豆芽組織의 ADP-ribosyl Transferase에 關한 硏究

ADP-ribosyl transferase, which catalyzes the transfer of ADP-ribose moiety of NAD to acceptor portein, has been reported only in bacteria nad animal tissues but not in plant tissues. The enzyme activity in soybean hypocotyl was demonstrated only in nuclear fraction, showing similar distribution with poly (ADP-ribose) synthetase of animal tissues. The growth curve of soybean hypocotyl showed the ideal sigmoid shape with maximal growth rate between 4 and 6 days. And the maximal activity of this enzyme was demonstrated at about 2cm fraction from the distal tip of soybean hpocotyl, where the growth and differentiation were most active. The enzyme activities in these fractions showed gradual increase with development and reached the maximum at 6th day. ADP-ribosyl transferase of soybean hypocotyl was very unstable, exhibiting only 30% of initial activity after 4 days storage at 4℃, but stabilized by the addition of MgCl₂. Inhibitors of poly (ADP-ribose) synthetase such as nicotinamide and benzamide showed little inhibitory effect on the enzyme activity, comparing with a profound inhibition of LaCl₃ and hgCl₂.
Cholesterol 代謝에 미치는 Lanthanum Chloride의 影響

Agents ingibiting calciru deposition into arteries are known to suppress atherosclerosis in animals. In this study, a calcium antagonist, lanthanum chloride was given in drinking water at a concentration of 50mg% to rabbits feeding cholesterol-enriched diet and its effect on the metabolism of cholesterol and triglyceride was observed. 1. Rabbits fed 2% cholesterol diet with and without lanthanum chloride for 10 weeks showed the marked increases in serum cholesterol level of 1,500 and 1,533mh% respectively. 2. Atherogenic diet resulted in the significant accumulation of cholesterol and triglyceride in aorta, liver and myocardium, but their accumulation in aorta and liver was prevented by lanthanum chloride treatment. 3. Electrophoretic pattern of serum lipoproteins showed the increased LDL/HDL ratio by cholesterol feeding, while no difference was observed by lanthanum chloride treatment.
Streptomyces alboniger 가 생산하는 Protease 의 특성에 관한 연구

Some physico-chemical properties of proteolytic enzyme produced from Streptomyces alboniger Hesseltine were characterized. The optimal pH and temperature of the enzyme; were around pH 9.0 and 40℃, respectively. The enzyme was stable between pH 6.0∼10.8 and the enzyme activity was not inactivated by heat treatment in lower temperature than 40℃. However the enzyme activity decreased by 70% of the initial activity for 10minutes at 70℃. The Km value was 7.1μM with Hammarsten casein and 333.3μM with cytochrom C. The activity of enzyme was inhibited by metal ions in the order of Fe^(+++)$gt;Hg^(++)$gt;Cu^(++$gt;Pb^(++)$gt;Zn^(++), whereas Ca++ increased the enzyme activity. There was no effect of Mg^(++) and Co+t on the enzyme activity. The enzyme was inhibited by EDTA strongly. When Ca^(++) was added to the EDTA-denaturated enzyme, the activity of enzyme was restored.
Vibrio vulnificus가 분비하는 Cytolysin에 관한 연구

The extracellular cytolysin produced by Vibrio vulnificus isolated from a patient with primary sepsis was partially purified and its properties were studied. 1. The cytolysin production in culture supernatant reached to the maximum of 800HU/ml at 5 hours' culture and was reduced by the addition of NaCl in culture media. 2. The cytolysin was heat-labile and completely inactivated when stored at 4℃ for 24 hours but the stability was greatly enhanced by the addition of glycerol(50%) or bovine serum albumin(mg/ml). 3. The cytolysin in culture supernatant was partially purified 40 folds to the specific activity of 32570HU/mg protein by ammonium sulfate precipitation, calcium-phosphate gel adsorption and DEAE-cellulose chromatography. 4. The cytolysin was lethal for mouse(18HU/mouse), but pretreatment with verapamil or Tavegyl 1 hour before the injection of cytolysin significantly prevented the lethal effect of cytolysin.
