      • 고삼(Sophora flavescens) 에틸아세테이트 추출물의 쥐 국소 뇌허혈-재관류 모델에서의 신경 보호 효과

        The aim of this study is to investigate the possible effect of EASF against cerebral ischemic injury using the middle cerebral artery occlusion (MCAO) model. Rats were randomly divided into two groups: MCAO group, and MCAO + EASF group. Neurological severity score tests (NSS) and infarct volumes were measured to determine the neuroprotective effects of EASF. Neuronal survival in striatum was examined by TUNEL staining. Caspase-3 like enzyme activity was measured by measuring the cleaved fluorescent products. Intraperitoneal injection of EASF reduced brain infact volume at 24h after MCAO. EASF decreased the score of neurological deficit and the TUNEL positive cells after MCAO. Furthermore, treatment with EASF declined the apoptosis related caspase-3 activity. These results suggest that intraperitoneal injection of EASF plays neuroprotective role through inhibition of apoptosis after MCAO in rats. EASF merits further investigation as a potential therapeutic agent in ischemic stroke patients.

        Effects of Psoralen from Psoralea corylifolia on Quinone Reductase, Ornithine Decarboxylase, and JB6 Cells Transformation Promotion

        The cancer chemopreventive effect of psoralen isolated from the seeds of Psoralea corylifolia was investigated in the induction of quinone reductase (QR) activity, intracellular detoxification enzyme, inhibition of 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced ornithine decarboxylase (ODC) activity, a key regulatory enzyme for polyamine metabolism, and tumor promotion in mouse epidermal JB6 cells, sensitive to tumor promoters (clone 415a P+ cells), which are related to suppress multistage carcinogenesis including initiation and promotion. Psoralen was isolated and identified from the ethyl acetate-soluble fraction of the methanolic extract from the seeds. Psoralen was active in induction of QR activity, the concentration of psoralen required to induce 1.5 fold QR activity was 14.8 μg/mL. Also, this pure compound inhibited TPA-induced ODC activity by 50% (designated IC_50) at the concentration 15.6 μg/mL and exhibited inhibition of TPA-induced tumor promotion in mouse epidermal JB6 cells with an IC_50 value of 17.1 μg/mL. Therefore, it is extrapolated that psoralen has the potential capable of inhibiting the initiation and/or promotion stage of carcinogenesis by induction of QR activity, inhibition of TPA-induced ODC activity and mouse epidermal JB6 cells tumor promotion.

      • Magnolialide의 면역글로블린-E 매개 I형 과민반응에 미치는 억제 효과

        The aim of this study was to investigate the inhibitory effects of magnolialide on basophil-mediated allergic reactions and interleukin (IL)-5-mediated B cell growth. Magnolialide was isolated from the leaves of Laurus nobilis L. (Lauraceae). The effects of magnolialide on mast cell stabilization were determined by measuring immunoglobulin (Ig) E-induced β-hexosaminidase activity in rat basophilic leukemia 2H3 (RBL-2H3) cells. Furthermore, IL-4 production, IL-5-induced Y16 pro-B cell proliferation, and the cytotoxic effects of magnolialide on RBL-2H3 cells were studied. Magnolialide inhibited the release of β-hexosaminidase by RBL-2H3 cells. The IC50 value of magnolialide (21.4 M) was lower than the ketotifen positive control (56.2 M). Additionally, magnolialide was able to inhibit IL-4 mRNA transcription and IL-4 production in IgE-induced RBL-2H3 cells with IC50 values of 17.8 μM and 15.2 μM, respectívely. Magnolíalíde also suppressed pro-B cell growth in IL-5-stimulated Y-16 cells in a dose-dependent manner. These results indicate that magnolialide inhibits IgE-sensitized RBL-2H3 cell de-granulation, IL-4 production, and IL-5 stimulated early during B-cell proliferation. These factors are all key parameters involved in the body’s response to a type 1 hypersensitivity reaction.

        사향함유 우황청심원액과 영묘향함유 우황청심원액의 혈압강하 작용 및 적출심장에 미치는 효과에 대한 약리효능 비교

        WooHwangChungSimWon is a traditional prescription formula for treating with hypertension, arteriosclerosis, coma, and stroke in China, Korea, and Japan. In the new prescription of WooHwangChungSimWon, the civet is substituted for the musk, the major component of WooHwangChungSimwon, because of the prohibition law (CITES) on the commercial use of musk. We have made a comparative study of the effects on the hypertension and contractile force of heart between the musk-containing and civet-containing WooHwangChungSimWon. The SHR rats were used for investigating the effects on the hypertension and the rabbits were used for investigating effects on the contractile force of isolated heart. The blood pressure which was recorded during the adminstration period showed that all the samples except the low dose of musk-containing WooHwangChungSimWon decreased the blood pressure, and the effects on the heart works of all the samples were higher than control group, which resulted from the examination of isolated heart. These results suggest that all the musk-containing and civet-containing WooHwangChungSimWon formula have similar protective effects on hypertension and palpitation.

        술류에 관한 연구(Ⅱ) : 한국산 창술과 백술의 사염화탄소 간독성에 대한 보간효과 비교

        Traditional herbal drugs of Korea, namely, Atractylodes Rhizome(蒼朮) and Atractylodes Rhizome White(白朮) which are derived from an old-grown rhizome and a newly-grown rhizome of Atractylodes japonica Koidz. (Compositae), respectively, were investigated their potential liver-protective activities against hepatoto xicity induced by CCl₄ intoxication in mice. Each herbal drug was extracted separately with chloroform, methanol and butanol. No extract of Atractylodes Rhizome showed a singnificant reduction of the duration of hypnosis produced by hexobarbital after CCl₄ intoxication in mice. In addition, the the treatments of Atractlyodes Rhizome extracts produced no decrease of aspartate aminotransferase(EC 2.6.1.1) activity in serum. However, the methanol extract of Atractylodes Rhizome White exhibited a marked protection from hepatotoxicity induced by CCl₄ intoxication in mice. It produced significant reductions of the duration of hypnosis and serum enzyme activity, but no other exract showed liver-protectivie activity against CCl₄-induced hepatotoxicity.

        Protection of Prenylated Flavonoids from Mori Cortex Radicis (Moraceae) against Nitric Oxide-induced Cell Death in Neuroblastoma SH-SY5Y Cells

        Seven prenylated flavanoids, licoflavone C (1), cyclomulberrin (2), neocyclomorusin (3), sanggenon I (4), morusin (5), kuwanon U (6) and kuwanon E (7), and three 2-arylbenzofurans, moracin P (8), moracin O (9), and mulberrofuran Q (10) were isolated from the MeOH extract of Mori Cortex Radicis. Among these, compounds 2-7 enhanced cell viability in a dose-dependent manner against sodium nitroprusside-induced cell death in neuroblastoma SH-SY5Y cells, which was measured by MTT reduction assay (EC50 values of 4.4, 5.6, 8.0, 6.4, 8.7, and 11.9 μg/mL, respectively). Among 10 compounds, C-3 prenylated flavones (2, 3, and 5) and prenylated flavanones (4, 6, and 7) showed cell protection. However, compound 1 which lacks the prenyl group at C-3 and three 2-arylbenzofurans (8-10) did not show protective effect. The order of cell protection was as follow: C-3 prenylated flavones (2, 3, and 5) > prenylated flavanones (4, 6, and 7) > 2-arylbenzofurans (8-10) and flavone (1). From this result, we show that some prenylated flavones and flavanones might protect neuronal cells against nitrosative stress-mediated cell death. Even though further evaluations are necessary in vitro and in vivo study, we carefully suggest that some prenylated flavonoids from Mori Cortex Radicis might protect neuronal cells from neurodegenerative diseases.

        Induction of Quinone Reductase Activity by Psoralidin Isolated from Psoralea corylifolia in Mouse Hepa 1c1c7 Cells

        Quinone reductase (QR) is a protective phase II enzyme against mutagens and carcinogens which is inducible by a number of chemical compounds in plants. This study was carried out to investigate effects of the fractions from the seeds of Psoralea corylifolia on the induction of QR with Hepa 1c1c7 murine hepatoma cell line. The ethyl acetate-soluble fraction of the methanolic extract from the seeds was found to induce QR and the concentration of 1.5 fold QR induction (1.5 FIC) was 1.2 μg/mL. We obtained as an active compound, psoralidin, isolated from the ethyl acetate-soluble fraction after further sequential fractionation with column chromatography and 1.5 FIC of psoralidin was 0.5 μg/mL. The seeds of Psoralea corylifolia and psoralidin might be a candidate for developing QR inducers.

        영묘향함유 우황청심원액과 사향함유 우황청심원액의 중추신경계에 대한 약리효과 비교시험

        WooHwangChungSimWon is a traditional medicine for treatment of hypertension, arteriosclerosis, coma, and stroke in China, Korea, and Japan. In the new prescription of WooHwangChungSimWon, the civet is substituted for the musk, the major component of WooHwangChungSimwon, because of the prohibition law about the musk. We have made a comparative study of the effects on the central nervous system between the musk containing and civet containing WooHwangChungSimWon. In order to investigate the effects on the central nervous system, we have examined spontaneous motor activities, anti-convulsion activities induced by chemicals or electric shock, sleep induced by hexobarbital, and antistress effects. In the examination of anti-convulsion effects against the electric shock and injection of pentetrazol, only the civet containing WooHwangChungSimWon showed the anti-convulsion effects, i.e. the duration of convulsion by electric shock was decreased (control: 104.0 seconds, low dose of civet containing WooHwangChungSimWon: 60.7 seconds) and the duration of suppression against pentetrazol induced convulsion was increased (control: 392.3 seconds, low dose of civet containing WooHwangChungSimWon: 574.0 seconds, high dose of civet containing WooHwangChungSimWon: 561.4 seconds). In the other examinations, all kinds of WooHwangChungSimWon showed sedation, anti-convulsion activities, and induction of sleeping. These results suggest that there are no significant differences between the musk containing and civet containing WooHwangChungSimWon except the anti-convulsion effects against the convulsions induced by electric shock and pentetrazol.

        Nitric oxide에 의해 산화적 스트레스를 받은 Neuronal cell에 항산화 효과를 가지는 수종 생약추출물의 검색

        The objective of this study is screening the anti-oxidative effects of several plant MeOH extracts against oxidative stress in Neuroblastoma cell. Oxidative stress has been implicated in the pathogenesis of many neurotoxicity, neurodegenerative disorders and cell death. This oxidative stress is generated by ROS (Reactive Oxygen Species) such as nitric oxide, nitrogen dioxide, peroxyl, superoxide (O2 - ), hydroxyl, alkoxyl. So, in the present study, we induced oxidative stress by treatment of sodium nitroprusside (2.5 mM) in human neuroblastoma SH-SY5Y cell which was treated samples before 24hr, and cell viability was measured by MTT reduction assay. Of those tested, the extracts of Paeonia japonica (roots), Eucommia ulmoides (炒)(barks), Paeonia japonica (曝乾)(roots), Phyllostachys bambusoides (stems), Polygala tenuifolia (去心, 炒)(roots), Paeonia japonica (roots), Polygala tenuifolia (roots), Machilus thunbergii (barks), Mallotus japonicus (leaves), Poria cocos (whole), Sophora flavescens (roots), Angelica tenuissima (roots), Angelica gigas (當歸尾)(roots) showed anti-oxidative effects[EC50<15.20 μg/ml(Carnosine:Positive control)]in dose dependent manner.
