        Terphthalic Acid를 분해한는 Pseudomonas sp. T-1의 분리 및 특성

        26 bacterial strains capable of growing on Terephthalic acid (TPA) in minimal medium were isolated from soil and wastewater by selective enrichment culture, and among them, one isolate which was the best in the cell growth and TPA degradation was selected and identified as Pseudomonas sp. T-1 by its characteristics. Cell growth almost revealed a stationary phase at 24 hrs after cultivation. Cell growth dramatically increased in a minimal medium containing 0.1% of TPA as a sole carbon source and TPA was not detected any more at 80 hrs after cultivation. Therefore, it is suggested that Pseudomonas sp. T-1 could be effectively used for the biological treatment of wastewater containing TPA.

        Wastewater in textile industries Shows high pH, high organic strength and wide variation of organic loading. These characteristics make trouble in the Operation of activated sludge process. The objective of this study was investigated to find the operating condition of treatment process and improve the efficiency of removal in aerovic biological treatment of wastewater in textile industries by mixed culture of strains degrading components of wastewater in textile industries. The COD_(Mn), COD_(Cr), and BOD_5 of the wastewater in textile industries were 480∼1103mg/ℓ, 90 4∼3357mg/ℓ and 992∼3122mg/ℓ, respectively. pH was 11.8∼12.3. In the hydraulic retention time(HRT) of 32hr, COD removal efficiency of effluents water were 94∼97% and 94∼98% by continuous experients in the activated sludge process when the volumn loading rate and F/M ratio were 0.68∼1.91kg · COD_(Cr)/㎥ · day, 0.23∼0.62kg COD_(Cr)/kg·MLSS day, respectively. Organic loading rate of 0.65∼2.34 kg BOD_5/㎥·day and 0.18∼0.68 kg BOD_5/kg·MLSS·day, the removal efficiencies of BOD was 99.8%. BOD concentration of effluent water is reduced to 10 mg/ℓ. Activated sludge process was superior for the removal of organic matter because carry out biodegradation in a aeration reactor. In spite of variation of influent quality, the activated sludge process by mixed culture of strains degrading components of wastewater in textile industries could obtain a stable effluent quality in range of short HRT

        Microbacterium laevaniformans 가 생성하는 Isocitrate lyase 의 정제

        Purification of the isocitrate lyase extracted from Microbacterium laevaniformans was investigated. The isocitrate lyase was purified 43.6 folds by the following continuous treatment with ammonium sulfate fraction, DEAE-cellulose, DEAE-sephacel and Sephadex G-200 chromatography. The purified isocitrate lyase was showed to be a single protein band by polyacrylamide gel electrophoresis. The molecular weight of the purified isocitrate lyase was estimated 54,000 Da by the SDS-polyacrylamide gel electrophoresis. The Km and Vmax values for isocitrate were estimated to be 0.83mM and 0.33units/㎖, respectively. Activity of isocitrate lyase was inhibited by cystein-HCl and glutathione.

        As a research for degradation pathway of wastewater from acetaldehyde plant by biological method. The isocitrate lyase extracted from Microbacterium laeuaniformans and properties of the enzyme were investigated. The optimum pH and temperature of the purified enzyme were pH 7.5 and 37℃, respectively. The enzyme was activited Mg and inhibited Mn, Ca, Cu, Zn and Co. The activity of isocitrate lyase was increased by Mg at 3mM.

      • 액체배양에 의한 Monascus rubber의 적색 색소 생산을 위한 최적 배양 조건에 관한 연구

        Monascus rubber KCTC 6122를 이용하여 액체 배양을 통한 실험균의 적색 색소의 생산을 위한 배양 조건의 최적화에 관한 결과는 다음과 같다. 실험 균주가 생산하는 적색 색소의 최적 배양 조건은 탄소원으로 rice powder 4% 첨가, 질소원으로 NaNO₃ 0.2%, 인산염으로 Na₂HPO₄의 농도가 0.3%, MgSO₄의 농도가 0.15%일 때 가장 높은 적색 색소 생성을 나타내었다. 배양 온도는 30℃, 초기 pH가 6.5, 진탕속도를 150 rpm, 배양 시간 8일 일 때 적색 색소의 생성능이 흡광도 70.29로 가장 우수하였다. The optimum cultural conditions for production of red pigment from Monascus rubber KCTC 6122 is liquid culture were studied. Monascus rubber KCTC 6122 was shown to give the maximum production of red pigment in the medium containing 4% rice powder, 0.2% NaNO₃, 03% NaHPO₄and 0.15% MgSO₄. The optimum culture conditions, temperature, initial pH and shaking speed were 30℃, 6.5 and 150 rpm, respectively. The red pigment production reached a maximum level at 8days of cultivation.

        1.2-Dichlorobenzene을 분해하는 Pseudomonas sp. DCB3의 분리 및 특성

        Four bacterial strains able to degrade dichlorobenzene as the sole source of carbon and energy were isolated from soil by selective enrichment culture, and among them, one isolation was the best in the cell growth and identified as Pseudomonas sp. DCB3 by its morphology and physiological properties. Cell growth dramatically increased in a minimal medium containing 500ppm of dichlorobenzene was not detected any more at 72 hours after cultivation. The optimal temperature and initial pH for the growth of the isolated strain were 30$\circ$C and 7.0, respectively. Cell growth was increased by supplementing $(NH_2)_2CO$ and 50 ppm yeast extract as additional nutrients. Therefore, it was suggested that Pseudomonas sp. DCB3 could be effectively used for the biological treatment of wastewater containing dichlorobenzene.

      • Pseudomonas sp. T-1에 의한 Terephthalic Acid의 분해

        It was found that the growth of strain T-1 after culture for 60hrs was increased according to the concentration of terephthalic acid up to 2% in the media. Degradation of terephthalic acid was confirmed with the UV scanning and HPLC during the culture of Pseudomonas sp.T-1 in the media containg 0.1% terephthalic acid. It was observed that 90~95% of chemical oxygen demand was eliminated after 5 days, when this strain was applied to the treatment of dye works wastewater whose terephthalic acid concentration was about 0.2% and COD_(Mn) was 1,620ppm by continuous culture with a hydraulic retention time of 37hr.

        Ethylene Glycol과 Terephthalic Acid 분해균주를 이용한 감량가공폐수처리

        Terephthalic acid and ethylene glycol resulting form the weight-reduction process of polyester make trouble in the operation of activated sludge process. Also, polyester weight loss wastewater shows high pH, high organic strength and wide variation of organic loading. Therefore, this study was conducted in order to improve treatment efficiency by activated sludge process with Pseudomonas sp degrading components of polyester weight loss wastewater. The CO DMn and BO S5 of the waste wastewater were 560~3,000 mg/l and 8000~3,000 mg/l, respectively. pH was 11.8~12.3. COD removal efficiency by activated sludge-coagulation process with Pseudomonas sp was 94.1~95.8% for 35 hr of hydraulic retention time. Total organic carbon removal efficiency was 97.1%. Ethylene glycol and terephthalic acid in the wastewater were completely degraded during 32 hr of hydraulic retention time.e.
