        For the development of antifungal agents. modification of naftifine which exhibits significant antimycotic activity was performed by replacing the naphthalene ring of it to hetero cyclic rings such as morpholine. benzothiazole. piperidine and pyridine derivatives. The synthesized compounds were tested in vitro antifungal activity against five different fungi with naftifine as a comparative antimycotic molecule. From the biological evaluation two compounds. (E)-N-(3-phenyl-2-propenyl)-N-(4-piperidinylmethyl)amine(3d) and (E)-N-(3-phenyl-2-propenyl)-N-(3-pyndylmethyl)amine(3D) showed relatively noticeable activity(MIC=50㎍/㎖). On the other hand. the other compounds had no activity.

        Some allylamine compounds which are benzothiazole substituants in stead of naphthylring in naftifine, antifungal agents, were sythesized as potential antimycotics. The intermediate Schiff bases that were obtained by condensation of 2-aminobenzothiazole and trans-cinnamaldehyde, were reduced to imine compounds to give allylamines(5a-5d) after mrthylation. These compounds which were tested in vitro against five fungal cell lines containing Trichophyton mentagrophytes. showed no activity in 0.1∼100 μg/ml range.

        혈청이 마우스 간 세포주 BNL CL.2의 Nitric Oxide 생성에 미치는 영향

        마우스 간 세포주인 BNL CL.2의 시험관내 배양에서 혈청과 IFN-γ가 세포주의 nitric oxide(NO) 생성과 세포 손상에 미치는 영향을 알아보기 위한 실험을 하였다. 혈청이 공급된 배양에서 IFN-γ에 의한 세포 생존율은 거의 변동이 없었으나, 혈청을 제거한 배양에서는 약 65%의 생존율이 유지되었으며, NO 생성 억제제인 N -monomethy-L-arginine (NMA)의 첨가는 농도 의존적으로 세포의 생존율을 감소시켰다. 혈청이 제거된 BNL CL.2세포주는 IFN-γ단독 처리에서도 NO 를 생성할 수 있었으며, IFN-γ와 lipopolysaccharide (LPS)의 복합 처리는 세포주의 NO 생성을 상승적으로 증가시켰다. 또한 protein tyrosine kinase (PTK) inhibitor인 herbimycin A와 genistein에 의해서 NO 생성이 억제되어 PTK의 활성이 혈청이 고갈된 BNL CL.2 세포에서 NO의 생성에 중요한 역할을 담당하고 있기 때문으로 판단된다. IFN-γ의 독성은 혈청을 제거시킬 때 NO 생성 억제제에 상승적으로 간 세포를 손상시키며, 이때 NO가 IFN-γ에 의해 유도된 손상을 어느 정도 억제시키는 것을 알 수 있었다. Nitric oxide (NO) plays an important role in immunologic defense, and influences upon the functioning of secretory tissues and cells. It also exhibits cytotoxic/cytostatic activity as one of major operating effectors of the cellular immunity system. We investigated the effects of serum on the cell damages and NO production in the mouse liver cell line BNL CL.2 to establish the role of NO. We observed that, when BNL CL.2 cells were cultured in serum-free medium, they were induced to cell damage by the stimulation of IFN-γalone or IFN-γplus LPS. Serumstarved cells showed large amount of nitrite accumulation and NO synthase (NOS) expression in response to IFN-γalone in dose-and time-dependent manners, but serum-supplied cells did not. The production of NO was blocked by protein tyrosine kinase (PTK) inhibitors, genistein and herbimycin. These results suggest that the deprivation of serum in the BNL CL.2 cell culture medium might primed with the cells to produce NO when the cells are triggered by IFN-γ and the involvement of PTK signal transduction pathway in the expression of NOS gene in murine hepatocytes.

      • 정신분열병에서 종양괴사인자 베타 유전자의 제한효소 절편길이 다형성

        연구목적: 정신분열병은 유전적 성향이 강한 정신 질환의 하나로, 최근에는 분자유전학적 기법을 이용하여 유전자 다형성과 질환사이의 연관성을 밝히는 연구가 정신분열병의 유전학 연구에서 중요한 부분을 차지하고 있다. 이에 본 연구는 중추신경계와 면역계간 상호작용에 관여하는 싸이토카인 중 종양괴사인자베타 (TNFβ) 유전자의 다형성을 분석하여 정신분열병에서의 유전적 관련성을 알아보고자 하였다. 방법: 환자군은 DSM-IV에 의하여 정신분열병으로 진단된 환자 146명을 선정하였고, 정상 대조군으로는 가톨릭 조혈모세포 정보은행에서 보유하고 있는 정상한국인 202명의 자료를 이용하였다. 전혈(whole blood)에서 DNA를 추출하고 TNFβ 유전자 부위를 증폭한 후 제한효소 NcoI과 반응시켰다. NcoI으로 전달되어 555bp와 185bp의 절편을 갖는 TNFB1과 NcoI 절단부가 없어 740bp의 절편을 갖는 TNFB2 등 2가지 대립유전자의 제한효소절편길이 다형성을 조사하였다. 모든 자료의 분석은 χ² 검증을 이용하였다. 결과: 정신분열병 환자군과 정상 대조군 간에 TNFB의 유전자형 TNFB1/1, TNFB1/2의 발 현 빈도에는 유의한 차이는 없었다. 정신분열병 환자군과 정상 대조군간 TNFB1과 TNFB2의 대립 유전자형의 분포는 유의한 차이가 있었다. 결론: 본 연구 결과 정신분열병의 후보 유전자로 종양괴사인자베타의 대립 유전자가 가능성이 있음을 확인하였고, 향후 연구에서는 보다 통합적이고 체계적인 연구가 이루어져야 할 것으로 생각되었다. Objective: Schizophrenia is known to have high genetic influences. Recently, the main focus of etiologic study in schizophrenia has been concentrated on molecular genetic approach including polymorphism analysis. This study was designed to investigate the relationship between schizophrenia and immunologic influences by analyzing polymorphism of TNFB that is involved in interaction between immunologic system and CNS. Method: 146 schizophrenic patients diagnosed by DSM-IV criteria were included and data of 206 normal population from the Catholic Hemopoietic Stem Cell Information Bank(Seoul,Korea) were used as a control group in this study. DNA was extracted from whole blood, thereafter amplified by polymerase chain reaciton, and digested by NcoI. We obtained and assessd RFLP of two alleles, TNFB1 which has a NcoI restriction site generating 555bp and 185bp fragments, and TNFB2 which lacks th NcoI restriction site. All data were analyzed by χ² test. Results: There were no significant differences in frequency of TNFB1/1, TNFB1/2, and TNFB2/2 between the schizophrenic patient and the control group. Alleric frequencies of TNFB1 and TNFB2 were significantly different between schizophrenic patient and control group. Conclusion: We found the possible association between alleles of TNFB and schizophrenia in this study. To clarify the influences of TNFB on schizophrenia, further systematic studies should be conducted.

      • 적외선 체열촬영을 이용한 당뇨병성 족부궤양의 평가

        To assess whether the development of plantar foot ulceration could be predicted from the mean plantar foot temperature(MFT), as assessed by digital infrared thermographic imaging(DITI), in patients with or without diabetic polyneuropathy or foot ulceration, 46 feet with no diabetic polyneuropathy(group Ⅰ), 40 feet with diabetic polyneuropathy(group Ⅱ), and 17 feet with diabetic foot ulcer(group Ⅲ) were assessed by DITI in six standard plantar sites and were correlated with laboratory findings (blood sugar, hemoglobin Alc, C-peptide). Total MFT was significantly higher in group Ⅲ(33.18±1.86℃) than group Ⅰ(29.13±1.63℃) or group Ⅱ(30.89±1.51℃). There were increasing tendency of focal MFT in the third metatarsal head and greater toe where revealed higher incidence of foot ulceration. There was a trend toward elevation of glycosylated hemoglobin(HbAlc) in groupⅢ. In conclusion, DITI is a simple, noninvasive method of idenfifying the neuropathic foot at increased risk of ulceration. Patients with high plantar foot temperature are at increased risk of neuropathic foot ulceration.

      • 농흉과 종격동염이 합병된 viridans streptococcus에 의한 인후농양 1예

        최근 저자들은 viridans streptococcus에 의한 인후농약으로 인하여 발생한 종격동염 및 농흉을 보인 47세 여자 환자를 적절한 배농과 항생제로 치료한 국내 최초의 1예를 치험하였기에 문헌 고찰과 함께 보고하는 바이다. Since the antibiotic era, retropharyngeal abscess became a rare disease entity nowadays. But it may often progress to lethal complications involving vital structure if diagnosis or treatment is delayed. We experienced a case of empyema and mediastinitis complicating retropharyngeal abscess caused by viridans streptococcus in a 47-year-old female patient with diabetes mellitus. She was successfully treated with appropriate antibiotic therapy combined with percutaneous abscess drainage. To our knowledge, such a complication of retropharyngeal abscess has not been reported in Korea yet and we present this unusual case herein followed by a review of the related literature.

      • 大學綜合試驗 出題에 關한 硏究

        The Comprehensive Examination for the undergraduate students is planned to adopt the Jeonbug National University for the purpose of enhancing the quality of its educational outputs. In order to effectively measure and evaluate the level of student progress toward the minimun standard as the students who have finished their general education and basic education for their major field study, the university has decided to make an item pool related to the subjects for the examination. This study is an attempt to make good items of five required courses among general education subjects which are Korean Language, English, Korean History, National Ethics. and Natural Science and are expected to be the subjects of the examination. Instead of making those items separately from the regular examination during the academic semesters, the university plans to collect and use good items from the usual mid-term and final examinations of those subjects. A key concern, in this study, therefore, is to analyze all the items used for the mid-term and final examinations of those subjects during the regular academic semesters. From the data of an about 100 sample in each subject at each time of total three tests, the difficulty index, discrimination index, the response rate of students on each item, and other general descriptive statistics are calculated and then good items are pooled for the future use for the Comprehensive Examination. Anhother important concern in this study is this study is given to the collection and analysis of controversial issues and problems on the process of operating the examination at other universities where the Comprehensive Examination is already adopted. The data from four institutions and from opinions of six participants of this study are used. The following main findidings are summarized : 1. The items have generally shown to be easy while the degree of discrimination is low. 2. Among the five subjects, Korean Language shows the highest rate of right answer (difficulty index) while English the strongest one in the discrimination index. 3. When comparing two types of test between the multiple choice and the short-answer, the latter seems to be powerful in both difficulty and discriminative. 4. The only one question of essay type test reveals clear limitations in scoring and evaluating students progress. 5. All items are constructed under the consideration of content-referenced base, not of the basis on behavioral aspect. 6. The analysis of issues and problems related to the Comprehensive Examination suggests that it is acceptable to test in terms of required subjects of general education and a few most basic subjects of major areas of study. 7. The examination can be functionally operated when it is held at the end of the first year of and before the beginning of the second year of the college years. The length of the examination is usally held from three to five hours. 8. Since the examination should be existed on the idea of criterion-referenced base rather than of norm-referenced one because of its testing the qualification of the entering behavior of students for the further study in their major field, the standard of the pass-and-failure should be set on at least 60 percent of correct answers in each subject. But there must be provided at least two additional opportunities for the make-up examination for those who failed in the first one. 9. A special committee must be established in order to deal with all the details related to the Comprehensive Examination.

        쑥 추출물의 항돌연변이 활성효과

        한국산 쑥으로부터 얻어진 ethanol 추출물과 각 용매획분에 대하여 in vitro로서 Salmonella typhimurium reversion assay 시험을 하여 자체 변이원성 유무와 4가지 양성 변이원 물질(3㎍/plate 2NF, 2㎍/plate AFB_1, 1㎍/plate Trp-P-1 그리고 1㎍/plate Trp-P-2)에 대한 항돌연변이 활성에 대한 쑥의영향을 고찰하였다. 쑥의 ethanol 추출물은 2NF, AFB_1, Trp-P-1 그리고 Trp-P-2 등으로 유도한 돌연변이 시험(Ames test)에서 변이원성을 보여주지 않았다. 돌연변이 억제시험에서는 frameshift type인 Sal. typhimurium TA 98에 대하여 직접 변이원인 2-NF에서는 19.7~22.9%, 간접 변이원인 AFB_1, Trp-P-1과 Trp-P-2에서는 각각 47.9~61.2%, 64.1~70.7% 그리고 67.4~78.7%의 억제효과를 나타내었다. 또한 쑥의 ethanol 추출물을 n-hexane, chloroform, ethyl acetate 그리고 water로 각각 분획하여 직접변이 원인 2-NF와 간접변이원인 AFB_1을 사용하여 변이원성과 항돌연변이원성 시험을 하였다. 돌연변이 시험에서 변이원성은 4가지 획분에서 모두 나타나지 않았다. 직접 변이원인 2-NF로 유도된 돌연변이에 대하여 n-hexane 획분은 9.4~33.7%, chloroform 획분은 21.6~26.5% 그리고 ethytl acetate 획분은 2.4~21.9%를 나타내었다. 간접 변이원인 AFB_1로 유도된 돌연변이에 대하여 n-hexane 획분, chloroform 획분 그리고 ethytl acetate 획분들은 각각 63.0~80.0%, 77.5~82.1%, 68.5~83.1%로 높은 억제 효과를 보여주었다. Mugwort has been know as a traditional substitutive foodstuff and as showing a physiologically beneficial function to a human being. Therefore, effect of mugwort extract in terms of mutagenicity and desmutagenicity was investigated to verify its function. Ethanol extract from mugwort did not exhibit any mutagenicity. On the contrary, inhibitory effects of the ethanol extract were observed on mutagenicity induced by aflatoxin B_1(AFB_1), 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole(Trp-P-1), 3-amino-1-methyl-5H-pyrido[4,3]indole(Trp-P-2) and 2-nitroflourene(2NF) using Salmonella typhimurium reversion assay. On direct-acting mutagen(2NF, 3 ㎍/plate), ethanol extract showed a slight inhibitory effect of 19.7%~22.9%, however on indirect-acting mutagen such as AFB1(2 ㎍/plate), Trp-P-1(1 ㎍/plate) and Trp-P-2(1 ㎍/plate), we observed higher inhibitory effect of 47.9~61.2%, 64.1~70.7%, 67.4~78.7%, respectively. Step-wise fractionation of the ethanol extract was done by using hexane, chlorofractions showed high inhibition of 63.0~80.0%, 77.5~82.1%, and 68.5~83.1%, respectively on the mutagenicity of AFB_1 in Sal. typhimurium TA98. Consequently, these results indicated that mugwort extract contains some compound(s) which may show desmutagenicity.
