        Effects of sodium diacetate and microbial inoculants on fermentation of forage rye

        Rye (Secale cereale L.) is a valuable annual forage crop in Korea but there is limited information about the impact of chemical and biological additives on fermentation characteristics of the crop. This experiment was conducted to investigate fermentation dynamics of wilted forage rye treated with the following six additives; control (no additive), sodium diacetate applied at 3 g/kg wilted forage weight (SDA3), 6 g/kg wilted forage weight (SDA6), inoculations (106 CFU/g wilted forage) of Lactobacillus plantarum (LP), L. buchneri (LB), or LP+LB. The ensiled rye sampled at 1, 2, 3, 5, 10, 20, 30, and 45 days indicated that the acidification occurred fast within five days of storage than the rest of the storage period. The microbial inoculants decline the pH of ensiled forage, more rapidly than the control or SDA treated, which accompanied by the decrease of water-soluble carbohydrates and increase of lactic acid. Compared with the control silage, all treatments suppressed ammonia-nitrogen formation below to 35 g/kg DM throughout the sampling period. Suppression of total microbial counting occurred in SDA6, LP, and LP + LB. The lactic acid production rates were generally higher in microbial inoculation treatments. Acetic acid concentration was lowest in the LP-treated silage and highest in the SDA- and LB-treated silages. The in vitro dry matter (DM) digestibility and total digestible nutrients were the highest in the silage treated with SDA (6 g/kg) at day 45 of ensiling. Based on lower ammonia-nitrogen concentrations and higher feed value, ensiling forage rye treated with SDA at 6 g/kg is promising through enhanced silage quality.

        Comparison of Productivity and Feed Value of Silage Corn according to the Cutting Height

        Corn silage is extensively utilized in ruminant feeding on a global scale, with substantial research efforts directed towards enhancing its nutritional worth and managing moisture content. The purpose of this study was to assess the impact of normal cutting height and elevated cutting height on whole-crop corn silage. Corn was harvested at heights of 15 cm and 45 cm above the ground, respectively, 45 days after heading. The harvested corn was cut into 2-3 cm lengths and packed into 20-liter plastic silos in triplicate. The results showed that dry matter (DM), crude protein (CP), water soluble carbohydrates (WSC), and in vitro dry matter digestibility (IVDMD) of C45 were significantly higher than those of the control, while the neutral detergent fiber (NDF) was significantly lower in C45 (p<0.05). The C15 had higher yields than C45 (p<0.05). There was no significant difference in the total digestible nutrients (TDN) yield of whole-crop corn silage. The increase in cutting height resulted in a larger change in moisture content and NDF per centimeter. After 60 days-ensiling, C45 showed significantly lower NH3-N concentrations. Moreover, C45 had significantly higher lactic acid concentration, lactic acid/acetic acid ratio, and lactic acid bacteria count compared to the control. Mold was not detected and the yeast count was less than 2 log10 cfu/g fresh matter in both control and C45. In summary, C45 improved the feeding value and fermentation quality of whole-crop corn silage at the expense of forage productivity.

        Effects of sodium diacetate or microbial inoculants on aerobic stability of wilted rye silage

        Objective: The primary goal was to identify the effectiveness of chemical or biological additives in delaying the deterioration of early-harvested wilted rye silage after exposure to air. Methods: Rye harvested as a whole plant at the early heading stage was wilted for 24 h. The wilted forage was divided into treatments including sodium diacetate (SDA) at 3 (SDA3) and 6 g/kg (SDA6), Lactobacillus plantarum (LP), L. buchneri (LB), or their equal mixture (LP+LB) at 1×106 colony-forming unit/g fresh matter. Results: After 60 d of conservation in 20-L silos, lactic acid was greater in LP and LP+LB silages than other treatments (102 vs 90.2 g/kg dry matter [DM]). Acetic acid was greatest in SDA6 (32.0 g/kg DM) followed by LB (26.1 g/kg DM) and was lowest in LP treatment (4.73 g/kg DM). Silage pH was lower with microbial inoculation and the lowest and highest values were observed in LP and untreated silages, respectively. After 60 d, neutral detergent fiber concentration was lowest in SDA6 silages, resulting in the greatest in vitro DM digestibility (846 g/kg DM). Aerobic stability was longest in SDA6 (176 h) followed by LB treatment (134 h). Instability after aerobiosis was greatest in LP silages (68 h), about 8 h less than untreated silages. After aerobic exposure, yeast and mold numbers were lowest in SDA6 silages, resulting in DM loss minimization. Exhaustion of acetic acid and lactic acid after aerobic exposure was lowest with SDA6 but greatest with untreated and LP silages. Conclusion: Treatment of early-cut wilted rye forage with SDA at 6 g/kg resulted in silages with higher feeding value and fermentation quality, and substantially delayed deterioration after aerobic exposure, potentially qualifying SDA at this load for promotion of silage quality and delaying aerobic spoilage of early-harvested (low DM) rye forage.       • No Association Between the USP7 Gene Polymorphisms and Colorectal Cancer in the Chinese Han Population

        Colorectal cancer (CRC), now the third most common cancer across the world, is known to aggregate in families. USP7 is a very important protein with an important role in regulating the p53 pathway, which is critical for genomic stability and tumor suppression. We here genotyped eight SNPs within the USP7 gene and conducted a case-control study in 312 CRC patients and 270 healthy subjects in the Chinese Han population. No significant associations were found for any single SNP and CRC risk. Our data eliminate USP7 as a potential candidate gene towards for CRC in the Han Chinese population.

        Flower cultivation regimes aff ect apocarotenoid accumulation and gene expression during the development of saff ron stigma

        The quality of commercial saff ron largely depends on apocarotenoid levels. Whether they are aff ected by diff erent fl owercultivation regimes remains unknown. In present study, apocarotenoid levels and the expression profi les of 12 genes relatedto apocarotenoid biosynthesis between fi eld- and indoor-cultivated Crocus sativus L. were analyzed during stigma development. Under both cultivation regimes, apocarotenoid accumulation and gene expression showed similar profi les, fi rstsharply increasing and then gradually decreasing with stigma development, which suggested the coordinated function ofgene expression and apocarotenoid accumulation. However, the rate of increase/decrease varied widely between two blossomregimes at diff erent developmental stages. Based on correlation analysis, βCH1 and ZDS seemed to play a more crucial rolein apocarotenoid accumulation under both cultivation regimes. At anthesis, the total apocarotenoid level in saff ron cultivatedin the fi eld was higher than that cultivated indoors. This may be due to lower gene expression levels in the apocarotenoidpathway, especially βCH1 , ZDS , and CCD2 , and higher CCD4a and NCED expression in the competitive pathway in saff roncultivated indoors than that in the fi eld, which indicated higher conversion in apocarotenoid biosynthesis fl ux from crocinsand picrocrocin synthesis to β-ionone and the ABA biosynthetic branch in saff ron cultivated indoors. These results providevaluable information for modifying planting mode to enhance saff ron quality.

      • Association of Single Nucleotide Polymorphisms in the Prostaglandin-endoperoxide Synthase 2 (PTGS2) and Phospholipase A<sub>2</sub> Group IIA (PLA2G2A) Genes with Susceptibility to Esophageal Squamous Cell Carcinoma

        Background: The prostaglandin-endoperoxide synthase 2 (PTGS2) and phospholipase A2 group IIA (PLA2G2A) genes encode enzymes that are involved in arachidonic acid and prostaglandin biosynthesis. Dysregulation of both genes is associated with inflammation and carcinogenesis, including esophageal squamous cell carcinoma (ESCC). We therefore hypothesized that there is an association between single nucleotide polymorphisms (SNPs) in these genes and susceptibility to ESCC. Methods: We performed a gene-wide tag SNP-based association study to examine the association of SNPs in PTGS2 and PLA2G2A with ESCC in 269 patients and 269 healthy controls from Taihangshan Mountain, Henan and Hebei Provinces, the rural area of China which has the highest incidence of esophageal cancer in the world. Thirteen tag SNPs in PLA2G2A and 4 functional SNPs in PTGS2 were selected and genotyped using a high-throughput Mass Array genotyping platform. Results: We found a modest increased risk of ESCC in subjects with the PTGS2 rs12042763 AA genotype (OR=1.23; 95% CI, 1.00-3.04) compared with genotype GG. For PLA2G2A, a decreased risk of ESCC was observed in subjects with the rs11677 CT (OR=0.51, 95%CI, 0.29-0.85) or TT genotype (OR=0.51, 95%CI, 0.17-0.96) or the T carriers (CT+TT) (OR=0.52, 95%CI, 0.31-0.85) when compared with the CC genotype. Also for PLA2G2A, rs2236771 C allele carriers were more frequent in the control group (P=0.02). Subjects with the GC (OR=0.55, 95%CI, 0.33-0.93) or CC genotype (OR=0.38, 95% CI, 0.16-0.94) or the C carriers (GC+CC) (OR=0.52, 95%CI, 0.32-0.85) showed a negative association with ESCC susceptibility. Conclusions: Our results suggest that PTGS2 and PLA2G2A gene polymorphisms may modify the risk of ESCC development.

        Butyrolactones Derivatives from the Fermentation Products of an Endophytic Fungus Aspergillus versicolor

        Two new butyrolactones, asperphenol A (1) and B (2), together with four known butyrolactones (3-6) were isolated from the fermentation products of an endophytic fungus Aspergillus versicolor. Their structures were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1-6 were also tested for their anti-tobacco mosaic virus (anti-TMV) activities. The results showed that compound 2 exhibited high anti-TMV activity with inhibition rate of 46.7%. The other compounds also exhibited potential anti-TMV activities with inhibition rates in the range of 21.8-28.4%.

        Two New Diphenylethylenes from Arundina graminifolia and Their Cytotoxicity

        Two new diphenylethylenes, gramniphenols H and I (1 and 2), together with six known diphenylethylenes (3-8), were isolated from Arundina graminifolia. The structures of 1-8 were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1 and 2 were evaluated for their cytotoxicity against five human tumor cell lines. Compound 1 showed cytotoxicity against PC3 cells with $IC_{50}$ value of 3.5 ${\mu}M$. Compound 2 showed cytotoxicity against NB4 and PC3 cells with $IC_{50}$ values of 3.6 and 3.8 ${\mu}M$, respectively.
