Isolation, characterization, and genomic analysis of the novel T4-like bacteriophage ΦCJ20

Pathogenic Escherichia coli infections havebeen consistently reported annually. The basic characteristicsand genome of the newly isolated UCJ20 from swinefeces was analyzed. To determine basic characteristics,dotting assays and double-layer agar assays were conducted. Bacteriophage particles were analyzed via transmissionelectron microscopy. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis was performed todetermine the sizes of major structural proteins. Thecomplete genome of the phage was analyzed. Bacteriophageparticles were identified as Myoviridae, with a headmeasuring 110.57 ± 1.89 nm and a contractile tail measuring107.97 ± 3.20 nm and were found to infect E. coli. Major structural proteins of UCJ20 showed two well-pronouncedbands of approximately 53.6 and 70.9 kDa. Thegenome size of UCJ20 was 169,884 bp, and 118 of 307open reading frames were annotated. This study provides abaseline for the development of E. coli infection treatmentstrategies.
Effects of rumen-protected amino acid prototypes on rumen fermentation characteristics in vitro

This study was conducted to evaluate the effects of rumen-protected amino acid (RPAA) prototypes, which were chemically synthesized, on in vitro rumen fermentation and protection rate outcomes. Several RPAA prototypes were incubated with timothy hay and concentrate. Treatments consisted of 1) control (CON; no RPAA prototype supplement), and prototypes of 2) 0.5% RP-methionine (RPMet), 3) 0.5% RP-tryptophan (RPTrp), 4) 0.5% RPvaline (RPVal), 5) 0.5% RP-phenylalanine (RPPhe), 6) 0.5% RP-leucine (RPLeu), 7) 0.5% RPhistidine (RPHis), 8) 20% RPMet, and 9) 20% RPTrp (w·w-1 feed). The inoculum (50 mL) prepared with rumen fluid and McDougall’s buffer (1 : 4) was dispensed in individual serum bottles and was anaerobically incubated for 0, 6, and 24 h at 39℃ in triplicate. The dry matter degradability did not differ among the groups, except for the 20% RPMet and the 20% RPTrp treatments at 6 and 24 h. The total volatile fatty acid concentration in the 20% RPMet was higher (p < 0.05) than the rest of the groups at 6 h, and 20% RPMet showed the highest molar proportion of acetate, whereas the lowest proportion of propionate was found at 6 h (p < 0.05). The protection rate of the RPAA prototypes ranged from 29.85 to 109.21%. at 24 h. In conclusion, the chemically synthesized RPAA prototypes studied here had no detrimental effects on rumen fermentation parameters. Further studies using animal models are needed for more accurate evaluations of the effectiveness of RPAA.
멧누에 ( Bombyx mandarina ) 난간단백질 유전자의 유전적 분석

Two multigene families, high-cysteine A(HcA) and high-cysteine B(HcB), are located within 140 kb region of chorion locus in Bombyx mori. Each family consists of 15 members and one of each members are transcribed in pairs. We report isolation and characterization of HcA and HcB genes of the wild type silkmoth, Bombyx mandarina. In this study, we examined 6 repeating units out of 15 HcA genes and 6 units of 15 HcB genes of B. mori and B. mandarina, respectively, and found that the arrangements and expression units of these are dramatically different. In addition, we found that most of the sequence variations were located within the C-variable region. These results suggest that the likely mechanism of concerted evolution is gene conversion-like event in HcA/HcB gene pairs.
