This study was performed to examine the effect of 2,3',4,4',5-pentachlorobiphenyl (PCB#118) on testis of male rats. PCB#118 (20 mg/kg/week) in corn oil was intraperitoneally injected to adult male rats for 2, 5, 8 weeks. The body and testicular weights were measured at 3, 6, 9 weeks of PCB treatment. The morphological changes in the rat testes were then analyzed by light microscopy (LM) and transmission electron microscopy (TEM). The results showed that PCB#118 caused significant change in the body weights and testicular weights. Moreover, the morphological studies that were conducted on the PCB-treated rats revealed that the number of spermatocytes and spermatids in their seminiferous tubules decreased than control group (LM). The nuclear membrane was damaged when PCB was administered to them for 9 weeks (TEM). These results suggest that the reproductive function of the adult male rats is sensitive to PCB#118, and that may affect the testicular morphology of adult male rats.
3'-methyl-4-diethylaminoazobenzene으로 유발된 랫트 hepatocellula carcinoma 모델에서 항암제의 항암효과에 대한 평가기법 개발

This study was carried out for investigating antitumor effects of 5-fluorouracil(5-FU), methotrexate(MTX) and retinoic acid(RA) on hepatocellular carcinoma induced in Sprague-Dawley rat. Antitumor effects were examined a flow cytometric DNA distributions by flow cytometry and stuied ATP/Pi using nuclear magnetic resorance, and the enzymatic activity of thymidylate synthetase and dihydrofolate reductase as well as contents of total collagen and sialic acid were measured with spectrophotometer. In this study, S phase fraction, contents of sialic acid and total collagen were decreased in the induced hepatocellular carcinoma treated with 5-FU and MTX, and synergistic effects of anticancer drugs were exhibited in the hepatocellular carcinoma treated with 5-FU and MTX simultaneously, and the inhibition of thymidylate synthetic and dihydrofolate reductase activity were shown in the hepatocellular carcinoma treated with 5-FU, MTX, and 5-FU and MTX simultaneously. On the other hand, the ratio of ATP/Pi were increased in all groups except group treated with RA. The experimental results suggest that above method may be valuable for evaluating antitumor effect of anticancer drugs.

A simple and rapid analytical method for the determination of tylosin in chicken, pork and muscle was established by High-Performance Liquid Chromatography(HPLC). Chicken, pork and beef muscle(5 g) were fortified by adding the $0.2{\mu}g/ml$ of standard tylosin and the drug was extracted from meats with 70% acetonitrile(ACN) and followed by liquid-liquid partition for clean-up procedure. Then $20{\mu}l$ portion of ACN elution was directly analyzed by HPLC with spectra 100 variable wavelength detector, and unfortified blank control were treated similarly. The average recovery rate of tylosin added to chicken, pork and beef muscle were $83{\pm}2.3$, $96{\pm}3.3$ and $92{\pm}1.6$(%) at the level 0.2 ppm, respectively. No tylosin residues in marketing meats. These results suggested that HPLC methodology could be acceptable for the extraction, determination and screening of tylosin residues in edible meats.

신령버섯균사체(Agaricus blazei mycelia: ABM)를 대두박이 함유된 액체배지에 배양하고, 이것을 자가분해(53 ℃, pH 5.5, 120 rpm, 3 hr)하여 항암성이 강한 isoflavone-conjugated glycoprotein (Gluvone 이라 명명)을 분리하였다. Gluvone 은 지금까지 알려진 당단백질과는 달리 분자량이 작고(9,400 Da), isoflavone이 결합되어 있다는 점이 다르다, Gluvone 은 60% 탄수화물(glucose, fructose, ribose), 31% 단백질 및 2% isoflavone (daidzein, genistein)으로 구성되어 있었다. 이 Gluvone은 S-180 복수암세포, MCF-7 인체유선암세포에 대한 독성이 강하였고, S-180 세포로 유발한 mouse 복수암을 강하게 억제하였다. Most beta-glucans obtained from various fruit bodies of mushrooms and mushroom mycelial cultures have high-molecular weight glycoproteins, conjugated with beta-glucans. We report that isoflavone- conjugated glycolproteins (designated as gluvone) were isolated and exhibited stronger anticarcinogenic activities. Agaricus blazei mycelia (ABM) was cultured in a liquid medium containing soybean flakes for 14 days. The liquid culture was autolyzed by incubating at 53℃ (pH 5.5) for 3 h. A crude glycoprotein (CGP) fraction with a cytotoxic effect on a mouse ascite cancer cell line (S-180) and a human breast cancer cell line (MCF-7) was isolated from the autolyzed ABM cultures by 80% ethanol treatment. Gluvone was isolated from the CGP with Sephadex G-75 column chromatography. It exhibited a stronger anticancer effect than CGP against the S-180 cell-induced female ICR mouse ascites carcinogenesis. Gluvone with 9,400 daltons was identified as a glycoprotein conjugated with isoflavone. According to HPLC and GC analysis, in conjunction with ¹H-NMR spectral analysis, it contained 60% carbohydrates (glucose, fructose, and ribose), 31% protein, and 2% isoflavone (daidzein and genistein), which is a novel material. These results indicate that a strong anticarcinogenic gluvone was isolated from the autolyzed product of a submerged liquid culture of ABM, suggesting that autolysis could be a useful tool to produce antitumor agents.
Salmonella 방제에 대한 한방사료첨가제의 야외적용 효과

The present study was conducted to investigate the preventive effect of feed supplemented with 1.0% oriental herbal medicine feed additives (OHMFA) on the colonization of Salmonella spp. and incidence of death in broiler chickens. The frequency of Salmonella spp. in feces samples treated with OHMFA (25/239; 10.5%) was significantly reduced (p<0.05) than that of control group (83/347; 23.9%). A total of 108 Salmonella spp, belonging to four different serotypes, were isolated from three broiler farms. Among the serotypes isolated, Salmonella typhimurium was the moot prevalent (60.2%), followed by S. enteritidis (20.4%), S. gallinarum (13.0%) and S. pullorum (6.4%). In incidence of death in chickens during experiment, the mortality of rate OHMFA group (3.9%) was significantly lower (p<0.01) than that of control group (5.7%). These results show that the administration of OHMFA may prevent the colonization of Salmonella in chickens.

In this study, antibacterial activity on the chicken feed supplemented with different concentrations of oriental herbal medicine feed additives (OHMFA) extract was tested for some organisms and their preventive effects on the colonization of Salmonella enteritidis (SE) in broiler chickens were examined. The growth of Bacillus cereus, listeria monocytogenes and Staphylococcus aureus was inhibited on the feed of 0.25% OHMFA and Salmonella spp. and Escherichia coli O157:H7 were inhibited on the feed containing 2.0% level. The broiler chickens fed a forage additived 1.0% level of OHMFA after inoculation of a $10^7CFU/ml$ of SE were increased in weight than the chicken fed forage without OHMFA. The incidence of SE in liver, spleen and cecum of the chickens fed forage with OHMFA was decreased on the 7th days of postinfection and SE was not isolated from the organs of the chickens after the 14th days of postinfection. Serum antibody titers of the chickens were lowered than the control group.

This study was designed to evaluate the possible DNA damaging effects of T-2 toxin using an alkaline single cell gel electrophoresis (SCGE) comet assay and also to investigate toxic effects in chickens. A total of 20 chickens were used in these experiments. Graded concentrations of dietary T-2 toxin (0, 4, 8, and 16 ㎍/g of diet) were given to groups of 5 broiler chickens. In comet assay, The DNA damage was analysed by the tail extent moment (TEM) and tail length (TL), which were used as markers of DNA strand breaks in SCGE. A significant dose-dependent increase in the extent of DNA migration as well as in the percentage of cells with tails was observed after treatment with T-2 toxin (P<0.05). Treatment with the low T-2 toxin (4 μ/g of diet) induced a relatively low level of DNA damage in comparison with the high T-2 toxin (16 μ/g of diet) group. The growth rate was significantly reduced by concentrations of 8, and 16 μ/g of diet (P < 0.05). The feed conversion ratio were significantly affected by any concentrations (P < 0.05). The relative weight of the spleen, and lung was decreased by the growth inhibitory concentrations. The bursa of Fabricius, thymus, and kidney were decreased in relative weight by concentrations of 16 μ/g of diet. The relative weight of the liver and heart were unaffected. The hemoglobin (Hb), hematocrit (HCT), and mean corpuscular hemoglobin (MCH) were decreased at concentration of 16 μ/g of diet. As compared with control chickens, there was no marked change in serum components except uric acid in T-2 treated chickens. All lymphoid tissues retained atrophic and lymphoid cell depletion throughout the three weeks trial.
마우스에서 diazinon, toxaphene 과 endrin 단독 혹은 그 혼합물 독성의 대사

The effects of mixtures of diazinon(DA;5mg/kg), toxaphene(TOX;40mg/kg) and/or endrin(END; 5mg/kg) on the hepatic mixed-function oxygenase(MFO) system were stuided in ICR mice(18~22g) by oral intubation daily for 7 days. In general, TOX and TOX-containing mixtures were found to induced the metabolism of aminopyrine(22~60%), aniline(42~85%), phenacetin(145~194%) and benzo [a]pyrene(158~210%), and pentobaribtal biotransformation in the 9,000g liver supernatants and to increased the hepatic cytochrome p-450 contents(47~89%). Results of these may be, at least in part, associated with the MFO system. TOX pretreatment increased the aliesterase activity in the serum and liver homogenates and supernatants by 23~145%. The toxicity of TOX and TOX-containing mixtures would be lower than that of diazinon because of TOX-induced increase in the metabolism of diazinon(DA) or diazioxon(DO) and capability of TOX to stimulate the metabolism of diazinon and diazioxon and provide a pool of non-critical enzymes. These results suggest that this information might be helpful in the evaluation of the potential hazard due to occupational and/or environmental exposures to pesticides and their mixtures.
