Electrical stimulation was carried out in the hypothalmus of 21 cats. Effects of stimulation parameters on the magnitude and maintenance of pressor and cardiac rate responses were studied. The decay time of these responses was quantitated by calculating the 50% responses decay time as the time in which response progressively decreased to one-half the initial maximum responses. It was found and concluded that the magnitude of these cardiovascular response was affected by stimulation frequency, intensity and pulse duration whereas maintenance of responses was predominantly frequency-dependent. A parametric set of 20/sec-3msec was as effective as 100/sec-1msec for elicitation of pressor and heart rate responses from the hypothalamus while the responses were best maintained with stimulation frequency up to 20/sec.
수종 생약 추출물이 동물의 혈압 및 항이뇨홀몬 수용체에 미치는 효과

As vasopressin, which increases significant reabsorption of water and therefore prevents loss of water into urine, regulates the water balance of animal and exers vasoconstrictor activity, it influences the arterial pressure of animals. In liver vasopressin controls the metabolism of glycogen. In this study, effect of methanol extracts of several herbs(Scrophularia buergeriana : MESc, Cirsium japonicum : MEC, Sophora flavescens : MES) on cAMP level of the animal cell and arterial pressure of cat. In recent years cDNA of vasopressin receptor was isolated and the structure of vasopressin receptor was elucidated. It has seven transmembrane domains and coupled with G protein. Therefore, vasopressin agonists increase the cellular cAMP throuth activation of adenylate cyclase and mobilize the cellular Ca²through activation of phospholipase C. Konig developed LVIP2. 0ZC cells whose color is changed by concentration of cAMP. Use of this cell line offers the easy way to detection of cellular cAMP concentration. In present study vasopressin receptor was expressed on the plasma membrane of LVIP2. 0ZC cells and change of cAMP level was measured. To record the effect of vasopressin on arterial pressure cat was pretreated with reserpine and atropine. And also effects of methanol extract of herbs on change of arterial pressure induced by vasopressin were recorded continuously with oscillograph. MESc (1mg/kg) injected into vein devreased arterial pressure of cat, could not change the cAMP level of LVIP2. 0ZC cells. However, MEC and MES aboished the increment of cAMP concentration induced by vasopressin. MES decreased the cAMP level hightened by forskolin also and MEC could not decreased it. MEC and MES lowered the arterial pressure of the cat increased by vasopressin. The results of present study suggest that MESc decrease the arterial pressure by the unkown mechanism and MEC and MES decrease the arterial pressure through the deactivation of vasopressin receptor and adenylate cyclase, respectively.
膽汁 및 數種 膽汁酸에 의한 血壓降下作用에 關한 比較 硏究

The Present study was undertaken in order to study the effects of intravenously administered bovine bile and several bile acids on blood pressure in cats. Depressor activity of Sigma bovine bile (Sigma Chemical Co. product) was compared with that of dried bile and depressor activity was compared between cholic acid, glycocholate, taurocholate and deoxycholic acid. Also studied were effects of atropine, propranolol and regitine pretreatment on the depressor responses elicited by intravenously administered bovine bile and bile acids. The results obtained are as follows; 1. After intravenous administration of Sigma bovine bile, depressor responses were observed invariably. Sigma bovine bile at doses of 10mg/kg and 20mg/kg elicited 55.4±3.0mmHg and 96.0±4.4mmHg depressor responses, respectively. 2. The maximum depressor responses to 20mg/kg and 30mg/kg of intravenously administered dried bovine bile were respectively 67.7±5.4mmHg and 85.3±3.9mmHg. And it was recognized that depressor activity of Sigma bovine bile was greater than that of dried bile. 3. From the results of present study it was suggested that generally depressor activity of bovine bile is greater than that of porcine bile. 4. No changes in depressor responses to Sigma bovine bile were observed in animals pretreated with propranolol while lowered blood pressure was partally blocked by the treatment of animals with either atropine or regitine. 5. Following intravenous administeration of trihydroxy bile acids (30mg/kg), mea arterial blood pressure was decreased approximaely by 40 ∼ 50mmHg. The depressor activity of bile acids were in the order, taurocholate > glycocholate > cholic acid. Depressor responses to DCA were found to be greater than cholates suggesting that dihydroxy bile acid is more effective than trihydroxy bile acid in the elicitation of hypotension. 6. Depressor responses to DCA and glycocholate were partially reduced by pretreatment of animal with regitine, while that to taurocholate was partially blocked by atropine as well. No changes in depressor responses to cholic acid was noticed following treatment of animals with above autonomic nerve blockers.
