        Complete genome sequence of Deinococcus puniceus DY1<sup>T</sup>, a radiation resistant bacterium

        이 연구에서는 5 kGy 의 감마선에 조사된 토양으로부터 분리된 Deinococcus puniceus $DY1^T$의 완전한 게놈서열을 분석하였다. 이 균주는 UVC 와 감마선에 대한 저항성을 보였으며, PacBio RS II platform 을 통해 시퀀싱을 진행하였다. 해당유전체의 분석결과 G + C 함량이 62.5%인 2,971,983 bp 크기의 원형 염색체를 확인하였으며, 해당 염색체는 2,617 개의 코딩 서열과 2,762 개의 유전자 그리고 88 개의 위유전자를 포함하고 있다. Cells of Deinococcus puniceus $DY1^T$ are Gram-positive, coccus-shaped, and crimson color-pigmented. Strain $DY1^T$ was isolated from soil irradiated with 5 kGy gamma radiation and showed resistance to UVC and gamma radiation. In this study, we report the complete genome sequence of a bacterium Deinococcus puniceus $DY1^T$ is consist of circular chromosome comprised of 2,971,983 bp, with the G + C content of 62.5%. The complete genome sequence was obtained using the PacBio RS II platform, it included 2,617 coding sequences (CDs), 2,762 genes, and 88 pseudogene.

        Complete genome sequence of Spirosoma montaniterrae DY10<sup>T</sup> isolated from gamma-ray irradiated soil

        그람 음성이며 황색인 긴 막대 모양의 세균 Spirosoma montaniterrae $DY10^T$는 전라북도 덕유산에서 분리가 되었다. 이 세균의 세포는 감마선에 대해 12 KGy의 $D_{10}$값을 보이며 극단적인 감마선 저항성을 보였다. $DY10^T$ 균주의 완전한 게놈서열은 5,116개의 유전자, 39개의 tRNA 유전자를 포함하는 원형 염색체(5,797,678 bp)로 구성되었다. 유전체 특징은 감마선 및 UVC에 대응하는 주요 효소를 포함하였다. A Gram-negative, yellow-pigmented, long-rod shaped bacterium Spirosoma montaniterrae $DY10^T$ was isolated from a soil sample collected at Mt. Deogyusan, Jeonbuk Province, Republic of Korea. Cells showed extreme gamma radiation resistance with the $D_{10}$ value of 12 KGy. The complete genome sequence of strain $DY10^T$ is consist of a circular chromosome (5,797,678 bp) encoding 5,116 genes, 9 rRNA genes and 39 tRNA genes. The genomic features contain the key enzymes for gamma and UVC radiation.

        A report of 9 unrecorded radiation resistant bacterial species in Korea

        Five bacterial strains, ES10-3-3-1, KKM10-2-2-1, Ant11, JM10-4-1-3, and KMS4-11 assigned to the genus Deinococcus were isolated from soil samples collected from Namyangju-si in Gyeonggi-do, Gangnam-gu and Dongdaemun-gu in Seoul, Korea. In addition, four bacterial strains, KKM10-2-7-2, JM10-2-5, JM10-2-6-2, and KKM10-2-3 assigned to the genus Hymenobacter were isolated from soil samples collected from Gangnam-gu and Dongdaemun-gu in Seoul, in South Korea. The five Deinococcus species were Gram-stain positive, pink-pigmented, and short-rod or coccus shaped. The four Hymenobacter species were Gram-stain negative, red-pigmented, and short-rod shaped. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strains ES10-3-3-1, KKM10-2-2-1, Ant11, JM10-4-1-3, and KMS4-11 were most closely related to Deinococcus citri NCCP-154T (with 99.8% similarity), Deinococcus grandis DSM 12784T (99.0%), Deinococcus marmoris DSM 12784T (98.8%), Deinococcus claudionis PO-04-19-125T (98.7%), and Deinococcus radioresistens 8AT (99.8%), respectively. KKM10-2-7-2, JM10-2-5, JM10-2-6-2, and KKM10-2-3 were most closely related to Hymenobacter algoricola VUG-A23aT (99.1% similarity), Hymenobacter elongatus VUG-A112T (99.1% similarity), Hymenobacter gelipurpurascens Txg1T (99.1% similarity), and Hymenobacter psychrotolerans Tibet-IIU11T (99.3% similarity), respectively. These nine species have never been reported in Korea; thus, five Deinococcus species are reported in the family Deinococcaceae, order Deinococcales, class Deinococci, phylum Deinococcus-Thermus and four Hymenobacter species are reported in the family Cytophagaceae, order Cytophagales, class Cytophagia, phylum Bacteroidetes.

        Complete genome sequence of Deinococcus swuensis, a bacterium resistant to radiation toxicity

        Deinococcus swuensis DY59T is a Grampositive, coccus-shaped bacterium. Most members of the genus Deinococcus are able to grow in the presence of high levels of chronic radiation toxicity and desiccation because they can protect enzymes from reactive oxygen species generated during ionizing radiation. The mechanisms behind the resistance to radiation toxicity and the genomic features of resistance could be useful to exploit Deinococcus swuensis in the biotechnological applications such as detoxification of xenobiotic contaminated with radioactive wastes. Strain DY59T showed resistance to gamma radiation with a D10 value (i.e. the dose required to reduce the bacterial population by 10-fold) in excess of 5 kGy. However, the genus Deinococcus is slightly characterized at the genome level, despite its potential importance. Thus, the present study determined the features of Deinococcus swuensis DY59T, as well as its genome sequence and annotation. The genome comprised of 3,531,443 bp with a G+C content of 67.4%, which included 3,305 protein-coding genes and 58 RNA genes. Based on the genome annotation, the strain DY59T undergoes prokaryotic type nucleotide excision repair pathway, restores the damaged gene, and resists the ionizing radiation toxicity.

        Complete genome sequence of Lactobacillus koreensis 26-25, a ginsenoside converting bacterium, isolated from Korean kimchi

        A Gram-positive, rod-shaped, ivory colored, and motile, Lactobacillus koreensis 26-25 was isolated from Korean kimchi. Strain 26-25 showed the ability of conversion from major ginsenosides into minor ginsenosides for which whole genome was sequenced. The whole genome sequence of Lactobacillus koreensis 26-25 consisted of one circular chromosome comprised of 3,006,812 bp, with a DNA G + C content of 49.23%. The whole genome analysis of strain 26-25 showed many glycosides hydrolase genes, which may contribute to identify the genes responsible for transformation of major ginsenosides into minor ginsenosides for its high pharmacological effects. 김치로부터 분리한 Lactobacillus koreensis 26-25 균주의 유전체서열을 분석하였다. 균주 26-25의 유전체는 G + C 비율이 49.23%이며, 2,720개의 유전자와 2,556개의 단백질 코딩 유전자, 85개의 위유전자 그리고 78개의 RNA 유전자를 포함한 단일 원형 염색체로 구성되었으면 그 크기는 3,006,812 bp였다. 균주 26-25는 인삼사포닌의 당 분해에 관여하는 여러 타입의 글라이코시다제 유전자를 가지고 있었다. 이러한 지놈 분석은 주요 진세노사이드가 우수한 약리학적 활성의 미량 진세노사이드로 전환하는데 관여하는 유전자 특징을 이해하는데 큰 기여가 되었다.

        Isolation and characterization of four unrecorded wild yeasts from the soils of Republic of Korea in winter

        The purpose of this study was to isolate and identify wild yeasts from the soil collected in Gwangju and Pocheon City, Gyeonggi Province, Republic of Korea. Among 10 strains, six strains were already reported, but four strains were unrecorded in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were performed using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation tests were carried out using API 20C AUX kit. The 10 strains were assigned to the phyla Basidiomycota (8 strains) and Ascomycota (2strains). The unrecorded four yeast strains, NH33, NH19, NH20, and YP416, belong to the phylum Basidiomycota and the genera Buckleyzyma, Leucosporidium, Holtermanniales, and Mrakia, respectively. All strains had oval-shaped and polar budding cells. In this research, the morphological and biochemical properties of four unreported yeast species were characterized intensively, which were not officially reported in Korea

        A report of eight unrecorded radiation resistant bacterial species in Korea isolated in 2018

        Eight bacterial strains assigned to the phylum Firmicutes were isolated from the soil samples in Korea. Phylogenetic analysis based on 16S rRNA gene sequences showed that strains 18JY14-16, 18JY14-35, 18JY42-5, 18JY12-20, 18JY35-8, 18JY76-9, 18JY39-1 and 18JY54-12 were most closely related to Paenibacillus lupini (MH497638; 99.4%), Paenibacillus illinoisensis (MH497643; 99.8%), Paenibacillus tundrae (MH497658; 99.7%), Paenibacillus selenitireducens (MH497639; 99.4%), Paenibacillus eucommiae (MH497640; 99.9%), Paenibacillus vini (MH497654; 99.4%), Paenibacillus gorillae (MH497647; 99.5%), and Paenibacillus macquariensis (MH497649; 99.9%) respectively. These Paenibacillus species were Gramstain-positive, rod-shaped and radiation resistant bacteria. This is the first report of these nine bacterial species in Korea.

        A report of four unrecorded Proteobacteria species isolated from soil in Korea

        In 2015 and 2017, the National Institute of Biological Resources has isolated four unrecorded prokaryotic species designated as R-1-5, R-2-13, R-2-1, and R-1-8 from the peatland soil of Yongneup. Phylogenetic analysis based on 16S rRNA gene sequence similarity determined the four strains (R-1-5, R-2-13, R-2-1, R-1-8) were most closely related to Curvibacter lanceolatus (99.93%), Massilia brevitalea (98.7%), Pseudomonas lini (99.54%), and Pseudomonas vancouverensis (99.93%), respectively. The four unrecorded strains belong to the phylum Proteobacteria, in which the genera Curvibacter and Massilia are assigned to the class Betaproteobacteria, and the genus Pseudomonas to the class Gammaproteobacteria. Since there are no publications or official reports on these four strains, these four species are new records to Korea. The strains were further characterized by Gram reaction, colony and cell morphology, basic biochemical properties, and phylogenetic position. Descriptive information of the four unrecorded species is provided.

        Complete genome sequence of Paenibacillus swuensis DY6<sup>T</sup>, a bacterium isolated from gamma-ray irradiated soil

        박테리아 종들은 정교한 효소 시스템의 존재로 인해 이온화 방사선 처리 후에 생존할 수 있는 것으로 보고되어 왔고 몇몇 내생 포자를 생산하는 박테리아 또한 두꺼운 포자껍질의 존재 때문에 방사선에 저항할 수 있다. 이 연구에서는 방사선이 조사된 토양의 시료에서 추출된 Paenibacillus swuensis $DY6^T$의 완전한 게놈 서열을 분석하였다. 이 게놈은 G+C 함량이 49.93%인 5,012,599 bp으로 구성되어 있고 단백질 정보를 암호화한 유전자 4,463개와 133개 RNA 유전자를 포함하고 있다. Several bacterial species have been reported to be surviving after the ionizing radiation treatment due to the presence of sophisticated enzymes systems and some endospores producing bacterial strains can also resist, due to the presence of thick spore coat. In this study, we report the complete genome sequence of a bacterium Paenibacillus swuensis $DY6^T$, isolated from an irradiated soil sample. The genome comprised of 5,012,599 bp with the G+C content of 49.93%, the genome included 4,463 protein coding genes and 133 RNA genes.

        Complete genome sequence of Flavisolibacter tropicus LCS9<sup>T</sup>, a radiation resistant bacterium

        Flavisolibacter tropicus $LCS9^T$은 한국 중서부에 위치한 서천 국립생태원 에코리움 내 열대관 토양에서 분리되었다. 이 연구에서 G + C 함량이 41.5%인 5,940,863 bp 의 원형 염색체로 구성된 Flavisolibacter tropicus $LCS9^T$ 의 완전한 게놈서열을 분석하였다. 이 완전한 게놈서열은 5,075 개의 유전자, 337개의 위유전자 그리고 59 개의 rRNA 유전자를 포함하고 있다. 유전체 특징은 감마선 및 UVC 에 대응하는 주요 효소를 포함하였다. Flavisolibacter tropicus $LCS9^T$ was isolated from a soil sample collected from tropical zone within the Ecorium of the National Institute of Ecology in Seocheon, central-western Korea. In this study, we report the complete genome sequence of the bacterium Flavisolibacter tropicus $LCS9^T$, which possesses a circular chromosome comprised of 5, 940,863 bp with the G + C mol content of 41.5%. The genome sequence annotation showed that the complete genome includes 5,075 genes, 337 pseudogenes, and 59 rRNA genes. The radiation resistance genes such as excinuclease UvrABC complex and UvdE were present in the genome.
