        Volatile Chemical and Carotenoid Profiles in Watermelons [Citrullus vulgaris (Thunb.) Schrad (Cucurbitaceae)] with Different Flesh Colors

        Twelve watermelon [Citrullus vulgaris (Thunb.)Schrad (Cucurbitaceae)] cultivars with different flesh colors were analyzed by HPLC, GC-FID, and GC-MS for their differences in carotenoid, soluble sugar, organic acid,and flavor. Results showed that all-trans violaxanthin, 9-cis-violaxanthin, and luteoxanthin were the main carotenoid esters in watermelons with yellow flesh. However,watermelons with red flesh were rich in all-trans lycopene and their cis-isomers. High concentrations of β-carotene and pro-lycopenes were found in watermelon with orangeyellow flesh. Large variations in the sucrose concentration were observed among the different watermelons. Sucrose and/or fructose were the dominant sugars, while citric acid and malic acid were the main organic acids in watermelon flesh. Limonene was detected in the watermelon flesh of all investigated genotypes. Interestingly, partial correlation analysis of the chemical concentrations revealed 2 significant (p<0.01) positive correlations between β-ionone and β-carotene, and between (E)-geranyl acetone and prolycopenes

        Analysis of Geographic and Pairwise Distances among Chinese Cashmere Goat Populations

        This study investigated the geographic and pairwise distances of nine Chinese local Cashmere goat populations through the analysis of 20 microsatellite DNA markers. Fluorescence PCR was used to identify the markers, which were selected based on their significance as identified by the Food and Agriculture Organization of the United Nations (FAO) and the International Society for Animal Genetics (ISAG). In total, 206 alleles were detected; the average allele number was 10.30; the polymorphism information content of loci ranged from 0.5213 to 0.7582; the number of effective alleles ranged from 4.0484 to 4.6178; the observed heterozygosity was from 0.5023 to 0.5602 for the practical sample; the expected heterozygosity ranged from 0.5783 to 0.6464; and Allelic richness ranged from 4.7551 to 8.0693. These results indicated that Chinese Cashmere goat populations exhibited rich genetic diversity. Further, the Wright's F-statistics of subpopulation within total (FST) was 0.1184; the genetic differentiation coefficient (GST) was 0.0940; and the average gene flow (Nm) was 2.0415. All pairwise FST values among the populations were highly significant (p<0.01 or p<0.001), suggesting that the populations studied should all be considered to be separate breeds. Finally, the clustering analysis divided the Chinese Cashmere goat populations into at least four clusters, with the Hexi and Yashan goat populations alone in one cluster. These results have provided useful, practical, and important information for the future of Chinese Cashmere goat breeding.

        Effect of Joint Stiffness on the Stability of Cable-braced Grid Shells

        Bolted joints used in cable-braced grid shells are typically semi-rigid joints, and the joint stiffness has a significant effect on the stability of cable-braced grid shells. The effect of joint stiffness on the stability of cable-braced grid shells is studied in this paper. Based on the experimental results of improved bolted joints, finite element models of elliptic paraboloid cable-braced grid shells with bolted joints are established, and spring elements are used to simulate the joint stiffness. The effect of the joint stiffness on the nonlinear buckling load is studied by changing the joint stiffness. The main conclusions are as follows. First, the joint rotational stiffness has a significant effect on the failure mode. When the joint rotational stiffness is small to a certain extent, the failure mode of cable-braced grid shells changes from overall buckling to local buckling. Second, the nonlinear failure mode is similar to the first-order eigenvalue buckling mode and the maximal compression stress distribution. The structural integrity is weakened, and the maximal steel tube compression stress decreases with the decrease of the joint rotational stiffness. The smaller the joint rotational stiffness, the lower the utilization rate of steel strength. The results suggest that the joint stiffness of elliptic paraboloid cable-braced grid shells should not be less than 20% of the rigid joint stiffness.

      • Selective miRNA Expression Profile in Chronic Myeloid Leukemia K562 Cell-derived Exosomes

        Background: Chronic myeloid leukemia (CML) is a myeloproliferative disorder of hematopoietic stem cell scarrying the Philadelphia (Ph) chromosome and an oncogenic BCR-ABL1 fusion gene. The tyrosine kinase inhibitor (TKI) of BCR-ABL1 kinase is a treatment of choice for control of CML. Objective: Recent studies have demonstrated that miRNAs within exosomes from cancer cells play crucial roles in initiation and progression. This study was performed to assess miRNAs within exosomes of K562 cells. Methods: miRNA microarray analysis of K562 cells and K562 cell-derived exosomes was conducted with the 6th generation miRCURYTM LNA Array (v.16.0). Gene ontology and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were also carried out. GO terms and signaling pathways were categorized into 66 classes (including homophilic cell adhesion, negative regulation of apoptotic process, cell adhesion) and 26 signaling pathways (such as Wnt). Results: In exosomes, 49 miRNAs were up regulated as compared to K562 cells, and two of them were further confirmed by quantitative real-time PCR. There are differentially expressed miRNAs between K562 cell derived-exosomes and K562 cells. Conclusion: Selectively expressed miRNAs in exosomes may promote the development of CML via effects on interactions (e.g. adhesion) of CML cells with their microenvironment.

        Low-dose diethylhexyl phthalate exposure does not impair the expressive patterns of epigenetics-related genes and DNA methylation of breast cancer-related genes in mouse mammary glands

        Backgrounds: Di-(2-ethylhexyl) phthalate (DEHP), as an endocrine-disrupting chemical (EDC), is widely used in plasticizer and other productions. Ubiquitous human exposure to DEHP has been proposed to be a potential risk to public health. Developmental exposure to DEHP could alter epigenetic programming and result in adult-onset disease. Methods: In this study, we investigated whether DEHP exposure to pregnant mice affected epigenetic changes as a result of increase in breast cancer incidence. Results: Our results showed that the expression of total 143 epigenetics-related genes in mammary gland cells, have no significantly altered after short time and low-dose treated with DEHP from 0.5 days post-coitum (dpc) to 3.5 dpc of pregnant mice. DNA methylation status of some neoplastic development genes, such as EGFr, Esr1, Pgr, Fos and Rassf5 also had no obvious change. Conclusion: These finding showed no impact of DEHP on the expressive patterns of epigenetics-related genes and DNA methylation of breast cancer-related genes in pregnant mouse mammary gland cells.

        Many-to-One Encryption and Authentication Scheme and Its Application

        This paper is to study a subclass of group-oriented cryptographic scheme: Many-to-one encryption and authentication scheme. The many-to-one encryption and authentication scheme is to solve a practical problem, i.e., the scenario that the number of the receivers is very small compared with the number of the senders and a receiver may serve millions of senders. Compared with the traditional methods, the burdens of the receiver and the KGC are reduced greatly. How to revoke a sender from his receiver's legitimate sender group is also proposed and it is efficient compared with some traditional methods. The proposed scheme is proven in the random oracle models. The computational complexity of our scheme is independent of the number of the senders. At the end of the paper, an example is given to show how to use our scheme in online software registration and update.

        Nanoarchitectonics of tannic acid based injectable hydrogel regulate the microglial phenotype to enhance neuroplasticity for poststroke rehabilitation

        Background Stroke is the second leading cause of mortality and disability worldwide. Poststroke rehabilitation is still unsatisfactory in clinics, which brings great pain and economic burdens to stroke patients. In this study, an injectable hydrogel in which tannic acid (TA) acts as not only a building block but also a therapeutic drug, was developed for poststroke rehabilitation. Methods TA is used as a building block to form an injectable hydrogel (TA gel) with carboxymethyl chitosan (CMCS) by multivalent hydrogen bonds. The morphology, rheological properties, and TA release behavior of the hydrogel were characterized. The abilities of the TA gel to modulate microglial (BV2 cells) polarization and subsequently enhance the neuroplasticity of neuro cells (N2a cells) were assessed in vitro. The TA gel was injected into the cavity of stroke mice to evaluate motor function recovery, microglial polarization, and neuroplasticity in vivo. The molecular pathway through which TA modulates microglial polarization was also explored both in vitro and in vivo. Results The TA gel exhibited sustainable release behavior of TA. The TA gel can suppress the expression of CD16 and IL-1β, and upregulate the expression of CD206 and TGF-β in oxygen and glucose-deprived (OGD) BV2 cells, indicating the regulation of OGD BV2 cells to an anti-inflammatory phenotype in vitro. This finding further shows that the decrease in synaptophysin and PSD95 in OGD N2a cells is effectively recovered by anti-inflammatory BV2 cells. Furthermore, the TA gel decreased CD16/iNOS expression and increased CD206 expression in the periinfarct area of stroke mice, implying anti-inflammatory polarization of microglia in vivo. The colocalization of PSD95 and Vglut1 stains, as well as Golgi staining, showed the enhancement of neuroplasticity by the TA gel. Spontaneously, the TA gel successfully recovered the motor function of stroke mice. The western blot results in vitro and in vivo suggested that the TA gel regulated microglial polarization via the NF-κB pathway. Conclusion The TA gel serves as an effective brain injectable implant to treat stroke and shows promising potential to promote poststroke rehabilitation in the clinic.

      • Anti-tumor Effects of Penfluridol through Dysregulation of Cholesterol Homeostasis

        Background: Psychiatric patients appear to be at lower risk of cancer. Some antipsychotic drugs might have inhibitory effects on tumor growth, including penfluridol, a strong agent. To test this, we conducted a study to determine whether penfluridol exerts cytotoxic effects on tumor cells and, if so, to explore its anti-tumor mechanisms. Methods: Growth inhibition of mouse cancer cell lines by penfluridol was determined using the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Cytotoxic activity was determined by clonogenic cell survival and trypan blue assays. Animal tumor models of these cancer cells were established and to evaluate penfluridol for its anti-tumor efficacy in vivo. Unesterified cholesterol in cancer cells was examined by filipin staining. Serum total cholesterol and tumor total cholesterol were detected using the cholesterol oxidase/p-aminophenazone (CHOD-PAP) method. Results: Penfluridol inhibited the proliferation of B16 melanoma (B16/F10), LL/2 lung carcinoma (LL/2), CT26 colon carcinoma (CT26) and 4T1 breast cancer (4T1) cells in vitro. In vivo penfluridol was particularly effective at inhibiting LL/2 lung tumor growth, and obviously prolonged the survival time of mice bearing LL/2 lung tumors implanted subcutaneously. Accumulated unesterified cholesterol was found in all of the cancer cells treated with penfluridol, and this effect was most evident in LL/2, 4T1 and CT26 cells. No significant difference in serum cholesterol levels was found between the normal saline-treated mice and the penfluridol-treated mice. However, a dose-dependent decrease of total cholesterol in tumor tissues was observed in penfluridol-treated mice, which was most evident in B16/F10-, LL/2-, and 4T1-tumor-bearing mice. Conclusion: Our results suggested that penfluridol is not only cytotoxic to cancer cells in vitro but can also inhibit tumor growth in vivo. Dysregulation of cholesterol homeostasis by penfluridol may be involved in its anti-tumor mechanisms.

        Analytical Perturbation Methods for Studying a Transversely Isotropic Medium in Multipole Acoustic Logging

        A new analytical perturbation method is developed in this study to investigate the general reflectioncoefficients in the frequency-wavenumber domain of the acoustic field in a fluid-filled boreholesurrounded by a transversely isotropic medium (TIM). The transversely isotropic medium with asymmetric axis parallel to the borehole axis, which is usually called a VTI medium, was adoptedbecause its exact solutions exists, and a corresponding isotropic medium was adopted as a referencestate of perturbation solution. The general reflection coefficients were originally calculated byusing the perturbation method and were compared with the analytical solutions. The zero-, firstandsecond-order perturbation solutions for the general reflection coefficients excited by monopole,dipole and quadrupole sources were investigated for a transversely isotropic elastic solid. The resultsshowed that the general reflection coefficients obtained by using the perturbation solutionsand the analytical solutions were similar for all three sources. In summary, our study demonstratedthat the perturbation method is valid and effective in acoustical logging. This work provided atheoretical foundation for extending perturbation analyses to complicated anisotropic acousticallogging applications.
