Vibrio fluvialis oligopeptide permease (oppA) 유전자 deletion에 의한 생리적 특성

Oligopeptide is known to be an essential nitrogen nutrient for bacterial growth. Oligopeptide can be transported into cytoplasm by a specific transport system, Opp system. Opp system is composed of five proteins, which are transcribed by an operon. These are responsible for oligopeptide binding protein (OppA), permease (OppB and OppC) and energy generation system (OppD and OppF), respectively. Previously, we isolated the opp operon from Vibrio fluvialis and constructed the oppA mutant by allelic exchange method. In this study, we investigated the growth pattern and biofilm production under the different growth condition. When the cells were cultivated using brain heart infusion(BHI) medium, the wild type was faster than the mutant in growth during the exponential phase. However, it showed that the growth pattern of two strains in M9 medium is very similar. The growth of wild type showed better than that of the mutant grown at pH 8. At pH 7, there was no an obvious difference in growth. After 5 mM $H_2O_2$ was treated to the cells $(OD_{600}=1.2)$, the cell survival was examined. The oppA mutation did not affect in survivability. In the presence of $10{\mu}g/ml$ polymyxin B, the biofilm production of the oppA mutant was higher than that of the wild type. 미생물이 이용할 수 있는 nitrogen source는 di-, tri-, oli- go-peptide 또는 amino acid의 형태로 세포내로 uptake되어 대사과정에 사용되고 있다. 이와같은 peptide는 특이한 transport system에 의해서 이동되고 있는데 oligo peptide(Opp) transport system에는 binding protein, permease protein, energy 생성을 위한 ATP 분해에 관여하는 protein 이 관여하고 있으며 염색체 상에서 이들 단백질들은 operon 형태의 유전자로부터 발현되고 있다. 본 연구는 gram 음성 세균이며 수해양 서식 세균인 V, fluvialis로부터 얻어진 Opp operon 유전자 가운데 oligopeptide binding protein을 coding하고 있는 oppA 유전자가 deletion된 mutant를 사용하여 여러 환경변화에 따른 생육을 wild type과 비교한 연구 결과 이다. 생육을 위한 완전배지인 brain heart infusion (BHI) 배지와 최소배지인 M9 minimal 배지를 사용한 결과 OppA protein의 생성 결핍에 따라 초기 및 대수증식기 과정 중에는 mutant의 생육이 늦어지고 있으나 Opp system이 아닌 다른 peptide전달 경로로 추정되는 system을 이용하여 대수 증식기 후반에서는 wild type과 거의 같은 생육 형태를 보여 주고 있었다. pH의 변화에 따른 생육은 pH 7에서는 생육정도가 비슷하였으나 약알칼리 부근에서는 oppA mutant의 생육이 wild type에 비하여 낮아지고 있었다. 또한 5 mM $H_2O_2$를 사용하여 $OD_{600}=1.2$농도의 세포들에 대한 영향을 검토한 결과 두 균 모두 높은 생존율을 보여 주었으며 이는 대수증식기 세포들을 사용한 결과와는 매우 다른 형태를 보여 주고 있었다. 항생제 내성에 대한 연구에서는 mutant가 streptomycin과 tetracycline 에 대해서는 wild type과는 다르게 매우 낮은 농도에서도 생육이 되고 있지 않으나 polymyxin B에 대해서는 wild type과 같이 $10{\mu}g/ml$의 농도에서도 잘 자라고 있었다.

빅터 터너(Victor Turner)의 의례이론의 예배 연구에의 적용

This paper consists of two parts. The first part is a brief explanation of Victor Turner`s ritual theory. The second part is its application to Korea, i.e., to the liturgical movement of "Koreanizing of worship that has been developing since late the 1980`s. In the latter part of this paper the relation between the renewal of worship practice and the social context has been investigated. Turner`s concept of social drama has been devised to describe and to analyze the context of ritual. According to Turner, context of ritual is characterized as a dynamic process, rather than a static condition. An example of a dynamic process is a social conflict. Social drama is a unit of the episode exposing social conflicts. A ritual has been defined as a redress in social drama, which consists of four phases: breach, crisis, redress and reintegration or separation. The concept of social drama is a useful device to explain the relation between ritual and a social context. Since late the 1980`s in Korea there has been a liturgical movement called "Koreanizing of worship." This movement has had a socio-cultural background which tried to restore Korean cultural identity. The liturgical movement in Korea is regarded as a redressive action in a social drama happening because of a conflict around the cultural identity in Korean society. According to Turner, a redressive action has two distinctive features: self-reflectivity and liminal quality. The Korean church and theology have practiced self-reflection on depreciation of the traditional Korean culture in the Korean church history. The form of renewed worship accepting Korean traditional cultural elements appeared to neither be totally koreanized, nor be handed down from the missionaries. This feature, "betwixt and between," is to be regarded as a liminal quality of redress. The liminal, according to its own quality, cites a possibility of becoming. Worship as a ritual function is a sphere of possibility. The possibility indicates and alternative. The Korean worship renewal shows an altemative cultural topos for the Korean society. From this point of view, the renewal of worship practice has a closecorrelation with the social context.

The contemporary discussion about death and its ritual is focused on the privatization of death ritual and the exclusion of death in every day life. The religious system is considered as a subsystem which supports the society by giving the holistic meaning of death. The privatization of death and its ritual may vanish the holistic approach to death and life from individuals. For this reason, the privatization of death and its ritual could be a obstacle which should be overcome for the holistic meaning of death. A death hurts the heart of the bereaved. However, the message of death can easily turn into that of resurrection in the Korean Protestant death ritual. If Protestants don’t mention about the reality of death even in this ritual, there is no chance to reflect seriously on death and life. Korean Protestants have to overcome the privatization of death ritual and the exclusion of death in every day life. For this purpose, they should resist the socioeconomic pressure to commercialize this ritual. Furthermore, they must enrich the meaning of a transition stage in death ritual as the rite of passage. 영아 보육교사의 자질 및 역할과 전문성에 관한 심층사례 연구

The purpose of this study was to investigate the basic materials for enhancing the professionalism of 51 infant/toddler teachers through in-depth interview about the actual condition of child care centers. The results were as follows: First, the infant/toddler teachers got less credits of young child care and education subjects than day care teachers did. The current criteria for qualification of infant/toddler teachers was not appropriate for providing the child care services. An efficient and substantial curriculum must be designed to facilitate the day care services for infant/toddler teachers. Second, the infant/toddler teachers recognized that their roles were similar to child care teachers. The curriculum and practice must encourage and nurture the characters, beliefs, and dispositions of infant/toddler teachers. Third, it is necessary to review the various problems to enhance the professionalism of infant/toddler teachers. Several suggestions for enhancing the professionalism are discussed.

영아 및 유아 보육교사의 개인적 특성, 근무여건, 사회적 지지가 소진에 미치는 영향

본 연구는 보육교사의 개인적 특성 및 근무여건과 사회적 지지와 소진이 관련성이 있는지를 살펴보고자 수행되었다. 보육교사의 개인적 특성으로는 결혼유무, 교직경력, 학력, 이직횟수가, 근무여건으로는 근무시간, 보수, 보육아동 연령이 포함되었다. 영아반 교사 102명과 유아반 교사 118명을 합한 총 220명의 자료를 분석한 결과, 소진의 수준은 기대한 것보다 크게 높지 않았다. 소진은 개인적 특성이나 근무여건에 따라 상이하였는데, 미혼이거나 교직경력이 낮고 유아반 교사가 소진을 더 많이 경험하였다. 또한 근무시간이 길고 보수가 적을수록 소진의 지각수준은 증가하였다. 그러나 보육교사가 직장 내 동료들이나 시설장으로부터 지지를 받는다고 지각할수록 소진 수준은 낮아졌다. 한편, 교사의 학력이나 이직을 한 횟수에 따른 소진의 차이는 나타나지 않았다. The purpose of this study was to examine a variety of factors which would be associated with burnout of child care teachers. A questionnaire was distributed to 220 teachers who worked in nonprofit, for-profit and company-operated child care centers. Results showed that there were differences in burn out in terms of marital status, work experiences of child care teachers. The reports of burnout were higher among single and young teachers. In addition, working hours, salary and social support were statistically related with burnout of child care teachers. The higher levels of burnout was found in a group of teachers who worked for more hours in average, a group of those who got the lower salary, and a group of those who had the lower level of social support group. Teachers for 3-through 5-year-olds were likely to report higher levels of burnout than teachers of infants and toddlers. Implications for diminishing burnout in child care teachers are discussed.

Comparison of Muscle Activity Ratio of Upper Trapezius to Serratus Anterior During Shoulder Elevation Between Subjects With and Without Pain Experienced in Upper Trapezius

The aim of this study was to compare the activity of the upper trapezius (UT) and serratus anterior (SA) and ratio of UT to SA during shoulder elevations. Ten subjects with UT pain (UTP) and 13 subjects without UTP participated in this study. Subjects with a UTP of over five in a pain intensity visual analogue scale (0-10 ㎝) for more than 2 months and latent myofascial trigger points (MTrPs) in the UT muscle were included in the UTP group. Electromyography (EMG) data of UT and SA at 1st and 10th elevations were analyzed. Two-way repeated analyses of variance were used to compare the EMG activity of UT and SA and the ratio of UT to SA during shoulder elevations between groups with and without UTP. There was a significant increase in UT/SA ratio in the group with UTP compared to the group without UTP (p=.01). The activity of UT and SA measured at the 10th elevation was significantly greater than that in the first elevation (p<.05). The activity of SA was significantly greater in the group without UTP than the group with UTP (p=.03). However, there was no significant difference between groups with and without UTP in terms of UT activity (p=.28). These results indicate that UTP may have relevance to the increased muscle activity ratio of UT to SA during shoulder elevations.
