Rice (Oryza sativa) is a major cereal crop that has been developed as a monocot model species. In past decades rice researchers have established valuable resources for functional genomics in rice, such as complete genome sequencing, high-density genetic maps, a full length cDNA database, genome-wide transcriptome data, and a large number of mutants. Of these, rice mutant lines are very important to definitively determine functions of genes associated with valuable agronomic traits. In this review we summarize the progress of functional genomics approaches in rice using T-DNA mutants. 주요 작물인 벼 (Oryza sativa)는 단자엽 모델식물이다. 현재까지 벼 연구자들은 전체 염기서열 분석을 완료하였으며, 고밀도 유전자 지도 작성, 전장 cDNA 유전자 은행구축, 게놈 규모의 유전자 발현 정보 및 대량의 돌연변이 집단을 육성하는 등 기능 유전체 연구를 위한 중요한 재료를 확립하였다. 특히 벼 돌연변이 라인은 유용 농업형질 유전자들의 기능을 최종적으로 규명하는데 매우 중요하게 사용되고 있다. 본 종설에서는 벼 T-DNA 돌연변이를 사용한 기능 유전체 연구에 대한 최근의 현황을 요약하고자 하였다.

The WRKY family proteins are transcription factors involved in the regulation of plant defense response pathways. In our present study, we characterized a rice WRKY gene, OsWRKY30 that had been previously isolated through systematic expression analysis of OsWRKYs during defense responses. The endogenous expression of OsWRKY30 was found to be abolished in salicylic acid (SA)-nonaccumulating NahG transgenic rice. OsWRKY30 activates transcription in maize protoplasts and its N-terminus retains transcription activation ability in yeast. OsWRKY30-GFP fusion proteins were found to localize in the nuclei of maize protoplasts. OsWRKY30 binds to the consensus W-box motif in yeast. Overexpression of OsWRKY30 in rice upregulates an SA-responsive gene OsWRKY45, and enhances disease resistance to the Xanthomonas oryzae pathovar oryzae. These results suggest that OsWRKY30 functions as a positive regulator of rice disease resistance via an SA signaling pathway.

The required for Mla12 resistance (RAR1) protein is essential for the plant immune response. In rice, a model monocot species, the function of Oryza sativa RAR1 (OsRAR1) has been little explored. In our current study, we characterized the response of a rice osrar1 T-DNA insertion mutant to infection by Magnaporthe oryzae, the causal agent of rice blast disease. osrar1 mutants displayed reduced resistance compared with wild type rice when inoculated with the normally virulent M. oryzae isolate PO6-6, indicating that OsRAR1 is required for an immune response to this pathogen. We also investigated the function of OsRAR1 in the resistance mechanism mediated by the immune receptor genes Pib and Pi5 that encode nucleotide binding-leucine rich repeat (NB-LRR) proteins. We inoculated progeny from Pib/osrar1 and Pi5/osrar1 heterozygous plants with the avirulent M. oryzae isolates, race 007 and PO6-6, respectively. We found that only Pib-mediated resistance was compromised by the osrar1 mutation and that the introduction of the OsRAR1 cDNA into Pib/osrar1 rescued Pib-mediated resistance. These results indicate that OsRAR1 is required for Pib-mediated resistance but not Pi5-mediated resistance to M. oryzae.
국내 축산물 중 헥사클로로부타디엔(HCBD) 오염도 조사

헥사클로로부타디엔(HCBD)은 할로겐화 지방족 불포화 탄화수소로, 2015년 스톡홀름 국제 협약에 의해 부속서 C(사용 및 유통금지)로 지정된 잔류성 유기오염물질(POPs)이다. 우리나라에서 HCBD는 식품 중 기준·규격 미관리 물질로 공정시험법은 없는 상황이므로, 국내유통 식품 중 오염실태조사 및 안전성 확보를 위해서는 별도의 시험법이 필요하다. 따라서 본 연구에서는 식품공전의 폴리브롬화비페닐 시험법과 환경부 공정시험기준 및 문헌 등을 토대로 추출 및 정제 등 전처리 방법과 기체크로마토그래프-질량분석기 (GC-MS)를 이용한 정성·정량 시험법을 확립하였다. HCBD의 검출한계 (LOD)는 63 pg/g, 정량한계(LOQ)는 200 pg/g 수준으로 각각 산출되었다. 확립된 시험법을 이용하여 국내 유통되고 있는 축산물(돼지고기, 소고기, 오리고기, 닭고기, 달걀, 메추리알)에 대한 오염도를 발표하고자 한다.
