        Monitoring of genetically close Tsaiya duck populations using novel microsatellite markers with high polymorphism

        Objective: A set of microsatellite markers with high polymorphism from Tsaiya duck were used for the genetic monitoring and genetic structure analysis of Brown and White Tsaiya duck populations in Taiwan. Methods: The synthetic short tandem repeated probes were used to isolate new microsatellite markers from the genomic DNA of Tsaiya ducks. Eight populations, a total of 566 samples, sourced from Ilan Branch, Livestock Research Institute were genotyped through novel and known markers. The population genetic variables were calculated using optional programs in order to describe and monitor the genetic variability and the genetic structures of these Tsaiya duck populations. Results: In total 24 primer pairs, including 17 novel microsatellite loci from this study and seven previously known loci, were constructed for the detection of genetic variations in duck populations. The average values for the allele number, the effective number of alleles, the observed heterozygosity, the expected heterozygosity, and the polymorphism information content were 11.29, 5.370, 0.591, 0.746, and 0.708, respectively. The results of analysis of molecular variance and principal component analysis indicated a contracting Brown Tsaiya duck cluster and a spreading White Tsaiya duck cluster. The Brown Tsaiya ducks and the White Tsaiya ducks with Pekin ducks were just split to six clusters and three clusters when K was set equal to 6 and 3 in the Bayesian cluster analysis. The individual phylogenetic tree revealed eight taxa, and each individual was assigned to its own population. Conclusion: According to our study, the 24 novel microsatellite markers exhibited a high capacity to analyze relationships of inter- and intra-population in those populations with a relatively limited degree of genetic diversity. We suggest that duck farms in Taiwan could use the new (novel) microsatellite set to monitor the genetic characteristics and structures of their Tsaiya duck populations at various intervals in order to ensure quality breeding and conservation strategies.

      • Grey Neural Network-Based Forecasting System for Vision-Guided Robot Trajectory Tracking

        This paper presents a grey neural network-based forecasting system (GNNFS) in solving the prediction problem. GNNFS adopts a grey model to predict the signal and a neural network (NN) to forecast the prediction error of the grey model. A sequential batch learning (SBL) is developed to adjust the weights of the NN. The proposed GNNFS is applied to a binocular robot, called an Eye-Robot, for human-robot interaction which involved predicting the trajectory of a participant’s hand and tracking the hand. By applying the SBL, the GNNFS can gradually learn to predict the trajectory of the hand and track it well. The experimental results show that the GNNFS can carry out the SBL in real-time for vision-guided robot trajectory tracking.

        Development of novel microsatellite markers to analyze the genetic structure of dog populations in Taiwan

        Objective: Alongside the rise of animal-protection awareness in Taiwan, the public has been paying more attention to dog genetic deficiencies due to inbreeding in the pet market. The goal of this study was to isolate novel microsatellite markers for monitoring the genetic structure of domestic dog populations in Taiwan. Methods: A total of 113 DNA samples from three dog breeds—beagles (BEs), bichons (BIs), and schnauzers (SCs)—were used in subsequent polymorphic tests applying the 14 novel microsatellite markers that were isolated in this study. Results: The results showed that the high level of genetic diversity observed in these novel microsatellite markers provided strong discriminatory power. The estimated probability of identity (P(ID)) and the probability of identity among sibs (P(ID)sib) for the 14 novel microsatellite markers were 1.7×10–12 and 1.6×10–5, respectively. Furthermore, the power of exclusion for the 14 novel microsatellite markers was 99.98%. The neighbor-joining trees constructed among the three breeds indicated that the 14 sets of novel microsatellite markers were sufficient to correctly cluster the BEs, BIs, and SCs. The principal coordinate analysis plot showed that the dogs could be accurately separated by these 14 loci based on different breeds; moreover, the Beagles from different sources were also distinguished. The first, the second, and the third principal coordinates could be used to explain 44.15%, 26.35%, and 19.97% of the genetic variation. Conclusion: The results of this study could enable powerful monitoring of the genetic structure of domestic dog populations in Taiwan.         An Improvement on Robust H<SUB>∞</SUB> Control for Uncertain Continuous-Time Descriptor Systems

        This paper proposes a new approach to solve robust H∞ control problems for uncertain continuous-time descriptor systems. Necessary and sufficient conditions for robust H∞ control analysis and design are derived and expressed in terms of a set of LMIs. In the proposed approach, the uncertainties are allowed to appear in all system matrices. Furthermore, a couple of assumptions that are required in earlier design methods are not needed anymore in the present one. The derived conditions also include several interesting results existing in the literature as special cases.

      • Immature Fruit Recognition using Exposure Fusion and Machine Learning

        Labor shortage is one of the most serious issues in agriculture industry. During harvest season, a lack of manpower may delay the best time for harvesting and significantly affects the postharvest processes and its quality, especially for perishable products, e.g. fruit and vegetables. Mechanical harvesting provides a solution to alleviate the situation by integrating mechanism design and machine vision to perform automatic harvesting. Precise identification and localization algorithm for targeted crops are keys in the system. However, uneven field illumination and high color similarity between target and background perturb the object identification in machine vision. In this study, exposure fusion, a newly developed method to capture a scene with higher dynamic range, is applied to overcome the influence of light for target images. Eleven exposure values will be tested to obtain a best fused image for further analysis. Color space transformation and machine-learning approaches, such as support vector machine (SVM) and linear discriminant analysis (LDA) are proposed to segment immature green fruits (e.g. cherry tomatoes and kumquats) from leafy background.

        An update of preimplantation genetic diagnosis in gene diseases, chromosomal translocation, and aneuploidy screening

        Preimplantation genetic diagnosis (PGD) is gradually widely used in prevention of gene diseases and chromosomal abnormalities. Much improvement has been achieved in biopsy technique and molecular diagnosis. Blastocyst biopsy can increase diagnostic accuracy and reduce allele dropout. It is cost-effective and currently plays an important role. Whole genome amplification permits subsequent individual detection of multiple gene loci and screening all 23 pairs of chromosomes. For PGD of chromosomal translocation, fluorescence $in-situ$ hybridization (FISH) is traditionally used, but with technical difficulty. Array comparative genomic hybridization (CGH) can detect translocation and 23 pairs of chromosomes that may replace FISH. Single nucleotide polymorphisms array with haplotyping can further distinguish between normal chromosomes and balanced translocation. PGD may shorten time to conceive and reduce miscarriage for patients with chromosomal translocation. PGD has a potential value for mitochondrial diseases. Preimplantation genetic haplotyping has been applied for unknown mutation sites of single gene disease. Preimplantation genetic screening (PGS) using limited FISH probes in the cleavage-stage embryo did not increase live birth rates for patients with advanced maternal age, unexplained recurrent abortions, and repeated implantation failure. Polar body and blastocyst biopsy may circumvent the problem of mosaicism. PGS using blastocyst biopsy and array CGH is encouraging and merit further studies. Cryopreservation of biopsied blastocysts instead of fresh transfer permits sufficient time for transportation and genetic analysis. Cryopreservation of embryos may avoid ovarian hyperstimulation syndrome and possible suboptimal endometrium.

      • Improved thin film morphology and bulk-heterojunction solar cell performance through systematic tuning of the surface energy of conjugated polymers

        <P>A detailed model study has shown that thin film morphology and bulk-heterojunction solar cell performance can be significantly improved by systematic tuning of the surface energy of the conjugated donor polymer through side-chain functionalization. Thiophene-flanked diketopyrrolopyrrole (DPP) moieties with different contents of cyanohexane side chains were incorporated into three low band-gap conjugated copolymers (<B>PIDTDPP1</B>, <B>PIDTDPP2</B> and <B>PIDTDPP3</B>) consisting of indacenodithiophene (IDT) donors and DPP acceptors. The resulting polymers possessed good solubility in common organic solvents and showed similar energy levels, bandgaps, and hole mobilities. However, the introduction of cyano groups onto the terminal of side-chains significantly changed their surface energy. Topographical images obtained from atomic force microscopy (AFM) proved that a better matched surface energy between polymer and PC<SUB>71</SUB>BM had led to enhanced miscibility, which resulted in better BHJ film morphology. Consistent with the surface energy enhancement, the performance of BHJ photovoltaic devices increased from 0.97% for <B>PIDTDPP1</B>, to 2.16% for <B>PIDTDPP2</B> then to 3.67% for <B>PIDTDPP3</B>.       • High-mobility low-bandgap conjugated copolymers based on indacenodithiophene and thiadiazolo[3,4-<i>c</i>]pyridine units for thin film transistor and photovoltaic applications

        <P>Two new semiconducting polymers based on indacenodithiophene and thiadiazolo[3,4-<I>c</I>]pyridine units were synthesized <I>via</I> Stille coupling polymerization. The polymers, PIDTPyT and PIDTDTPyT, exhibited main absorption bands in the range of 550–800 nm while their absorption maxima were located at around 700 nm in films. With two additional thiophene spacers, PIDTDTPyT showed a broader absorption band but a 20 nm blue-shifted maximum peak compared to that of PIDTPyT. Both of the polymers possess low bandgaps (∼1.6 eV) and deep energy levels for both the highest occupied molecular orbital (HOMO) and the lowest unoccupied molecular orbital (LUMO). Organic field-effect transistors (OFETs) device measurements indicate that PIDTPyT and PIDTDTPyT have high hole carrier mobilities of 0.066 and 0.045 cm<SUP>2</SUP> V<SUP>−1</SUP> s<SUP>−1</SUP>, respectively, with the on/off ratio on the order of 10<SUP>6</SUP>. Bulk heterojunction photovoltaic devices consisting of the copolymers and PC<SUB>71</SUB>BM gave power conversion efficiencies (PCE) as high as 3.91% with broadband photo-response in the range of 300–800 nm.
