        HPLC-ELSD를 이용한 발효유 제품 중의 Isomaltooligosaccharides 분석법 개발

        본 연구를 통해 현행 이소말토올리고당 분석법의 한계를 극복하고 원료 시럽뿐만 아니라 발효유 중의 미량의 이소말토올리고당 함량을 신속, 정확하게 분석하기 위한 새로운 분석법을 개발하였다. IDF method와 dSPE 기술을 적용하여 전처리 방법을 개선하였고, 당 전용 컬럼과 ELSD를 이용하여 기기분석조건을 최적화하였다. 새롭게 개발된 분석법은 유효성 검증 절차에 의해 선택성, 직선성, 검출한계 및 정량한계, 회수율, 정확성 및 정밀성이 유효함을 확인하였다. 또한 시장에서 유통 중인 시럽 및 발효유 제품을 분석한 결과 이전에 발표된 연구결과와 일치하는 결론을 얻었으며(Goffin et al., 2011), 이소말토올리고당을 구성하는 성분 중 panose, isomaltose 및 isomaltotriose가 가장 많은 비율을 차지하는 것을 확인하였다. 본 연구 결과는 지방 및 단백질이 많고 유화의 특성을 가진 발효유 중에서의 이소말토올리고당 함량을 신속, 정확하게 분석할 수 있는 기술이 될 것으로 기대된다. 이러한 분석 기술은 향후 식품산업현장에는 물론 발효유를 소비하는 소비자들에게 발효유의 기능성에 대한 정확한 평가를 가능하게 하고, 이소말토올리고당 뿐만 아니라 당 분석을 위한 기초 연구자료로 활용될 것으로 기대된다. A rapid and simple analytical method for the determination of 9 isomaltooligosaccharides (IMO) species in yoghurts was developed using dispersive solid phase extraction (dSPE) clean-up technic and high performance liquid chromatography with evaporative light-scattering detector (HPLC-ELSD). In this study, 9 IMO were extracted from samples simply with chemical reagent using ISO22662 IDF198 method and additional dSPE clean-up. The optimum instrument conditions for the determination were used carbohydrate ES $5{\mu}$ column with gradient elution of water and acetonitrile and ELS detector. The linearity of this method was expressed as the correlation coefficient ($r^2$), the results of IMO 9 species were shown in 0.9999. LOD and LOQ were respectively 7.9-22.1 mg/kg, 25.9-72.8 mg/kg. The accuracy of intra- and inter-day measurements were in the range from $84.3{\pm}4.5$ to $104.9{\pm}6.5%$, and the preceision of the intra- and inter-day measurements were in the range from 0.8 to 7.7%. The recoveries were from $84.3{\pm}4.5$ to $104.9{\pm}6.5%$. The determination results of IMO 9 species for the 9 yoghurts circulated in the market were in the range from $0.317{\pm}0.007$ to $1.624{\pm}0.050$ g/100 g. The newly developed method is appropriate for the determination of IMO in yoghurts, is a rapid and simple method with excellent resolution in compared with previous method.

        이온선택성 전극법을 이용한 조제분유중의 염소 및 요오드 이온함량 분석방법 연구

        This study was carried out to develop an accurate and rapid analysis method to measure the contents of iodide and chloride in infant formula using an ion selectivity electrode. The infant formula SRM 1846 (Standard Reference Material 1846, NIST, USA) was used as a CRM (Certified reference material). Samples were dissolved in water with 3% acetic acid and filtered through filter paper and adjusted to pH 7.0 with 0.1N NaOH. At pH 7.0, the iodide content of the CRM was $1.04{\pm}0.03\;mg/kg$ compared to the specification of $1.11{\pm}0.1\;mg/kg$ of CRM, and the recovery was $93.7{\pm}2.7%$. The chloride content was $5114.3{\pm}240\;mg/kg$ compared to the specification of $4,920{\pm}300mg/kg$ of CRM, and the recovery was $103.9{\pm}4.9%$. Ion selectivity electrodes could be successful1y used to determine the concentration of iodide and chloride ions in infant formula by a simple and rapid pretreatment of sample matrixes.

        Development of Vitamin D Determination in Infant Formula by Column-Switching HPLC with UV Detector

        This study was carried out to develop an analytical method for the determination of vitamin D in infant formula. Vitamin D was determined by column-switching high-performance liquid chromatography (HPLC) equipped with a reversed phase column and UV detector after saponification and extraction of the formula with an organic solvent. A preseparation column (C8), focusing column (C18), analytical column (C18) and UV-Vis detector (254 nm) were used. The limits of detection (LOD) and the limits of quantification (LOQ) for vitamin D were estimated to be 1.51 μg/kg and 4.95μg/kg, respectively. The linearity, recovery, precision and accuracy of the analytical method for vitamin D were evaluated through the application of a SRM (Standard Reference Material) 1846 (National Institute of Standard &Technology, USA). The linearity of this method was calculated with a value of the coefficient of determination (r2)≥0.9999. The recovery of vitamin D was 85.20±3.00%. The intra-assay precision for vitamin D was between 1.68±0.03%and 5.75±0.33%, and the inter-assay precision for vitamin D ranged from 1.73±0.03% to 2.96±0.09%. The intra-assay accuracy for vitamin D was between 100.03±2.77% and 102.01±0.59%, and the inter-assay accuracy for vitamin D ranged from 99.00±1.53% to 102.01±3.04%. The proposed method is optimal for the separation and quantification of vitamin D from infant formula.

        This paper presents design of meter for estimation Committed Information Rate(CIR) in Differentiated Services(DiffServ) networks. The DiffServ is a target model rather than a specification that contains detailed information about the required implementation. DiffServ provides a moderate level of quality differentiation without strict guarantees[1]. A DiffServ router consists of different components including classifier, meter, marker, dropper, shaper and scheduler. In this paper, we use the benefits of the fuzzy logic controller to design a fuzzy based traffic conditioner for DiffServ[2]. Simulations show that the approach is efficient and promising.

      • 스키에 의한 슬관절 인대 손상

        목적: 스키손상에서슬관절인대손상의형태와영향을줄수있는인자를분석하고, 수상전후의슬관절기능평가및스포츠활동도변화를알고자하였다. 대상및방법: 슬관절인대손상환자30명을대상으로설문및이학적검사, X선및MRI 촬영을통한평가를하였고, 기능평가는Lysholm Knee Scoring Scale, 스포츠활동도는Tegner activity score로평가하였다. 결과: 내측측부인대손상을동반한전방십자인대손상이11명으로가장많았고, 내측측부인대손상이9명으로그다음이었다. 수상기전은유령발현상(phantom foot phenomenon)과외전-외회전기전이각각13례와12례로83 $\%$를차지했다. 최종추시에서평균Lysholm score는93.4점으로매우양호하였으며, 활동도는수상전Tegner activity score 6.1점에서수상후4.2점으로감소되었다. 기능과활동도평가에서내측측부인대단독손상군이경과가가장양호하였으며, 내측측부인대손상을동반한전방십자인대손상군이경과가불량하였다. 스키손상에영향을미칠수있는요인으로는스키를지속적으로2시간이상탄경우, 바인딩이풀리지않는경우, 적절한스키강습프로그램을받지않은경우였다. 결론: 환자들의수상후Lysholm score는매우양호했지만, 수상후스포츠활동도에서는수상전과뚜렷한차이를보여적극적이고체계적인스포츠복귀프로그램이필요함을알수있었다. Purpose: This study is to analyse the injury patterns of knee ligament and the factors influencing ligament injuries of knee, and to evaluate the changes of knee function and activity after ski injury. Materials and Methods: Thirty cases of ligament injuries of knee were studied with a questionaire, stress radiographs, magnetic resolution imaging, and physical examination. Mean age was 28.6 years old and mean follow up was 29.6 months. Clinical evaluation of Lysholm knee scoring scale was used for knee function and Tegner activity score was used for activity change after ski injury. Results: The anterior cruciate ligament (ACL) injury accompanied with medial collateral ligament(MCL) injury was most in 11 cases and isolated MCL injury was the next in 9 cases. The common types of injury mechanism were Phantom foot phenomenon (13 cases, 43$\%$) and valgus external rotation injury (12 cases, 40$\%$), which constitute 83$\%$ of all case. At the last follow up, the mean Lysholm score was 93.4 and mean Tegner activity score was 4.2. The reduced Tegner activity score after injury was 1.9. Among several injury groups, the evaluation of knee function and activity was best in the isolated MCL injury group and worst in the ACL injury accompanied with MCL injury group. The factors to influence ski injury were participation to ski class, release of binding, and skiing long time more than 2 hours. Conclusions: Lysholm score at last follow up revealed good grading, but sports activity after ski injury was reduced when compared with pre-injury state. It seems to need a active, systemic sports rehabilitation program after sports injury.

      • Sliding Mode Congestion Control of Differentiated-services Networks

        In this paper, we propose sliding mode congestion controller for differentiated-services network. Two important issue in differentiated-services architecture are bandwidth guarantee and fair sharing of unsubscribed bandwidth among TCP flows with and without bandwidth reservation. We use tight upper and lower bounds for various settings of differentiated-services parameters using the loss-bounded model. The Sliding mode congestion controller scheme is designed using nonlinear control theory based on a nonlinear model of the network that is generated using fluid flow consideration. The methodology used is general and independent of technology, as for example TCP/IP or ATM. The sliding mode congestion controller methodology has been applied to an TCP network. We use NS-2 simulation to demonstrate that the proposed control methodology achieves the desired behavior of the network, and possesses important attributes. as e.g, stable and robust behavior, high utilization with bounded delay and loss, together with good steady-state and transient behavior.

        제2배부 중수골 유경 혈관 이식과 일시적 삼중 주상골 고정에 의한 Kienböck 질환의 치료

        목적: Kienbock 질환 치료에 있어 월상골의 재혈관화를 위한 유경혈관 이식술은 유용한 치료법의 하나로 알려져 있다. 이에 저자들은 Kienbock 질환 환자에서 유경 혈관 이식 및 일시적 삼중 주상골 고정술의 결과와 예후를 알아보고자 하였다. 대상 및 방법: 1999년 3월부터 2003년 5월까지 Kienbock 질환으로 유경 혈관 이식 및 일시적 삼중 주상골 유합술을 받은 9명의 환자를 대상으로 추시 관찰하였다. 추시 기간은 15개월에서 54개월로 평균 35개월이었고 수술 당시 2명이 Lichtman stage Ⅱ, 6명이 stage Ⅲa, 1명이 stage Ⅲb였으며 모든 환자에서 척골 중립변위였다. 수술은 요골 동맥의 제2배부 중수골 분지와 동반 정맥을 이용한 유경 혈관 이식 및 일시적 삼중 주상골 고정술을 시행하였다. 결과: 요측 편위를 제외한 술 후 운동범위와 악력은 유의하게 증가하였다. 이전의 직업으로 복귀하는데 걸린 시간은 평균 22주였다. 단순 방사선 검사상 경화가 호전되는 소견은 8예에서 보였으나 월상골의 함몰 이 6예에서 증가된 소견을 보였다. Lichtman의 기준에 의한 평가는 7예에서는 만족, 비교적 고연령인 2예에서는 불만족이었으며 Nakamura 평가법에 의하면 5예에서 양호, 3예에서 보통, 1예에서는 불량이었다. 결론: 척골 중립변위인 Lichtman stage IIIa 이하의 Kienbock 질환 환자에서 일시적 삼중 주상골 고정술을 동반한 유경 혈관 이식술은 시도할 만한 가치가 있는 치료법이다. 하지만 재혈관화 술식 시 환자의 나이를 고려해야하며 향후 일시적 삼중 주상골 고정의 기간에 대한 체계적인 연구가 필요할 것으로 생각된다. Purpose : Revascularization is a useful method for treating Kienbock's disease. The aim of this study was to evaluate the use of the second dorsal metacarpal vascular pedicle to treat Kienbock's disease. Materials and Methods : A retrospective study was carried out on 9 patients who had undergone vascular pedicle graft for Kienbock's disease between 1999 and 2003. The mean follow up period was 35 months. At the time of surgery, 2 patients were graded as stage Ⅱ, 6 as Ⅲa and 1 as Ⅲb. The ulnar variance was neutral in all cases. A vascular pedicle graft using second dorsal metacarpal vessel were performed and temporary scaphotrapeziotrapezoid (STT) fixation were applied for 6 months. Results : Wrist motion except for the radial deviation and grip strength had improved significantly. The patients returned to work after an average of 22 weeks. Resorption of the sclerosis was observed in 7 cases, but lunate collapse was observed in the simple radiograph in 6 cases. According to Lichtman's criteria, 7 cases were satisfactory but 2 cases with a relative old age were unsatisfactory. According to Nakamura's classification, 5 cases were good, 3 cases were fair and 1 case was poor. Conclusion : In Kienbock's disease with neutral ulnar variance, a revascularization procedure of vascular pedicle graft with temporary STT fixation is a good treatment method but the patient's age should be considered when determining the revascularization procedure. A prospective study will be needed to determine the optimal duration of temporary STT fixation.
