Multiplex PCR을 이용한 유전자변형 카네이션의 검출법

유전자변형 카네이션(Moonvista, Moonaqua, Moonshadow, Moonlite, Moonshade, 및 Moodust) 개발에 이용된 형질전환 벡터(pCGP1470, pCGP1991)를 동시에 검출할 수 있는 multiplex PCR이 개발되었다. 유전자변형 카네이션을 스크리닝하기 위해 acetolactate synthase(SuRB/ALS) terminator의 염기 서열로부터 한 쌍의 primer를 제작하였다. Anthocyanidin synthase(ANS)를 카네이션의 내재유전자로 PCR 검사법에 이용하였다. 100bp의 PCR 산물을 얻을 수 있는 ANS-KF/KR primer가 ANS 유전자를 증폭시키는데 이용되었고, 이것을 이용하여 11개의 다른 식물에 대해서 PCR한 결과 카네이션에서만 특이적인 증폭산물이 확인되었다. C1991-KF/KR primer가 형질전환 벡터 pCGP1991에 삽입된 Dihydroflavonol reductase(DFR) 유전자와 DFR terminator 사이의 염기서열을 확인할 수 있도록 제작되었고, 형질전환 벡터 pCGP1470를 확인하기 위해 C1470-KF/KR primer가 Mac promoter와 DFR 유전자 사이의 염기서열을 확인할 수 있도록 제작되었다. 본 연구에서 개발된 multiplex PCR의 검정한계치는 약 1%이며, 이러한 multiplex PCR이 국내 미승인 유전자변형 카네이션을 모니터링 하는데 유용하게 사용될 수 있을 것이다. A multiplex PCR method was developed to simultaneously detect two transformation vectors (pCGP1470 and pCGP1991) used in the development of GM carnation plants (Moonvista, Moonaqua, Moonshadow, Moonlite, Moonshade, and Moodust). To screen for GM carnation plants, one specific primer was designed from acetolactate synthase (SuRB/ALS) terminator sequence. The anthocyanidin synthase (ANS) was also used as an endogenous reference gene of carnation in PCR detection. The primer pair ANS-KF/KR producing 100 bp amplicon was used to amplify the ANS gene and no amplified product was observed in any of the 10 different plants used as a template. The primer pair C1991-KF/KR was designed to amplify the junction sequence between dihydroflavonol reductase (DFR) gene and DFR terminator in pCGP1991. To confirm pCGP1470, the primer pair C1470-KF/KR was designed to cover the junction sequence between Mac promoter and DFR gene. The detection limit of the multiplex PCR method is approximately 1%. This result indicates that this multiplex PCR method could be a useful tool for monitoring unauthorized GM carnation in Korea.

TKR-TSR 연결을 위한 나진(Rajin)-하산(Khasan) 프로젝트의 발전방향과 전략수립

나진-하산 프로젝트(Rajin-Khasan Porject)는 TKR-TSR 연결을 위한 시범사업이기 때문에 본 프로젝트가 갖는 의미는 향후 우리나라의 대북사업 및 TSR의 성공적인 연결을 위한 지표가 될 수 있다. 따라서 본 연구에서는 2008년 이후 멈춘 TKR의 사업 현황과 TKR과 TSR 연결이 갖는 의미를 살펴보며 TKR-TSR 연결을 위한 나진-하산 프로젝트의 발전방향을 제시하였다. 본 프로젝트의 기대효과를 보면 러시아와 북한에 막대한 물류비용 수익증가, 새로운 경제권의 형성, TSR 이용 운임에 관한 물류협상의 주체, 수출선의 다변화, 북한의 개혁ㆍ개방을 촉진을 기대할 수 있다. 이러한 효과가 기대되는 나진-하산 프로젝트의 성공적인 발전을 위해서 본 연구는 다음과 같은 발전방향을 제시하였다. 첫째, 물량의 안정적인 확보와 둘째, 물류시설의 현대화, 셋째, 북한과 러시아간의 입출항 절차의 신속성, 넷째, 국민의 공감대 형성, 다섯째, 북한의 건전한 비즈니스 파트너쉽 마지막으로 TKR의 연결과 같은 발전방향을 제시하였다. 본 연구는 초기상태인 나진-하산 프로젝트에 대한 기대효과와 발전방향에 대한 시사점을 고려하였다는데 실무적 시사점을 찾을 수 있으며 향후 실증분석을 위한 변수의 제시에서 학문적 시사점을 찾을 수 있다. Purpose : This paper is about expectation effectiveness and direction of improvement of Rajin-Khasan Project regarded as demonstration business for instructing between TKR and TSR. Research design, data, methodology : For suggesting effectiveness and direction, this paper reviews the overviews of the TKR, TSR and Rajin-Khasan Project. Moreover, this study analyzed the factors that have influenced on the modal shift toward railroad, which are become reference for diffusion of railroad. Results : We suggest the directions for the success of the project as follows; cargoes stabilization, modernization of distribution facilitates, expedience procedures of arrival and depature between north Korea and Russia, form a social consensus, closer partnership between north Korea and south Korea and linkage of TKR. Conclusions : This study suggests managerial implications and strategic problems for Rhajin-Khasan Project of initial stage.

위성항법 지상기반보강시스템(GBAS) 기술 및 개발 동향

국제적인 항행계획에 따라 기존의 ILS, VOR, 그리고 DME와 같은 기존의 항행시설을 위성항법시스템 기반으로 대체할 이행계획이 수립되고, 각 국가에서는 이를 이행하기 위한 준비를 수행하고 있다. 대표적인 위성항법시스템 기반 항행기법으로는 ABAS, SBAS, 그리고 GBAS가 있다. GBAS는 공항 주변 지상에 설치된 기준국을 활용하여 보정 및 무결성 정보를 생성하여 접근중인 항공기에 제공하여 주는 시스템이며, 곡선 접근 및 다수의 활주로를 지원할 수 있다는 장점을 가진다. 2009년, 최초로 CAT-I급 GBAS(Honeywell SLS-4000)가 제품인증을 받았으며, 최근에는 CAT-II/III급 GBAS 장비 및 표준 개발이 전 세계적으로 활발히 진행중에 있다. 본 논문에서는 국제 GBAS 기술개발 동향과 함께, 현재 진행 중인 GBAS 표준화 및 향후 전개될 GBAS 발전 방향에 대해 제시하고자 한다. According to the global air navigation plan from ICAO, the implementation plan to substitute the existing ILS, VOR, and DME by satellite navigation system has been completed and many countries are preparing the plan to implement it. Representative techniques for air navigation based on satellite navigation system are ABAS, SBAS, and GBAS. GBAS provides the correction and integrity information generated from ground facilities installed around the airport to aircraft for supporting the precision approach and landing. In 2009, FAA granted system design approval to Honeywell on its GBAS CAT-I equipement. Recently, CAT-II/III GBAS research and development activities with its standarization are in progress actively. This paper presents the global status of GBAS research and development and the advanced GBAS standardization and techniques.

Presacral air insuffulation with tomogram demonstrated the presence of a enlarged left sided suprarenal tumor, confirmed at operation and histologically proven to be adenoma. I emphasized that the relief of hypertension of primary aldosteronism can -be accomplished frequently by surgical means.
