        Induction of erythroleukaemia by F - MuLV and Purification of Erythropoietin

        Friend Murine Leukemia Virus (F-MuLV) is affective against 2-day old new borns in NFS/n Swiss mice. Infected babies suffer from splenomegaly and anemia at the age of 4-5 weeks beyond which the condition worsens and is fatal. However, the spleen size can be reduced by hypertransfusion for 8 weeks. If left alone after this treatment, the spleen enlarges and becomes palpable again. This time the spleen cells are transplantable and inflict the same disease if injected to normal adults. While these leukemic cells were considered to be hormone-independent, addition of erythropoietin (EPO) was found to be essential for maximum growth in culture. Influence of EPO on the in vivo growth of leukemic cells was indicated by our findings that administration of the hormone caused a more severe leukemia and rapid death while, transfusion of red blood cells, which lowers endogenous EPO, led to increased survival of leukemic mice. It is suggested that hormone-associated therapy may be efficacious in the treatment of this and perhaps, other leukemias. Several erythroleukemic cell lines released factors which stimulated erythropoiesis in vivo and in vitro. A simple five step scheme has been devised which allows purification of this erythropoietic activity to apparent homogeneity. The methods employed included lectin affinity chromatography (wheat germ agglutinin), gel filteration (ultro gel ACA 44), ion exchange, hydroxylapatite, and high performance liquid chromatography, Following polyacrylamide gel electrophoresis, biological activity was recovered in an area corresponding to a molecular weight of 35,000 daltons. Silver staining of a polyacrylamide gel after electrophoresis of our most purified preparation revealed a single band of 35,000 daltons.

      • Industrial Demand and Integrated Material Flow of Terbium in Korea

        The integrated material flow analysis methodology uses bottom-up flow analysis for primary and secondary resources and top-down flow analysis for the distribution structure. By combining the advantages of the top-down and bottom-up methods, the integrated material flow analysis Methodology can overcome the limitations of each method. Using the IMFAM, this study investigated the material flow of terbium in 2011, in Korea. Terbium was used to produce 3-wevelength fluorescent lamps, liquid crystal display back light unit, plasma display panel and so on. 7,239 kg of terbium was required to produce the intermediate product in Korea. Among total terbium used to produce intermediate products, 2,592 kg the part of the fluorescent lamp, 4,984 kg the part of flat panel display, which were used, 107 kg, was exported and 46 was put in collect stage. In the part of the 3-wevelength fluorescent lamp, terbium’s use was expected to gradually increase for needs of consumers. But in the part of flat panel display, terbium’s use is expected to gradually decrease. The use of terbium is expected to decrease in others. This MFA results represent the materials flow of terbium in Korea and can be used to predict demand through investigations on the demand industry.

        보리산업의 현황과 수매중단에 따른 대응방안 - I. 보리가공제품 생산실태 및 품질과 가격조사 -

        The current state of barley processing enterprises and actual conditions of processed products containing barley in circulation have been investigated. This survey was done in order to set up countermeasures for stable supply and demand of domestic barley consequential to the abolition of procurement system of barley. Liquor-excepted barley processing companies were 153, and 17 items of processed products containing barley and 240 processed products were examined. Mixed grain was the most found among the processed products containing barley, followed with polished waxy barley. The price of polished barley products increased as the grain was more processed and waxy. Eco-friendly products were more expensive than normal products on an average of 41% with a range variation of 12% to 93%. Some processed products containing barley were four times more expensive than other same products made by different companies but actually there weren’t any special difference among the products in terms of quality. Only 31% of the products known as waxy barley could be accepted as 'actual' waxy barley. The rest of them were mixed with non-waxy barley and some of them were even entirely consisted of non-waxy barley. In case of barley malt products, more than a half of them were below the standard of the diastatic power (150 DP). About 24% of the products were below 100 DP which means the product were in a really bad condition. However, there weren't any relation between the quality and price as some bad products were even more expensive than others. Therefore, it can be said that the introduction of a quality grading system is needed for proper quality control of waxy barley or barley malt products. 국내보리의 수매중단에 따른 원료곡 수급의 대책을 마련하기위한 기초자료로 보리를 이용한 가공제품을 생산하고 있는 전국의 가공업체 현황과 시중에 유통되고 있는 가공제품의 종류와 가격을 조사하고 이들의 품질정도를 분석한 결과는 다음과 같다. 1. 전국적으로 수집된 보리가공제품(주류제외)에 의해 조사된 보리가공업체의 수는 153개였으며 시도별로는 경기 >충북 >전남 >전북 >경북 >경남 =충남 >강원 >서울 순이었다. 2. 보리함유 가공제품(주류제외)의 품목 수는 17개였으며 가공제품의 수는 240종이었다. 3. 보리가공제품의 품목별 수는 혼합곡 >찰보리쌀 >일반보리쌀 >미숫가루 >보리차 >엿기름 >압맥 >보리과자류 순이었다. 4. 보리가공업체가 생산하는 제품의 종류는 평균 1.6종이었으며 전체업종의 68%가 1 ~ 2 종류를 생산하며, 5종 이상을 생산하는 업체도 13%정도 되었다. 5. 보리쌀 제품류별 유통가격은 압맥(찰성) >흑맥(메성) >할맥(메성) >찰보리쌀 >압맥(메성) >메보리쌀 순이었다. 6. 친환경제품은 일반제품에 비해 평균 41%정도 가격이 높았으며 제품에 따라 12~93%의 가격 차이를 보였고, 차이가 큰 순으로는 보리차 >찰보리쌀 >압맥 >미숫가루 >혼합곡이였다. 7. 동일품목 내에서도 가공업체에 따른 차이는 미숫가루가 가장 커서 최고가가 최저가의 4배 이상이고, 보리차음료(PET병), 보리차(티백), 찰보리쌀(친환경)은 3배 이상의 차이를 보였다. 8. 찰보리쌀 제품 42종 중 찰성순도가 높은 제품이 13종, 찰성과 메성이 혼합된 제품이 26종, 완전 메성인 제품이 3종이었으나, 찰성순도와 이들의 가격 간에는 관련이 없었으며 오히려 찰성순도가 낮은 제품이 비싼 경우도 있었다. 9. 메보리쌀 제품 34종 중 표시가 잘못된 찰성 보리쌀 제품이 1종 있었고, 찰성과 메성이 혼합된 제품이 2종 있었다. 10. 보리 엿기름제품 30종의 평균 효소력가는 131oL로 낮은 편이었으며 150˚L미만의 품질이 낮은 제품이 절반이상이었다. 11. 보리 엿기름의 효소력가와 제품가격 간에 관련이 없었으며 오히려 품질이 낮은 제품이 비싼 경우도 있었다.

        Effects of Size, Impurities, and Citrate Capping on the Toxicity of Manufactured Silver Nano-particles to Larval Zebrafish (Danio rerio)

        Objectives: This study was conducted to identify factors determining the toxicity of manufactured silver nanoparticles (AgNPs) on aquatic organisms. Methods: For this purpose, we prepared several AgNPs with varied characteristics, including hydrodynamic size (nano-ABCAgCit vs -sized-ABCAgCit), impurities (ABCAg stock vs ABCAg), and citrate capping (ABCAgCit), using a commercially available manufactured AgNP (ABCAg stock). Acute tests were conducted using larval zebrafish (Danio rerioI). In addition, in order to determine the ecotoxicological potentials of various capping agents, toxicity tests were conducted with microbes, waterfleas, and fish for eight different capping agents that are used for NPs. Results: The toxicity of AgNPs in terms of 96 h fish LC50 increased in the following order: ABCAg stock < ABCAg = ABCAgCit = nano-ABCAgCit < μ-sized-ABCAgCit < AgNO3. After removing impurities by dialysis, 96 h LC50 value decreased significantly from 126.6 μg/L (95% confidence intervals [CI]: 107.0-146.2) (ABCAg stock) to 78.6 μg/L (CI: 72.7-84.8) (ABCAg). For μ-sized-ABCAgCit (ranging between 3.9 and 40.6 nm) and ABCAgCit (40.6 nm and 9.1 μm), the 96 h LC50 of the former (43.9 μg/L, CI: 36.0-51.7) was approximately two-fold lower than that of the latter (87.0 μg/L, CI: 73.5-100.3). Conclusions: In this study, we found that for acute lethality, the contribution of impurities and particle size was significant, but that of citrate was negligible.

        Multidisciplinary Functional MR Imaging for Prostate Cancer

        Various functional magnetic resonance (MR) imaging techniques are used for evaluating prostate cancer including diffusion-weighted imaging, dynamic contrast-enhanced MR imaging, and MR spectroscopy. These techniques provide unique information that is helpful to differentiate prostate cancer from non-cancerous tissue and have been proven to improve the diagnostic performance of MRI not only for cancer detection, but also for staging, post-treatment monitoring, and guiding prostate biopsies. However, each functional MR imaging technique also has inherent challenges. Therefore, in order to make accurate diagnoses, it is important to comprehensively understand their advantages and limitations, histologic background related with image findings, and their clinical relevance for evaluating prostate cancer. This article will review the basic principles and clinical significance of functional MR imaging for evaluating prostate cancer.

        Morphological Oogenesis in the otu1 Mutant of Drosophila melanogaster Meigen (Diptera: Drosophilidae)

        The sex-linked, recessive otu1 mutant of Drosophila melanogaster was studied cytologically to reveal the effect of the otu gene mutation on oogenesis. Consequently, the germarium of the otu1 mutant was found to be larger as compared to the normal fertile one and contains many undifferentiated tumor cells resembling cystocytes. Each ovary consists of three types of ovarioles; ovarioles with only a germarium, ovarioles with a germarium and one tumor chamber, and ovarioles with a germarium and two tumor chambers. In the otu1 germarium, the mesodermal follicle cells invade the clusters of different number of tumor cells at the posterior region of the germarium in order to make the tumor chamber and the dividing frequency of the tumor cells in the .rst tumor chamber was six times more than in the second tumor chamber. Follicle cells surrounding the tumor chamber are morphologically normal, but the number of the follicle cells becomes less as the tumor chambers move down contrary to the fertile one. The otu1 mutant female never lays the egg and the formation of the vitelline membrane is incomplete. The otu1 mutant tumor cells also showed characteristics of certain rapidly dividing cells and its nucleus is lobulated.
