        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.       • Protective Effects of Hyperoside against Carbon Tetrachloride-Induced Liver Damage in Mice

        <P>In this study, the hepatoprotective effects of hyperoside (<B>1</B>), a flavonoid glycoside isolated from <I>Artemisia capillaris</I>, have been examined against carbon tetrachloride (CCl<SUB>4</SUB>)-induced liver injury. Mice were treated intraperitoneally with vehicle or <B>1</B> (50, 100, and 200 mg·kg<SUP>−1</SUP>) 30 min before and 2 h after CCl<SUB>4</SUB> (20 μL·kg<SUP>−1</SUP>) injection. Levels of serum aminotransferases were increased 24 h after CCl<SUB>4</SUB> injection, and these increases were attenuated by <B>1</B>. Histological analysis showed that <B>1</B> prevented portal inflammation, centrizonal necrosis, and Kupffer cell hyperplasia. Lipid peroxidation was increased and hepatic glutathione content was decreased significantly after CCl<SUB>4</SUB> treatment, and these changes were reduced by administration of <B>1</B>. Protein and mRNA expression of tumor necrosis factor-α (TNF-α), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and heme oxygenase-1 (HO-1) and nuclear protein expression of nuclear factor erythroid 2-related factor 2 (Nrf2) significantly increased after CCl<SUB>4</SUB> injection. Compound <B>1</B> suppressed TNF-α, iNOS, and COX-2 protein and mRNA expression and augmented HO-1 protein and mRNA expression and Nrf2 nuclear protein expression.       • Anti-Proliferative Effect of an Aqueous Extract of <i>Prunella vulgaris</i> in Vascular Smooth Muscle Cells

        <P>The abnormal proliferation of vascular smooth muscle cells (VSMCs) in arterial walls is an important pathogenic factor of vascular disorders such as diabetic atherosclerosis. We have reported the anti-inflammatory effect of an aqueous extract from <I>Prunella vulgaris</I> (APV) in vascular endothelial cell. In the present study, APV exhibited inhibitory effects on high glucose-stimulated VSMC proliferation, migration, and invasion activities, inducing G<SUB>1</SUB> cell cycle arrest with downregulation of cyclins and CDKs and upregulation of the CKIs, p21<SUP>waf1/cip1</SUP> and p27<SUP>kip1</SUP>. Furthermore, APV dose dependently suppressed the high glucose-induced matrix metalloproteinase activity. High glucose-induced phosphorylation of ERK, p38 MAPK, was decreased by the pretreatment of APV. NF-<I><I>κ</I></I>B activation by high glucose was attenuated by APV, as an antioxidant. APV attenuated the high glucose-induced decrease of nuclear factor E2-related factor-2 (Nrf2) translocation and heme oxygenase-1 (HO-1) expression. Intracellular cGMP level was also increased by APV treatment. These results demonstrate that APV may inhibit VSMC proliferation via downregulating ROS/NF-<I><I>κ</I></I>B /ERK/p38 MAPK pathways. In addition, APV has a beneficial effect by the interaction of Nrf2-mediated NO/cGMP with HO-1, suggesting that <I>Prunella vulgaris</I> may be useful in preventing diabetic atherosclerosis.</P>

        Purpose: The consequences of precipitously rising allergic skin inflammation rates worldwide have accelerated the risk of atopic dermatitis (AD). Natural product-based agents with good efficacy and low risk of side effects offer promising prevention and treatment strategies for inflammation-related diseases. We have already reported that Platycodon grandiflorum root-derived saponins (Changkil saponins, CKS) have many pharmacological effects, including anti-inflammatory and anti-allergic effects, but its influence on AD remains unclear. Therefore, we evaluated the inhibitory effect of CKS, mainly platycodin D, on AD-like skin symptoms in mice and the possible mechanisms in cells. Methods: Mice were sensitized and challenged with 2,4-dinitrochlorobenzene (DNCB). Four weeks after challenge, mice were treated with oral administration of CKS for 4 weeks. In addition, cells were used to evaluate the effect of CKS, mainly platycodin D, on the TARC expression regulated mechanism. Results: CKS attenuated DNCB-induced dermatitis severity, serum levels of IgE and TARC, and mRNA expression of TARC, TNF-α, IFN-γ, IL-4, IL-5, and IL-13 in mice. Histopathological examination showed reduced thickness of the epidermis/dermis and dermal infiltration of inflammatory cells and mast cells in the ears. Moreover, CKS and platycodin D inhibited TNF-α/IFN-γ-induced TARC expression through the suppression of NF-κB and STAT1 and induction of Nrf2/ARE-mediated hemeoxygenase-1 (HO-1) expression in cells. Conclusion: We suggest that CKS and platycodin D inhibited the development of AD-like skin symptoms by regulating cytokine mediators and may be an effective alternative therapy for AD-like skin symptoms.ⓒ2014 Elsevler Gmbh.       • 어린 브리티쉬 숏헤어 고양이에서 발생한 비대성심근병증

        A l0-month-old, neutered male, British shorthair cat was referred to the Veterinary Medical Teaching Hospital of Chungnam National University with the history of tachypnea, anorexia and depression which onset were two days ago. On physical examination, increased respiratory rate and no murmur were found. A complete blood count indicated thrombocytosis. Results from a serum biochemical profile demonstrated elevations in CK, ALP, BUN and Ca, and decreases in sodium, potassium and chloride. Thoracic radiographs revealed patchy interstitial opacity increase and left atrium enlargement. Echocardiographs showed the evidences of hypertrophic cardiomyopathy which were hypertrophy of diastolic interventricular septum and left ventricular free wall. Other findings on echocardiography were the reverse E:A ratio(tranmitra1 inflow pattern) and absence of systolic anterior motion of mitral valve. Electrocardiographs demonstrated wide P and R wave, which mean left atrium enlargement and left bundle branch block, respectively. Medications including furosemide (2mg/kg), enalapril (0.25mg/kg), and spironolactone (1mg/kg) were prescribed twice a day and orally. Client education about exercise restriction and possibility of aggressive progress was given. Clinical signs suggestive of thromboembolism were developed on 6th day after showing tachypnea; right forelimb lameness, bilateral hindlimb lameness, loss of femoral pulse and severe pain. After all, euthanasia was performed at local animal hospital, according to the owner's request.

        8년간(1992~1999) 산업안전보건연구원에 의뢰된 직업병 심의 사례 분석

        Objectives : Pneumoconisis and noise-induced hearing loss (NIHL) have been reported as main occupational diseases by the Special Health Examination. The Industrial Accident Compensation Insurance has reported various work-related diseases, however, these two diseases occupied almost a half of compensated cases. Therefore, it was not well known about the status of occupational diseases other than pneumoconiosis, NIHL, and cardio-cerebrovascular accident (CVA). This study was conducted to analyze claimed cases as an occupational disease, that was requested to the Korea Industrial Safety and Health Agency (KOSHA). Methods : The local office of the Korea Labor Welfare Corporation (KLWC) has asked the KOSHA for confirmation of claimed cases as an occupational disease. We analyzed 379 cases requested from KLWC, the Ministry of Labor, employers, unions and occupational health agencies from 1992 to 1999. Results : Male was 80.7 % of the requested cases. Their mean age was 42 years old and 75.5 % of them were more than 35 years old. The requested cases were increased rapidly from 25 cases in 1992 to 108 cases in 1999 and the accept rate was 50.7 %. The majority of requested cases were respiratory diseases(22.4%), cancers(18.5%), Neuropsychiatric problems(14.5%), and musculoskeletal problems(13.5%). The accept rate was high in reproductive, respiratory, musculoskeletal and digestive disorders and low in neuropsychiatric, renal and otologic problems and occupational cancers. 73.6% of them were caused by chemical agents, especially 28.5 % were by organic solvents. 67% of them were clinically confirmed at university hospitals. A half of the cases were from KyongIn area, even the request came from the whole country. Conclusions : A claim was common in workers whose age was over 35 years old and exposure history was over 10 years. The respiratory diseases and neuropsychiatric disorders were still main problems in occupational health and occupational cancers was increasing even though its accept rate was not high yet.

        It has been recognized as impossibility to have a solution which dominates the complete stress distribution in theoretical aspect with the exception of a few cases in which the boundary conditions are simple enough. In this paper, bars having regular polygonal cross sections under pure tortion was thoroughly examined experimentally using membrane analogy method utilizing Laser beam. When this experimental method was applied to the solution-known simple sections, ie; square and circular, it was confirmed that the results were more than satisfactory in terms of coincidence with the solution derived from the elastic theory. Moreover, in dealing with the n-polygonal sections, it was also observed that the experiment oriented solution was exactly coincided with that of circular section when n approaches to infinite. Thus, the experimentally derived fomula pertaining to the factor of , proved itself to be quite applicable with great accuracy to the stress distributional solution of any regular-polygonal sections.         4MAT System 학습방법이 초등학생의 창의력, 과학과, 학업성취도 및 과학관련태도에 미치는 효과

        본 연구는 McCarthy가 개발한 4MAT 프로그램을 과학과 학습에 투여하여 창의력, 학업성취도와 과학에 대한 태도의 향상에 대한 효과를 분석하였다. 4MAT 학습 프로그램은 학습양식이론을 기초로 하여 좌우반구의 인지기능 특성을 종합한 전뇌학습 방법이다. 인천 지경의 초등학교 5학년과 6학년 학생을 대상으로 4MAT 프로그램을 투여하여 그 효과를 검증하였다. 실험연구에서 4MAT 학습은 연구대상 학년 모두에게 창의력을 향상시키는데 효과적임을 알 수 있었다. 과학과 학업성취도 변화에 있어서, 5학년에서 통계적 유의한 차이는 없었으나 6학년에서는 유의한 차이가 있었다. 과학 관련 태도 변화에 있어서도 5학년에서 통계적 유의한 차이는 없었으나 6학년에서는 유의한 차이가 있었다. The purpose of this study was to explore effects of 4MAT system program on the creativity, science achievement and science-related attitudesof elementary school students. 4MAT learning program is a model based on the whole brain learning including the learning styles and the cognitive specialization of left and right hemispheres. The study was conducted for 10 weeks during the 1998 second semester in H elementary school which is located in Incheon. 4MAT learning program was applied to the 5th and 6th grade students. One hundred 5th grade students and eighty three 6th grade students are selected for this study. The experimental group was taught in applying the 4MAT system, and the control group was taught by the traditional manner. The results of this study were as follows: There was a significant difference on creativity between the instruction by applying 4MAT system and the traditional instruction. In the 6th grade, there was a statistically significant difference on science achievement and science-related attitude. In the 5th grade, there was slightly increased, but statistically nonsignificant. In conclusion, the instruction by applying 4MAT system was more effective in developing student's creativity than the traditional instruction in both grades.

        유전치 치근단 병소에의한 계승 영구치배의 변위

        유치의 치수 감염을 적절히 치료하지 않으면 영구 계승치의 저형성이나 저석회화 같은 법랑질 형성부진, 치배의 위치변화, 맹출 부진이나 맹출 정지 같은 많은 합병증을 초래할 수 있다 이 중에서도 계승 영구치배의 위치변화는 선행 유치의 변성핀치수로 인한 위치변화가 가장 많다. 본 증례는 조선대학교 치과병원 소아치과에 내원한 환아 중 유전치의 치근단 병소로 인해 계승영구전치 치배의 위치변화가 초래된 것으로 사료되어 유치 치료의 중요성을 문헌고찰과 함께 보고하는 바이다. The effect of primary tooth infection on permanent teeth was reported by many previous study. It is seen histologic change in the enamel forming organ and in the surrounding sac, thus produce the defect as either hypoplasia or hypocalcification and alter the eruption of the permanent teeth. The periapical inflammation cause permanent tooth displacement in various direction and can be classified in-to the three categories : rotation, deflection, inversion. This study reported case which displacement of succedaneous teeth may be caused by periapical infection of primary dentition and concluded that treatment procedure of infected primary tooth is selected and performed by importance to retain the tooth for space maintain, possibility to successfully restored, evidence of abnormal developement of the succedaneous tooth.

      • 回轉圓板의 應力擴大係數에 관한 硏究 : Stress Freezing Method of Photoelastic Experiment

        It is well known fact that the stress distributions in the vicinity of crack tip and fracture problems are governed by stress intensity factor which is presented as a function of the crack shape, boundary condition and loading mode etc. In. the determination of the stress intensity factor, there are many theoritical and experimental methods. Among them the stress freezing method of photoelastic experiment has been considered to be one of the most accurate experimental methods which have been used for stress analysis. In this paper, this method was applied to the rotating disk with inner arbitrary crack. The results of photoelastic experiment for disk with various straight cracks, radial and others which are oblique to the radical direction were as follows. In case of radial crack, in comparison with the results of photoelastic experiment obtained by stress freezing method and the theoritical solutions of Blauel, stress freezing method is fully available for analysis of uniform dynamic status.
