Decursin is a major biological active component of Angelica gigas Nakai and is known to induce apoptosis of metastatic prostatic cancer cells. Recently, other reports have been commissioned to examine the anticancer activities of this plant. In this study, we evaluated the inhibitory activity and related mechanism of action of decursin against glioblastoma cell line. Decursin demonstrated cytotoxic effects on U87 and C6 glioma cells in a dose-dependent manner but not in primary glial cells. Additionally, decursin increased apoptotic bodies and phosphorylated JNK and p38 in U87 cells. Decursin also down-regulated Bcl-2 as well as cell cycle dependent proteins, CDK-4 and cyclin D1. Furthermore, decursin-induced apoptosis was dependent on the caspase activation in U87 cells. Taken together, our data provide the evidence that decursin induces apoptosis in glioblastoma cells, making it a potential candidate as a chemotherapeutic drug against brain tumor.

<P>Betulinic acid (BA), a plant-derived pentacyclic triterpenoid, may interact with the members of the organic anion transporting polypeptide 1B subfamily. Here, we investigated the interactions of BA and its analogs with OATP1B1/3 and rat Oatp1b2 in vitro and in vivo. BA inhibited the activity of OATP1B1/3 and rat Oatp1b2 in vitro. Systemic exposure of atorvastatin was substantially altered with the intravenous co-administration of BA (20 mg/kg). Preincubation (incubation with inhibitors, followed by washout) with BA led to a sustained inhibition of OATP1B3, which recovered rapidly in the media containing 10% fetal bovine serum. The addition of albumin to the media decreased intracellular concentrations of BA and expedited the recovery of OATP1B3 activity following preincubation. For asunaprevir and cyclosporin A (previously known to inhibit OATP1B3 upon preincubation), the addition of albumin to the media shortened recovery time with asunaprevir, but not with cyclosporin A. Overall, our results showed that BA inhibits OATP1B transporters in vitro and may incur hepatic transporter-mediated drug interactions in vivo. Our results identify BA as another OATP1B3 inhibitor with preincubation effect and suggest that the preincubation effect and its duration is impacted by altered equilibrium of inhibitors between intracellular and extracellular space (e.g., albumin in the media). Background: The aim of this study is to evaluate the influence of immunosuppressants on in-hospital mortality from sepsis. Methods: Using data of the Health Insurance Review & Assessment Service, we collected data from patients who were admitted to the hospital due to sepsis from 2009 to 2013. Based on drugs commonly used for immunosuppression caused by various diseases, patients were divided into three groups; immunosuppressant group, steroid-only group, and control group. Patients with no history of immunosuppressants or steroids were assigned to the control group. To identify risk factors of in-hospital mortality in sepsis, we compared differences in patient characteristics, comorbidities, intensive care unit (ICU) care requirements, and immunodeficiency profiles. Subgroup analysis according to age was also performed. Results: Of the 185,671 included patients, 13,935 (7.5%) were in the steroid-only group and 2,771 patients (1.5%) were in the immunosuppressant group. The overall in-hospital mortality was 38.9% and showed an increasing trend with age. The steroid-only group showed the lowest in-hospital mortality among the three groups except the patients younger than 30 years. The steroid-only group and immunosuppressant group received ICU treatment more frequently (p < 0.001), stayed longer in the hospital (p < 0.001), and showed higher medical expenditure (p < 0.001) compared to the normal group. Univariate and multivariate analyses revealed that age, male gender, comorbidities (especially malignancy), and ICU treatment had a significant effect on in-hospital mortality. Conclusions: Despite longer hospital length of stay and more frequent need for ICU care, the in-hospital mortality was lower in patients taking immunosuppressive drugs than in patients not taking immunosuppressive drugs.

This study was conducted to develop the simultaneous analysis of veterinary pharmaceuticals (VPs) in rice straw. The method for VPs in rice straw was developed and validated by LC-QTOF-MS. The method was established using solid phase extraction (SPE) cartridges. Efficiency of cartridges was tested in view of recovery and relative standard deviation (RSD). Hydrophilic lipophilic balance (HLB) cartridge was showed the higher result than that of tested other SPE cartridges. Recovery and limit of quantitation (LOQ) of tested 9 VPs in rice straw were 72.0-90.4% and 0.076-7.6 μg/L, respectively. The precision was reliable since RSD percentage (1.7-4.6%) was below 20, which was the normal percent value. The results showed that the developed method might be used the monitoring of VPs in rice straw.
비교란 Lysimeter를 활용한 논 토양에서의 살충제 Chlorpyrifos-methyl의 용탈특성

This study was conducted to investigate the leaching characteristics of insecticide chlorpyrifos-methyl in paddy soil using undisturbed lysimeter. Soil horizons of sandy loam, loam, and silty clay loam lysimeter were composed of 6, 6, and 5, and clay contents were ranged from 53.1-63.2, 40.2-47.7, and 32.6-39.2 %, respectively. Bulk densities of each lysimeter were 1.38, 1.30, and 1.35 Mg/m3, respectively. Chlropyrifos-methyl was treated to each lysimeter at 15 days after rice planting, and water was supplied according to rice cultivation method. Pore volumes of sandy loam, loam, and silty clay loam lysimeter were calculated to 718.87, 764.15, and 735.85 L, and pore volume numbers of supplied water for rice cultivation were 0.98, 1.08, and 0.24, respectively. Movement of chlorpyrifos-methyl in each lysimeter was investigated through the residue of it in 30, 55, 85, and 125 cm soil water from topsoil and leaching water. Gas chromatography (GC) was used for chlorpyrifos-methyl analysis in water sample. Recovery and limit of quantitation (LOQ) of chlorpyrifos-methyl in water were 112.1-115.5% and 3 μg/L. Changes of moisture tension, humidity, and temperature in sandy loam, loam, and silty clay loam lysimeter was not significantly different during experiment period. Weight of silty clay loam lysimeter was higher than other lysimeter because of less leaching. Leaching volume of sandy loam, loam, and silty clay loam lysimeter were 705.9, 828.7, and 174.5 L, respectively. Total leaching content of chlorpyrifos-methyl (treatment content 770.57 mg) during experiment period in sandy loam, loam, and silty clay loam lysimeter were 0, 0, and 0.03 mg, respectively. These results showed that the movement of chlorpyrifos-methyl in sandy loam, loam, and silty clay loam paddy soil was very low.
