우리나라의 생명공학기술 수준은 선진국에 비해 93년도 50%, 97년도 80% 수준으로 평가된다. 연구인력은 97년도 기준 4천4백35명이며 시장규모는 4천2백억원으로 세계시장에 비해 1.6% 수준이다. 최근 성공한 복제소 영롱이의 개발은 우리의 유전공학분야가 세계수준으로 진입했음을 증명해주는 쾌거이며 이제 우리나라도 세계 5번째 동물 복제국이 되었다.

인체 인터루킨 - 2 - ( IL - 2 ) Peptide 의 단일클론 항체 생산

Monoclonal antibody against Interleukin-2 peptide was generated. One hundred four hybridomas which were survived in HAT media produced anti-IL2 antibody. Among 104 hybridomas, eleven clones has been identified to produce IgG1 type monoclonal antibody which has kappa light chain after ELISA subtyping analysis. After purification of IgG1 monoclonal antibody, heavy and light chain of immunoglobulin has been identified whose molecular weights are 55 kD and 25 kD, respectively. These IL-2 antibodies were confirmed using immunoblotting techniques. IgGl type IL-2 monoclonal antibody were bound to 133 amino acids IL-2 antigen whose M.W. is 15 kD. These monoclonal antibodies were used for purifying IL-2 peptides. IL-2 ELISA systems were generated using immuno-affinity purified IL-2 as antigens.

All six clones has been identified to produce IgM-type monoclonal antibody which has kappa light chain after ELISA subtyping analysis. After purification of IgM monoclonal antibody, heavy and light chain of immunoglobulin has been identified whose molecular weights are 79KD and 26.5KD, respectively. These pre-S2 antibodies were confirmed using immunoblotting techniques. IgM type pre-S2 monoclonal antibody were bound to 55 amino acid pre-S2 antigen whose M.W. is 55KD.

대장균에서 재조합 발현된 Hepatitis B virus 표면항원 Pre-$S_2$ peptide로 면역시킨 Balb/c 마우스 비장세포와 SP2/O-Ag 14 미에로마 세포를 융합시켜 Pre-$S_2$에 대한 단일클론 항체를 생산하였다. HAT 선택배지에서 자란 70개의 융합세포군 중 Pre-$S_2$에 대한 항체를 생산하는 6개의 융합세포군을 제한 희석법으로 클로닝하였으며 ELISA 방법을 이용하여 이들 6클론을 isotype 한 결과 모두 kappa light chain을 갖는 IgM이었다. 이들 항체의 항원특이성은 western bolt를 이용하여 55 amino acid Pre-$S_2$ peptide와 결합함을 확인하였다. Rubus crataegifolius (RC)는 장미과에 속하는 전통적인 아시아 약용 식물이다. RC 열매는 항산화 작용을 통해 성인병을 예방하는 것으로 알려져 있다. 본 연구에서는 RC 열매 추출물(RCex)이 비만과 비알코올성 지방간 질환(NAFLD)에 미치는 영향을 동물 모델을 통해 평가하였다. 28마리의 수컷 C57BL/6J 마우스에 8주간 비만을 유도한 후, 추출물을 8주간 경구 투여하였다. 그룹 1은 일반 대조군으로 표준사료를 섭취하였다. 그룹 2는 HFD 대조군으로, 그룹 3에는 심바스타틴(6.5 mg/kg/일)을, 그룹 4에는 RCex (200 mg/kg)을 투여하였다. RCex투여는 실험 마우스의 체중, 지방 조직, 간 무게를 감소시켰으며, 또한 지질 대사(ALT, AST, TC, TG, LDL, HDL)를 포함한 생화학적 바이오마커를 개선하였다. AMPK의 활성화는 지방생성 유전자(LXR, SREBP-1c, FAS, ACC1)의 발현을 감소시켰으며, RCex에 의한 CPT 활성 증진 효과를 검증하였다. RCex는 또한 에너지 소비 및 신진대사와 관련된 호르몬(adiponectin 및 leptin)의 혈장 수준에도 영향을 미쳤다. 또한, RCex가 HFD로 유도된 비만 mice의 포도당 불내성을 개선했음을 확인 하였다. RCex는 AMPK의 인산화를 통해 지방산 산화 및 지방산 합성을 조절함으로써 항비만 및 항NAFLD 효과를 가짐을 처음으로 입증하였다. 이는 R. crataegifolius가 비만 및 관련 NAFLD 예방에 좋은 보충제가 될 수 있음을 시사한다. Rubus crataegifolius (RC) is a traditional Asian medicinal plant belonging to the Rosaceae family. The fruits of RC are known to prevent adult diseases through antioxidants. In this study, the effects of RC extract (RCex) on obesity and nonalcoholic fatty liver disease (NAFLD) were evaluated in animal models. Twenty-eight male C57BL/6J mice were induced to become obese for 8 weeks and then the extract was orally administered for 8 weeks. RCex reduced body weight, adipose tissue, liver weight. RCex improved biochemical biomarkers including lipid metabolism (alanine aminotransferase (ALT), aspartate aminotransferase (AST), plasma triglyceride (TG), total cholesterol (TC), high-density lipoprotein (HDL) cholesterol and low-density lipoprotein (LDL) cholesterol). The activation of AMP-activated protein kinase (AMPK) reduced the expression of adipogenesis genes (liver x receptor (LXR), sterol regulatory element-binding protein-1c (SREBP-1c), fatty acid synthesis (FAS), acetyl-CoA carboxylase 1 (ACC1) and the effect of enhancing carnitine palmitoyltransferase (CPT) activity by RCex was verified. RCex also influence on plasma production of hormones (adiponectin & leptin) related on energy expenditure and metabolism. In addition, we confirmed that RCex improved glucose intolerance in HFD-induced obese rats. RCex was first demonstrated to have anti-obesity as well as anti-NAFLD effects by regulating fatty acid oxidation and fatty acid synthesis by phosphorylation of AMPK. This suggests that RCex could be a good supplement for the prevention of obesity and related NAFLD.

노인의 생활체육활동 참가와 신체적, 정신적 기능상태의 관계

이 연구는 노인의 생활체육활동 참가와 신체적, 정신적 기능상태의 관계를 구명하는데 그 목적이 있다. 본 연구의 조사 대상은 2008년 현재 서울지역에 거주하는 만 65세 이상 노인을 모집단으로 설정한 후, 유층집락 무선표집법(systematic stratified cluster random sampling)의 방법을 이용하여 양로원, 노인정, 노인대학 집단 등을 중심으로 표집틀(sampling framework)을 작성한 후, 생활체육참여자 180명, 비참여자 180명 총 360명의 표본을 추출하였다. 독립변인으로는 여가스포츠활동 참가여부와 참가정도(기간, 빈도, 강도)로 분류하였으며, 그리고 종속변인인 신체적 기능상태는 정경희 등(1998), 김양례(2005a)등이 사용한 문항을 토대로 본 연구에 맞게 수정, 보완하여 사용하였다. 정신적 기능상태는 인지기능 척도와 우울척도를 사용하였다. 먼저, 인지기능 척도는 개인이 지능을 이용하여 당면문제를 해결하는 정도로써, 한국판 MMSE-K(Mini Mental State Examination-Korea)를 사용하였으며, 우울증 척도는 Beck(1978)가 개발하고 이영호, 송종용(1991)이 번안한 설문지를 조규황(2003)이 한국 실정에 맞게 수정한 설문지를 본 연구의 목적에 맞게 수정ㆍ보완하여 사용하였다. 연구결과의 분석을 통해서 다음과 같은 결론을 얻었다. 첫째, 노인의 생활체육활동 참가여부에 따라 신체적ㆍ정신적 기능상태에 부분적으로 영향을 미친다. 즉, 생활체육활동에 참가하는 노인집단이 참가하지 않는 노인집단보다 일상생활수행능력ㆍ인지기능과 우울증 하위변인 중 인지적ㆍ신체적ㆍ행동적 요인이 높았다. 둘째, 노인의 생활체육 참가정도는 신체적ㆍ정신적 기능상태에 부분적으로 영향을 미쳤다. 즉, 생활체육 참가정도에 따라서 신체적, 정신적 기능상태의 하위변인인 일상생활수행능력에서는 참가기간, 참가강도가 인지기능에서는 참가기간, 그리고 우울증의 하위변인 중 인지적ㆍ정서적 요인에는 참가기간, 참가빈도가 영향을 미쳤고, 신체적ㆍ행동적 요인에는 참가기간, 참가빈도, 참가강도 모두 영향을 미치는 것으로 나타났다. The purpose of this study was to study the Relationships between Sport for All Participation and Physical and Mental Function affecting the Elderly's Self-perceived Health. For accomplishing this purpose of this study, the 308 subjects were selected by the multi-stage stratified cluster sampling method from the Seoul, in the years of 2008. presently. Out of the 360 questionnaires responded to, only 308 were used for data analysis because 52 were responded to incompletely or incorrectly. The survey questionaries were used to collect data. The questionary for physical function state were developed and standardized by kung-hee chung(1993), yang-lae Kim(2005). And mental function state were divided two parts; self-perceived scale and depress scale. The questionary for self-perceived scale were based on MMSE-K(Mini Mental State Examination-Korea). The questionary for Depression were based on Depression Scale developed by Beck(1978), adapted in Korean and standardization by Lee Young-ho & Song Jong-yong(1991), regulated by Jo Gyu-whang(2004). From the analysis of this study, the following conclusions were obtained: First, according to the elderly people's participation in leisure sport activities, it partially affected physical and mental function state. Second, according to the degree of elderly people's participation in leisure sport activities, it partially affected physical and mental function state.

손가락 끝 박리법을 이용한 손을 이용한 복강경 부분신절제술의 초기 경험

Purpose: We reported the initial experience of hand-assisted laparoscopic partial nephrectomy (HALPN) for kidney neoplasm using finger-tip dissection technique. Materials and Methods: A total 11 patients with exophytic renal mass who had undergone HALPN for kidney neoplasms. After mobilizing the entire hilum thoroughly, the renal artery was hung with a rubber catheter. When massive bleeding occurred, it was clamped. Surgeon’s index finger-tip was used for the mass dissection. After making the incision line 5-10mm from the mass using scissors, the resection plane was gradually deepened bluntly by the finger-tip, which was directed centrally along the grain of the renal pyramids. If large vessels and calyxes were felt by the finger-tip during dissection, they were secured by clipping. The mass was completely removed by pinching off using the fingers. Results: Mean patient age was 51.4 years (37-72 years), mean tumor size was 2.1cm (1.0-3.0cm), the mean operation time was 163.4min (120-310min). In only five patients, renal artery was clamped. In these cases, mean ischemic time was 24min (15-30min). All patients had negative resection margins. Mean estimated blood loss was 304.8ml (63-900ml) and two patients required a post-operative blood transfusion. No serious complications occurred. Conclusions: The finger-tip dissection technique of renal mass in HALPN is feasible. This technique can allow the easy and safe dissection by the maximizing the tactile sense.
