        지역장애인구강진료센터의 역할 및 전담치과위생사의 적정역할에 대한 고찰

        Objectives: This study aims to define the role of the local dental clinic for people with disabilities and the appropriate role of the dental hygienist to resolve oral health inequality with people with disabilities. Methods: From September 12, 2022 to November 11, 2022, 16 experts who voluntarily participated in the research, among the experts recommended by the related institutes, were selected through stratification analysis. The results of this study and improvement plans are as follows. Results: First, as a result of measuring the importance of the role of a regional oral care center for people with disabilities in the analytic hierarchy process (AHP) middle category, check-up was found to be the most important (0.364, C.I=0.01). For examination, a facility visit checkup and regular checkups were required. Second, as a result of measuring the importance of the role of a dedicated dental hygienist in the AHP middle category, the institutional-linked activity was found to be the most important (0.417, C.I=0.02). As for institutional-linked activities, linkage with local community organizations, patient management, and public relations activities were required. Conclusions: Considering these results, the role of a regional oral care center for people with disabilities is to perform examinations and preventive treatments, primary treatments (simple), education and counseling, and establish a smooth medical return system to serve as a base for dental treatment for people with disabilities. Education and counseling are provided through professional patient response and the arrangement of a dedicated dental hygienist, and oral health care is provided to people with disabilities through professional treatment assistance for patients and treatment support and screening, linkage with local community institutions, and patient management. Specialized dental hygienists should play a role in supporting the operation of oral health care for people with disabilities.

        20대 비만여성의 유방형태 분석

        The purpose of this study is to provide for some basic data useful to the development of the brassieres comfortable and more suitable for obese women. The results of this study can be summarized as follows; 1. Obese women were categorized using Rohrer index into three groups, and thereupon, their breast forms were analyzed. As a result, it was found that group 1 and 2 had similar small breasts, while group 3 had the largest size of breasts. 2. The forms of obese women's breasts could be determined by such factors as breast obesity, factors related with breast split, protrusion and volume, factors related with breast area and drooping, factor affecting the breast protrusion and factor defining the lower form of breast. 3. The actual breast types of obese womwn could be classified into cone type, protusion type, drooping type Ⅰand Ⅱ. The more obese a woman was, her breast tended more to droop.

        Mitochondrial pyruvate dehydrogenase phosphatase 1 regulates the early differentiation of cardiomyocytes from mouse embryonic stem cells

        Mitochondria are crucial for maintaining the properties of embryonic stem cells (ESCs) and for regulating their subsequent differentiation into diverse cell lineages, including cardiomyocytes. However, mitochondrial regulators that manage the rate of differentiation or cell fate have been rarely identified. This study aimed to determine the potential mitochondrial factor that controls the differentiation of ESCs into cardiac myocytes. We induced cardiomyocyte differentiation from mouse ESCs (mESCs) and performed microarray assays to assess messenger RNA (mRNA) expression changes at differentiation day 8 (D8) compared with undifferentiated mESCs (D0). Among the differentially expressed genes, Pdp1 expression was significantly decreased (27-fold) on D8 compared to D0, which was accompanied by suppressed mitochondrial indices, including ATP levels, membrane potential, ROS and mitochondrial Ca2+. Notably, Pdp1 overexpression significantly enhanced the mitochondrial indices and pyruvate dehydrogenase activity and reduced the expression of cardiac differentiation marker mRNA and the cardiac differentiation rate compared to a mock control. In confirmation of this, a knockdown of the Pdp1 gene promoted the expression of cardiac differentiation marker mRNA and the cardiac differentiation rate. In conclusion, our results suggest that mitochondrial PDP1 is a potential regulator that controls cardiac differentiation at an early differentiation stage in ESCs.

        Combination treatment with 2-methoxyestradiol overcomes bortezomib resistance of multiple myeloma cells

        Bortezomib is a proteasome inhibitor used for the treatment of relapsed/refractory multiple myeloma (MM). However, intrinsic and acquired resistance to bortezomib has already been observed in MM patients. In a previous report, we demonstrated that changes in the expression of mitochondrial genes lead to changes in mitochondrial activity and bortezomib susceptibility or resistance, and their combined effects contribute to the differential sensitivity or resistance of MM cells to bortezomib. Here we report that the combination treatment of bortezomib and 2-methoxyestradiol (2ME), a natural estrogen metabolite, induces mitochondria-mediated apoptotic cell death of bortezomib-resistant MM KMS20 cells via mitochondrial reactive oxygen species (ROS) overproduction. Bortezomib plus 2ME treatment induces a higher level of cell death compared with treatment with bortezomib alone and increases mitochondrial ROS and Ca2þ levels in KMS20 cells. Pretreatment with the antioxidant N-acetyl-L-cysteine scavenges mitochondrial ROS and decreases cell death after treatment with bortezomib plus 2ME in KMS20cells. Moreover, we observed that treatment with bortezomib plus 2ME maintains the activation of c-Jun N-terminal kinase (JNK) and mitogen-activated protein kinase kinase kinase 4/7 (MKK4/7). Collectively, combination treatment with bortezomib and 2ME induces cell death via JNK-MKK4/7 activation by overproduction of mitochondrial ROS. Therefore, combination therapy with specific mitochondrial-targeting drugs may prove useful to the development of novel strategies for the treatment of bortezomib-resistant MM patients.

        Voluntary stand-up physical activity enhances endurance exercise capacity in rats

        Involuntary physical activity induced by the avoidance of electrical shock leads to improved endurance exercise capacity in animals. However, it remains unknown whether voluntary stand-up physical activity (SPA) without forced simulating factors improves endurance exercise capacity in animals. We examined the eff ects of SPA on body weight, cardiac function, and endurance exercise capacity for 12 weeks. Twelve male Sprague-Dawley rats (aged 8 weeks, n=6 per group) were randomly assigned to a control group (CON) or a voluntary SPA group. The rats were induced to perform voluntary SPA (lifting a load equal to their body weight), while the food height (18.0 cm) in cages was increased progressively by 3.5 every 4 weeks until it reached 28.5 cm for 12 weeks. The SPA group showed a lower body weight compared to the CON group, but voluntary SPA did not affect the skeletal muscle and heart weights, food intake, and echocardiography results. Although the SPA group showed higher grip strength, running time, and distance compared to the CON group, the level of irisin, corticosterone, genetic expression of mitochondrial biogenesis, and nuclei numbers were not affected. These findings show that voluntary SPA without any forced stimuli in rats can eff ectively reduce body weight and enhance endurance exercise capacity, suggesting that it may be an important alternative strategy to enhance endurance exercise capacity.
