(1-3)-$\beta$-glucanase 동위효소의 발현 양상을 등 전점 전기영동 젤에서 직접 검출 확인할 수 있는 방법을 개발하였다. 개발된 방법은 시판되고 있는 (1-3)-$\beta$-glucanase활성 측정용 염료착색 기질을 이용하였다. 본 방법은 신속, 간편하며 보리 종자에서 발현되는 것으로 알려져 있는 모든 (1-3)-$\beta$glucanase 동위효소를 검출할 수 있을 정도로 민감하고 특이적이었다. 시판되고 있는 Penicillium(1-3)-$\beta$-glucanase에 대한 활성 검출 한계단위는 50 $\mu$U 정도로 추정되었다. 따라서, 본 방법은 특별한 시설이나 연구 인력을 확보하고 있지 않는 연구실에서 식물체의 (1-3)-$\beta$-glucanase발현에 대한 단백질 수준에서의 연구를 수행하는데 유용하게 이용될 수 있을 것으로 생각된다. A procedure for the direct detection of (1-3)-$\beta$-glucanase isozymes in electrophoresis gels was developed. The procedure employed the commercial preparation of AZCL-pachyman as a chromogenic substrate for (1-3)-$\beta$-glucanases. The procedure detected the three basic isozymes which have been known to be expressed in germinating barley kernels. A major acidic and a minor isozymes were also detected in germinating kernels. The procedure was proved to be fast, simple and sensitive enough to be used for the analysis of the expression of (1-3)-$\beta$-glucanase isozymes in plant tissues. The detection limit of the procedure for the commercial preparation of Penicillium (1-3)-$\beta$-glucanase was estimated to be as low as 50$\mu$U. The procedure could be used for the investigation of (1-3)-$\beta$-glucanases in laboratories facilitated with ordinary equipments and research personnel.

Maximum Power Point Tracking (MPPPT) techniques are employed in PV systems to make full utilization of PV array output power which depends on solar irradiation and temperature. Recently, many MPPT algorithms and control schemes of PV system have been proposed, but different control methods and different PID controller parameters lead to different output efficiency and dynamic response. Therefore, in this paper, a PSCAD/EMTDC user defined module of the solar array is established to easily simulate its inherent characteristics with the basic specification data. Moreover, we also compare the advantages and weakness of the existing MPPT control methods by simulations to give a reference to the Grid-Connected Photovoltaic System (GCPS) designers.

<P>The purpose of this study is to examine the patient-reported outcomes (PRO) after discontinuing nonsteroidal anti-inflammatory drugs (NSAIDs) and clinical factors associated with a favorable outcome in patients with rheumatoid arthritis (RA) in remission or with low-disease activity (LDA). A 16-week prospective open-label trial was conducted at eight rheumatology clinics in Korea. RA patients with 28-joint disease activity score based on erythrocyte sedimentation rate (DAS28-ESR) < 3.2 who were on NSAIDs for more than a month were enrolled, and NSAIDs were discontinued. Acetaminophen (AAP) was used as the rescue medication, and NSAIDs were restarted when joint pain was intolerable with AAP. The endpoint was to analyze the group of patients who continued to withdraw NSAIDs. Among 109 enrolled patients, 105 completed the 16-week follow-up. Eighty-nine (84.8%) patients remained without restarting NSAIDs. In these patients, there was a slight increase in their pain levels compared with baseline (median 14.0 versus 19.0 using the pain-visual analog scale, p = 0.010). However, changes in DAS28-ESR (p = 0.638) and routine assessment of patient index data 3 (RAPID-3) (p = 0.128) were insignificant. Moreover, 66 (62.9%) patients showed sustained effectiveness on PRO without restarting NSAIDs. In the multivariate regression models, joint swelling was the detrimental factor in NSAID withdrawal (odds ratio [OR] 0.149, 95% confidence interval [CI] 0.033-0.680, p = 0.014) and sustained effectiveness (OR 0.284, 95% CI 0.091-0.883, p = 0.030). Joint pain in RA patients in remission or with LDA can be well managed without NSAIDs, especially in those without swollen joints at the time of cessation.</P>

Purpose: Peritoneal carcinomatosis in gastric cancer (GC) patients results in extremely poor prognosis. Malignant ascites samples are the most appropriate biological material to use to evaluate biomarkers for peritoneal carcinomatosis. This study identified exosomal MicroRNAs (miRNAs) differently expressed between benign liver cirrhosis-associated ascites (LC-ascites) and malignant gastric cancer-associated ascites (GC-ascites), and validated their role as diagnostic biomarkers for GC-ascites. Materials and Methods: Total RNA was extracted from exosomes isolated from 165 ascites samples (73 LC-ascites and 92 GC-ascites). Initially, microarrays were used to screen the expression levels of 2,006 miRNAs in the discovery cohort (n=22). Subsequently, quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) analyses were performed to validate the expression levels of selected exosomal miRNAs in the training (n=70) and validation (n=73) cohorts. Furthermore, carcinoembryonic antigen (CEA) levels were determined in ascites samples. Results: The miR-574-3p, miR-181b-5p, miR-4481, and miR-181d were significantly downregulated in the GC-ascites samples compared to the LC-ascites samples, and miR-181b-5p showed the best diagnostic performance for GC-ascites (area under the curve [AUC]=0.798 and 0.846 for the training and validation cohorts, respectively). The diagnostic performance of CEA for GC-ascites was improved by the combined analysis of miR-181b-5p and CEA (AUC=0.981 and 0.946 for the training and validation cohorts, respectively). Conclusions: We identified exosomal miRNAs capable of distinguishing between non-malignant and GC-ascites, showing that the combined use of miR-181b-5p and CEA could improve diagnosis.

폐불가사리를 이용하여 고순도의 산화칼슘, 인산일수소 칼슘(DCPD, DCPA)과 파이로인산 칼슘(CPP) 등의 칼슘제제를 제조하였으며, 제조된 인산일수소 칼슘(DCPA)은 순도가 96.8%, 입도가 5.5㎛, 비표면적이 10.3㎡/g이었으며, 94%의 백색도를 가지는 양질의 분말인 것으로 평가되었다. 또한 DCPA를 하소하여 제조한 파이로인산 칼슘(CPP, Ca_2P_2O_7)의 순도는 97.9%, 입도가 4.4㎛, 비표면적인 1.5㎡/g이었으며, 백색도는 98%, pH는 7.0±0.2이었다. Calcium Oxide(CaO), Dicalcium Phosphates Dihydrate(DCPD), Dicalcium Phosphates Anhydrate(DCPA) and Calcium Pyrophosphates(CPP) of High purity were prepared from starfish by heat treatment and chemical reaction. It was characterized that Dicalcium Phosphates Anhydrate(DCPA) was the purity of 96.8%, the particle sized of 5.5㎛, specific surface area of 10.3㎡/g and standard of white color of 94%. It was characterized that Calcium Pyrophosphates(CPP) prepared from Dicalcium Phosphates Anhydrate(DCPA) was the purity of 97.9%, the particle size of 4.4㎛, the specific surface area of 1.5㎡/g, standard of white color of 98% and 7.0±0.2pH.

<P> Pancreatic β-cells play a crucial role in glucose homeostasis, and the failure of these cells to function results in the development of type 1 diabetes (T1D). The MIN6 cell line, which closely resembles pancreatic β-cells, was used to unravel the relationship between pancreatic β-cell function and the antioxidant enzyme PRX-1. PRX-1 was knocked down in MIN6 cells using a shPRX-1 lentiviral construct, and a mixture of inflammatory cytokines was administered to challenge the MIN6 cells. Nitric oxide (NO) production and inducible NO synthase (iNOS) expression were elevated in shPRX-1 compared with the control. Also, shPRX-1 transduced cells showed higher levels of NF-κB nuclear translocation, suggesting that PRX-1 has a regulatory role in NF-κB nuclear translocation and iNOS expression. In correlation with NO levels, decreased anti-apoptotic gene Bcl-xl level and elevated pro-apoptotic gene Bim levels were observed in shPRX-1 cells compared with scramble, and cell viability decreased accordingly. A rescue experiment was performed subsequently using an iNOS inhibitor to confirm NO as the cause of cell death. Overall, the results of this study suggest possible protective roles of the antioxidant enzyme PRX-1 in the insulinoma cell line MIN6 and possibly in pancreatic β-cells under T1D conditions. </P>
