        전사인자를 이용한 질병특이 유도전능줄기세포

        The recent years have seen great advances in reversal of programming of differentiated somatic cells towards pluripotency by methods not involving nuclear transfer. Although differentiation of cells involves complex genetic and epigenetic changes, it is now possible to generate cells with many properties of pluripotent embryonic stem cells by transduction of differentiated cells with only four transcription factors: Oct3/4, Sox2, Klf4 and c-Myc. In advance, the protocol for improving the efficiency of generating induced pluripotent stem(iPS) cells and establishing clinically applicable cells has been developed. Recently, the generation of iPS cells from patients with a variety of genetic diseases with either Mendelian or complex inheritance including X-linked adrenoleukodystrophy, amyotrophic lateral sclerosis, Duchenne and Becker muscular dystrophy, Parkinson's disease, Huntington's disease, Gaucher disease type III, Down syndrome is also possible. Such disease-specific iPS cells offer an unprecedented opportunity to recapitulate both normal and pathologic human tissue formation in vitro, thereby enabling disease investigation, drug development and cell therapy.

        This study on the stability of chlorophyll in sea mustard, Undaria pinnatifida, during blanching, salting and air drying was carried out. Raw sea mustard was blanched for 30 seconds in the various temperature. The blanched product was salted with table salt after centrifuged after 48 hours for dewatering. In addition, sea mustard was dried in a drier to make clear the dehydration mechanism during the forced air drying. The drier was designed to controll the temperature, relative humidity and velocity of air. The most effective blanching temperature and time for maximal residual amount of chlorophyll was 90℃ and 30 seconds. The soaking in 1% chemical solution for 20 minutes showed more desirable results than blanching in 1% chemical solution. Oyster shell powder, reed ash and 0.5% Ca(CH₃CO₂)₂with 0.5% MgCO₃solutions were more effective than the 1% solution of other chemicals. The most reasonable ratio of added salt to dewater the product for 48 hours was 25% in w/w. The amount of total organic acid and volatile acids revealed no correlation with the amount of added salt. The optimal condition during drying at 80℃ and 90℃ with different relative humidities was effective at 80℃ with 30% relative humidity.

        N/A In Nb, V and Ti added steels, carbo-nitrides are formed due to their strong interaction with C and N. The formation of carbo-nitrides has an important role to control the microstructure as well as mechanical properties by grain size refinement and precipitation hardening. However, the quantitative analysis of distribution of precipitates and the effect of precipitates on the phase transformation and mechanical properties are still far from satisfactory. In this study, the quantitative analysis of precipitates in austenite was investigated using the fact that the formation of precipitates in Nb, V and Ti added steels accelerates austenite/ferrite transformation. The formation of precipitates was controlled by adjusting holding temperature and time in austenite region. transformed Volume fractions were measured by dilatometer during slow cooling. Iso-precipitation kinetics were determined by comparing 5% and 50% volumes transformed at various conditions respectively. The result was compared with the calculated.

      • 새우젓 품질향상을 위한 제조방법의 개선

        Changes of chemical components and compositions of fatty acid in fermented shrimp added additives such as ethyl alcohol and sorbitol to improve the quality were discussed. The ranges of moisture, lipid and protein contents in products during fermentation at 15℃ for 120 days were 60.3-64.8%, 1.4-2.0% and 10.3-12.3%, respectively and the remarkable change of those contents in fermentation period was not observed. Lipid oxidation in the fermented shrimp with 5% of ethyl alcohol and 5% of sorbitol decreased slightly as compared with that of control during fermentation period. The major fatty acids were 16:0, 22:6, 20:5, 18:1, 16:1, and 18:0 in raw sample and products. In the changes of fatty acid compositions, saturates were increased while polyenes were decreased slightly during fermentation period.

        선체 청소로봇 자동화를 위한 광 변위센서 기반의 위치추정 방법

        본 논문에서는 선체 청소로봇의 자동화를 위한 새로운 위치추정 방법을 제안하였으며, 제안한 위치추정 방법을 실제 선체 청소로봇에 적용 가능성을 평가하기 위해 동일한 주행방법을 가지는 소형로봇에 적용하여 위치추정 실험을 수행하였다. 위치추정 실험을 통해 광 변위센서를 사용한 위치추정 방법이 회전 엔코더를 사용한 방법보다 더 정확하게 위치를 추정하는 것을 확인하였으며, 더불어 제안한 위치추정 방법을 통해 로봇 주행방향 또한 기존의 회전 엔코더 방식보다 정확하게 계산되는 것을 확인하였다. 이후의 연구에서 제안한 위치추정 방법에 오차보정을 위한 센서를 추가하여 위치추정 정확도를 보완하고, 이를 실제 선체 청소로봇에 적용하여 사용할 계획이다. This paper presents the new position estimation method which contains the optical displacement sensor and the dead reckoning based position estimation algorithm for automation of hull cleaning robot. To evaluate feasibility of the proposed position estimation method on the hull cleaning robot, it was applied on the small scale robot model which has an identical drive method with the hull cleaning robot and then a set of the position estimation experiments were performed. The experimental results of the position estimation demonstrate that the estimated results with the optical displacement sensors is more accurate than used rotary encoder method. In addition, it continuously calculated the robot position quite close to the real robot driving path. In a follow-up study, the proposed position estimation method will be complemented and exploited on the actual hull cleaning robot by adding additional sensor modules that correct measurement errors.

        도파 반응성 근육긴장이상, Segawa병 1례

        도파 반응성 근육 긴장이상으로서, Segawa병은 10대에 발병하며 현저한 주간 변동을 보이는 유전성 진행형 근육 긴장이상 증상을 보이며 GCH I 유전자의 결함에 의한 dopamine 합성의 장애로 발생한다. 저용량 levodopa로도 부작용 없이 정상생활을 유지할 수 있고 혹 Segawa병과 관련된 저신장 또한 levodopa의 조기 투여로 정상 신장으로 회복 될 수 있는 바, 조기 발견에 의한 치료가 중요하며, 국내에서도 Segawa병과 관련된 근육 긴장 이상 질환들에 대한 임상적, 분자 생물학적 연구와 관심이 필요하다. 저자들은 전형적인 Segawa병의 임상 증상과 뇌척수액 neopterin 감소로 확진되어 저용량 levodopa로 정상 생활을 유지하는 여자 환아 1례를 경험하였기에 보고한다. Segawa disease, hereditary progressive dystonia with marked diurnal fluctuations or defined dopa-responsive dystonia has age-dependent clinical courses, which are characterized with marked progression in the first one and half decades, its subsiding in the third decade and almost stationary courses after the fourth decade. Also, it has characteristic diurnally fluctuating symptoms, aggravated towards the evening and alleviated after sleep. This autosomally dominantly inherited dystonia is caused by abnormalities of the gene of GTP cyclohydrolase I. The heterozygotic gene´s abnormality induces partial decrement of tetrahydrobiopterin and affects synthesis of tyrosine hydroxylase(TH) rather selectively. The reduction of TH induces decrement of dopamine and disfacilitates the Dl receptor-striatal direct pathway. The pathognomonic finding in biochemical examination is the decrease of neopterin in the cerebrospinal fluid(CSF). Levodopa, by replacing dopamine contents at the terminal, alleviates motor symptoms completely and the effects sustain without any side effects. We experienced a girl diagnosed as Segawa disease with typical clinical courses and a decrease of neopterin in the CSF.

        소아 난치성 무긴장성발작에서의 Deflazacort의 병용투여 효과

        목적:난치성 무긴장성 발작을 보이는 소아 환자에서 corticosteroid 성분의 deflazacort 병용 투영의 발작 조절 효과와 부작용을 알아보고자 본 연구를 시행하였다. 방법:2000년 6월부터 2000년 12월까지 연세의대 세브란스 병원 소아신경과에 6개월 이상의 무긴장성발작 또는 무긴장성발작을 동반한 Lennox-Gastaut 증후군으로 내원한 환아 중, 항간질제를 두가지 이상 최대 용량으로 투여하였음에도 난치성을 보이는 48명의 환아를 대상으로 전향적 개방형 연구를 시행하였다. 약물 투여 전 4주간의 관찰기 이후 최초 4주간은 deflazacort 2 mg/kg/d를 3회 분복하여 투여하였으며 이후 발작 조절을 관찰하면서 20주간은 deflazacort 2 mg/kg/q.o.d로 총 24주간 투여하였다. 4주 간격으로 간질 발작 빈도와 함께 부작용 여부를 측정하였고 deflazacort의 유효성을 평가하기 위한 주결과 변수로 관찰기 동안과 약물 투여 6개월간의 발작회수감소율의 중앙값을 통계적으로 검정하였으며 보조 결과 변수로 간질 발작 빈도가 50% 이상 감소한 환아들과 발작이 완전히 소실된 환아들의 비율을 조사하였다. 더불어 Lennox- Gastaut 증후군에 동반된 무긴장성발작과 무긴장성발작만을 발현하는 예 간에 발작회수감소율의 차이를 분석하였으며 50% 이상 발작 감소를 보인 반응군과 비반응군 간의 임상 양상의 차이를 비교 관찰하였다. 결과:48명의 환아를 대상으로, 관찰기와 약물투여 6개월 기간 중 발작회수감소율의 중앙값은 42.7%로 통계적으로 유의하게 감소됨을 확인할 수 있었다. Lennox- Gastaut 증후군의 발작회수감소율의 중앙값은 48.9%였으며 무긴장성발작 형태로만 발현하는 경우는 39.3%로 Lennox- Gastaut 증후군에서 다소 효과가 높은 경향이 있었으나 통계적으로 의미는 없었다. 간질 발작 빈도가 50% 이상 감소한 예는 22명(45.8%)이였으며, 간질 발작이 완전히 소실된 경우는 4명(8.3%)이였다. 발작 빈도가 50% 이상 감소한 반응군과 비반응군 간의 임상 양상에 있어 통계적으로 의미 있는 차이는 없었다. 부작용은 20명(41.6%)에서 나타났으며 행동장애가 4명(8.3%),체중 증가와 쿠싱 형상이 16명(33.3%)에서 보였다. 하지만 약물을 중단하지는 않았으며 추적 관찰중이다. 결론:항간질제에 난치성을 보이는 소아 무긴장성발작에서 deflazacort의 병용 투여는 간질 발작 조절에 효과가 있었으며, 부작용으로 경한 행동장애와 쿠싱 형상이 있었으나 약물 투여 중단을 요하지는 않았다. PURPOSE: This is a clinical study to evaluate the efficacy and adverse reactions of deflazacort as adjunctive therapy in childhood intractable atonic seizure including Lennox- Gastaut syndrome. METHODS: This is a clinical prospective, add-on, and open-label study performed for 6 months from Jun. 2000 to Dec. 2000 at the pediatric neurology clinic of Severance Hospital. Subjects were selected according to the following criteria, 1) Patients were diagosed as refractory atonic seizure disorder including Lennox-Gastaut syndrome during more than 6 months, 2) Patients had been on maximal doses of at least 2 anticonvulants including sodium valproate and clonazepam or clobazam. We observed seizure frequency of 4 weeks and 24 week medication period as well as adverse reactions every 4 weeks. Those data were analysed primarily for median seizure frequency reduction rate and other efficacy variables such as responder rate with frequency reduction more than 50% and seizure free rate. We also compared the clinical aspects between responder and non responder group. RESULTS: 48 patients were evaluated for efficacy and adverse reactions. Median seizure frequency reduction rate was 42.7%, responders were 22 patients(45.8%) and seizure free patients were 4(8.3%). In Lennox-Gastaut syndrome, median seizure frequency reduction rate was 48.9% and in atonic seizure only 39.3%. However, there were no statistically significant differences in efficacy. We compared clinical aspects between responder and non responder groups, but couldn't find any difference. The number of patients manifesting adverse reactions was 20(41.6%) in an descending order of frequency, weight gain in 16 patients(33.3%), and irritability in 4 patients(8.3%). CONCLUSION: Deflazacort is believed to be an effective and safe anticonvulsant when used as adjunctive therapy for atonic seizure including Lennox-Gastaut syndrome. However, long term follow up is required to evaluate relapse rate and its adverse reactions.
