg), the lowest at ?Hot6? as (0.98±0.05 mg/kg). P content was the highest on ?Hot6?(1.34±0.05 mg/kg) and detected at ?Red Bull?. Enamel surface hardness variation of the energy drinks before and after immersion showed rapid decrease at ?Red Bull? (66.65±35.60), and ?Volt Energy? (61.96±31.42), ?Burn Intense? (58.53±24.84), ?Hot6? (53.99±60.26) was in order. Distilled water, the control group, showed significant difference with the experimental group (p<0.05). But there was no significant difference between energy drinks. At SEM observation and analysis, ?Burn Intense? was the most severe demineralization, ?Volt Energy?, ?Hot6?, ?Red Bull?, distilled water was in order. In the above results, taken together there were no statistically differences between energy drinks but significant difference in comparison with distilled water. In addition, at SEM observation and analysis all energy drink caused dental erosion, especially ?Burn Intense?, has the lowest acidity, was serious. Thus, it is believed to provide a variety of oral health education and information about energy drinks that can affect the erosion of the teeth so public have the right to be recognized and reasonable dental care.

        This paper explains compressive behaviors of sandwich composite laminates with adhesively bonded patches. The sandwich composite laminate is used for a train carbody structure and is of an aluminum honeycomb core and CF1263 woven fabric carbon/epoxy faces. The sandwich composite laminates were damaged by low velocity impact. The damaged sandwich composite laminate was repaired using scarf repair method. Then, the strength restoration of it was assessed by compressive test. From the test, it could be known that the compressive strength was restored up to 91% of undamaged one.
