Background: Oridonin isolated from Rabdosia rubescens, a plant used to treat cancer in Chinese folk medicine, is one of the most important antitumor active ingredients. Previous studies have shown that oridonin has antitumor activities in vivo and in vitro, but little is known about cell cycle effects of oridonin in gastric cancer. Materials and Methods: MTT assay was adopted to detect the proliferation inhibition of SGC-7901 cells, the cell cycle was assessed by flow cytometry and protein expression by Western blotting. Results: Oridonin could inhibit SGC-7901 cell proliferation, the $IC_{50}$ being $15.6{\mu}M$, and blocked SGC-7901 cell cycling in the $G_2/M$ phase. The agent also decreased the protein expression of cyclinB1 and CDK1. Conclusions: Oridonin may inhibit SGC-7901 growth and block the cells in the $G_2/M$ phase by decreasing Cdk1 and cyclinB1 proteins.

Background: Both alcohol and aqueous extracts of Sauromatum giganteum(Engl.) Cusimano and Hett, the dried root tuber of which is named Baifuzi in Chinese, have been used for folklore treatment of cancer in Northeast of China. However, little is known about which is most suitable to the cancer therapy. Materials and Methods: Serum pharmacology and MTT assays were adopted to detect the effects of ethanol and aqueous extracts of Sauromatum giganteum(Engl.) Cusimano and Hett, prepared by heat reflux methods, on proliferation of different cancer cells. Results: Cancer cells treated with medium supplemented with 10%, 20%, 40% serum(v/v) containing ethanol extract had a decline in viability, with inhibition rates of 7.69%, 21.8%, 41.9% in MCF-7 cells, 42.8%, 48.1%, 51.8% in SGC-7901 cells, 44.1%, 49.2%, 53.7% in SMMC-7721 cells, 6.8%, 15.2%, 39.8% in HepG2 cells, 7.57%, 16.3%, 36.2% in HeLa cells, 6.24%, 12.5%, 27.4% in A549 cells, and 7.20%, 17.5%, 31.3% in MDA-MB-231 cells, respectively. Viability in the aqueous extract groups was no different with that of controls. Conclusions: An ethanol extract of Sauromatum giganteum(Engl.) Cusimano and Hett inhibited the proliferation of SMMC-7721, SGC-7901 and MCF-7 cells, which supports the use of alcoholic but not aqueous extracts for control of sensive cancers, which might include hepatocarcinoma, gastric cancer and breast cancer.

2C-methyl-D-erythritol 2, 4-cyclodiphosphate synthase (MECPS, EC: 4.6.1.12) is the fifth enzyme of the non-mevalonate terpenoid pathway for isopentenyl diphosphate biosynthesis and is involved in the methylerythritol phosphate (MEP) pathway for ginkgolide biosynthesis. The full-length mecps cDNA sequence (designated as Gbmecps) was cloned and characterized for the first time from gymnosperm plant species, Ginkgo biloba, using RACE (rapid amplification of cDNA ends) technique. The full-length cDNA of Gbmecps was 874 bp containing a 720 bp open reading frame (ORF) encoding a peptide of 239 amino acids with a calculated molecular mass of 26.03 kDa and an isoelectric point of 8.83. Comparative and bioinformatic analyses revealed that GbMECPS showed extensive homology with MECPSs from other species and contained conserved residues owned by the MECPS protein family. Phylogenetic analysis indicated that GbMECPS was more ancient than other plant MECPSs. Tissue expression pattern analysis indicated that GbMECPS expressed the highest in roots, followed by in leaves, and the lowest in seeds. The color complementation assay indicated that GbMECPS could accelerate the accumulation of $\beta$-carotene. The cloning, characterization and functional analysis of GbMECPS will be helpful to understand more about the role of MECPS involved in the ginkgolides biosynthesis at the molecular level.

We construct the empirical Bayes (EB)estimation of the parameter in two-side truncated distribution families with asymmetric Linex loss using negatively associated (NA) samples. The asymptotical optimality and convergence rate of the EB estimation is obtained. We will show that the convergence rate can be arbitrarily close to $O(n^{-q}),\;q\;=\;{\lambda}s(\delta\;-\;2)/\delta(s\;+\;2)$.

Study on load distribution ratio of composite pre-tightened tooth joint by shear nonlinearity

Load distribution has a great influence on the mechanical properties of composite pre-tightened multi-tooth connection. To obtain the load distribution mechanism of composite pre-tightened multi-tooth joints, the multi-tooth joints were studied by experimental and theoretical methods. First, an experimental study was conducted on three-tooth specimens with different tooth depths and tooth lengths, and the failure mode, bearing capacity and load distribution mechanism of the specimens were obtained. Then, based on the nonlinear constitutive of interlaminar shear, an analytical model for load distribution of composite pre-tightened multi-tooth joint was proposed to research the multi-tooth load distribution mechanism. Finally, the theoretical and experimental results were compared. The research showed: (1) The theoretical results of the multi-tooth load distribution ratio were in good agreement with that of the experimental results, the maximum error between the theoretical value and the experimental value of the three-tooth joint was 17.44%, and the minimum error was only 2.35%; (2) The load distribution ratio of composite pre-tightened multi-tooth was uneven, for three-tooth joints, the values of load distribution ratio from large to small were: the first tooth, the third tooth and the second tooth.; (3) Multi-tooth load distribution ratio changed with the change of external load. The change of load distribution ratio was obvious in the early stage of loading, and tended to be gentle in the later stage of loading.

<P>Conventional learning-based methods for segmenting prostate in CT images ignore the relations among the low-level features by assuming all these features are independent. Also, their feature selection steps usually neglect the image appearance changes in different local regions of CT images. To this end, we present a novel semi-automatic learning-based prostate segmentation method in this article. For segmenting the prostate in a certain treatment image, the radiation oncologist will be first asked to take a few seconds to manually specify the first and last slices of the prostate. Then, prostate is segmented with the following two steps: (i) Estimation of 3D prostate-likelihood map to predict the likelihood of each voxel being prostate by employing the coupled feature representation, and the proposed Spatial-COnstrained Transductive LassO (SCOTO); (ii) Multi-atlases based label fusion to generate the final segmentation result by using the prostate shape information obtained from both planning and previous treatment images. The major contribution of the proposed method mainly includes: (i) incorporating radiation oncologist's manual specification to aid segmentation, (ii) adopting coupled features to relax previous assumption of feature independency for voxel representation, and (iii) developing SCOTO for joint feature selection across different local regions. The experimental result shows that the proposed method outperforms the state-of-the-art methods in a real-world prostate CT dataset, consisting of 24 patients with totally 330 images, all of which were manually delineated by the radiation oncologist for performance evaluation. Moreover, our method is also clinically feasible, since the segmentation performance can be improved by just requiring the radiation oncologist to spend only a few seconds for manual specification of ending slices in the current treatment CT image.</P>

RV-23 is a melittin-related antibacterial peptide (MRP) with lower cytotoxicity than either melittin or AR-23, another MRP. The aim of this study was to explore the mechanism of RV- 23`s antibacterial selectivity and its hemocompatibility. The results showed that all the peptides exhibited lytic activity against Staphylococcus aureus and Escherichia coli, with RV-23 showing the highest potency. Moreover, RV-23 had lower cytotoxicity than melittin or AR-23 at their minimal inhibitory concentration. In addition, CD experiments showed that melittin, RV-23, and AR-23 all had a typical α-helical structure, and RV-23 had the lowest α-helix content. The structural information showed that RV-23 has the lowest hydrophobicity and highest hydrophobic moment. Because hydrophobicity and α-helix content are believed to correlate with hemolysis, the results indicate that the selective lytic activity against bacteria of RV-23 may be due to its low hydrophobicity and α-helicity, which lead to low cytotoxicity without affecting antibacterial activity. Furthermore, RV-23 did not affect the structure and function of blood components such as red blood cells, platelets, albumin, and the blood coagulation system. In conclusion, RV-23 is a cell-selective antibacterial peptide with high hemocompatibility due to its unique structure.
