        Targeted Doxorubicin Delivery Based on Avidin-Biotin Technology in Cervical Tumor Cells

        Targeting antitumor drugs to specific tissues or cells has attracted considerable interest in the recent tumor therapy because it can reduce the side effects in the body and the treated drug dosage through homing drug to the desired tissues. To utilize the high affinity of biotin to avidin as a drug carrier for doxorubicin (DOX), biotinylated targeting material (biotin-polyethylene glycol (PEG)-folic acid, BPF) and biotinylated drug with a bioreducible linker (biotin-SS-PEG-DOX, BSPD) were designed in this study. The in vitro cellular uptake of the avidin/biotinylated drug complexes showed that the complexes with BPF were internalized dose-dependently into HeLa cells, while their uptake was not detected in the absence of BPF. Although the anti-tumor activity of the drug complexes with BPF and BSPD were similar to that of free DOX in cervical cancer cells, the cytotoxic effect of DOX was significantly reduced in normal cells. Their targeting antitumor effect revealed the significant inhibition of the increasing tumor size in HeLa-xenograft mouse model. These results show that the strong complexing between avidin and biotin acted as a targeting moiety and the antitumor drug is an adaptable tool in the fields of tumor therapy.

        Robert Frost made his apperance in the second literary Renaissance of the United States, And grew in the rural area of New England breathing with his nature. A characteristic of Frost's poem is Dualism, and Frost greatly succeeded in it by effors in Form and meaning of his poetic expression. He gradually expressed his deep ideas in his poems and his expresson of nature and thing are being made a symbolic feature. He took an attitude to collate rivalry matters and ideas, thus containing. Mysticism in his Dualism, Frost expressed these thoughts by the means of poetic construct. The major theme of Frost' s poem whose forms and meanings are rooted in Dualism are developing Misticism. Its back ground, facts, jobs, Persons, dialogues and actions are described by lively sounds of nature. They generates colorful images. They are symbolizing an opposition and harmony between the nature and human life. Frost' s poetic effort begins to discover and define the Seperation and integration from the condition of human life, and seeks to find out universal truth, and exds the clarification of life. In these regard, We can point out "love and A question", "Home burial", "Death of the hiredman", and "Mending Wall", etc. Frost' s poetic thoughts are started as a realist. Frost had described the phenomena of natural world and human life based on naturalism. At one time he had stayed in scepticism. He also was an optimist effected by thc Frontier Mind. Yet above all these, the main thought of Robert Frast is Transcendental Mysticism. And his Mysticism was deeply effected by R. W. Emerson and H.D.Thoreau. We can read Transcendentalism in "Nature", the worka of Emerson and "Walden" of Thoreau. Frost' s Mysticism seeks the sense of things and living creatures which are growing and vanishing in the universe as well as man and nature. Searching attitude is sincere, warm harted and full of humanity.(The end)

      • 海上保險에서 重複因果關係와 保險者의 補償責任에 관한 硏究

        The purpose of this paper is to consider how to decide the cause of loss or damage to the transport goods when maritime accident occurs. In marine insurance, the underwriter is liable for any loss or damage proximately caused by a risk insured(MIA Art.55). So it is very important to determine the proximate cause of loss or damage to ascertain whether it is to be recoverable under the policy. But there is no definite conception or rule what is the proximate cause. It was left to the tribunal as a question of fact. In this paper, I will suggest the general rules to determine the proximate cause of loss or damage of the transport goods in marine insurance. First, in MIA 1906, there is the rule of proximate causation and it has been established the effective causation by cases since 1918. Second, in Institute Cargo Clauses(B) & (C), there are rules of considerably relaxed standards to determine the causation of loss of or damage using the "attributable to" and "caused by" basis. Third, it is noted, under the complex causation situation, there are difference basises to decide the liability of underwriters between the case of successive occurrence of single risk and the case of concurrent occurrence of several risks. Forth, in practice, it couldn't be ascertained the underwriter's liability by a definite rule and it should be fully considered the circumstances and conditions of the loss.

        pH-Dependent Mode of Antibacterial Actions of Low Molecular Weight Water-Soluble Chitosan (LMWSC) against Various Pathogens

        This study examined the antibacterial mode of action of low molecular weight water-soluble chitosan (LMWSC; MW1, MW3, MW5 and MW10) using a combination of approaches, including antibacterial assays, culture in media with different pH values, bactericidal kinetics, cellular leakage measurements, depolarization, and electron microscopy, as well as an in vivo study utilizing a wound infection and wound healing model. The antibacterial activity of LMWSC was related inversely to pH, with higher activities being observed at lower pH values. In addition, evaluation of the effect of bactericidal concentrations of LMWSC on the morphology of Bacillus megaterium and Escherichia coli O-157 revealed that it induces filamentation. Furthermore, the degree of depolarization (in Escherichia coli, pH 7.4) and calcein leakage (lipid composition; L-α-phosphatidylethanolamine (PE)/L-α-phosphatidyl-DL-glycerol (PG)=7/3 (w/w), pH 5.4 and 7.4) were evaluated. Moreover, cells cultured at various pHs were evaluated by confocal microscopy (pH 5.4 and 7.4). These results showed that treatment with LMWSC with different molecular weights had different effects on bacteria. In particular, LMWSC (MW10) cause more visible damage to the bacterial cell membrane than lower molecular weight LMWSC, which is due most likely to the penetration of cells by the lower weight molecules. Interestingly, MW5 was found to attack the membrane and penetrate the cells. In addition, scanning electron microscopy showed that MW10 caused significant morphological changes to the surface of the bacteria. Finally, an in vivo study utilizing a wound infection or healing model showed that LMWSC had the potential for use as a lead compound for the development of a novel anti-infective or healing compound.

        돼지를 이용한 표준화된 화상 모델의 구현

        A standardized animal model with an identical burn wound depth is necessary to carry out a study on burn wound healing. It should be easy to operate, reliable and reproducible. The authors used pigs because they histologically resembles human skin most. The method of getting the pigs burned embodied a standardized burn models by using a digitally controlled aluminum thermal block designed by the authors. Nine pigs were burned by maintaining the heat at 70℃ for 10 seconds, 15 seconds and 20 seconds, respectively, and at 80℃, 85℃, 90℃, 95℃ and 100℃, every 5 seconds, 10 seconds and 15 seconds, for a total of 54 contact burn wounds. And another six pigs were burned at same contact time and temperature (n=3). Another three pigs were burned by maintaining the heat at 80℃ for 10 seconds to produce 30 more contact burn wounds (n=30). After wounding, the burned skin was biopsied, stained with H&E and observed under microscopy. Two pathologists measured the vertical distance between the basement membrane and dermal tissue of the deepest burned tissue. The depth of burn increased at a regular phase in proportion to the temperature and the time of increasing temperature. The depth of 30 biopsies of the burned tissue at 80℃ for 10 seconds was similar statistically. Therefore, the standardized burn model of a pig, made by using digitally controlled aluminum thermal block is highly simple, reliable and reproducible for a standardized burn model, and will be very helpful in the study of burn wound healing.
