Grown in the secondary broth of production media, the strain Streptomyces albulus has increased more the production of its metabolite ${\varepsilon}$-poly-L-lysine, one of poly(amino acid)s used as disinfecting food additives, than the strain in the primary culture of growth nutrients. Having the strain removed, the large concentrate obtained by ultrafiltrating the secondary culture broth. The concentrated production broth exchanged into followed by detecting in UV flowcell at 220nm the peptide bond of the components eluting the adsorbed proteins and polylysine with NaCl salt of gradient concentration, and has separated into five components. Among them the component in the fourth peak fraction has proved to be the pure ${\varepsilon}$-poly-L-lysine after the portion being hydrolyzed the fraction with HCl into amino acid followed by being the composing amino acid analysis.

Streptomyces albulus 세정균체를 이용하여 항세균물질인 폴리리신의 대량생산 가능성에 대하여 조사하였다. 발효기 배양에서 배양액 pH는 6.8에서 3.2로 감소하는 동안에 pH 4.0에서 4.5사이에서 폴리리신이 최대로 축적되므로 대수말기의 균체를 멸균증류수로 세척하여 마련한 세정균체를 아미노산 생산을 향상할수 있는 구연산을 2%첨가하고 pH를 최적인 4.2로 조정한 배지로 옮겨 배양하였다. 이러한 2단계공정으로 세정균체를 배양한 경우 배양액내의 폴리리신은 일반 1단계 단순 배양에 비하여 최대 6배의 축적량을 나타내었다. The feasibility of mass production of polylysine, an antibacterial substance, employing a washed mycelium of Streptomyces albulus has been investigated. Since the maximum accumulation of polylysine had appeared between 4.0 to 4.5 of pH during fermentor cultivation of which the pH of the broth has declined to 3.2 from 6.8, the strain growing in the broth on late log phase had been washed with sterilized water, then the sole mycelium had been transferred to another medium containing 2% citric acid having pH adjusted with NaOH to optimum of 4.2 to improve the production of the amino acid. The total accumulation of polylysine by washed mycelium of Streptomyces albulus from this two step procedure has exhibited 6 times more than the normal production of the simple one step fermentation.

연자성 세라믹 분말이 길항미생물의 생장과 저온 활성에 미치는 영향

The possibility of employment of ceramics, known to be emitting far infrared waves, to keep the growth and the viability of the antagonistic microorganisms was examined. Among four kinds of commercial ceramics, the ceramic powders composed of soft ferrite as a main component has exhibited to grow Pseudomonas cepacia and Saccharomyces cerevisiae better. The ceramic powder in the growth medium has increased the number of cells of P. cepada about ten times and that of S. cereuisiae two to five times more than that in the control at zrc. The viability of the microorganism at low temperature which was measured from the regrowth behavior at 27℃ after five days store at 4℃ has shown that the lag time of the two microorganisms reduced about three hours without any defect in the rate of logarithmic growth. These results demonstrated that the ceramic powders was available to the growth and viability at low temperature of antagonistic microorganisms.<br/> <br/> <br/>

송이 균사체(Tricholoma matsutake mycelium)의 화학영양성분 추구성

Tricholoma matsutake is a fungus symbiotically related with pine tree and it is a representative mushroom of basidiomycota formatting fruiting body in autumn. Tricholoma matsutake, a ectotrophic mycorrhiza of the root of pine tree, is a symbionts. This study measured chemotaxis of carbon source of Tricholoma matsutake mycelium to find host-finding ability of root of pine tree on mycelia growth. For this study, identification and isolation of Tricholoma matsutake mycelium was carried out. Mycelia supplied by KCTC and Forest Research Institute(FRI) and isolated in our laboratory were confirmed by the RAPD(Random Amplified Polymorphisms DNA) using TMF, TMR primers to equalize DNA band with tissue of Tricholoma matsutake basidiocarp collected in Yangyang. From the result of RAPD, all of the mycelia sequenced the same DNA band with basidiocarp to be the 370 base pair except two mycelia isolated in our laboratory. Among them T. matsutake FRI91009, the best grown mycelia, was used for this study. The experiment for each carbon source to measure chemotaxis of T. matsutake FRI91009 showed the best groth 18.2mm/30days and recognition of carbon source in starch. The growth of mycelia was excellent in glucose and in sorbitol, 15.3mm/30days and 12.4mm/30days, respectively.
