        miR-139-5p promotes neovascularization in diabetic retinopathy by regulating the phosphatase and tensin homolog

        Pathological retinal neovascularization is adriver of the progression of diabetic retinopathy (DR). Thepresent study sought to identify the microRNAs (miRNAs)that are diff erentially expressed during the progression ofDR as well as to explore the specifi c regulatory mechanismof those miRNAs in retinal neovascularization. Using amicroarray data set and a diabetic mouse model, it was determinedthat miR-139-5p was signifi cantly upregulated duringthe progression of DR. The in vitro investigation revealedan elevation in the miR-139-5p level in both the high glucose(HG)-treated mouse retinal microvascular endothelialcells (mRMECs) and the HG-treated human RMECs(hRMECs). The miR-139-5p overexpression elevated cellmigration, facilitated tube formation, and increased vascularendothelial growth factor (VEGF) protein level in thehRMECs. While the angiogenic eff ect of miR-139-5p overexpressionwas halted by an anti-VEGF antibody. Meanwhile,the miR-139-5p knockdown eliminated the VEGFinducedcell migration and tube formation in the hRMECs. The phosphatase and tensin homolog (PTEN) was the targetgene of the miR-139-5p. PTEN overexpression removed theangiogenic eff ect of miR-139-5p overexpression, which ledto reduced cell migration and tube formation. In the diabeticmice, the miR-139-5p antagomir eff ectively decreased theacellular capillaries and suppressed the formation of aberrantblood vessels in the retinal tissues. Taken together, miR-139-5p promotes retinal neovascularization by repressingPTEN expression.

        Design-Oriented Stability of Outer Voltage Loop in Capacitor Current Controlled Buck Converters

        Due to the inherent feedforward of load current, capacitor current (CC) control shows a fast transient response that makes it suitable for the power supplies used in various portable electronic devices. However, considering the effect of the outer voltage loop, the stable range of the duty-cycle is significantly diminished in CC controlled buck converters. To investigate the stability effect of the outer voltage loop on buck converters, a CC controlled buck converter with a proportion-integral (PI) compensator is taken as an example, and its second-order discrete-time model is established. Based on this model, the instability caused by the duty-cycle is discussed with consideration of the outer voltage loop. Then the dynamical effects of the feedback gain of the PI compensator and the equivalent series resistance (ESR) of the output capacitor on the CC controlled buck converter with a PI compensator are studied. Furthermore, the design-oriented closed-loop stability criterion is derived. Finally, PSIM simulations and experimental results are supplied to verify the theoretical analyses.

        Effect of Ionic Liquids on the Fibril-Formation and Gel Properties of Grass Carp (Ctenopharyngodon idellus) Skin Collagen

        Self-assembled environment of collagen is one of the important factors for improving and regulating the properties of collagen-based biomaterials. This study aimed to investigate the effect of ionic liquids (ILs) on the fibril-formation and gel properties of grass carp (Ctenopharyngodon idellus) skin collagen. Fibrillogenic kinetics analysis showed that the collagen self-assembly can be suppressed by the introduction of ILs, and the inhibitory effect is influenced by concentration and types of ILs. Scanning electron microscopy test indicated that the assembled collagen fibrils in the presence of ILs had bigger diameters than that in the conventional buffer. Differential scanning calorimetry analysis revealed that the thermal stability of collagen fibrils can be significantly increased when self-assembly is performed in the presence of ILs. Moreover, the introduction of ILs enhanced the mechanical strength of collagen gels. Finding from this work provides a new idea for improving the performance of fish-sourced collagen biomaterials.

        Role of Dehydrocorybulbine in Neuropathic Pain After Spinal Cord Injury Mediated by P2X4 Receptor

        Chronic neuropathic pain is one of the primary causes of disability subsequent to spinal cord injury. Patients experiencing neuropathic pain after spinal cord injury suffer from poor quality of life, so complementary therapy is seriously needed. Dehydrocorybulbine is an alkaloid extracted from Corydalis yanhusuo. It effectively alleviates neuropathic pain. In the present study, we explored the effect of dehydrocorybulbine on neuropathic pain after spinal cord injury and delineated its possible mechanism. Experiments were performed in rats to evaluate the contribution of dehydrocorybulbine to P2X4 signaling in the modulation of pain-related behaviors and the levels of pronociceptive interleukins and proteins after spinal cord injury. In a rat contusion injury model, we confirmed that chronic neuropathic pain is present on day 7 after spinal cord injury and P2X4R expression is exacerbated after spinal cord injury. We also found that administration of dehydrocorybulbine by tail vein injection relieved pain behaviors in rat contusion injury models without affecting motor functions. The elevation in the levels of pronociceptive interleukins (IL-1β, IL-18, MMP-9) after spinal cord injury was mitigated by dehydrocorybulbine. Dehydrocorybulbine significantly mitigated the upregulation of P2X4 receptor and reduced ATP-evoked intracellular Ca2+ concentration. Both P2XR and dopamine receptor2 agonists antagonized dehydrocorybulbine’s antinociceptive effects. In conclusion, we propose that dehydrocorybulbine produces antinociceptive effects in spinal cord injury models by inhibiting P2X4R.

        Cloning and Expression Analysis of a Chitinase Gene Crchi1 from the Mycoparasitic Fungus Clonostachys rosea (syn. Gliocladium roseum)

        Clonostachys rosea (syn. Gliocladium roseum) is a well-known biocontrol agent and widely distributed around the world. In this study, an endochitinase gene Crchi1 was isolated from the mycoparasitic fungus C. rosea using the DNA walking strategy. The Crchi1 ORF is 1,746 bp long and interrupted by three introns. The cloned gene Crchi1 encodes 426 amino acid residues and shares a high degree of similarity with other chitinases from entomopathogenic and mycoparasitic fungi. Several putative binding sites for transcriptional regulation of Crchi1 in response to carbon (5'-SYGGRG-3') and nitrogen (5'-GATA-3') were identified in the upstream of Crchi1. Expression of Crchi1 gene in different carbon sources was analyzed using real-time PCR (RT-PCR). We found that the Crchi1 expression was suppressed by glucose but strongly stimulated by chitin or solubilized components of the cell wall from Rhizoctonia solani. Phylogenetic analysis of chitinases from entomopathogenic and mycoparasitic fungi suggests that these chitinases have probably evolved from a common ancestor.

        Long Non-Coding RNA RMRP Contributes to Sepsis-Induced Acute Kidney Injury

        Purpose: This study aimed to explore the role of the long non-coding RNA (lncRNA) RNA component of mitochondrial RNAase P (RMRP) in sepsis-induced acute kidney injury (AKI). Materials and Methods: Venous blood was collected from septic patients and healthy people. C57BL/6 mice who underwent cecal ligation and puncture (CLP) were used as in vivo models of septic AKI. Lipopolysaccharide (LPS)-induced HK-2 cells were employed as in vitro models of AKI. Flow cytometry analysis was conducted to detect cell apoptosis. Enzyme-linked immunosorbent assay and Western blot assays were used to detect levels of pro-inflammatory cytokines. Results: RMRP was upregulated in sera from patients with AKI and in LPS-induced cells. Knockdown of RMRP inhibited cell apoptosis and reduced production of inflammatory factors in LPS-induced cells, as well as alleviated AKI in CLP mice. RMRP facilitated inflammation by activating NACHT, LRR, and PYD domains-containing protein 3 (NLRP3) inflammasome. We found that microRNA 206 (miR-206) binds with and is negatively regulated by RMRP: miR-206 directly targets the 3’ untranslated region of DEAD-box helicase 5 (DDX5) and negatively regulates DDX5 expression. By binding with miR-206, RMRP upregulated DDX5 expression. Rescue assays revealed that overexpression of DDX5 counteracted the effect of RMRP inhibition on cell apoptosis and inflammatory response in LPS-induced cells. Conclusion: The lncRNA RMRP contributes to sepsis-induced AKI through upregulation of DDX5 in a miR-206 dependent manner and through activation of NLRP3 inflammasome. This novel discovery may provide a potential strategy for treating AKI.
