        사람치아 단백질을 분리 흡착한 PVDF막의 생체반응에 관한 연구

        Purpose: Human tooth proteins are highly heterogeneous, comprising diverse proteins derived from a number of genes. The attempts to identify protein for activity of tooth matrix proteins have been defied by several factors. First, the amount of proteins within teeth is very small relative to many extracellular matrix proteins of other tissues. Second, the bioassay system is tedious and needed for long time. Therefore we tried to find easy techniques, which increase the product rate, and an assay of small proteins, with which amino acid sequence is possible without additional procedures. Materials and Methods: Total protein were extracted from 300 g enamel removed teeth and 600 g teeth with 4 mol/L guanidine HCl and purified by gel chromatography. Aliquot of proteins was implanted into muscle pouches in Sprague-Dawley rats for bioassay. By SDS-PAGE and membrane blotting, molecular weight of each protein was estimated and a partial amino acid sequence was obtained. Each fraction blotted on the membrane was cut out and inserted in rat ectopic model. Results: In dissociative method, total tooth proteins were obtained 1mg/ml from enamel removed teeth and 3.5 mg/ml from teeth. In SDS-PAGE, four clear bands at the sites corresponding to 66, 40, 20 and 18 kD. Especially The 66 kD band was clearly exhibited. Amino acid sequencing from tooth could be possible using PVDF membrane blotting technique. In amino acid sequencing, 66 kD protein was identified as albumin. Conclusion: Compared with conventional method for extraction of teeth protein and bioassay of proteins, the methods in this study were easy, time-saving and more productive technique. The matured tooth proteins omitting additional procedure of mechanical removal of enamel were simply analyzed using blotted PVDF membrane. This method seems to make a contribution as a technique for bioassay and amino acid sequencing of protein.

        코로나19에 스마트매체를 활용한 어린이집 교육경험의 현상학적 연구

        목적 본 연구는 코로나19 시기에 스마트매체를 활용한 어린이집 교육경험과 체험한 현상 속에 담겨진 본질을 심층적 기술로 이해하며 개선방안에 대하여 알아봄으로써 영유아 교육현장에 효과적인 지원방안을 제안하는데 있다. 방법 서울, 인천, 경기지역 어린이집에서 코로나19 기간 중 어린이집 교육경험이 있는 원장과 교사 12인을 연구참여자로 선정하였다. 자료수집은 2021년 11월부터 2022년 1월까지 표적집단면접법(포커스그룹인터뷰)와 개별면담을 통해 이루어졌고 질적연구자료 분석절차에 맞추어 해석, 코딩, 분류하며 현상학적 연구방법에 의해 분석하였다. 결과 코로나19 시기에 스마트매체를 활용한 어린이집 교육경험은 ‘수업방식의 변화와 예상치 못한 문제점’, ‘ZOOM을 통한 운동회, 부모교육’, ‘스마트매체로 현장학습 출발’, ‘스마트매체로 지역사회 연계활동’, ‘교사의 업무편의 고려 필요’, ‘스마트매체사용 부모의 걱정 증가’, ‘상상력과 창의력 수업의 자율성’, ‘가정과의 협력 가능한 기자재 지원 필요’, ‘부모상담도 스마트매체로’로 나타났고, 개선방안 제안으로는 ‘보건복지부 차원의 동일한 기자재 보급과 교사교육’, ‘동일한 재난상황 발생시 사전 부모교육으로 이해 필요’, ‘연령에 맞는 스마트 교육프로그램 개발’, ‘어린이집에 스마트교육 전문가 지원’, ‘스마트 수업 준비 시간과 보조교사 인력확충’으로 나타났다. 결론 본 연구는 코로나19 시기에 스마트매체를 활용한 어린이집 원장과 교사의 교육경험에 주목하고 체험한 현상의 본질적 의미를 심층탐구하고 개선방안을 제안했다는 점에서 연구의 의의를 가진다. 영유아 교육현장에 효과적인 지원방안을 마련하는데 도움이 되기를 기대하며 스마트매체의 보편적인 활용을 위한 국가차원의 보급과 교육 방향에 대한 후속 연구를 제안하였다. Objectives The purpose of this study is to suggest effective support measures in the field of early childhood education by understanding the essence contained in the experience and experienced phenomena of daycare center education using smart media in the time of Corona 19 with in-depth technology and finding out ways to improve it. Methods At daycare centers in Seoul, Incheon, and Gyeonggi Province, 12 principals and teachers with experience in daycare during the COVID-19 period were selected as research participants. Data collection was conducted from November 2021 to January 2022 through the target group interview method (focus group interview) and individual interviews. Results The experience of daycare education using smart media in the time of Corona 19 is ‘Changes in class methods and unexpected problems’, ‘Sports day through ZOOM, parent education’, ‘Starting field trips with smart media’, ‘Community connection with smart media’ Activities’, ‘Teachers need to consider the convenience of work’, ‘Increasing concerns of parents using smart media’, ‘Autonomy of imagination and creativity classes’, ‘Need support for equipment that can cooperate with families’, ‘Parent counseling as a smart medium’ ‘Same equipment supply and teacher education at the level of the Ministry of Health and Welfare’, ‘In case of the same disaster situation, it is necessary to understand the parents in advance’, ‘Development of smart education programs suitable for age’, ‘Smart education experts at daycare centers' Support’, smart class preparation time and expansion of assistant teachers' manpower. Conclusions This study has significance in that it paid attention to the educational experiences of daycare center directors and teachers using smart media during the COVID-19 period, and in-depth explored the essential meaning of the experienced phenomenon and suggested improvement measures. In the hopes that it will be helpful in preparing effective support measures in the field of early childhood education, a follow-up study on the distribution and education direction at the national level for the universal use of smart media was proposed.

        골 재생을 위한 생물학적 유도막

        The purpose of this study was to evaluate the stability and efficacy of biologic membrane made of freeze-dried cartilage as a barrier to facilitate guided bone regeneration in experimental non-healing bone defects in the rat mandible. Nine adult Sprague-Dawley rats(400-500g) were used in experiment. 5.0mm in diameter were created on the mandibular angle area by means of slow-speed trephine drill. In microscopic examination, dynamic immature bone forming at 2 weeks and its calcification at 4 weeks were observed. The membrane made of lyophilized cartilage taken from human costal cartilage seems to be very effective for guided bone regeneration as a biologic membrane and the scaffold for attachment of cells or local drug delivery system of growth factor, which may meet the ideal requirement of a barrier membrane and graft materials

        하악전돌증 환자에서 내측 익돌근 근력측정 및 분석

        The purpose of this study was to evaluate the resistant force of medial pterygoid muscles against the mandibular advancement and distraction to anterior, and inquire into the relationship between medial pterygoid muscles and cephalometric variables. Sixty six patients with class III malocclusion underwent bilateral sagittal splitting of ramus with intraoralvertico-sagittal ramus osteotomy for mandibular set-back. The spring scale was used to measure the resistance of medial pterygoid muscles after splitting of ramus. Skeletaldental cephalometric analysis was made and statistic package was used for correlation between resistance and cephalometric variables. The resistant force of the right medial pterygoid muscle was greater than the left one in Koreans with class III malocclusion, and the force had a linear regression relationship with facial depth. The results suggested that facial depth has significant correlation with the resistance of medial pterygoid muscle, which can be acquired from patient’s cephalometric analysis.

        MG-63 세포주에서 Vascular Endothelial Growth Factor (VEGF) mRNA 발현에 대한 Insulin-like Growth Factor I (IGF-I)의 효과에 대한 연구

        Purpose: To determine the role of Insulin-like Growth Factor-I (IGF-I) in the regulation of Vascular Endothelial Growth Factor (VEGF) expression in MG-63 cells and then to find the mechanism b which this regulation occurs. Materials and methods: MG-63 cells were grown to confluence in 60-mm dishes. To determine the effects of IGF-I on expression of VEGF mRNA according to time and concentration, the cells were treated with 10 nM IGF-I, following isolation of total RNA and Northern blot analysis after 1, 2, 4, 8, 12, 24 hours and after 2 hours of treatment with 0.5, 2, 10, 25, 50 nM IGF-I respectively, isolation of total RNA and Northern blot analysis were followed. To determine the mechanism of action of IGF-I, inhibitors such as hydroxyurea (76.1 ㎍/㎖), actinomycin D (2.5 ㎍/㎖), cycloheximide (10 ㎍/㎖) were added 1 hour after treatment of 10 nM IGF-I. Results: 1. the expression of VEGF mRNA was increased with treatment of IGF-I. 2. The expression of VEGF mRNA was increased according to time- and concentration dependent manner of IGF-I. 3. The effect of IGF-I was decreased by hydroxyuera, actinomycin D, but not by cycloheximide. Conclusion: IGF-I regulate the expression of VEGF mRNA in the level of DNA synthesis and transcription. These results could suggest that IGF-I plays an important role in angiogenesis in the process of new bone formation and remodeling.

        다양한 소독방법이 PPDO-hybrid-PLGA nanofiber scaffold의 형태와 세포부착에 미치는 영향에 관한 연구

        Objectives : the effect of different sterilization methods on the surface morphology of PPDO-hybrid-PLGA nanofiber scaffold and attachments of PC12 cell were investigated. Methods : Poly (p-dioxone)-hybrid-Poly (lactide-glycolide) (PPDO-hybrid-PLGA) nanofiber scaffold, fabricated in a tube form with 1.5 mm internal diameter, 0.2 mm thickness and 5 mm length, was prepared using electrospinning method. To study the surface morphology using SEM, The study group and control group in respective were; Control:Non-sterilized scaffold, Group I:scaffold sterilized with 70% Alcohol, Group II: scaffold sterilized with Ethylene Oxide at 65 ℃ , and Group Ⅲ: scaffold sterilized with Ethylene Oxide at 37 ℃. To investigate viability of the PC12 cell on the scaffold, The study group and control group in respective were; Control: sterilized with 70% Alcohol, Group I: sterilized with Ethylene Oxide at 65℃, and Group II: sterilized with Ethylene Oxide at 37 ℃. Results : 1. The surface morphology was slightly changed in Group I, II and GroupⅢ, compared with control. 2. The attachment of PC12 cells in Group I, II was not higher than in control Discussion : The attachment of PC12 cell is not influenced by different sterilization methods.
