      • 한약 탕제를 이용한 항 Herpes virus 제제의 개발 연구

        In order to search for anti-HSV agents from natural complex products, we extended the number of specimens. Both methanol extract and boiling water extract of the natural complex products were screened to detect anti-HSV activity by MTT assay. Anti-HSV activities of thirteen natural complex products extracted by methanol and boiling water were screened. Three of 13 natural complex products extracted by methanol showed efficacy against HSV. Natural complex products showing anti-HSV activities as methanol extracts were No.3, 6, 11 and their Sl were 323.809, 2811.041 and 708.20. As water boiling extracts, No.8 and No.11 have displayed Sl of 16.45 and 60.39 respectively. Especially anti-HSV activities of natural complex products extracted by methanol No.6 was stronger than other ones.

        소목 추출물의 세포독성 효과와 Topoisomerase I 억제 활성에 관한 연구

        To evaluate cytotoxic effect and topoisomerase I inhibition activity of Caesalpinia sappan L., both water and methanol extracts were examined using in vitro assay. The cytotoxic effect of Caesalpinia sappan L. examined using MTT and SRB assay and $IC_{50}$ values were measured against U937, HL60, HepG2, SNU-1, SNU-16 cancer cell lines. Among them the representative cytotoxic results are shown as follows; water extract (U937=13.39 ${\mu}g/ml$, HL60=8.65 ${\mu}g/ml$, HepG2=38.48 ${\mu}g/ml$, SNU-1=7.72 ${\mu}g/ml$, SNU-16=25.49 ${\mu}g/ml$), methanol extract (U937=13.35 ${\mu}g/ml$, HL60=9.43 ${\mu}g/ml$, HepG2=25.67 ${\mu}g/ml$, SNU-1=8.37 ${\mu}g/ml$, SNU-16=28.64 ${\mu}g/ml$). The inhibitory concentration of DNA topoisomerase I activity against water extract was 100 ${\mu}g/ml$ and the inhibitory concentration of DNA topoisomerase I against methanol extract was 400 ${\mu}g/ml$.

        온도변화에 기인한 토양미생물 우점종의 변화에 관한 연구

        Today, the weather is changing continually, due to the progress of global warming. As the weather changes, the habitats of different organisms will change as well. It cannot be predicted whether or not the weather will change with each passing day. In particular, the biological distribution of the areas climate change affects constitutes a major factor in determining the natural state of indigenous plants; additionally, plants are constantly exposed to rhizospheric microorganisms, which are bound to be sensitive to these changes. Interest has grown in the relationship between plants and rhizopheric microorganisms. As a result of this interest we elected to research and experiment further. We researched the dominant changes that occur between plants and rhizospheric organisms due to global warming. First, we used temperature as a variable. We employed four different temperatures and four different sites: room temperature (27℃), +2℃, +4℃, and +6℃. The four different sites we used were populated by the following species: Pinus deniflora, Pinus koraiensis, Quercus acutissima, and Alnus japonica. We counted colonies of these plants and divided them. Then, using 16S rRNA analysis we identified the microorganisms. In conclusion, we identified the following genera, which were as follows: 10 species of Bacillus, 2 Enterobacter species, 4 Pseudomonas species, 1 Arthrobacter species, 1 Chryseobacterium species, and 1 Rhodococcus species. Among these genera, the dominant species in Pinus deniflora was discovered in the same genus, but a different species dominated at 33℃. Additionally, that of Pinus koraiensis changed in both genus and species which changed into the Chryseobacrterium genus from the Bacilus genus at 33℃.

Additionally, those of Pinus koraiensis and Quercus acutissima changed in both genus and strains which changed into the Bacillus genus from the Paenibacillus genus at 33℃.

        Herpes simplex Virus Type-1 Thymidine Kinase 유전자의 크로닝

        Herpes simplex virus type-1의 vero 세포에서의 증식기작을 규명하기 위해 전자현미경으로 관찰하였고, 유전학적 특성을 규명하기 위해 유전자도서관을 작성하였고, thymidine kinase (TK) 유전자를 클로닝을 하였다. 감염 48시간 후 많은 수의 바이러스 nucleocapsid가 핵뿐만 아니라 세포질에서 관찰되었다. 바이러스는 세포에 감염된 후 핵내에서 복제증식한 후 세포질내로 이동하였으며, 이때 핵막을 통과하면서 외투막을 갖고 세포질로 이동하여 세포밖으로 나가는 것을 관찰할 수 있었으며, 또한 일부 nucleocapsid는 세포막을 출아하여 비리온으로 출아되었다. HSV-1의 DNA를 BamHI과 BglII 제한효소로 각각 절단하여 DNA의 절단 양상을 조사하였다. BamHI에 의해 절단된 단편은 27개 이었고, 그들 분자량의 범위는 1.1 - 14 kb이었으며, BglII에 의해 절단된 단편은 16개이었고, 분자량의 범위는 4.5 - 20.1 kb이었다. Southern blot 방법으로 TK 유전자를 포함하고 있는 단편을 확인하였는데, pHLA-12와 pHLB-14클론에 포함되어 있었고 각 단편의 분자량은 3.74와 6.41 kb이었다. Multiplication of Herpes simplex virus type-1 was observed by electronmicroscopy, a gene library of the genome was constructed and thymidine kinase gene was cloned. Vero cells infected with the virus were lysed 48 h p.j. and multinucleated giant cells were observed approximately at 72 h p.i. The nucleocapsids were observed in nuclei and cytoplasm, and the assembled nucleocapsids were budded out through the vacuole and cytoplasmic membranes, and then virions were released from the cells. HSV-1 genome DNA was digested with BamHI and BglII enzymes and then the gene library of the genome fragments were constructed. The BamHI cleaved the genome DNA into twenty-seven fragments in the range of 1.1 - 14 kb, and BglII cleaved the genome DNA into sixteen fragments in the range of $4.5{\sim}20.1\;kb$. The pHLA-12 and pHLB-4 recombinant plasmids were contained TK gene by Southern blot analysis. The molecular sizes of the fragments which contained the TK gene were 3.74 in pHLA-12 and 6.41kb in pHLB-4 recombinant plasmid, respectively.

      • Cyclophosphamide로 빈혈을 유도한 쥐에서 가미육미환의 조혈효과

        항암제 치료에서 비롯된 조혈조직의 일반적 기능저하를 경감시키는 항빈혈제의 개발이 요구되고 있다. 본 연구에서는 실험용쥐를 이용하여 가미유기미환의 조혈효과를 생체내 실험으로 시험하였다. 시클로포스파미드를 쥐의 복막내에 1회 주사하여 골수기능저하를 유도하였다. 가미육미환 추출액은 시클로포스파미드 주사후 8일 경과 시점에서 3일 동안 경구투여하였다. 체중, 장기중향, 생화학적 검사치, 혈액학적 검사치를 측정하엿다. 시클로포스파미드 처치군에서 적혈구수, 헤모글로빈, 적혈구용적율이 감소되는 빈혈현상이 관찰되었다. 가미육미환 처치군에서 적혁구, 헤모글로빈, 적혈구용적율이 대조군에 비하여 경미하게 상승하였으며 혈소판의 수준도 가미육미환 처치군에서 상승하였다. 이러한 결과는 빈혈을 유도한 쥐 모델에서 가치육미환 부분적으로 효과가 있다는 증거를 제시한다.

        한약단미제 및 탕제의 항 Herpes Simplex Virus Type-1 활성탐색

        향약집성방 및 동의보감등 한의학 서적을 기초로 하여 선택한 탕제 45종과 단미제 80종의 메탄올과 열수추출물을 가지고 MTT assay를 실시하여 항Herpes simplex virus-1 (HSV-1)에 대한 활성을 조사하였다. 열수추출 탕제 45종 중 백사전방, 현람방I, 현람방II, 반천청방 4종에서 항 HSV-1 활성을 나타냈고, 이들의 SI (selective index)값은 $2.1{\pm}0.5$에서 $11.8{\pm}2.2$ 범위의 값을 가졌으며 메탄올추출 탕제 45종 중에서는 단치시호탕III, 반천청방, 정향울금방, 대황오배자고, 홍인락삼등방, 호장해독탕에서 활성을 보였고 이들의 SI값은 $1.7{\pm}0.2$에서 $10.5{\pm}3.1$ 범위의 값을 나타냈다. 열수추출 단미제 중계지, 관중, 구인분, 대황, 자화지정, 포공영, 호장근, 황백등 8종에서 항 HSV-1 활성이 나타났고, 이들의 SI값은 $1.6{\pm}0.1$에서 $10.2{\pm}0.7$ 범위의 값을 가졌으며 메탄올추출 탕제 중에서는 계지, 목방기, 상지, 호장근에서 활성을 보였고 이들의 SI 값은 $2.9{\pm}1.5$에서 $9.3{\pm}0.5$ 범위의 값을 나타내었다. 처방제 중 반천청방은 열수추출물과 메탄올 추출물 모두에서 항바이러스 활성을 나타냈고, 단미제 중 계지와 호장근도 열수와 메탄올 추출물 모두에서 항바이러스 활성을 보였다. 이들 탕제 및 단미제는 분획 및 분석실험을 실시하여 활성성분을 추적하고 있다. In order to search for anti-Herpes simplex virus (HSV) type-1 agents from Korean medicinal herbs and Korean traditional prescriptions (herb complexes), we selected 80 medicinal herbs and 45 prescriptions, based on a review of the Korean traditional medicine books. Both methanol extracts and boiling-water extracts were tested by means of the MTT assay (tetrazolium based colorimetric assay). Ten of the 125 methanol extracts: CM-11, CM-18, CM-19, CM-21, CM-22, CM-39, MM-3, MM-18, MM-29, MM-73 (see explanation of nomenclature below), showed efficacy against HSV-1. Twelve of the water extracts: CW-2, CW-3-I, CW-3-II, CW-18, MW-3, MW-5 MW-6, MW-12, MW-47, MW-69, MW-73 and MW-79 were active. #3 (individual herb) and #73 (individual herb) were interesting because both water and methanol extracts were active. Especially, #3 is a part of composition of Hong-il-$laksamd{\check{u}}ngbang$ and Hojanghaedokt'ang which have anti-HSV-1 activitives. The SI value of MW-69 and CW-18 was relative high as $10.2{\pm}0.7$ and $11.8{\pm}2.2$. The cytotoxic effect on Vera cells of $Panch'{\check{o}}nch'onch'{\check{o}}ngbang$, Taraxacum platycarpum H. Dahlst. and acycloguanosine was determined by MTT assay. Water extracts of $Panch'{\check{o}}nch'onch'{\check{o}}ngbang$ (prescription) and Taraxacum platycarpum H. Dahlst. showed very weak cytotoxic effects on Vero cells at > $100\;{\mu}g/ml$ but acycloguanosine showed strang cytotoxic effects on Vera cells at > $100\;{\mu}g/ml$. As a result, #3, #73, MW-69 and CW-18 are considered as potentially useful for anti-HSV-1 agent and will be the focus of further research. Abbreviations: CM - methanol extracts of traditional prescriptions; CW - water extracts of traditional prescriptions; MM - methanol extracts of individual herbs; MW - water extracts of individual herbs.

      • 한약 탕제분획의 항 Herpes simplex virus 활성에 관한 연구

        Herpes simplex viruses(HSV) are one of the most common infectious virus of man. Though chemotherapies and antibiotics against HSV have been developed in many countries, but anti-HSV agents were not satisfactory to mankind by their toxic reaction and side effects. In order to search for anti-HSV agents from Korean traditional prescriptions, we extended the number of specimens. Both methanol extract and boilling water extract of the Korean traditional prescriptions were screened to detect anti-HSV activities by MTT assay. Korean traditional prescriptions showing anti-HSV activities as methanl extracts were Paekyopsan, Chesupwilyungtang, Yongdamsagantang, and prescription 11. Four methanol extracts showing anti-HSV activities were freationated by hexane and their efficacies were tested. Hexane freationations of Paekyopsan, Chesupwilyungtang, and prescription 11 showed in anti-HSV activities both haxane and methanol fractionation.

        한약처방제의 인체 위암 세포주에 대한 세포독성 효과에 관한 연구

        ln order to search for antigastric cancer agents from Korean traditional prescriptions. We selected 41 traditional prescriptions, based on a review of the Korean traditional medicine books. Both boiling water and methanol extracts were tested, by means of the Sulforhodamine B (SRB) protein assay. Six of the 41 water extracts; #3, #34, #35, #38, #40, #41 showed efficacy against gastric cancer cell (AGS: Human gastric carcinoma, ATCC HTB 103). #3 inhibited 50% cancer cell growth1 at the concentration of $152\;{\mu}g/ml$, #34, #35, #38, #40 and #41 inhibited 50% cancer cell growth at the concentration of $145\;{\mu}g/ml$, $129\;{\mu}g/ml$, $173\;{\mu}g/ml$, $10\;{\mu}g/ml$ and $19\;{\mu}g/ml$ respectively. Ten of the 41 methanol extracts; #1, #3, #32, #33, #35, #36, #37, #38, #41 were active. #1 inhibited 50% cancer cell growth at the concentration of $206\;{\mu}g/ml$, #3, #32, #33, #35, #36, #37, 738, #40, #41 inhibited 50% cancer cell growth at the concentration of $133\;{\mu}g/ml$, $159\;{\mu}g/ml$, $199\;{\mu}g/ml$, $147\;{\mu}g/ml$, $113\;{\mu}g/ml$, $187\;{\mu}g/ml$, $130\;{\mu}g/ml$, $9\;{\mu}g/ml$, $15\;{\mu}g/ml$ respectively. Prescription #3, #35, #38, #40, #41 were also interesting because both methanol and water extracts were active.
