Cell Surface Tissue Transglutaminase-induced Activation of Phosphoinositol 3-Kinase/Akt Pathway

Tissue transglutaminase (tTGase)는 lysine과 glutamine residue를 서로 연결하여 Ca²?의 조절 하에 Nε-(γ-L-glutamyl)-L-lysine (GGEL) isopeptide 결합을 형성한다. tTGase는 세포 안과 밖의 단백질들을 서로 결합하는 작용을 하므로 이와 같은 기능을 통해 상피조직의 기능성 방벽 또는 apoptosis와 같은 여러 가지 생명 현상에 관여하는 것으로 알려져 있다. tTGase는 주로 세포질 내에 존재하지만 세포에 따라서는 상당한 양의 tTGase가 핵에서 발견되기도 하며, 세포표면과 세포외기질(extracellular matrix)에서 관찰된다. 본 연구는 세포표면부착 tTGase가 세포의 밖에서 세포막을 통과하여 세포질 내로 전달되는 과정에서 어떤 역할을 하는지 관찰하고자 하였다. 사람의 tTGase 단백질을 과발현 시킨 H9c2 세포주(H9c2/tTGase)를 이용하여 세포면역형광염색, western blot 분석 방법을 이용하여 세포표면의 tTGase 단백질의 증가를 확인한 후 항 tTGase 항체를 처리하여 세포 외부로부터 내부로 전달되는 신호를 증폭하는 방법을 이용하여 세포표면의 tTGase가 관여하는 신호전달체계를 조사하였다. 과발현된 tTGase 단백질은 세포내부뿐만 아니라 세포표면에서도 증가하였으며, 세포배양액으로의 분비량 역시 증가하였다. H9c2/tTGase 세포의 세포표면에 부착된 tTGase 단백질을 통해 증폭된 신호를 세포내부로 전달한 결과 세포질 내부의 tTGase 발현이 감소하였다. 이와 같은 tTGase 단백질의 감소는 phosphoinositol 3-kinase/Akt (PI-3K/Akt) 신호전달계를 통해 mitogen-activated protein kinase (MAP kinase)에 전달된 신호에 의해 일어나는 것으로 사료된다. 본 연구의 결과들은 활성화된 세포로부터 유리된 tTGase가 세포표면에 부착됨으로써 세포내부의 신호전달체계에 영향을 미치는 것으로 사료되며, 이와 같은 결과는 암 발생과 전이 등의 세포부착과 관련된 질병현상의 이해를 돕기 위한 기초자료가 될 것이다. Multifunctional tissue transglutaminase (tTGase) is found in the cytoplasm and cell surface, as well as in the extracellular matrix. However, it is difficult to determine the exact function of tTGase in each cell compartment. This study focused on the potential role of cell surface tTGase in the process of “outside-in” signal transduction. Immunofluorescence study and western blotting was performed to localize the overexpression of tTGase. tTGase-overexpressed H9c2/tTGase cells were treated with anti-tTGase antibody to evaluate the potential functions of tTGase on the outside-in signal process. The tTGase level markedly increased in each cell compartment and the culture media of H9c2/tTGase cells that show overexpression of tTGases. Anti-tTGase monoclonal antibody reduced tTGase levels in the whole lysate of H9c2/tTGase cells, and concomitantly increased the activity of the Akt. The results suggest that the cell surface expression of tTGase may be associated with an intracellular signaling pathway via the phosphoinositol-3 kinase/Akt. Phosphorylation of mitogen activated protein kinase family, ERK1/2, and Jun N-terminal Kinase (JNK), was also inhibited in the anti-tTGase antibody-treated H9c2/tTGase cells. These results suggest that cell surface tTGase may regulate intracellular signaling pathways in an autocrine or paracrine manner.
동시증착에 의한 Si(111) - 7×7 기판 위에 TiSi₂ 에피택셜 성장

초고진공에서 기판 Si(111)-7×7 위에 Ti : Si = 1 : 1 또는 1:2 의 조성비로 Ti와 Si을 동시 증착한 후 in situ 열처리하여 TiSi₂ 박막을 에피택셜 성장시켰다. XRD와 XPS 분석결과 동시증착된 혼합 층에서 C49-TiSi₂ 박막의 성장은 핵형성에 의함을 확인하였으며, 양질의 C49-TiSi₂ 박막은 Ti를 증착한 후 Ti와 Si를 동시 증착한 (Ti+2Si)/(Ti)/Si(111)-7×7 구조의 시료를 초고진공에서 500℃에서 열처리하여 얻을 수 있었다. 형성된 C49-TiSi₂/Si(111)의 계면은 깨끝하였고, HRTEM 분석 결과 C49-TiSi₂/Si(111)의 계면은 약 10℃의 편의를 가지면서 TiSi₂[211]││Si[110], TiSi₂(031)││Si(111)의 정합성을 가졌으며, 시료의 전 영역에 에피택셜 성장되었다. TiSi₂ film was grown epitaxially on the Si(111) substrate by means of the coevaporation of Ti and Si (Ti : Si= 1 : 1 to 1 : 2) on Si(111)-7×7 surface followed by in situ annealing in ultrahigh vacuum. X-ray diffraction and X-ray photoelectron spectroscopy showed that silicide formation was initiated by the nucleation of C49-TiSi₂ crystallites in the deposited layer. High-quality epitaxial C49-TiSi₂ layer was obtained by in situ annealing (Ti+2Si)/Ti/Si(111)-7×7 sample at 500℃ for 10 min in UHV. TiSi₂/Si(111) interface was clear and the orientation relationship between the C49-TiSi₂ and the Si(111) substrate deduced from the HRTEM lattice image was TiSi₂[211]││Si[110], TiSi₂(031)/Si(111) with a misorientation angle of 10˚. The whole area of the sample was covered with the epitaxial C49 structure.

Takotsubo cardiomyopathy is a clinical syndrome characterized by chest pain, transient left ventricular dysfunction, and specific electrocardiographic changes induced by physical or emotional stress. We describe a rare case of this syndrome associated with acute mitral valve bacterial endocarditis in a young female.
헤어리베치 신품종 조생종 콜드그린과 중만생종 청파의 생육특성 및 수량성

본 시험의 목적은 대구 경북에 자생하는 헤어리베치로 다수성 조생품종과 중만생품종을 개발하기 위하여 수행하였다. 경북 성주와 경남 사천에서 1998년부터 2006년까지 자료수집 및 순화, 생육이 왕성한 조생 및 중만생계통선발, 상호교잡의 단순순환선발법으로 조생종인 콜드그린과 중만생종인 청파를 육성하였다. 육성종자는 2004년부터 2006년까지 경북성주와 경남사천에서 생육특성과 수량을 평가하였다. 줄기의 털은 콜드그린과 청파는 있었으나, 헤이메이커프러스와 라티고는 없었다. 잎의 모양은 콜드그린은 엽폭이 넓은 장타원형이고, 청파, 헤이메이커프러스, 라티고는 좁은 장타원형이었다. 꽃색은 콜드그린은 자주색이고 청파는 보라색이었다. 내한성은 성주에서 콜드그린, 청파, 라티고가 헤이메이커프러스 보다 높았으며 사천에서는 차이가 적었다. 개화기는 콜드그린과 헤이메이커프러스가 조생종으로 유사했고 중만생종인 청파는 라티고 보다 다소 빨랐다. 성주에서 건물수량은 콜드그린이 다른 품종 보다 높았으며 청파와 헤이메이커프러스는 라티고 보다 높았다(P<0.05). 2005년 사천에서는 콜드그린과 헤이메이커프러스는 다른 품종보다 높았으며, 청파는 라티고 보다 높았다(P<0.05). 베치의 조단백질 함량은 높고 ADF의 함량은 낮았다. The objective of this research was to develop a high yielding, early and medium-late flowering new hairy vetch(Vicia villosa Roth) varieties derived from an accession in Korea. Most vigorous early and medium-late flowering hairy vetch lines were selected and crossed by open pollination and their seeds were bulked and plants were reselected at Seongju in the Keongbuk and at Sacheon in the Keongnam. A performance trial was conducted to evaluate agronomic characteristics, forage quality and dry matter(DM) yield of new hairy vetch early-maturing 'Cold green' and medium-late maturing variety 'Cheong pa' at Seongju and Sacheon. 'Cold green' and 'Cheong pa' are hair but 'Haymaker plus' and 'Latigo' are nearly hairless. 'Cold green' has purple flowers while 'Cheong pa' has violet. The cold tolerance of 'Cold green,' 'Cheong pa' and 'Latigo' was higher than 'Haymaker plus' at Seong-ju, Keongbuk province, in inland region but there were a little differences at Sacheon, Keongnam province in southern coast region. Fifty percent-flowering dates of 'Cold green' and 'Haymaker plus' had earlier than others and 'Cheong pa' was earlier than 'Latigo'. The DM yield of 'Cold green' was higher than that of others, but 'Cheong pa' and 'Haymaker plus' had higher than 'Latigo' at Seong-ju(P<0.05). In Sacheon, The DM yield of 'Cold green' and 'Haymaker plus' was higher than others and 'Cheong pa' had higher than 'Latigo'.
Supra Pubic Catheter를 施行한 腦卒中 患者 1例에 對한 證例報告

According to the literary study on catheterization of bladder in one case of patient with C.V.A. who was treated by supra pubic catheterization, the results were as follows. 1. If not immediately treat ischuria, emergency disease, very great danger to life can happen by loss of kidney function. So we must treat ischuria instantly by consideration of treatment from the factor. 2. Catheterization of bladder generally acted in emergency room, is emergency treatment only in the lower part of urethral obstruction. 3. Catheterization of bladder was put on record in Son Saimiao's Qianjinyaofang, which is the first book on it. It had been developed by Luo Qianyi, Zhang Jiepin, and Wang Dao. 4. Ancient times, an urethral catheter was made of green onion leaf, pig bladder, goose feather and so on. 5. Nelaton is catheterization of bladder in functional or momentary ischuria. Indwelling catheterization is treatment keeping in urethral canal for a long time. 6. Supra pubic catheterization is a surgical operating catheterization when urethral catheterization is impossible, is very easy and can decrease risk of infection. 7. After catheterization, for decreasing risk of the infection, a complication of catheterization, it is must be taken precedence, ascetic treatment, constantly management and sterilization, anti-biotic medication. 8. At times, increasing patient of emergency disease, as C.V.A etc., the needing of catheterization of bladder is increasing also for preventing infection and bed sore by ischuria and incontinence of urine. Therefore a specialty knowledge and skill about it is necessary.
경피내시경하 위루술(PEG)을 시행한 중풍후유증 환자 1례에 대한 증례보고

According to the case study of patient with seguela of C.V.A who was treated by Percutaneous Endoscopic Gastrotomy(PEG), his chief complaint is the dysphagia. We treated him by a viewpoint of oriental medicine and got the good clinical symtom.

Gullian Barre Syndrome is an acute symmetric polyradiculoneuropathy that typically presents as a progressive flaccid paralysis. The pathology is believed to be caused by both cellular and humoral immune processes The possibility of death is 3-5% and About 20% of patients require mechanical ventilaton. Recently We have experienced a patient who had suspected Gullian Barre Syndrome for 60days, his condition weakness and pain was impoved through the herb and acupucture Therefore I reported it for the treatment.

In patients with Cerebral vascular accident, Voiding Dysfunction is very usual, but if with no improvement for a long time, it can cause many complexed-problems as urinarytract injury, infection or inflamation and patients suffering from pain, inconvenience and anxiety. In this case 58-year-old female with Voiding Dysfunction with Neurogenic Bladder after Cerebellum Hemorrhage, for 6 monts there was no interval change on symptom, We prescribed appropeiate herbal medication, and perform acupuncture and moxa. 5 days later after admission in Taejeon Oriental Hospital, The patien was able to have self-voiding and gradually improved. So we report it for the better treatment.
