        Isoniazid와 Nitrofurantoin으로 유발되는 간 세포독성에 대한 식물추출물의 보호효과

        Isoniazid was discovered in 1950’s and since then it has been widely used as a synthetic bactericidal agent in the treatment of tuberculosis. However, the adverse effect of isoniazid has been reported to show significant hepatotoxicity in approximately 1-2% of patients. Nitrofurantoin {1-(5-nitro-2-furfurylideneamino)-hydantoin} is a synthetic nitrofuran that is commonly used for the treatment and prophylaxis of urinary tract infections, but its use is associated with liver cirrhosis and fatal liver necrosis. Therefore, studies for natural products with protective effect on the isoniazid- and/or nitrofurantoin-in duced hepatotoxcity would be valuable as the potential therapeutic use. 107 plants sources were collected at Mt. Baekdu, and extracte d with methanol. These extracts had been screened for the protective effects against isoniazid- and/or nitrofurantoin-induced cyt otoxicity in HepG2 cells at the both 100 and 300 µg/ml. Five methanolic extracts, Acanthopanax senticosus, Acer mono, Asparagus schoberioides, Fagopyrum tataricum, Potentilla centigrana, showed significant protective effects against isoniazidinduced hepatotoxicity. Two methanolic extracts, Acer mono and Leonurus artemisia, showed significant protective effects against nitrofurantoin-induced cytotoxicity in HepG2 cells.

        결명자의 타크린으로 유발한 간 세포독성 보호 성분

        is an acetylcholinesterase inhibitor that is approved for the treatment of Alzheimer’s disease. However,tacrine treatment for Alzheimer’s disease results in reversible hepatotoxicity in 30-50% of patients, which seriously limits itsclinical use. Accordingly, the identification of constituents in natural products that have protective efects on tacrine-inducedhepatotoxicity would be valuable. In the present study, an immortalized human hepatoma cell line, HepG2 was employed toscreen for agents that protect against tacrine-induced hepatotoxicity. The bioassay-guided fractionation of water extract of Cas-siae Semen furnished two anthraquinones, aurantio-obtusin (1) and obtusifolin (2). Compounds 1 and 2 showed hepato-protective effects with the protection ratio values of 55.3 +/ 0.5% and 41.2 +/ 0.8% at 160M, respectively.Key words Cassia obtusifolia L, aurantio-obtusin, obtusifolin, tacrine, hepatoprotection, HepG2

        타크린으로 유발한 간 세포 독성에 대한 백두산 식물 추출물의 보호 효과

        The present study represents the screening of the protective eftìεcts of herbal methanolic extracts from Baekdu mountain against tacrine-induced cytotoxicity in HepG2 cells. Tacrine is an acetylcholinesterase inhibitor, and used for the treatment of Alzhleimer’s disease. However, administration oftacrine for the treatment of Alzhleimer’s disease results in a reversible hepatotoxicity in 30 -50% of patients, consequently limiting clinical usε. Therefore, studies for natural products with protective effect on the tacrine-induced hepatotoxicity would be valuable as providing potential therapeutic use. 109 plant sources were collected in Baekdu mountain, and extracted with methanol. These extracts had been screened the protective effects against tacrine-induced cytotoxicity in HepG2 cells at the 100 and 300 μg/ml. Of these, ten methanolic extracts, roots of Ampelosis japonica, aerial parts of Berberis amurensis, aerial parts of Sedum aizoon, aerial parts of Lespedeza tomentosa, aerial parts of Lespedeza juncea, aerial parts of Hypenricum ascyron, stem barks of Syringa reticulata, fruits of Gleditsia japonica, aerial parts of Chamaenerion angustifolium, branches of Ginkgo biloba, showed significant protective effects against tacrine-induced cytotoxicity in HepG2 cells.

        도깨비바늘 꽃 성분의 항산화 및 간보호 효과

        Activity-guided fractionation of the EtOH extract of flowers of Bidens bipinnata L. have been furnished five flavonoids, sulfuretin(1), butein(2), 7,8,3',4'-tetrahydroxyflavanone(3), maritimetin(4) and okanin(5). All of the compounds showed significant activities on both linoleic acid peroxidation inhibition and DPPH radical scavenging effect. The evaluation for protective effect of isolated compounds against tacrine-induced cytotoxicity in human liver-derived Hep G2 cells was conducted. Compounds 1, 3, 4 and 5 showed significant protective effects with the EC50 values of 36.1±0.9, 23.3±0.7, 41.0±1.0 and 29.8±1.1 µM, respectively. Silybin, one of the well-known hepatoprotective agents, used as a positive control, and also showed protective effect with an EC50 value of 84.3±0.7 µM.

        산화적 스트레스에 대한 췌장 베타 세포 보호활성 식물추출물 탐색

        Diabetes mellitus is metabolic disorder characterized by hyperglycemia caused by insufficient insulin secretion or insulin receptor insensitivity to endogenous insulin. It is well-known that hyperglycemia is one of the main causes of oxidative stress in both type 1 and 2 diabetes. Oxidative stress is related by death of pancreatic β cell and dysfunction of β cell. Although β cell death or dysfunction is induced by many substances or molecules, increased evidences that oxidative stress plays a crucial role in β cell death or dysfunction. Considering the importance of oxidative stress in the pathogenesis of diabetes mellitus, we investigated the cytoprotective effects against hydrogen peroxide-induced oxidative stress in pancreatic β cell line RIN-m5F cell. 110 Plant sources were collected in Mt. Baek-du, and extracted with methanol. These extracts had been screened the protective effects against hydrogen peroxide-induced oxidative damage in RIN-m5F cells at 50 and 200 μg/ml. Of these, ten methanolic extracts, aerial part of Erigenron cannadensis, aerial part of Lespedeza juncea, whole plant of Alopecurus aequalis, fruit of Lycium chinense, leaf of Morus alba, rhizome of Polygonatum odoratum, root of Ampelosis japonica, whole plant of Ranunculus japonicus, aerial part of Polygonum sieboldii, rhizome of Arisaema amurense var. violaceum showed significant protective effects against hydrogen peroxide-induced oxidative damage in pancreatic β cell line RIN-m5F cell.
