        Brown guinea pig에서 백지 열수추출물의 미백효과

        The purpose of this study was to investigate the whitening effect of Angelicae dahuricae Radix water extract (ADWE) by its application on artificial tanning spots which were induced by 1,500 $mJ/cm^2$ of UVB radiation on the backs of brown guinea pigs weighing approximately 450~500 g. Thirty ${\mu}l$ of ADWE, at each application, were applied twice a day, 5 days a week, for 8 weeks to the guinea pigs. The artificial tanning spots were divided into 3 groups which were vehicle control group [propylene glycol: ethanol: water (5:3:2)], positive control group (2% hydroquinone) and experimental group (2% ADWE). The visible whitening effect and changes in melanin index were evaluated once a week. On completion of the experiment, the animals were sacrificed under anesthetization, and the artificial tanning spots were excised by biopsy punch and stained with Hematoxylin and Eosin (H&E) to observe histological change and also stained with Fontana-Masson's silver (F-M) and S-100 to observe melanin pigmentation and melanocytes. In the gross observation, the experimental group showed higher pigmentation than the positive control group but lower pigmentation than the vehicle control group. Histological observation confirmed that ADWE had a positive whitening effect by showing a lower distribution of melanin and melanocytes in the epidermis of experimental group than in the vehicle control group.

        백지 에탄올추출물의 항산화 활성에 관한 연구

        본 연구는 백지 에탄올추출물의 항산화 활성을 알아보기 위하여 총 포리페놀 함량, 총 플라보노이드 함량, 전자공여능을 독립적으로 3회 이상 반복 실시하여 측정 하였고, brown guinea pig(450∼500g)의 등 부위에 1500mJ/㎠ 광량의 자외선 조사로 유발시킨 피부(6areas per group)에 1일 2회, 주 5일, 매회 30㎕씩 총 5주간 시료를 도포한 후 염산 케타민으로 마취, 시료를 도포한 피부 부위를 직경12mm의 biopsy punch로 절취하여 10%의 중성 포르말린 용액 에 12시간 실온에서 고정한 후 Hematoxylin and eosin(H&E) 염색으로 표피 및 진 조직을 관찰하였다. 백지 에탄올추 출물의 총 폴리페놀 함량과 총 플라보노이드 함량을 측정한 결과, 각각 20.7㎎/g, 19.5mg/g으로 확인되어 항산화물질 함량이 양호하게 나타났다. 전자공여능을 측정한 결과, 백지 에탄올추출물은 농도가 증가함에 따라 전자공여능 또한 유의하게 증가하였고, 500㎍/㎖과 1000㎍/㎖농도에서 각각 14.8%, 19.8%의 전자공여능을 보여 양성대조군으로 사용한 Dibutylated hydroxytoluene(BHT)보다 공여능이 낮았으나 의미 있는 항산화 효과를 보였다. H&E 염색 결과, 대조군의 표피에서 경미한 비후도가 나타났으나, 백지 에탄올추출물 도포군은 피부조직 배열이 규칙적이며 염증관련 세포 침윤 등의 별다른 이상 징후는 발견되지 않았다. 이상의 결과를 종합하면 백지 에탄올추출물은 천연 항산화제로서 이용가 치가 있을 것으로 판단된다. In order to evaluate the anti-oxidative capabilities of Angelica dahurica Radix ethanol extract (ADEE), we analyzed the contents of polyphenol and flavonoid compounds and the electron donating ability from ADEE. For animal experimentation, the test agent was topically applied to the artificial tanning spots which were induced by 1,500 mJ/㎠ of ultraviolet B radiation on the backs of brown guinea pigs weighing approximately 450 ̴500g. The test agent of 30㎕ was applied (6areas per group) twice a day, five days a week, for five weeks. On completion of the experiment, the animals were sacrificed under anesthetization, and the artificial tanning spots were obtained by biopsy punch and stained with H&E to observe the histological change in the epidermis and dermis. As a result, the contents of polyphenol and flavonoid compounds in ADEE were 20.7㎎/g and 19.5mg/g respectively. As the for electron-donating capability of ADEE, it was observed that ADEE displays a dose-dependent antioxidative capacity of 14.8% and 19.8% at the concentration of 500 and 1,000 ㎍/ ㎖ respectively. Tissue staining with H&E revealed that the epidermis of the control group was slightly thicker than that of the other groups. However no inflammation or any other undesirable effect on the skin tissue due to ADEE was observed. These results indicate that ADEE is of value as a natural antioxidant.

        보문 : 브라운 기니피그와 melan-a 세포에서 백지에탄올 추출물의 미백 효과

        The whitening effect of Angelica dahurica Radix ethanol extract (ADEE) was investigated with in vitro and in vivo tests. For in vitro tests, ADEE was treated at concentrations of 6.25, 12.5, 25, and 50 μg/mL to melan-a cells in order toe valuate ADEE`s inhibitory effects on melanin synthesis and tyrosinase activity. Arbutin was used as positive control. For in vivo tests, the test agent was topically applied to hyperpigmented areas on the back skins of brown guinea-pigs (weight: 450~500 g). The hyperpigmented areas were generated by 1,500 mJ/cm2 of ultraviolet B (UVB) irradiation. The test agent of 30 μL was applied twice a day, five days a week, for five weeks. The animals were divided into 6 groups: normal (N), control (C), vehicle control (VC), positive control (PC, 2% hydroquinone), experimental 1 (E1, 1% ADEE) and experimental 2 (E2, 2% ADEE). ADEE effectively inhibited melanin synthesis and tyrosinase activity of melan-a cells in a dose-dependent manner. The pigmentation of brown guinea-pigs was clearly lightened in the PC, E1 and E2groups compared to the C and VC groups. At the fifth week, the melanin indices of the ADEE groups were significantly lower (p/0.00l) than that of the VC group. Visual and quantitative analyses revealed that the degree of melanin pigmen-tation and S-100 protein expression levels were significantly decreased (p/0.00l) in ADEE treated groups compared to control groups.

        백지 에탄올추출물의 미백효능 연구

        본 논문은 백지 에탄올추출물의 미백효능을 조사하기 위하여 melan-a cell, 브라운 기니피그 및 HMB-45 염 색을 사용하였다. melan-a 세포에 시료를 6.25, 12.5, 25, 50, ㎍/㎖ 농도로 처치하여 세포 생장율의 변화 관찰 및 멜라 닌세포의 형태학적 변화를 관찰하였다. 또한 brown guinea pig (450 ̃500 g)등 부위에 1500 mJ/㎠ 광량의 ultraviolet B 조사로 유발시킨 인공색소반 (ф 2 mm)에 1일 2회, 주 5일, 매회 30 ㎕씩 총 5주간 시료를 도포하여 주 1회 육안적 변화를 관찰 하였고, 실험 5주째 되는 날 실험동물은 염산 케타민으로 마취 후 시료를 도포한 인공색소반 부위를 biopsy punch로 절취하여 HMB-45 염색으로 멜라닌소체 내 glycoprotein(gp100) 단백질의 조직학적 변화를 관찰 하였 다. 세포생장율 측정에서 시료처치 6.25~50 ㎍/㎖에서의 세포생장율은 높았고, 멜라닌세포의 형태학적 변화에서는 시 료처치 농도가 증가할수록 멜라닌합성 및 수지상 돌기가 적게 관찰 되었다. 동물 실험 육안적 관찰에서 시료 도포군 이 용매대조군에 비해 멜라닌 색소침착 정도가 확연하게 밝아졌고, gp100 단백질의 조직학적 관찰에서 시료 도포군은 gp100 단백질 발현 정도가 현저하게 줄어들었으며, 영상분석에서도 시료 도포군이 용매대조군에 비해 유의하게 (p<0.001) 감소하였는데 이러한 결과는 현미경 관찰 결과와 일치하였다. 이상의 결과를 종합하면 백지 에탄올추출물은 우수한 미백효과가 있는 것으로 판단된다. To investigate whitening Effects of Angelica dahurica Radix Ethanol Extract (ADEE), we used melan-a cell line, brown guinea pig, and HMB-45. We treated with ADEE of 6.25, 12.5, 25, and 50 ㎍/㎖ concentration in order to evaluate the effect of ADEE on cell viability and on morphological observation of melan-a cells. Also we were induced the artificial tanning spots by 1,500 mJ/㎠ of ultraviolet B radiation on the backs of brown guinea pigs (approximately 450 ̴500g) and then the test agent of 30㎕ was applied on the spots twice a day, five days a week, for five weeks respectively. The visible whitening effect was evaluated once a week. At the end of the experiment, the animals were sacrificed under anesthetization. The artificial tanning spots were obtained by biopsy punch and stained with HMB-45 to observe the gp100 proteins which were melanosomes. Our results show that cell viability was not reduce at ADEE concentrations between 6.25 and 50 ㎍/㎖, melanin synthesis and melanocyte dendricity were decreased in ADEE treated melan-a cells increasing ADEE concentration. In the gross observation, ADEE treated groups had lower pigmentation than the vehicle control groups. And in the histological observation, ADEE treated groups had lower melanocytes than the vehicle control groups. Also in the quantitative analysis of the gp100 proteins using image analysis software, ADEE treated groups had a significantly lower value (p<0.00l) than the vehicle control group and this results agreed with the results of observation under microscope. From these results, we concluded that ADEE had positive whitening effect.

        라벤더 오일을 이용한 손 마사지가 입원노인의 스트레스 및 우울 완화에 미치는 효과

        This study was carried out to evaluate the effects of aroma hand massages using lavender oil on the alleviation of stress and depression in hospitalized elderly patients. Results show that the stress scale scores of the experimental group (17 people receiving aroma hand massages using lavender oil) were significantly improved (p<0.05) compared to the control group (15 people receiving hand massages using jojoba oil). Overall, when comparing stress and depression levels before and after treatments, results show that the experimental group had significant improvements in: i) subjective measures of stress (stress scale score (p<0.001)) and depression (p<0.01), and ii) objective measures, including saliva cortisol concentrations (p<0.01). Therefore, the aroma hand massages using lavender oil can be effective in relieving stress and depression for hospitalized elderly patients. Interestingly, when comparing before and after measures of stress and depression in the control group, we observed a significant improvement in subjective, but not objective measures of stress (p<0.001) and depression (p<0.05). Therefore, general hand massage (ie, without the use of lavender oil) may also afford a more limited ability to relieve depression and stress for hospitalized elderly patients.

        Hairless 마우스에서 제라늄 오일의 피부노화 억제 효과(1) -육안적 관찰을 중심으로-

        To evaluate the skin aging inhibition effect of geranium oil, skin aging animal model was produced by both the irradiation of UVB and the application of squalene-OOH to SHR-1 hairless mice for 4 weeks. And at the same time 3% of geranium oil diluted in jojoba oil was applied topically. The skin erythema index for the positive control group and geranium oil application group were lower than that of the control. Whereas, both the lipid capacity and water capacity for the positive control group and geranium oil application group were higher than those of the control. Wrinkles for the control group were formed as a pattern of deep furrows and thick crests. Whereas, wrinkles for the positive control group and experimental group were formed as a pattern of shallow furrows and thin crests which were similar to that of the normal group. As for the both absolute and relative weight of the spleen, the positive control group were significantly higher than the other groups. In conclusion, geranium oil can be used practically for the prevention or improvement of skin aging in terms of health promotion and beauty for the people.

        The purpose of this study was to investigate the effects of frankincense oil in skin aging animal model. Skin aging was induced by both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) to the back of experimental animals for 4 weeks. And at the same time experimental materials were applied topically. Six to seven weeks female SHR-1 hairless mice were divided into five groups including nor-mal (N: saline), control (C: UVB+Sq-OOH+saline), vehicle control (VC: UVB+Sq-OOH+jojoba oil), positive control (PC: UVB+Sq-OOH+0.01% retinoic acid) and experimental (E: UVB+Sq-OOH+3% Frankincense oil) groups, five animals each group. Lipid lamella and lipid content in stratum corneum of the E group were almost intact with a regular arrangement which were similar to the N group. Collagen fibers in dermis of the E group were almost intact with a regular arrangement which were similar to the N group. Relatively much less number of mast cells and inflammatory cells were found in the E group compared to the C group. The activities of XO, SOD and CAT were no significant difference between the E and N groups. In conclusion, the application of frankincense oil to the skin aging animal model reduced both the generation of free radicals and the damage of skin tissues. Therefore, frankincense oil can be used practically for the prevention or improvement of skin aging in terms of health promotion and beauty for the people.
